Antibacterial activity and inhibition against Staphylococcus aureus NorA efflux pump by ferulic acid and its esterified derivatives

objective:To evaluate the inhibitory activity of ferulic acid and four of its esterified derivatives(methyl,ethyl,propyl,and butyl)against resistance mechanisms in Staphylococcus aureus ***:Ferulic acid derivatives were obtained by esterification with methanol,ethanol,propanol,and butanol,and then characterized by hydrogen and carbon-13 nuclear magnetic resonance *** minimum inhibitory concentrations(MIC)of ferulic acid and its esterified derivatives,ethidium bromide,and norfloxacin were obtained using the microdilution test,while the efflux pump inhibition test was conducted by examining reduction in the MICs of norfloxacin and ethidium *** docking was also carried out using the Schrodinger Suite 2015 molecular modeling software.A three-dimensional model of NorA efflux pump was generated using *** best scoring model was used as a receptor for ligand-receptor ***:The methyl and butyl ester derivatives did not demonstrate significant antimicrobial ***,a significant synergic effect was evidenced when norfloxacin was combined with the ethyl and propyl esterified *** docking study demonstrated favorable energy of interaction between ferulate derivatives and NorA,and amino acid residues TYR57,TYR58,and LEU255 were present commonly in stabilizing all *** PCA analysis corroborated the docking hypothesis that the lipophilic character and hydrogen bond interactions were the most relevant characteristics involved with NorA *** pharmacokinetic parameters of ferulic acid derivatives showed good ADMET properties,demonstrating that they can be easily absorbed and have no effect or inhibit the cytochrome P450 enzyme complex,revealing their potential as drug ***:This study provides strong evidence that the molecular basis for this activity is potentially due to the NorA efflux pump.

Immune response to inactivated bacterial vector carrying the recombinant K39 antigen of Leishmania infantum in mice

objective:To evaluate the immunological response elicited by an inactivated bacterial vector carrying the K39 antigen of Leishmania infantum,and a purified ***:Mice were subjected to the following treatments:(1)Purified recombinant K39(rK39)protein at a 20μg dose with complete Freund’s adjuvant;(2)Inactivated Escherichia coli(BL21 DE3)carrying the K39 protein at an equivalent total protein content of 200μg;(3)Inactivated bacteria lacking the K39 protein;(4)Non-immunized control *** monitoring was *** groups were challenged by intraperitoneal injection of 10^(7) Leishmania infantum *** euthanasia,the liver and spleen were collected to analyze the levels of TNF,IFN-γ,IL-12,IL-4,and ***:Mice immunized with purified rK39 or the inactivated bacterial vector carrying the K39 antigen of Leishmania infantum showed a long-lasting immune response with high levels of polyclonal antibodies specifically recognizing the recombinant *** IgG1 subclass was the predominant immunoglobulin;however,the induction of IgG2a and the profile of cytokines produced were indicative of the induction of a mixed-type ***:The inactivated bacterial vector carrying the K39 antigen,as well as the purified antigen can induce a long-lasting immune response in immunized mice,predominantly favouring a Th2 profile response.

Fire is an exceptional action that may occur during the life of a ***,it must be considered when designing a building *** standards provide several types of design methods for that propose,used for single elements,parts of structure or the structure as a *** fire design of columns is important both for new project as for remodel buildings and also for verification of the residual resistance of columns that have suffered a fire *** this way,the aim of this work is to analyze numerically different ways of fire exposure to check the compressive strength of the columns when subjected to fire and the influence of the adjacent walls to a column in case of *** thermal advanced analysis of the sections columns was performed using the finite element software,Abaqus CAE,where the standard fire curve,ISo 834(International organization for Standardization 834),was used,with 4 h of fire *** was possible,with the two methods used in this work,to compare them to verify which model is more conservative and which is closer to the advanced numerical model,for calculating temperatures in the column *** was checked that the walls act as thermal insulators,protecting part of the columns from the convection and radiation of the ***,the effects of raising the temperature over the compressive resistance of the reinforced concrete column,were reduced.

Helicobacter pylori in dental plaque and stomach of patients from Northern Brazil

AIM: To establish whether virulence factor genes vacA and cagA are present in Helicobacter pylori (H. pylori) retrieved from gastric mucosa and dental plaque in pa-tients with dyspepsia. METHoDS: Cumulative dental plaque specimens and gastric biopsies were submitted to histological exami-nation, rapid urease test and polymerase chain reac-tion (PCR) assays to detect the presence of cagA and vacA ***: Detection of H. pylori from dental plaque and gastric biopsy samples was greater by PCR com-pared to histological examination and the rapid ure-ase test. DNA from H. pylori was detected in 96% of gastric mucosa samples and in 72% of dental plaque samples. Sixty-three (89%) of 71 dental plaque sam-ples that were H. pylori-positive also exhibited identical vacA and cagA genotypes in gastric mucosa. The most common genotype was vacAs1bm1 and cagA positive, either in dental plaque or gastric mucosa. These viru-lent H. pylori isolates were involved in the severity of clinical ***: These pathogenic strains were found simultaneously in dental plaque and gastric mucosa, which suggests that gastric infection is correlated with the presence of H. pylori in the mouth.

Hospitalization and occurrence of Death in Frail older Adults: An Integrative Review

objective: To identify the relationship between hospitalization and occurrence of death among frail older adults in the scientific productions on the topic. Method: An integrative literature review is conducted by elaborating the research question, searching in the databases using descriptors, presenting and interpreting the results, and synthesizing the knowledge. Results: The search performed in April 2023 yielded 5804 studies, of which seven remained after applying the inclusion/exclusion criteria. There was an association between frailty and postoperative mortality in the older adults, p p Conclusion: An association was verified between frailty and in-hospital death, signaling frailty as a prognosis for negative outcomes. It is considered fundamental to assess frailty in frail aged people.

Salvia hispanica L. is a herbacia plant that originates from Mexico and Guatemala, and it is currently known by the popular name of chia. Currently, chia seeds have been considered to be of great importance for human health and nutrition because they have a high concentration of polyunsaturated fatty acids. They contain the largest known percentage of fatty α-linolenic acid (ALA) in plants—approximately 68%. Furthermore, they are an excellent source of protein, dietary fiber, calcium, magnesium, iron, vitamin B and phenolic compounds that have antioxidant properties. However, despite the high nutritional value present in the food and the possible health benefits of its nutrients, there is a need to evaluate the bioaccessibility of its micronutrients to measure their effectiveness. Thus, we evaluated the chemical composition of chia seeds from different producers, their lipid profiles and the bioaccessibility of some of their minerals.

Motivational and Methodological Factors Involved in the Helping Behavior Test in Laboratory Rodents

Empathy allows humans and other animals to share the emotional state of another, adopting that individual’s perspective on a given situation. This ability is fundamental for species living in groups. Helping behavior in laboratory animals has been used to study empathy. In this test, subjects are exposed to a conspecific that is trapped and learn to open the cage to release the other animal. However, the interpretation of helping behavior as an emphatically motivated action is still controversial. Here we review the studies that use the helping behavioral test proposed by Ben-Ami Bartal and colleagues in 2011 to better understand motivational factors for this behavior. In addition, we compare methodological aspects of these studies. In conclusion, helping behavior can be driven by empathy, but other factors such as the desire for social contact and learning components cannot be ruled out as motivators. In addition, studies focused on evaluating neurobiological mechanisms underlying helping behavior in laboratory rodents can help elucidate the factors involved in releasing the trapped co-specific.

An Application of the Taguchi Method (Robust Design) to Environmental Engineering: Evaluating Advanced oxidative Processes in Polyester-Resin Wastewater Treatment

This paper presents the Taguchi Method, a statistical design modelling for experiments applied in environmental engineering. This method was applied to optimize the treatment conditions of polyester-resin effluent by means of Advanced oxidative Processes (AoPs) using chemical oxygen demand (CoD) as response parameter. The influence of each independent parameter including respective interactions was evaluated by Taguchi Method, which allowed determining the most statistically significant variables and conditions to best fit the process. Results showed that Taguchi Method is a very useful tool for environmental engineering field and possible simplifications of analysis and calculations through commercially available software.

To evaluate the characteristics and synthesis activity of osteoblasts extracted from the calvaria of offspring of rats exposed to maternal *** adult Wistar rats were divided into two groups,one control and one treated with daily administration of L-thyroxine by an orogastric tube(50μg/animal/day)during *** days after delivery and confirmation of the mothers’hyperthyroidism,the offspring were euthanized for the extraction of osteoblasts from the *** 7,14,and 21 days,proliferation activity was assessed using MTT assay,while alkaline phosphatase(ALP)activity was assessed by the BCIP/NBT *** 21 days,the total area of the mineralized matrix stained by von Kossa was evaluated by *** expression of gene transcripts for Runx2,Bmp2,Fgfr1,collagen type 1(Col1),osteocalcin(oc),and osteopontin(op)were evaluated by real-time *** were compared using the Student’s *** activity was significantly higher at 14 and 21 days in cultures of osteoblasts extracted from offspring exposed to maternal hyperthyroidism,while MTT conversion was significantly lower at 21 days in this *** cultures of neonates exposed to maternal hyperthyroidism also showed a larger total area of mineralized matrix and greater expression of gene transcripts for oc and *** hyperthyroidism increases the activity of matrix synthesis,alkaline phosphatase activity,and expression of gene transcripts for osteocalcin and osteopontin in the osteoblasts,extracted from the calvaria of the offspring,which may be one of the mechanisms of premature fusion of cranial sutures.

Drought stress is major abiotic stress that affects soybean ***,it is widely desirable that soybean becomes more tolerant to *** provide insights into regulatory mechanisms of the stress response,we compared the global gene expression profiles from leaves of two soybean genotypes that display different responses to water-deficit(BR 16 and Embrapa 48,drought-sensitive and droughttolerant,respectively).After the RNA-seq analysis,a total of 5335 down-regulated and 3170 up-regulated genes were identified in the *** the other hand,the number of genes differentially expressed was markedly lower in the Embrapa 48,355 up-regulated and 471 down-regulated ***,induction and expression of protein kinases and transcription factors indicated signaling cascades involved in the drought ***,the results suggest that the metabolism of pectin is differently modulated in response to drought stress and may play a role in the soybean defense mechanism against *** occurs via an increase of the cell wall plasticity and crosslink,which contributed to a higher hydraulic conductance(Kf)and relative water content(RWC%).The drought-tolerance mechanism of the Embrapa 48 genotype involves remodeling of the cell wall and increase of the hydraulic conductance to the maintenance of cell turgor and metabolic processes,resulting in the highest leaf RWC,photosynthetic rate(A),transpiration(E)and carboxylation(A/Ci).Thus,we concluded that the cell wall adjustment under drought is important for a more efficient water use which promoted a more active photosynthetic metabolism,maintaining higher plant growth under drought stress.
