Morphological comparison and molecular marker screening of three skeletonema species found in Changjiang(Yangtze)River Basin

In our recent investigations of diatom diversity,we studied three species,namely,skeletonema costatum,skeletonema subsalsum,and skeletonema *** they have been found frequently in Changjiang(Yangtze)River Basin,their morphological and molecular identification is difficult in ***,to integrate morphological and molecular biological approaches,we compared systematically their morphological characters and performed phylogenetic *** strains of skeletonema were collected and isolated from shanghai and Jiangsu,China,and their morphological characteristics were examined by light microscopy(LM)and the scanning electron microscopy(sEM).Based on morphological comparison,we determined that *** is easy to distinguish from the other two *** heavily silicified areolae,undulated or cleft distal ends of terminal fultoportula processes(TFPPs),absence of basal pores of fultoportula processes(FPPs),the rootlike protrusions of FPPs,and no interlocking connection are the stable characteristics that can be used to identify ***,there are only two features that can distinguish *** from ***,namely the location of terminal rimoportulae(TRPs)and the distal shape of *** addition,we amplified and sequenced nine common genetic markers from the strains,from which 101 sequences were obtained,constructed phylogenetic trees based on the nine genes and evaluated that seven genes can be used to identify ***,and revealed that *** is the closest known relative of ***,and only ATP synthetase beta-subunit gene(atp B)is able to distinguish them from each other,which strongly support that it is an effective molecular marker for *** work provided a theoretical basis for the taxonomic study of skeletonema.

针对香蕉胚性悬浮细胞培养过程中存在的胚性愈伤组织的诱导率低、周期过长、胚性易丢失等问题,以"苹果蕉"及"威廉斯"香蕉的未成熟雄花为试验材料,对香蕉胚性愈伤组织的诱导、悬浮培养的理化条件进行改进。结果表明:(1)光照条件较暗培养更有利于诱导理想的香蕉胚性愈伤组织;(2)M1培养基中加入酪蛋白和脯氨酸有利于胚性愈伤组织的诱导;(3)用微孔板代替三角瓶悬浮培养,有利于悬浮细胞分裂增殖,而且可减少污染的可能。

H7N9 subtype avian influenza virus poses a great challenge for poultry *** disease virus(NDV)-vectored H7N9 avian influenza vaccines(NDV_(vec)H7N9)are effective in disease control because they are protective and allow mass *** note,these vaccines elicit undetectable H7N9-specific hemagglutination-inhibition(HI)but high IgG antibodies in ***,the molecular basis and protective mechanism underlying this particular antibody immunity remain ***,immunization with an NDV_(vec)H7N9 induced low anti-H7N9 HI and virus neutralization titers but high levels of hemagglutinin(HA)-binding IgG antibodies in *** residues(s150,G151 and s152)in HA of H7N9 virus were identified as the dominant epitopes recognized by the NDV_(vec)H7N9 immune *** transferred NDV_(vec)H7N9 immune serum conferred complete protection against H7N9 virus infection in *** NDV_(vec)H7N9 immune serum can mediate a potent lysis of HA-expressing and H7N9 virus-infected cells and significantly suppress H7N9 virus *** activities of the serum were significantly impaired after heat-inactivation or treatment with complement inhibitor,suggesting the engagement of the complement ***,mutations in the 150-sGs-152 sites in HA resulted in significant reductions in cell lysis and virus neutralization mediated by the NDV_(vec)H7N9 immune serum,indicating the requirement of antibody-antigen binding for complement ***,antibodies induced by the NDV_(vec)H7N9 can activate antibody-dependent complement-mediated lysis of H7N9 virus-infected cells and complement-mediated neutralization of H7N9 *** findings unveiled a novel role of the complement in protection conferred by the NDV_(vec)H7N9,highlighting a potential benefit of engaging the complement system in H7N9 vaccine design.

A functional gel polymer electrolyte based on PVDF-HFP/gelatin toward dendrite-free lithium metal batteries

The leakage of liquid electrolyte and the formation of lithium dendrites pose challenges to safety and stability of lithium metal batteries(LMBs).The appearance of gel polymer electrolyte(GPE)has obviously improved the safety of traditional ***,the limited inhibition of GPE on lithium dendrites is detrimental to the safety of ***,a kind of poly(vinylidene fluoride-co-hexafluoropropylene)(PVDF-HFP)/gelatin(GN)GPE with high ionic conductivity,high-temperature resistance,and flame-retardancy,was prepared by electrospinning and soaking *** the electrospinning network of PVDF-HFP,its affinity to liquid electrolytes,makes this GPE more beneficial to ions transport and the formation of ***,the GN with sol–gel properties,enhances the mechanical property(13.5 MPa)of HFP-GN ***,X-ray photoelectron spectroscopy(XPs)and density functional theory(DFT)suggest that the attraction of polar groups of GN to Li+can regulate the distribution of Li+and protect Li ***,the application of HFP-GN GPEs to LMBs with cathodes of LiFePO_(4) and LiCoO_(2) deliver excellent electrochemical performances:after 300 cycles,the LiFePO_(4)/HFP-GN GPE/Li battery keeps a low capacity decay rate of 0.09%at 5 C;after 400 cycles at 2 C,the LiCoO_(2)/HFP-GN GPE/Li cell retains a high capacity retention of 74%.This GPE is demonstrated for the application prospect of safe LMBs.

Intraseasonal variability of the Kuroshio observed via mooring at 18°N

Insufficient observations near the origin of the Kuroshio have led to incomplete understanding of the intraseasonal variability(IsV)of the *** measurements of the Kuroshio velocity were performed with an array of three profiler moorings(122.7°E,123°E,and 123.3°E)along 18°N from January 2018 to February *** IsV of the Kuroshio at 18°N was investigated based on a combination of mooring observations and global high-resolution HYbrid Coordinate Ocean Model reanalysis *** estimated time-averaged transport in the upper 350 m across the observation transect was 6.5±2.6 sv(1.0 sv=10^(6)m^(3)/s).Two significant IsV peaks at 50-60 and~100 d were recognized in the power spectra of the meridional velocity and *** analysis indicated that the IsV at 50-60 d was caused by westward-propagating eddies at average propagation speed of~13 cm/s and wavelength of~635 *** IsV peak at~100 d was mainly caused by northward-propagating eddies generated in the North Equatorial Current *** investigation indicated that the IsV of the Kuroshio at 18°N is dominated by meridional transport,rather than by the zonal migration of the Kuroshio main *** findings provide a better understanding of the IsV of the Kuroshio east of Luzon Island.
