Endotoxins(also known as lipopolysaccharides(LPs)) are undesirable by-products of recombinant proteins,purified from Escherichia coli.LPs can be considered stable under a wide range of temperature and pH,making their removal one of the most difficult tasks in downstream processes during protein purification.The inherent toxicity of LPs makes their removal an important step for the application of these proteins in several biological assays and for a safe parenteral administration.Immobilized metal affinity chromatography(IMAC) enables the affinity interactions between the metal ions(immobilized on the support through the chelating compound) and the target molecules,thus enabling high-efficiency separation of the target molecules from other components present in a mixture.Affinity chromatography is applied with Ca2+-iminodiacetic acid(IDA) to remove most of the LPs contaminants from the end product(more than90%).In this study,the adsorption of LPs on an IDA-Ca2+ was investigated.The adsorption Freundlich isotherm of LPs-IDA-Ca2+ provides a theoretical basis for LPs removal.It was found that LPs is bound mainly by interactions between the phosphate group in LPs and Ca2+ ligands on the beads.The factors such as pH(4.0 or 5.5) and ionic strength(1.0 mol/L) are essential to obtain effective removal of LPs for contaminant levels between endotoxin' concentration values less than100 EU/mL and 100 000 EU/mL.This new protocol represents a substantial advantage in time,effort,and production costs.

Ischemic heart disease (IHD) is the leading cause of sudden cardiac death (sCD) and often non-thrombosed severe coronary stenoses with or without myocardial scars are detected.Left dominant arrhythmogenic cardiomyopathy (LDAC) is a life-threating rare disease which has been more thoroughly studied in the last 10years.The macroscopic study of an sCD victim was conducted and re-evaluated 9years later.The cardiological work-up in his firstdegree relatives initially comprised an electrocardiogram (ECG) and an echocardiogram.When they were re-evaluted 9years later,a cardiac magnetic resonance,an ECG-monitoring,an exercise testing and a genetic study were performed and the pedigree was extended accordingly.In 2008,an IHD was suspected in the sports-triggered sCD of a 37-year-old man upon the postmortem (75% stenosis of the left main and circumflex coronary arteries;the subepicardial left ventricular fibrofatty infiltration with mild myocardial degeneration was assumed to be a past myocardial infarction).No cardiomyopathy was identified in any of the two proband's sisters.Nine years thereafter,distant relatives were diagnosed with LDAC due to a pathogenic desmoplakin mutation.The reanalysis of the two sisters showed ventricular arrhythmias in one of them without structural heart involvement and the reviewed postmortem of the proband was reclassified as LDAC based on the fibrofatty infiltration;both were mutation carriers.The completion of the family study on 19 family members yielded one sCD due to LDAC (the proband),three living patients diagnosed with LDAC (two with a defibrillator),one mutation carrier without structural ventricular involvement,and 14 healthy relatives (who were discharged) with a very good co-segregation of the mutation.Although rare,LDAC exists and sometimes its differential diagnosis with iHD has to be faced.Modifying previous postmortem misdiagnoses can help family screening to further prevent sCDs.

Retinitis pigmentosa(RP) is a group of inherited retinal disorders that lead to photoreceptor loss.RP has been reported to be related to oxidative stress,autophagy,and inflammation.(-)-Epigallocatechin gallate(EGCG),the most abundant catechin-based flavonoid in green tea leaves,has significant antioxidant,anti-carcinogenic,antimicrobial,and neuroprotective properties.EGCG,given its low molecular weight and hydrophilic properties,can cross the blood-retinal barrier and is able to reach different ocular tissues such as the lens,cornea,and retina.EGCG has been shown to provide retinal protection against ischemia;sodium nitroprusside-,N-methyl-D-aspartate-,lipopolysaccharide-,light-,sodium iodate-,or H2 O2-induced damage and diabetic retinopathy.This suggests that systemic EGCG administration has the potential to protect against retinal degenerative or neurodegenerative diseases such as RP.The aim of this work was to investigate whether EGCG can protect against RP progression in the animal P23 H line 1,the model of RP.Albino P23 H rats were crossed with pigmented Long Evans rats to produce offspring exhibiting the clinical features of RP.Pigmented P23 H rats were treated via intraperitoneal injection with saline or EGCG at a dose of 25 mg/kg every week from P100 to P160 and then compared to wild-type Long Evans rats.Rats treated with EGCG showed better visual and retinal electrical function with increased contrast sensitivity and b-wave values compared with those observed in P23 H rats treated with vehicle.EGCG reduced lipid peroxidation and increased total antioxidant capacity and catalase and superoxide dismutase activities.No differences were observed in visual acuity,nitrate levels,nitrite levels or glutathione s-transferase activity.In conclusion,EGCG not only reduced the loss of visual function in P23 H rats but also improved the levels of antioxidant enzymes and reduced oxidative damage.This study was approved by the Institutional Animal Care and Use Committee(CEICA) from the University of Zaragoza under proje

The E2F/DP pathway is a widely conserved regulatory mechanism in pluricellular organisms.The family of E2F and DP transcription factors was originally described having a role in the transition from the G1 to the s phase of the cell cycle.However,the discovery of hundreds of possible gene targets and their involvement in many other biochemical processes,soon showed that they participated in cell development and differentiation,chromatin remodeling,DNA repair and others.The E2F/DP transcription factors can act as either activators or repressors of transcription depending on their association to other regulatory proteins,particularly the retinoblastoma protein,or even depending to their protein structure that can define their role.In plants the E2F/DP pathway also regulates endoreduplication,a process present along the life cycle,from organ elongation to root differentiation and reproduction.These transcription factors also help plant cells to respond to environmental disturbances such as those caused by different types of radiation,or by pathogens.This review focuses on the“so called”non-canonical functions of the E2F/DP family proteins in animal and plant cells,that are in fact essential activities that connect regulatory circuits among multiple metabolic pathways by means of their atypical functions.

The number of people with food allergy(FA)has increased in recent decades,as has the number of foods to which they are allergic.Estimates of FA prevalence vary widely,likely because of different definitions,evaluated foods and geo-graphic variations.In developed countries,overall preva-lence estimates range from 1.9%to 7.2%[1,2].In Latin America,the prevalence of FA in children is between 1.4%and 5.3%[3-6],with important age-based variations(from 1.9%in infants to 0.4%in preschoolers)[7].FA is a complex trait in which the interplay of genetic and environmental fac-tors results in disease expression.Well-known factors that increase atopic susceptibility include sex[8,9],age[10],and socioeconomic status[11-13].Furthermore,the asso-ciation between family history of allergies and childhood FA has been consistently reported in recent years.Overall,a maternal history of allergic disease appears to be more strongly associated with FA than paternal history of atopy[14].

Aims Deforestation and biodiversity loss are two alarming,closely related problems,and the main factors triggering changes in land use.Indigenous agricultural practices in the western Amazon Basin are known as chakras,and their structure and dynamics are seemingly optimal for forest management.However,the variability in tree spe-cies and the degree of forest recovery after abandonment is poorly documented in this agroforestry system(AFs).The goals of this study were:(i)to investigate whether the different AFss(chakras)preserve similar levels of forest diversity,(ii)to determine the effect of trans-formation of mature forests(MF)to chakras,in particular,forest alpha and beta diversity levels,and(iii)to investigate whether native tree species recovery leads to the original forest structure following chakra abandonment.Methods We assessed the floristic composition in three AFss(cassava,corn,and cocoa),the secondary forest(sF),and the forest remnants in the buffer zone of the Northern Ecuadorian Amazon(NEA).All tree spe-cies with a diameter at breast height(dbh)≥10 cm were inventoried in 61 plots(0.28 ha average)representing 17.44 ha.Alpha diversity was calculated in all systems to determine the levels of variabil-ity using species richness and the shannon diversity index.Also,beta diversity was examined to evaluate the degree of dissimilarity among all AFss with the MF in order to analyze changes in floristic composition.The divergence between the sF and the MF was ana-lyzed to ascertain forest recovery after chakra abandonment.Important Findings A total of 4,060 trees(dbh≥10 cm)representing 109 species,96 genera,and 43 plant families were inventoried in 17.44 ha sampled in five systems in the buffer zone of the NEA.The most dominant plant families were Arecaceae,Myristicaceae,Fabaceae,Meliaceae,and Malvaceae,and the most representative genera included Iriartea,Virola,Guarea,Ocotea,Cordia,Chrysophyllum,and Inga.The MF in this zone is composed of 81 tree species circumscribed in 74 genera and 30 plant families.Transf

Ultrasonic spray pyrolysis deposition of snse and snse_2 using a single spray solution

Thin films of snse and snse2 have been deposited using the ultrasonic spray pyrolysis（UsP） technique.To the best of our knowledge this is the first report of the deposition of snse and snse2 thin films using a single spray solution.The use of a single spray solution for obtaining both a p-type material,snse,and an n-type material,snse2,simplifies the deposition technique.The snse2 thin films have a bandgap of 1.1 eV and the snse thin films have a band gap of 0.9 eV.The Hall measurements were used to determine the resistivity of the thin films.The snse2 thin films show a resistivity of 36.73 Ωcm and n-type conductivity while the snse thin films show a resistivity of 180 Ωcm and p-type conductivity.

The effect of non-pharmaceutical interventions on COVID-19 outcomes:A heterogeneous age-related generalisation of the sEIR model

successive generalisations of the basic sEIR model have been proposed to accommodate the different needs of the organisations handling the sARs-CoV-2 epidemic and the assessment of the public health measures adopted and named under the common um-brella of Non-Pharmaceutical Interventions(NPIs).so far,these generalisations have not been able to assess the ability of these measures to avoid infection by the sARs-CoV-2 and thus their contribution to contain the spread of the disease.This work proposes a new generalisation of sEIR model and includes a heterogeneous and age-related generation of infections that depends both on a probability that a contact generates the transmission of the disease and a contact rate.The results show(1)thanks to the universal wearing of facial coverings,the probability that a contact provokes the transmission of the disease was reduced by at least 50%and(2)the impact of the other NPI is so significant that otherwise Portugal would have gone into a non-sustainable situation of having 80%of its population infected in the first 300 days of the pandemic.This situation would have led to a number of deaths almost twenty times higher than the number that was actually recorded by December 26th,2020.Moreover,the results suggest that even if the requirement of universal wearing of facial coverings was adopted sooner jointly with closing workplaces and resorting to teleworking would have postponed the peak of the incidence,altought the epidemic path would have result in a number of infections hardly managed by the National Health system.Complementary,results confirm that(3)the health authorities adopted a conservative approach on the criteria to consider an infected individual not infective any longer;and(4)the most effective NPIs and stringency levels either impacting on self-protection against infection or reducing the contacts that would eventually result in infection are,in decreasing order of importance,the use of Facial coverings,Workplace closing and stay at home requirements.

About the Thermodynamics and Aging of self-Organizing systems

We know that the total daily energy dissipation increases in complex organisms like the humans. It’s very probable that this increase in total energy dissipation is related to the progressive increase in mass. But we also know that day by day the dissipation of energy per unit mass decreases in these organisms. We intend to verify if this decrease is only an expression of the second law of thermodynamics, or if it is related to the increase in mass that occurs in these organisms. For this, we set ourselves the following objectives: verify the correlation between total energy dissipation and the evolution of body mass, and verify the correlation between the dissipation of energy per unit of mass and the evolution of body mass. As a result of the data analysis, we found a high degree of correlation between total energy dissipation and the evolution of body mass. And we also found a high correlation between the energy dissipated per unit of mass and the evolution of body mass. We can conclude that self-organization produces not only an increase in mass, but also a decline in energy dissipation per unit mass beyond what is expected by the second law of thermodynamics.

Aging and Biological Oscillation: A Question of Geometry

Previous studies in different ethnic groups show changes in heart rate, respiratory rate, cortisol cycle, and sleep-wake cycle throughout life. Our purpose is to verify such changes by comparing the values of each variable before and after puberty. Puberty is associated with the end of growth and is an important point in our theoretical framework: when growth ends, changes occur in the geometry of the biological system. At the same time, this causes phase changes in the oscillatory variables, which are seen as chronodisruption. The results confirm the changes found by other authors in the evolution of the variables throughout life. Then, we can conclude that the variables studied present phase changes when growth ends, in accordance with the proposed theoretical framework.
