AIm:The functional integrity of the blood-testis barrier (BTB) in male mice exposed to Cr(V) was studied in order to clarify the mechanism underlying testicular injury.mETHoDS:Adult male mice were subcutaneously injected repeated doses of 8.02 micromol (0.5 ml) of Cr/*** for 5 days. Animals receiving a similar volume of bis(hydroxyethyl)-aminotris(hydroxymethyl)methane buffer (BT) were used as controls. The animals were sacrificed on day 6 and small fragments of seminiferous tubules, approximately 8-10 mm length, were incised and sutured at both ends. They were exposed in vitro to horseradish peroxidase-containing culture medium for 10 minutes. Tissues were then fixed and processed for ultrastructural studies.RESULTS:Controls and Cr(V)-treated group resulted in the uptake of the tracer by Sertoli cells. However, the major finding consisted in the permeability of the BTB only in the Cr(V)-group, as evidenced by the presence of the tracer within the junctions between the neighbouring Sertoli cells.CoNCLUSIoN:The BTB is disrupted in mice submitted to Cr(V). The permeability of the BTB is a crucial feature to be investigated for the understanding of lesions within the seminiferous tubule.

Despite improvements in adjuvant therapies for gastric cancer in recent years, the disease is characterized by high recurrence rates and a dismal prognosis. The major improvement in the treatment of recurrent or metastatic gastric cancer in recent years has been the incorporation of trastuzumab, a monoclonal antibody that inhibits human epidermal growth factor receptor 2(HER2) heterodimerization, after the demonstrated predictive value of the overexpression and/or amplification of this receptor. Beyond HER2, other genetic abnormalities have been identified, and these mutations may be targetable by tyrosine kinase inhibitors or monoclonal antibodies. The demonstration of four distinct molecular subtypes of gastric cancer by the Cancer Genome Atlas study highlight the enormous heterogeneity of the disease and its complex interplay between genetic and epigenetic alterations and provide a roadmap to implement genome-guided personalized therapy in gastric cancer. In the present review, we aim to discuss, from a clinical point of view, the genomic landscape of gastric cancer described in recent studies, the therapeutic insights derived from these findings, and the clinical trials that have been conducted and those in progress that take into account tailored therapies for gastric cancer.

The inflammatory pattern during Helicobacter pylori(***)infection is changeable and *** childhood,it is possible to observe a predominantly regulatory response,evidenced by high concentrations of key cytokines for the maintenance of Treg responses such as TGF-β1 and IL-10,in addition to high expression of the transcription factor *** the other hand,there is a predominance of cytokines associated with the Th1 and Th17 responses among ***-positive *** the last few years,the participation of the Th17 response in the gastric inflammation against *** infection has been highlighted due to the high levels of TGF-β1 and IL-17 found in this infectious scenario,and growing evidence has supported a close relationship between this immune response profile and unfavorable outcomes related to the ***,this cytokine profile might play a pivotal role in the effectiveness of *** *** is evident that age is one of the main factors influencing the gastric inflammatory pattern during the infection with ***,and understanding the immune response against the bacterium can assist in the development of alternative prophylactic and therapeutic strategies against the infection as well as in the comprehension of the pathogenesis of the outcomes related to that microorganism.

Altitude as environmental filtering influencing phylogenetic diversity and species richness of plants in tropical mountains

Elucidating how multiple factors affect biodiversity and plant community assembly is a central issue in ecology,especially in vulnerable ecosystems such as tropical *** studies are more relevant in global warming scenarios that induce the upward displacement of plant species towards reduced habitats and hostile environments in tropical *** study aimed to analyze how altitude affects taxonomic and phylogenetic diversity in plant communities of tropical ***,we tested if(i)increased altitude works as an environmental filtering promoting decreased species richness,decreased phylogenetic diversity,and increased phylogenetic clustering in these tropical mountains;and if(ii)plant communities of high altitude in tropical mountains are also result of recent diversification with plant species recently split shortening phylogenetic distances between closest related *** tested effects of altitude on species richness and phylogenetic metrics using linear mixed-effects *** Haleakala presented 114 species,mount Kilimanjaro presented 231 species and mount Purace presented 280 *** found an environmental filtering effect with increasing altitude causing phylogenetic clustering,decreased phylogenetic diversity and decreased species *** decreasing phylogenetic distances between closest relatives are congruent with neo-endemics,suggesting recent plant diversification in high altitudes of tropical mountains,possibly driven by geographic isolation and environmental *** of global warming should be monitored in tropical mountains focusing on distribution shifts.

Pine-fruit shell (PFS) is a lignocellulosic residue derived from the fruit of Araucaria angustifolia, a coniferoustree native of South America, part of a whole vegetation of the Atlantic Forest, found in the South and Southwestof Brazil. In this work PFS will be characterized and used in the production of PFS-based polyols throughoxypropylation. Three series were chosen (PFS/propylene oxide (Po) (w/v, g/mL) of 30/70, 20/80 and 10/90)with four catalyst levels (5%, 10%, 15% and 20%, (w/w, PFS based)). oxypropylation occurred at moderateconditions of temperature, pressure and time giving rise to liquid polyols with a homopolymer content (PPo)ranging from 4-65%, a hydroxyl number (IoH) between 257-605 mg KoH/g and viscosities (V) varying from0.76 Pa.s to 373.90 Pa.s (20°C) for the series 20/80 and 10/90, while for the series 30/70, the viscosity reachedvalues higher than 500 Pa.s, 20°C. The unreacted PFS (UR) varied between 3.6% and 77.4% (PFS-basis). Afterthe PFS-based polyols production and characterization, a principal component analysis (PCA) was performed inorder to evaluate the established interactions between the used formulation variables and the obtained polyolproperties. The PCA analysis allowed to clarify the interactions between PFS and Po contents and the finalbiopolyol properties (PPo, IoH, V and UR). This approach showed to be a simple method to rationally analyzethe influence of the input formulation variables on the final polyol properties.

objective:To evaluate short-term markers of outcome in diabetic macular edema(DmE).methods:Prospective interventional case series included 122 eyes of 122 patients with recently diagnosed *** were treated with a 3-monthly loading dose of ranibizumab or aflibercept and pro re nata *** enhanced deep imaging SD-oCT high resolution scans were used to measure subfoveal choroidal thickness(SFCT)and central retinal thickness(CRT).Anatomic(10%CRT decrease)and functional responses(best corrected visual acuity,BCVA gain≥5 letters)were assessed at 3 months and 6 months using univariate and multivariate *** tested were gender,duration of diabetes,HbA1c,hypertension,CRT,SFCT,BCVA,ellipsoid zone(EZ)status,subfoveal neuroretinal detachment(SND),anti-VEGF used and laser naivety.A logistic regression model was applied to find independent markers ***:BCVA increased,CRT and SFCT decreased at 3 months and 6 *** metabolic control(p=0.003),intact baseline EZ(p=0.030),EZ re-grading at 3 m(pociated with better functional *** multivariate linear regression model showed that baseline SND and CRT are predictors of anatomic response,while lower baseline BCVA and intact EZ are predictors of functional ***:The presence of SND predicts anatomic response only,while an intact EZ is critical to achieve a good functional outcome in DmE.

This work aimed to study and compare the effect of the incorporation of agro-industrial by-products(chestnut shell,grapeseed,and pomegranate peel)into yoghurts to fortify them while assessing their potential to replace synthetic *** each by-product,phenolic extracts were obtained and *** extracts demonstrated antioxidant and antibacterial properties,and the capacity to inhibitα-*** shell extract stood out regarding antioxidant capacity,displaying values of 1128 and 972 mgT_(rolox•gextract)^(-1),for the assays with DPPH and ABTS,*** yoghurts were produced(negative control,positive control with 0.1%of sorbic acid,two for each extract with 0.1%and 0.2%of extract,a mixture of extracts and a mixture of extracts with sorbic acid),and stored at 4℃.The incorporation of the extracts into the yoghurts maintained their physical-chemical properties and microbiological *** samples inhibited the growth of Escherichia coli and Staphylococcus *** analysis proved that higher concentrations of extract had similar results to synthetic *** results showed the viability to fortify yoghurts with the incorporation of by-product extracts,developing a value-added ***,revealed the possibility of these extracts replacing synthetic preservatives and antioxidants.

Brazilian industry produces huge amounts of tannery sludge as residues,which is often disposed by landfilling or land ***,consecutive amendment of such composted industrial wastes may cause shifts in soil microbial biomass(SmB) and enzyme *** study aimed to evaluate SmB and enzyme activity after 3-year consecutive composted tannery sludge(CTS) amendment in tropical sandy *** amounts of CTS(0.0,2.5,5.0,10.0,and 20.0 t ha^(-1)) were applied to a sandy *** C and N contents of SmB,basal and substrate-induced respiration,respiratory quotient(qCo_2),and enzyme activities were determined in the soil samples collected after CTS amendment for 60 d at the third *** 3 years,significant changes were found in soil microbial properties in response to different CTS amounts *** organic matter and Cr contents significantly increased with increasing CTS *** and soil respiration peaked following amendment with 10.0 and 20.0 t ha-1 of CTS,respectively,while qCo_2was not significantly affected by CTS ***,soil enzyme activity decreased significantly with increasing CTS *** CTS amendment for 3 years showed inconsistent and contrasting effects on SmB and enzyme *** decrease in soil enzyme activities was proportional to a substantial increase in soil Cr concentration,with the latter exceeding the permitted concentrations by more than ***,our results suggest that a maximum CTS quantity of 5.0 t ha^(-1) can be applied annually to tropical sandy soil,without causing potential risks to SmB and enzyme activity.
