A new perspective on intervertebral disc calcification—from bench to bedside

Disc degeneration primarily contributes to chronic low back and neck ***,there is an urgent need to understand the spectrum of disc degeneration phenotypes such as fibrosis,ectopic calcification,herniation,or mixed *** these phenotypes,disc calcification is the least *** calcification,by definition,is the pathological mineralization of soft tissues,widely studied in the context of conditions that afflict vasculature,skin,and ***,disc calcification is associated with poor surgical outcomes and back pain refractory to conservative *** is frequently seen as a consequence of disc aging and progressive degeneration but exhibits unique molecular and morphological characteristics:hypertrophic chondrocyte-like cell differentiation;TNAP,ENPP1,and ANK upregulation;cell death;altered Pi and PPi homeostasis;and local *** studies in mouse models have provided a better understanding of the mechanisms underlying this *** is essential to recognize that the presentation and nature of mineralization differ between AF,NP,and EP ***,the combination of anatomic location,genetics,and environmental stressors,such as aging or trauma,govern the predisposition to ***,the systemic regulation of calcium and Pi metabolism is less important than the local activity of PPi modulated by the ANK-ENPP1 axis,along with disc cell death and differentiation *** there is limited understanding of this phenotype,understanding the molecular pathways governing local intervertebral disc calcification may lead to developing disease-modifying drugs and better clinical management of degeneration-related pathologies.

Ni-based catalysts obtained from perovskites oxides for ethanol steam reforming

Perovskites as host structures of cations were used in order to generate in situ active and stable catalysts for ethanol steam reforming. For this purpose,La_(1-x)Mg_xAl_(1-y)Ni_yO_3(x = 0.1; y = 0,0.1,0.2,0.3) perovskites were synthetized by the citrate *** segregation is evident for a substitution level higher than 0. 2. The segregation of Ni as NiO generated species interacts with different metal-support after the reduction step. The y = 0.1 catalyst presents the highest H_2 yield value about 85% during reaction time,with low mean values of CH_4 and CO selectivities of 3.4% and 11%,respectively and a low carbon formation. The better performance of y = 0.1 catalyst could be attributed to the minor proportion of segregated phases,thus a controlled expulsion of Ni is successfully reached.

The high penetration of distributed generation(DG)has set up a challenge for energy management and consequently for the monitoring and assessment of power quality(PQ).Besides,there are new types of disturbances owing to the uncontrolled connections of non-linear *** stochastic behaviour triggers the need for new holistic indicators which also deal with big data of PQ in terms of compression and scalability so as to extract the useful information regarding different network states and the prevailing PQ disturbances for future risk assessment and energy management *** and continuous monitoring would guarantee the report to claim for damages and to assess the risk of PQ *** this context,we propose a measurement method that postulates the use of two-dimensional(2D)diagrams based on higher-order statistics(HOSs)and a previous voltage quality index that assesses the voltage supply waveform in a continous monitoring *** suitable for both PQ and reliability applications,the results conclude that the inclusion of HOS measurements in the industrial metrological reports helps characterize the deviations of the voltage supply waveform,extracting the individual customers’pattern fingerprint,and compressing the data from both time and spatial *** method allows a continuous and robust performance needed in the SG ***,the method can be used by an average consumer as a probabilistic method to assess the risk of PQ deviations in site characterization.

We analyzed the allelic and genotypic frequencies of the glutathione-S-transferase P1 （GSTPI） rs1695 single nucleotide polymorphism （SNP） in 290 patients with multiple sclerosis （MS） and in 310 healthy controls. We found no significant association between the rs1695 variant and MS. Among MS patients, there was no relationship between the rs1695 variant and either gender, clinical type of MS or the age of onset of MS.

Effects of Silver Nanoparticles on the Activities of Soil Enzymes Involved in Carbon and Nutrient Cycling

Soils are continuously exposed to large amounts of engineered nanoparticles, especially silver nanoparticles (agNPs), which can affect the activity, stability, and specificity of microbial enzymes. Therefore, the measurement of specific enzyme activity can be used to identify major changes in soil environments. Accordingly, the aim of the present study was to investigate the effects of agNPs on soil enzymes that play critical roles in mineralizing carbon and nutrients in soil. Soil samples (silt loam and sandy loam) were collected from the surface layer (0–15 cm) of a field at the George Washington Carver Farm, Lincoln University of Missouri, USA. The soils were then treated with agNP solutions at 0, 1 600, or 3 200 μg ag kg^(-1) dry soil, using either 10- or 50-nm agNPs and a randomized complete block design, with three replicates per treatment. The agNP-treated soil samples were homogenized and incubated for one month, and soil acid phosphatase, β-glucosaminidase, β-glucosidase, and arylsulfatase activities were measured after one hour, one week, and one month of incubation. The activities of all four enzymes were reduced by agNP treatment after one hour and one ***, agNP size had no effect. After one month of incubation, the agNP treatments had mixed effects, which suggests that soil enzymes are only affected on a short-term basis. Further studies are required to determine the mechanisms by which agNPs reduce soil enzyme activity.

Breast cancer is the most common type of malignant tumor among women and their second leading cause of cancer-related *** most common method for screening and diagnosis is ***,two main problems have been ***,the dose of radiation received during the test prevents the method from the use on women who are 〈 40 years ***,there can be mammogram failure owing to the lack of tumor contrast with the fibrous ***,there is a need for screening methods that will help to identify high-risk *** developed a biological marker test that can help to identify *** levels of sialic acid （SA） in saliva are known to correlated with breast *** this study,we evaluated the feasibility of Raman spectroscopy as a method for quantification of SA in saliva,using citrate-reduced silver nanoparticles （cit-ag-NPs） as a surface-enhanced Raman spectroscopy （SERS） *** of SA was accomplished by measuring its intensity in saliva and comparing it with a calibration curve of SA *** mean SA concentration in saliva was found to be significantly higher among 100 breast cancer patients （18.3 ＆#177; 9.4 mg＆#183;dL-1;mean ＆#177; SD） than among 106 healthy controls （3.5 ＆#177; 1.0 mg＆#183;dL-1）.The SERS test showed sensitivity of 94% and specificity 98% for detection of patients with breast cancer,assuming that SA concentration 〉 7 mg＆#183;dL-1 is a cutoff for positive test *** findings prove the usefulness of this SERS technique as a simple,convenient,and highly sensitive method of quantitative analysis of SA in *** simplicity of this nanotechnological test may help to substantially reduce the mortality among patients with breast cancer by providing women with a simple,noninvasive screening test that can be applied regardless of age or density of breast tissue.

Abscisic Acid, High-Light, and Oxidative Stress Down-Regulate a Photosynthetic Gene via a Promoter Motif Not Involved in Phytochrome- Mediated Transcriptional Regulation

In etiolated seedlings, light perceived by phytochrome promotes the expression of light-harvesting chlorophyll a/b protein of photosystem Ⅱ （Lhcb） genes. However, excess of photosynthetically active radiation can reduce Lhcb expression. Here, we investigate the convergence and divergence of phytochrome, high-light stress and abscisic acid （ABA） signaling, which could connect these processes. Etiolated Arabidopsis thaliana seedlings bearing an Lhcb promoter fused to a reporter were exposed to continuous far-red light to activate phytochrome and not photosynthesis, and treated with ABA. We identified a cis-acting region of the promoter required for down-regulation by ABA. This region contains a CCAC sequence recently found to be necessary for ABI4-binding to an Lhcb promoter. However, we did not find a G-box-binding core motif often associated with the ABI4-binding site in genes promoted by light and repressed by ABI4. Mutations involving this motif also impaired the responses to reduced water potential, the response to high photosynthetic light and the response to methyl viologen but not the response to low temperature or to Norflurazon. We propose a model based on current and previous findings, in which hydrogen peroxide produced in the chloroplasts under high light conditions inter- acts with the ABA signaling network to regulate Lhcb expression. Since the mutation that affects high-light and methyl viologen responses does not affect phytochrome-mediated responses, the regulation by retrograde and phytochrome signaling can finally be separated at the target promoter level.

Molecular Determinants of Antibiotic Resistance in the Costa Rican Pseudomonas aeruginosa ag1 by a Multi-omics Approach:A Review of 10 Years of Study

BACKGROUND Inflammatory bowel diseases(IBD)have been associated with a low quality of life(QoL)and a negative impact on work productivity compared to the general *** about disease control,patient-reported outcomes(PROs),treatment patterns and use of healthcare resources is relevant to optimizing IBD *** To describe QoL and work productivity and activity impairment(WPAI),treatment patterns and use of healthcare resources among IBD patients in *** A multicenter cross-sectional study included adult outpatients who were previously diagnosed with moderate to severe Crohn’s disease(CD)or ulcerative colitis(UC).At enrolment,active CD and UC were defined as having a Harvey Bradshaw Index≥8 or a CD Activity Index≥220 or calprotectin>200μg/g or previous colonoscopy results suggestive of inadequate control(per investigator criteria)and a 9-point partial Mayo score≥5,*** PRO assessment included the QoL questionnaires SF-36 and EQ-5D-5L,the Inflammatory Bowel Disease Questionnaire(IBDQ),and the WPAI *** about healthcare resources and treatment during the previous 3 years was collected from medical ***-square,Fisher’s exact and Student’s t-/Mann-Whitney U tests were used to compare PROs,treatment patterns and the use of healthcare resources by disease activity(α=0.05).RESULTS Of the 407 patients in this study(CD/UC:64.9%/35.1%,mean age 42.9/45.9 years,54.2%/56.6%female,38.3%/37.1%employed),44.7%/25.2%presented moderate-to-severe CD/UC activity,respectively,at *** in median values for CD/UC,respectively,the SF-36 physical component was 46.6/44.7 and the mental component was 45.2/44.2,the EQ-visual analog scale score was 80.0/70.0,and the IBDQ overall score was 164.0/*** to severe activity,female gender,being unemployed,a lower educational level and lower income were associated with lower QoL(P<0.05).Median work productivity impairment was 20%and 5%for CD and UC patients,re
