Impact of a Dietary Supplement Containing Rosemary and Daylily on Biochemical Markers of Cognitive Health, Sleep Quality and Related Variables in Men and Women

Background: We investigated the influence of a botanical agent to improve sleep quality and associated measures in men and women with self-reported difficulty sleeping. Methods: 32 individuals were randomly assigned in double blind manner to ingest a botanical agent (CLOCK?, containing Rosemary [Rosmarinus officinalis] and Daylily [Hemerocallis fulva]) or a placebo over a 6-week intervention. During weeks 1 and 2, subjects ingested one serving of the assigned condition, followed by a two-week washout. During weeks 5 and 6, subjects ingested two servings of the condition. The Leeds Sleep Evaluation Questionnaire was used as an outcome measure, as were subjective measures of sleep quality, energy level, and mood. Blood samples collected pre- and post-intervention were assayed for acetylcholine (ACH), brain-derived neurotrophic factor (BDNF), irisin, and melatonin. Results: No differences were noted between conditions in measures of sleep (p > 0.05). While no differences of statistical significance were noted in subjective feelings, during weeks 5 and 6 as compared to baseline, subjects assigned to the supplement noted an 8% increase in attentiveness, an 11% increase in alertness, a 12% increase in focus, a 14% increase in feeling energetic, a 12% increase in enthusiasm, a 23% increase in feeling well rested, an 11% decrease in feeling sluggish, and a 16% decrease in feeling depressed, without the same improvement observed for subjects in the placebo group. All biochemical measures were increased from pre- to post-treatment with two servings of the supplement;the largest percent increase noted for BDNF (27%) and the largest effect size noted for irisin (d = 1.36). Biochemical values for the placebo condition were unchanged. Conclusions: CLOCK? may have an impact on certain measurements of mood, with a significant impact on the biochemical marker, BDNF. Evidence for colorectal sarcomatoid carcinoma arising from tubulovillous adenoma

Sarcomatoid carcinomas of the colorectum are rare tu- mors that display both malignant epithelial and stromal components. Clinically, they are aggressive tumors with early metastasis. Due to their infrequent occurrence, the pathogenesis is poorly understood. We report a case of a 52-year-old woman who presented with a rectal mass and intermittent hematochezia. Superficial biopsies during colonoscopy revealed a tubulovillous adenoma with high-grade dysplasia. Endoscopic ultra- sonography confirmed an invasive nature of the mass, and deeper biopsies revealed the presence of neoplasm with mixed histological components. The surgically- excised specimen demonstrated the presence of poorly differentiated spindle cells underneath the tubulovillous adenoma and an intermediate stage of invasive adeno- carcinoma. Based on the histological appearance and immunohistochemical studies, a diagnosis of sarcoma- toid carcinoma was made. Only nine cases of sarcoma- toid carcinomas of the colorectum have been reported to date. As a result, the terminology and pathogenesis of sarcomatoid carcinoma remain speculative. To the best of our knowledge, this is the first report of co- existence of sarcomatoid carcinoma and invasive ad- enocarcinoma with tubulovillous adenoma; all stages represented within the same tumor. This observation supports the "monoclonal theory" of pathogenesis with an adenoma-sarcoma progression with or without an intermediate stage of carcinoma.

In 1992,The American Naturalist published a special supplement entitled:“Sensory *** Sensory Drive Biology Bias or Constrain the Direction of Evolution?” organized by john *** supplement contained a seminal paper on “sensory drive” by Endler (1992a) as well as several other well-cited papers on sensory exploitation (ryan and Keddy-Hector 1992),background matching with respect to motion (Fleishman 1992),chemical cues in mammals and amphibians (Alberts 1992;Roth et al.1992),and the relationship between auditory processing and call properties in frogs (Narins 1992).The paper by Endler was especially important;it has been cited over 1,200 times and has inspired research at many levels of ecology and evolution across diverse taxa and sensory *** this paper and associated ones,Endler laid out the primary components that influence the evolution of signaling systems,placing a large emphasis on the environmental conditions under which signaling occurs (Endler 1992a,1992b,1993).Twenty-five years later,the American Society of Naturalists held a symposium on “25 Years of Sensory Drive” at the 2017 Evolution meetings in Portland,Oregon,organized by Becky *** special column in Current Zoology summarizes the work presented there as well as other contributions made for the *** this editorial,we first review sensory drive and the state of the field when it *** then summarize the work in this special column and suggest fruitful ways forward.

Osteomyelitis is a devastating disease caused by microbial infection of bone. While the frequency of infection following elective orthopedic surgery is low, rates of reinfection are disturbingly high. Staphylococcus aureus is responsible for the majority of chronic osteomyelitis cases and is often considered to be incurable due to bacterial persistence deep within bone. Unfortunately, there is no consensus on clinical classifications of osteomyelitis and the ensuing treatment algorithm. Given the high patient morbidity,mortality, and economic burden caused by osteomyelitis, it is important to elucidate mechanisms of bone infection to inform novel strategies for prevention and curative treatment. Recent discoveries in this field have identified three distinct reservoirs of bacterial biofilm including: Staphylococcal abscess communities in the local soft tissue and bone marrow, glycocalyx formation on implant hardware and necrotic tissue, and colonization of the osteocyte-lacuno canalicular network(OLCN) of cortical bone. In contrast, *** intracellular persistence in bone cells has not been substantiated in vivo, which challenges this mode of chronic osteomyelitis. There have also been major advances in our understanding of the immune proteome against S. aureus, from clinical studies of serum antibodies and media enriched for newly synthesized antibodies(MENSA), which may provide new opportunities for osteomyelitis diagnosis, prognosis, and vaccine development. Finally, novel therapies such as antimicrobial implant coatings and antibiotic impregnated 3D-printed scaffolds represent promising strategies for preventing and managing this devastating disease. Here, we review these recent advances and highlight translational opportunities towards a cure.
