Impact of COVID-19 pandemic on the ocular surface

BACKGROUND There have been increased reports of dry eyes in the coronavirus disease 2019(COVID-19) pandemic era.AIM To analyze the differences in tear film properties from pre-and post-pandemic of the COVID-19 era.METHODS It was a retrospective comparative study.Patients were divided into three groups according to the data of multimodal ocular surface evaluation:(1) Group 1 if it was before Portugal lockdown decision(from August 2019 to March 2020);(2) Group 2 if it was after Portugal lockdown decision but without mask mandate(from April 2020 to October 2020);and(3) Group 3 if it was after Portugal lockdown but with mask mandate in health public highway(from November 2020 to April 2021).The following variables were analyzed:Lipid layer thickness,blink rate,Schirmer test,tear meniscus height,tear osmolarity,non-invasive break-up time,and loss area of the meibomian glands.RESULTS The study included 548 eyes of 274 patients,aged 18 years to 89 years,with a mean age of 66.15 ± 13.40 years at the time of multimodal ocular surface evaluation.Compared to group 1:(1) Mean lipid layer thickness was better in group 2(P = 0.001) and group 3(P < 0.001);(2) Schirmer test was similar in group 2(P = 0.576) and better in group 3(P = 0.002);(3) Tear osmolarity and loss area of the meibomian glands were worse in group 2(P = 0.031 and P < 0.001,respectively) and in group 3(both with P < 0.001);(4) Blink rate and tear meniscus height were similar in group 2(P = 0.821 and P = 0.370,respectively) and worse in group 3(P < 0.001 and P = 0.038,respectively);and(5) Non-invasive break-up time was worse in group 2(P = 0.030) and similar in group 3(P = 0.263).CONCLUSION Our study demonstrated that differences existed in tear film properties comparing data from the pre-and post-pandemic of the COVID-19 era.

Given the frequent co-existence of an aggressive tumor and underlying chronic liver disease,the management of hepatocellular carcinoma(HCC)patients requires experienced multidisciplinary team discussion.Moreover,imaging plays a key role in the diagnosis,staging,restaging,and surveillance of HCC.Currently,imaging assessment of HCC entails the assessment of qualitative characteristics which are prone to inter-reader variability.Radiomics is an emerging field that extracts high-dimensional mineable quantitative features that cannot be assessed visually with the naked eye from medical imaging.The main potential applications of radiomic models in HCC are to predict histology,response to treatment,genetic signature,recurrence,and survival.Despite the encouraging results to date,there are challenges and limitations that need to be overcome before radiomics implementation in clinical practice.The purpose of this article is to review the main concepts and challenges pertaining to radiomics,and to review recent studies and potential applications of radiomics in HCC.

With the expansion of eucalyptus crops to areas with severe water limitations,physiological studies involving eucalyptus clones to identify those that are tolerant to water stress become important.The objective of this study was to assess morphological and physiological responses by eucalyptus clones subjected to drought stress and rehydration.The experiment consisted of three eucalyptus clones:VC865,I224 and I144 and two water regimes:control and water stress followed by rehydration,with six replicates.Leaf water potential,gas exchange,maximum quantum efficiency of photo systemⅡand plant height and stem diameter were evaluated under drought stress and rehydration.After6 d of rehydration,the number of leaves,leaf area and dry mass of root,leaf,stem and their total were evaluated.All clones showed intense reduction of gas exchange during the drought stress period,and only VC865 and 1144 showed rapid recovery with 3 d of rehydration.Clone 1224 showed greater reduction in height,stem diameter,number of leaves,water potential at midday(Ψ_(w)_(Midday)),and maximum quantum efficiency of photosystemⅡ(F_(v)/F_(m)).Clones VC865and T144 showed lower reductions inΨ_(wMidday)and F_(v)/F_(m) under stress.VC865 had lower reductions in leaf number,leaf area and higher leaf dry mass,while clone I144 had higher height and lower reduction in root dry mass under.Both these clones showed higher water use efficiency with 3d of rehydration.These different phenotypic plasticities gave the clones VC865 and 1144 efficient mechanisms of acclimatization to stress and more drought tolerance,enhancing their greater capacity for recovery after stress,which allowed lower dry mass reduction.Clone 1224,however,was more susceptible to drought stress,undergoing greater physiological damage with only partial recovery during rehydration.

Quantification of theophylline or paracetamol in milk matrices by high-performance liquid chromatography

A simple, accurate and sensitive high-performance liquid chromatography （HPLC） method was developed, validated and applied to the determination of either theophylline or paracetamol in milk-based samples. The method allowed drug quantification in fresh and powdered milk with a relatively short run time of analysis and it was also successfully applied to the quantification of the drugs in solid dosage forms intended for pediatric use. Moreover, the main significant advantages over other published works are the simplicity of the sample preparation, reduced assay time and sample loss. The method meets the International Conference on Harmonization guideline for analytical methods validation regarding specificity, linearity, accuracy, precision, specificity and robustness as required by health authorities and applied by industry while designing and marketing new drug products. The technique encompasses the separation of the analytes with a reverse phase C18 column under isocrafic conditions and UV detection at 272 nm and 243 nm, respectively, for theophylline and paraeetamol. The lower limit of quantification for both drugs was determined as 0.2 μg/mL and the between-batch accuracy was 99.7%. This HPLC method allows quantification of theophylline and paracetamol in milk matrices and it can be applied in the design, development and production of milk-based pediatric dosage forms.

Background：Due to high neutral lipids accumulation in the cytoplasm,in vitro-produced embryos from Bos primigenius indicus and their crosses are more sensitive to chilling and cryopreservation than those from Bos primigenius taurus.The objective of the present study was to evaluate the effects of trans-10,cis-12 conjugated linoleic acid（CLA） on the development and cryotolerance of crossbred Bos primigenius taurus x Bos primigenius indicus embryos produced in vitro,and cultured in the presence of fetal calf serum.Bovine zygotes（n = 1,692）were randomly assigned to one of the following treatment groups：1） Control,zygotes cultured in Charles Rosenkrans 2 amino acid（CR2aa） medium（n = 815） or 2） CLA,zygotes cultured in CR2 aa medium supplemented with 100 μmol/L of trans-10,cis-12 CLA（n =877）.Embryo development（cleavage and blastocyst rates evaluated at days 3 and 8 of culture,respectively）,lipid content at morula stage（day 5） and blastocyst cryotolerance（re-expansion and hatching rates,evaluated 24 and 72 h post-thawing,respectively） were compared between groups.Additionally,selected mRNA transcripts were measured by Real-Time PCR in blastocyst stage.Results：The CLA treatment had no effect on cleavage and blastocyst rates,or on mRNA levels for genes related to cellular stress and apoptosis.On the other hand,abundance of mRNA for the 1-acylglycerol-3-phosphate0-acyltransferase-encoding gene（AGPAT）,which is involved in triglycerides synthesis,and consequently neutral lipid content,were reduced by CLA treatment.A significant increase was observed in the re-expansion rate of embryos cultured with trans-10,cis-12 CLA when compared to control（56.3 vs.34.4%,respectively,P = 0.002）.However,this difference was not observed in the hatching rate（16.5 vs.14.0%,respectively,P=0.62）.Conclusions：The supplementation with trans-10,ds-12 CLA isomer in culture medium reduced the lipid content of in vitro produced bovine embryos by reducing the gene expression of 1-acylglycerol 3-phosphate 0-acyltransferase（AGPAT） enzyme.However,a possibl

This study aims at investigating the effects of foliar application of aqueous solutions of La3+and Ce3+nitrates on soybean plants(Glycine max).First,we observe that Triton HW 1000 surfactant at 0.01 vol%reduces droplets contact angle and increases their drying time.Under greenhouse conditions,the foliar treatments do not affect chlorophyll content,plant height,number of leaves,number of pods,number of seeds per pod,and average seed weight.However,the treatments induce phytotoxicity since foliar injuries appear after the spraying.Microprobe X-ray fluorescence spectroscopy combined to scanning electron microscopy shows that the leaf lesions are positively correlated to accumulation of Ce and La on the leaf surface and also promote structural alteration to the epidermal cells.X-ray absorption near edge structure shows that the La and Ce nitrates are partially bio transformed into oxides by the leaves which might explain the harmful effects.

AIM: To analyze therapeutic changes in Crohn's disease(CD) patients following video capsule endoscopy(VCE) and to assess the usefulness of Lewis score and the Patency Capsule.METHODS: Patency Capsule was performed in every patient that had indication for VCE, and those with negative patency did not undergo VCE. Patients with established CD that underwent VCE between January 2011 and February 2014 were selected for this study; those with suspected CD were excluded, independent of VCE results, since our purpose was to address differences in therapeutic regimen in CD patients before and after VCE. Patients with inconclusive VCE were also excluded. Patients had to be free of non-steroidal anti-inflammatories for at least 1 mo. Those patients who met these criteria were allocated into one of three groups: Staging group(asymptomatic CD patients that underwent VCE for staging of CD), Flare group(patients with active CD), or Post-op group(CD patients evaluated for post-operative recurrence). Lewis score was calculated for every VCE procedure. Statisticalanalysis was performed to address the impact of VCE findings on the therapeutic management of CD patients and to evaluate the utility of the Lewis score.RESULTS: From a total of 542 VCEs, 135 were performed in patients with CD. Patency capsule excluded nearly 25% of the patients who were supposed to undergo VCE. No videocapsule retention during VCE was reported. From these 135 patients, 29 were excluded because CD diagnosis was not established at the time of VCE. Therefore, a total of 106 patients were included in the final analysis. From these, the majority were in the Staging group(n = 73, 69%), and the remaining were in the Flare(n = 23, 22%) or Post-op(n = 10, 9%) group. Median time between diagnosis and VCE was 5.5 years. Overall, VCE determined changes in the treatment of 40% of patients: only 21% remained free of immunosuppressors after VCE compared to 44% before VCE(P < 0.001).
