Cigarette smoking is the main cause of chronic obstructive pulmonary disease (COPD). Diaphragm injury is observed in patients with COPD. However, the potential role of smoking in triggering or perpetuating muscle injury is unknown. The present study was aimed at evaluating the potential role of commercial tobacco smoke as a direct cause of skeletal muscle injury in experimental conditions. Seventy Wistar rats (170 - 250 g) were assigned to smoking (n = 49) or non-smoking (n = 21) groups. The smoking groups were submitted to a single or multiple (i.e., five or thirty) daily sessions of cigarette smoking in an inhalatory chamber (time length: 2 h each session). The level of exposure was constant and assessed by CO concentrations (50 ppm) and serum cotinine analysis. Animals submitted to a single smoke exposure and the corresponding controls were euthanized in groups at 0 h, 2 h, 4 h, 24 h or 48 h after completing the exposure. Animals submitted to multiple exposures were euthanized at 0 h after smoking. Samples from vastus lateralis muscle were obtained and processed for assessing cell injury and selected protein expression. Monoclonal anti-albumin antibodies were used to identify muscle fibers with sarcolemmal (membrane) injury. Subcellular muscle injury was assessed using transmission electron microscopy (EM). MyoD, myogenin and α-tubulin were immunodetected using western blot techniques. Exposure to cigarette smoke associated with significant membrane damage (mean relative difference (MRD) with controls: +181%, p = 0.004) and sarcomere disruptions (MRD: +226%, p = 0.001). Expression of MyoD and myogenin (normalized to α-tubulin) were significantly increased at 4 h and remained increased at 48 h post-exposure. We conclude that not only a single but also consecutive exposure to tobacco smoke have acute deleterious effects on peripheral muscle structure. Genetic Diversity of Great Dane Breed Using Ten Microsatellites: Impact of Breeding Control over the Breeding Line

The American Kennel Club and the Fédération Cynologique Internationale recognize two phenotypic variants of Great Dane breed denominated American and European varieties. Historically, these varieties have been segregated according to morphological characteristics. In an attempt to obtain a better characterization, breeders have been interested in the genetic parameters that could evaluate the within-breed diversity. In this document, we studied the genetic structure of Great Dane breed with 10 STR markers in 88 dogs using capillary electrophoresis. Cluster analysis, population differentiation and phylogenetic analyses revealed that American and European varieties are genetically independent. Nevertheless, within the American variety a genetic stratification was found. Additionally, a high misclassification (28%) was detected, which could be due to wrong registration or false paternity. Our results support the importance to deem genetic markers as useful tools in breeding control. Similarly, these studies serve as an accurate reference to establish standards by dog breeding associations and for choosing among dogs for inter-breeding. Nevertheless, genetic tools are only a complement of morphological methods, since both are reshuffling the control over the breeding line. Notwithstanding, this database provides an overall and scape concerning the impact of genetic diversity within-breed. Unmistakably, more databases are needed to increase the quality of the breeding line as well as the number of STR in order to study, with more detail, the genetic structure in the Great Dane race.

*** strongest locus which associated with type 2 diabetes(T2D)by the common variant rs7903146 is the transcription factor 7-like 2 gene(TCF7L2).We aimed to quantify the interaction of diet/lifestyle interventions and the genetic effect of TCF7L2 rs7903146 on glycemic traits,body weight,or waist circumference in overweight or obese adults in several randomized controlled trials(RCTs).*** October 2016 to May 2018,a large collaborative analysis was performed by pooling individualparticipant data from 7 *** RCTs reported changes in glycemic control and adiposity of the variant rs7903146 after dietary/lifestyle-related interventions in overweight or obese *** treatment interaction models which used the genetic effect encoded by the allele dose and common covariates were applicable to individual participant data in all *** the joint analysis,a total of 7 eligible RCTs were included(n=4,114).Importantly,we observed a significant effect modification of diet/lifestyle-related interventions on the TCF7L2 variant rs7903146 and changes in fasting *** with the control group,diet/lifestyle interventions were related to lower fasting glucose by-3.06(95%CI,-5.77 to-0.36)mg/dL(test for heterogeneity and overall effect:I^(2)=45:1%,p<0:05;z=2:20,p=0:028)per one copy of the TCF7L2 T risk ***,regardless of genetic risk,diet/lifestyle interventions were associated with lower waist ***,there was no significant change for diet/lifestyle interventions in other glycemic control and adiposity traits per one copy of TCF7L2 risk *** findings suggest that carrying the TCF7L2 T risk allele may have a modestly greater benefit for specific diet/lifestyle interventions to improve the control of fasting glucose in overweight or obese adults.

Traditional mining activities are usually correlated with high levels of soil pollution, which is a major environmental concern. Extensive mining activities have taken place in the San Joaquin region in the State of querétaro, México resulting in high levels of mercury soil pollution (up to 1532 ± 300 mg/kg). We isolated mercury-resistant fungal strains from the San Joaquin region soils and identified them through morphologic characteristics and ITS rDNA region sequence analysis. We determined that fungi isolated belong to the genus Trichoderma. All the isolates selected showed the ability to catalyze the volatilization of Hg. For air sampling, an active sampling device was constructed and using acid KMnO4 as an absorbent, the concentration of mercury in solution was determined through the cold vapor atomic absorption method. The results show mercury volatilization from the fungal species assay, with a maximum of 213.04 ± 32.6 μg/m3 while mycelium accumulation ranged from less than 17.5 ± 2.9 to 20.0 ± 3.4 μg/g. The fungal isolates were also evaluated for their ability to reduce mercury uptake in Arabidopsis thaliana. These observations suggest the utility of Trichoderma for the mobilization of mercury in those contaminated soils.
