Rat cardiomyocyte H9c2(2-1)-based sulforhodamine B assay as a promising in vitro method to assess the biological component of effluent toxicity

The treatment of wastewaters is crucial to maintain the ecological status of receiving waters,and thereby guarantee the protection of aquatic life and human *** quality evaluation is conventionally based on physicochemical parameters,but increasing attention has been paid to integrate physicochemical and biological ***,the regulatory use of fish in biological testing methods has been subject to various ethical and cost concerns,and in vitro cell-based assays have thus become an important topic of ***,the present study intends:(a) to evaluate the efficiency of two different sample pre-concentration techniques (lyophilisation and solid phase extraction) to assess the toxicity of municipal effluents on rat cardiomyoblast H9c2(2-1) cells,and (b) maximizing the use of the effluent sample collected,to estimate the environmental condition of the receiving *** gathered results demonstrate that the H9c2(2-1) sulforhodamine B-based assay is an appropriate in vitro method to assess biological effluent toxicity,and the best results were attained by lyophilising the sample as *** to its response,the H9c2(2-1) cell line might be a possible alternative in vitro model for fish lethal testing to assess the toxicity of municipal *** physicochemical status of the sample suggests a high potential for eutrophication,and iron exceeded the permissible level for wastewater discharge,possibly due to the addition of ferric chloride for wastewater *** general,the levels of carbamazepine and sulfamethoxazole are higher than those reported for other countries,and both surpassed the aquatic protective values for long-term exposure.

Measuring ammonia(NH_(3))volatilization from urea-fertilized soils is crucial for evaluation of practices that reduce gaseous nitrogen(N)losses in *** small area of chambers used for NH_(3)volatilization measurements compared with the size of field plots may cause significant errors if inadequate sampling strategies are *** aims were:i)to investigate the effect of using multiple open chambers on the variability in the measurement of NH_(3)volatilization in urea-amended field plots and ii)to define the critical period of NH_(3)-N losses during which the concentration of sampling effort is capable of reducing *** use of only one chamber covering 0.015%of the plot(51.84 m^(2))generates a value of NH_(3)-N loss within an expected margin of error of 30%around the true *** reduce the error margin by half(15%),3–7 chambers were required with a mean of 5 chambers per *** the sampling efforts in the first two weeks after urea application,which is usually the most critical period of N losses and associated errors,represents an efficient strategy to lessen uncertainty in the measurements of NH_(3)*** strategy enhances the power of detection of NH_(3)-N loss abatement in field experiments using chambers.

Effects of Alumina on cristobalite crystallization and Properties of Silica-Based ceramic cores

In this work, the influences of alumina addition on cristobalite crystallization and properties of injec- tion molded silica-based ceramic cores were investigated. X-ray diffraction （XRD） was used to characterize phase transformations in the samples, and the XRD result indicated that the addition of alumina pro- moted crystallization of fused silica during sintering at 1180-1220 ℃ and thus increases the amount of cristobalite. The increased amount of cristobalite as well as alumina addition led to much more thermal dilation due to their higher coefficients of thermal expansion than that of fused silica. The flexural strengths at room temperature and 1500 ~c were tested, and it was shown that alumina addition could not affect room temperature strength, but decreased the flexural strength at 1500 ℃. In addition, deflection resis- tance during heating to high temperatures was investigated, and the result indicated that alumina addition speeded up high temperature softening of the samples. XRD and scanning electron microscopy equipped with energy dispersive spectrometry （SEMJEDS） analysis suggested that this softening behavior was related with viscous flow sintering which could be accelerated by the reaction of alumina and silica with a product of mullite.

Mesenchymal stromal cells (MScs) are multipotent and self-renewing stem cellsthat have great potential as cell therapy for autoimmune and inflammatorydisorders, as well as for other clinical conditions, due to their immunoregulatoryand regenerative properties. MScs modulate the inflammatory milieu by releasingsoluble factors and acting through cell-to-cell mechanisms. MScs switch theclassical inflammatory status of monocytes and macrophages towards a nonclassicaland anti-inflammatory phenotype. This is characterized by an increasedsecretion of anti-inflammatory cytokines, a decreased release of pro-inflammatorycytokines, and changes in the expression of cell membrane molecules and inmetabolic pathways. The MSc modulation of monocyte and macrophage phenotypesseems to be critical for therapy effectiveness in several disease models, sincewhen these cells are depleted, no immunoregulatory effects are observed. Here,we review the effects of living MScs (metabolically active cells) and metabolicallyinactive MScs (dead cells that lost metabolic activity by induced inactivation) andtheir derivatives (extracellular vesicles, soluble factors, extracts, and microparticles)on the profile of macrophages and monocytes and the implications forimmunoregulatory and reparative processes. This review includes mechanisms ofaction exhibited in these different therapeutic appro-aches, which induce the antiinflammatoryproperties of monocytes and macrophages. Finally, we overviewseveral possibilities of therapeutic applications of these cells and their derivatives,with results regarding monocytes and macrophages in animal model studies andsome clinical trials.

Pillcam coLoN 2~ in crohn's disease:A new concept of pan-enteric mucosal healing assessment

AIM: To evaluate mucosal healing in patients with small bowel plus colonic crohn's disease(cD) with a single non-invasive examination, by using Pillcam coLoN 2.(Pcc2).METHoDS: Patients with non-stricturing nonpenetrating small bowel plus colonic cD in sustained corticosteroid-free remission were included. At diagnosis,patients had undergone ileocolonoscopy to identify active cD lesions, such as ulcers and erosions, and small bowel capsule endoscopy to assess the Lewis Score(LS). After ≥ 1 year of follow-up, patients underwent entire gastrointestinal tract evaluation with Pcc2. The primary endpoint was assessment of cD mucosal healing, defined as no active colonic cD lesions and LS cluded(7 male;mean age: 32 years), and mean follow-up was 38 *** majority of patients(83.3%) received immunosuppressive therapy. Three patients(25%) achieved mucosal healing in both the small bowel and the colon,while disease activity was limited to either the small bowel or the colon in 5 patients(42%). It was possible to observe the entire gastrointestinal tract in 10 of the12 patients(83%) who underwent ***: only three patients in sustainedcorticosteroid-free clinical remission achieved mucosal healing in both the small bowel and the colon, highlighting the limitations of clinical assessment when stratifying disease activity, and the need for pan-enteric endoscopy to guide therapeutic modification.

Inhibitory Mechanisms of Water Extract of oplopanax elatus on Lipopolysaccharide-lnduced Inflammatory Responses in RAW 264.7 Murine Macrophage cells

objective:To study the anti-inflammatory action and cellular mechanism of oplopanax ***:A hot water extract of oE(WoE)was prepared and a major constituent,syringin,was successfully *** content in WoE was found to be 214.0 p.g/g dried plant(w/w).Their anti-inflammatory activities were examined using RAW 264.7 macrophages and a mouse model of croton oil-induced ear ***:In lipopolysaccharide(LPS)-treated RAW 264.7 cells,a mouse macrophage cell line,WoE was found to significantly and strongly inhibit cyclooxygenase-2(coX-2)-induced prostaglandin E2(PGE2)production[half maximal inhibitory concentration(Ic50)=135.2μg/mL]and inducible nitric oxide synthase(iNoS)-induced No production(Ic50=242.9μg/mL).In the same condition,WoE was revealed to inhibit No production by downregulating iNoS expression,mainly by interrupting mitogen activated protein kinases(MAPKs)/activator protein-1(AP-1)*** activation of all three major MAPKs,p38 MAPK,extracellular signal-regulated kinase(ERK),and c-Jun N-terminal kinase,was inhibited by WoE(50-300μg/mL).on the other hand,WoE reduced PGE2 production by inhibiting coX-2 enzyme activity,but did not affect coX-2 expression *** addition,WoE inhibited the production of proinflammatory cytokines such as interleukin-6 and tumor necrosis factor-α.In croton oil-induced ear edema in mice,oral administration of WoE(50-300 mg/kg)dose-dependently inhibited edematic ***:Water extract of oE exhibited multiple anti-inflammatory action mechanisms and may have potential for treating inflammatory disorders.

Effects of cr addition(0,3,and 6 wt%) on charpy impact properties of Fe-c-Mn-cr-based steels were studied by conducting dynamic compression tests at room and cryogenic *** room temperature,deformation mechanisms of charpy impacted specimens were observed as twinning induced plasticity(TWIP) without any transfo rmation induced plasticity(TRIP) in all the *** cryogenic temperature,many twins were populated in the cr-added steels,but,interestingly,fine ε-martensite was found in the ocr steel,satisfying the Shoji-Nishiyama(S-N) orientation relationship,{111}γ//{0002}ε and γ//ε.Even though the cryogenic-temperature staking fault energies(SFEs) of the three steel were situated in the TWIP regime,the martensitic transformation was induced by Mn-and cr-segregated *** the ocr steel,SFEs of low-(Mn,cr) bands lay between the TWIP and TRIP regimes which were sensitively affected by a small change of *** dynamic compressive test results well showed the relation between segregation bands and the *** of cr were known as not only increasing the SFE but also promoting the carbide *** order to identify the possibility of carbide formation,a precipitation kinetics simulation was conducted,and the predicted fractions of precipitated M23c6 were negligible,0.4-1.1×10-5,even at the low cooling rate of 10℃/s.
