Combined influence of basal media and fibroblast growth factor on the expansion and differentiation capabilities of adipose-derived stem cells

Background: Interest in adipose-derived stem cells (ASCs) has increased in recent years due to their multi-linagedifferentiation capabilities. While much work has been done to optimize the differentiation media, few studies havefocused on examining the influence of different expansion media on cell behavior. In this study, three basal media(low glucose Dulbecco’s modified Eagle’s medium (DMEM), high glucose DMEM and DMEM-F12) supplementedwith or without fibroblast growth factor 2 (FGF) were examined to assess their suitability for expanding ***: Flow cytometry, colony-forming unit assays (CFU-Fs) and differentiation assays were utilized to study cellbehavior. High glucose media CFU-Fs produced fewest colonies while the addition of FGF increased colony *** passage 2, the majority of cells were positive for CD44, 45, 73, 90 and 105 and negative for CD14, 31 and 45,indicating a mesenchymal phenotype. A sub-population of CD34 positive cells was present among passage 2 cells;however, by passage 4 the cells were negative for CD34. FGF has a negative effective on passage 4 ASC adipogenesisand high glucose media plus FGF-enhanced osteogenic capacity of passage 4 ASCs. FGF supplemented basal mediawere most suitable for chondrogenesis. High glucose media plus FGF appeared to be the most beneficial for primingASCs to induce a keratocyte ***: These findings demonstrate the reciprocal effect FGF and basal media have on ASCs. This research hasimplications for those interested regenerating bone, cartilage, cornea or adipose tissues.

Background: The negative impacts of the exotic tree, Ailanthus altissima(tree-of-heaven, stink tree), is spreading throughout much of the Eastern United States. When forests are disturbed, it can invade and expand quickly if seed sources are ***: We conducted studies at the highly dissected Tar Hollow State Forest(THSF) in southeastern Ohio USA,where Ailanthus is widely distributed within the forest, harvests have been ongoing for decades, and prescribed fire had been applied to about a quarter of the study area. Our intention was to develop models to evaluate the relationship of Ailanthus presence to prescribed fire, harvesting activity, and other landscape characteristics, using this Ohio location as a case study. Field assessments of the demography of Ailanthus and other stand attributes(e.g., fire, harvesting, stand structure) were conducted on 267 sample plots on a 400-m grid throughout THSF,supplemented by identification of Ailanthus seed-sources via digital aerial sketch mapping during the dormant season. Statistical modeling tools Random Forest(RF), Classification and Regression Trees(CART), and Maxent were used to assess relationships among attributes, then model habitats suitable for Ailanthus ***: In all, 41 variables were considered in the models, including variables related to management activities, soil characteristics, topography, and vegetation structure(derived from LiDAR). The most important predictor of Ailanthus presence was some measure of recent timber harvest, either mapped harvest history(CART) or LiDARderived canopy height(Maxent). ObjectiveToinvestigate whether depression, anxiety and stress increase the risk for delirium and poor quality of life （QOL） after co-ronary artery bypass （CABG） *** total of 180 CABG patients （mean age of 63.5 ± 10.1 years, 82.2% males） completed baseline and postoperative self-report questionnaires to assess distress and QOL. Incident delirium was diagnosed postoperatively with a structured clinical interview and patients were monitored every day post-operatively for confusion and disturbance in *** Delirium developed in 63 persons （35% of sample）. After adjustment for covariates, delirium was significantly associated with depression [odds ratio （OR）： 1.08; 95% confidence interval （CI）： 1.03-1.13,p = 0.003], anxiety （OR： 1.07; 95% CI： 1.02-1.13,p= 0.01） and stress （OR： 1.05; 95% CI： 1.00-1.09,p= 0.03）. preoperative depression scores were associated with poorer QOL including bodily pain （β =-0.39,p = 0.013）, vitality （β=-0.32,p = 0.020）, social functioning （β=-0.51,p≤0.001）, emotional role function （β=-0.44,p = 0.003） and general health （β=-0.33,p = 0.038）. Among the covariates, harmful levels of alcohol use was consistently associated with poorer *** Depression and harmful levels of alcohol use were consistently associated with poorer QOL whereas depression, anxiety and stress were associated with delirium risk. These findings point to further research examining depression and harmful levels of alcohol use in coronary heart disease populations undergoing coronary revascularization.

Water disinfection is an essential process that provides safe water by inactivating pathogens that cause waterborne diseases. However, disinfectants react with organic matter naturally present in water, leading to the formation of disinfection by-products(DBps). Multi-analyte methods based on mass spectrometry(MS) are preferred to quantify multiple DBp classes at once however, most require extensive sample pre-treatment and significant resources. In this study, two analytical methods were developed for the quantification of 32 regulated and unregulated DBps. A purge and trap(p&T) coupled with gas chromatography mass spectrometry(GC-MS) method was optimized that automated sample pre-treatment and analyzed volatile and semi-volatile compounds, including trihalomethanes(THMs), iodinated trihalomethanes(I-THMs), haloacetonitriles(HANs), haloketones(HKTs) and halonitromethanes(HNMs). LOQs were between 0.02-0.4 μg/L for most DBps except for 8 analytes that were in the low μg/L range. A second method with liquid chromatography(LC) tandem mass spectrometry(MS/MS) was developed for the quantification of 10 haloacetic acids(HAAs) with a simple clean-up and direct injection. The LC-MS/MS direct injection method has the lowest detection limits reported(0.2-0.5 μg/L). Both methods have a simple sample pre-treatment, which make it possible for routine analysis. Hyperchlorination and uniform formation conditions(UFC) formation potential tests with chlorine were evaluated with water samples containing high and low TOC. Hyperchlorination formation potential test maximized THMs and HAAs while UFC maximized HANs. Ascorbic acid was found to be an appropriate quencher for both analytical methods. Disinfected drinking water from four water utilities in Alberta, Canada were also evaluated.

Background:Vascular inflow occlusion(VIO)during liver resections(pringle manoeuvre)can be applied to reduce blood loss,however may at the same time,give rise to ischemia-reperfusion injury(IRI).The aim of this study was to assess the characteristics of hepatic microvascular perfusion during VIO in patients undergoing major liver ***:Assessment of hepatic microcirculation was performed using a handheld vital microscope(HVM)at the beginning of surgery,end of VIO(20 minutes)and during reperfusion after the termination of *** microcirculatory parameters assessed were:functional capillary density(FCD),microvascular flow index(MFI)and sinusoidal diameter(SinD).Results:A total of 15 patients underwent VIO;8 patients showed hepatic microvascular perfusion despite VIO(partial responders)and 7 patients showed complete cessation of hepatic microvascular perfusion(full responders).Functional microvascular parameters and blood flow levels were significantly higher in the partial responders when compared to the full responders during VIO(FCD:0.84±0.88 vs.0.00±0.00 mm/mm2,ppectively,and MFI:0.69–0.22 vs.0.00±0.00,ppectively).Conclusions:An interpatient heterogeneous response in hepatic microvascular blood flow was observed upon *** may explain why clinical strategies to protect the liver against IRI lacked consistency.
