ASFV transcription reporter screening system identifies ailanthone as a broad antiviral compound

African swine fever(ASF)is an acute,highly contagious and deadly viral disease in swine that jeopardizes the worldwide pig industry.Unfortunately,there are no authoritative vaccine and antiviral drug available for ASF control.African swine fever virus(ASFV)is the etiological agent of ASF.Among the ASFV proteins,p72 is the most abundant component in the virions and thus a potential target for anti-ASFV drug design.Here,we con-structed a luciferase reporter system driven by the promoter of p72,which is transcribed by the co-transfected ASFV RNA polymerase complex.Using this system,we screened over 3200 natural product compounds and obtained three potent candidates against ASFV.We further evaluated the anti-ASFV effects and proved that among the three candidates,ailanthone(AIL)inhibits the replication of ASFV at the nanomolar concentration(IC_(50)=15 nmol/L).Our in vitro experiments indicated that the antiviral effect of AIL is associated with its inhibition of the HSP90-p23 cochaperone.Finally,we showed the antiviral activity of AIL on Zika virus and hepatitis B virus(HBV),which supports that AIL is a potential broad-spectrum antiviral agent.

An Efficient Proxy Ring Signature Without Bilinear Pairing

Proxy ring signature is a proxy signature designed to protect the privacy of the proxy signer. In this paper, we propose a new proxy ring signature based on the Schnorr’s signature. Our scheme combines the idea of the Schnorr’s signature with the method of existing structure of proxy ring signature. In order to improve the performance, we eliminate the use of bilinear pairing operation. We only use the hash function in the signature verification phase. Because of above features, the proxy ring signature scheme we proposed is more efficient in computation. We also prove that our scheme is unforgeable against all kinds of adversaries in the random oracle model.

Shanghai center project excavation induced ground surface movements and deformations

Empirical data on deep urban excavations can provide designers a significant reference basis for assessing potential deformations of the deep excavations and their impact on adjacent structures. The construction of the Shanghai Center involved excavations in excess of 33-m-deep using the top-down method at a site underlain by thick deposits of marine soft clay. A retaining system was achieved by 50-m-deep diaphragm walls with six levels of struts. During construction, a comprehensive instrumentation program lasting 14 months was conducted to monitor the behaviors of this deep circular excavation. The following main items related to ground surface movements and deformations were collected：（1） walls and circumferential soils lateral movements; （2） peripheral soil deflection in layers and ground settlements; and （3） pit basal heave. The results from the field instrumentation showed that deflections of the site were strictly controlled and had no large movements that might lead to damage to the stability of the foundation pit. The field performance of another 21 cylindrical excavations in top-down method were collected to compare with this case through statistical analysis. In addition, numerical analyses were conducted to compare with the observed data. The extensively monitored data are characterized and analyzed in this paper.

Research on pavement cracking possibility based on the load mechanical response

Cavities under roads are one of the main reasons for early structural damage to pavements. It is necessary to conduct a structural analysis of road sections with cavities and evaluate the possibility of pavement cracking caused by different cavity sizes. In this study, an analysis method for evaluating the possibility of pavement cracking based on the load-mechanical response is proposed.An example library of the mechanical response of asphalt concrete(AC) pavements was established by numerical simulation.Based on the tensile cracking characteristics of pavements in the mechanical response research, the tensile strain at the bottom of the AC layer was selected as the key analysis parameter. Sensitivity analysis of the tensile strain was conducted, and the main factors controlling pavement cracking were determined. A tensile strain response prediction model was established using multiple linear regression, and its reliability was verified. The cavity influence coefficient(CIC) and pavement cracking factor(PCF) were constructed to analyze the cracking possibility. The variation in PCF with the cavity size and pavement structure parameters was studied. A quantitative relationship between the depth and length of the cavity for a given PCF was obtained.This law conforms to a power function. The possibility of pavement cracking can be determined by measuring the cavity size.Compared to the existing cavity management system, the proposed method provided analysis results of the cracking possibility that were more consistent when the cavity depth was small and the length was long. The findings of this study provide new insights for evaluating the possibility of pavement cracking.

Multi-dimensional visualization for the morphology of lubricant stearic acid particles and their distribution in tablets

The shapes of particles and their distribution in tablets, controlled by pretreatment and tableting process, determine the pharmaceutical performance of excipient like lubricant. This study aims to provide deeper insights to the relationship of the morphology and spatial distribution of stearic acid(SA) with the lubrication efficiency, as well as the resulting tablet property. Unmodified SA particles as flat sheet-like particles were firstly reprocessed by emulsification in hot water to obtain the reprocessed SA particles with spherical morphology. The three-dimensional(3 D) information of SA particles in tablets was detected by a quantitative and non-invasive 3 D structure elucidation technique, namely, synchrotron radiation X-ray micro-computed tomography(SR-μCT). SA particles in glipizide tablets prepared by using unmodified SA(GUT), reprocessed SA(GRT), as well as reference listed drug(RLD) of glipizide tablets were analyzed by SR-μCT. The results showed that the reprocessed SA with better flowability contributed to similarity of breaking forces between that of GRT and RLD. SA particles in GRT were very similar to those in RLD with uniform morphology and particle size, while SA particles in GUT were not evenly distributed. These findings not only demonstrated the feasibility of SR-μCT as a new method in revealing the morphology and spatial distribution of excipient in drug delivery system, but also deepened insights of solid dosage form design into a new scale by powder engineering.

Single-wavelength-excited fluorogenic nanoprobe for accurate realtime ratiometric analysis of broad pH fluctuations in mitophagy

Mitophagy has a critical role in maintaining cellular homeostasis through acidic lysosomes engulfing excess or impaired mitochondria,thereby pH fluctuation is one of the most significant indicators for tracking mitophagy.Then such precise pH tracking demands the fluorogenic probe that has tailored contemporaneous features,including mitochondrial-specificity,excellent biocompatibility,wide pH-sensitive range of 8.0–4.0,and especially quantitative ability.However,available molecular probes cannot simultaneously meet all the requirements since it is extremely difficult to integrate multiple functionalities into a single molecule.To fully address this issue,we herein integrate two fluorogenic pH sensitive units,a mitochondria-specific block,cellpenetrating facilitator,and biocompatible segments into an elegant silica nano scaffold,which greatly ensures the applicability for real-time tracking of pH fluctuations in mitophagy.Most significantly,at a single wavelength excitation,the integrated pHsensitive units have spectra-distinguishable fluorescence towards alkaline and acidic pH in a broad range that covers mitochondrial and lysosomal pH,thus enabling a ratiometric analysis of pH variations during the whole mitophagy.This work also provides constructive insights into the fabrication of advanced fluorescent nanoprobes for diverse biomedical applications.

Dynamic Response Analysis of Semi-Submersible Floating Wind Turbine with Different Wave Conditions

To address the problem of poor wave resistance of existing offshore floating wind turbines,a new type of semisubmersible platform with truncated-cone-type upper pontoons is proposed by combining the characteristics of offshore wind turbine semi-submersible floating platforms.Based on the coupled hydrodynamic,aerodynamic,and mooring force physical fields of FAST,the surge,heave,pitch,and yaw motions responses of the floating wind turbine under different wave heights and periods are obtained,and the mooring line tension responses are also obtained;and compare the dynamic response of the new semi-submersible platform with the OC4-DeepCwind platformat six degrees of freedom.The results show that different wave conditions have obvious effects on the heave and pitch motions of the new floating wind turbine,and fewer effects on the surge and yaw motions;the tensegrity response of the mooring system is more affected by the wave conditions;compared with the OC4-DeepCwind floating wind turbine,the pitch and roll response of the new floating wind turbine has been significantly reduced and has good stability.

Constructal design of a rectangular parallel phase change microchannel in a three-dimensional electronic device

Based on constructal theory, a rectangular parallel phase change microchannel model in a three-dimensional electronic device(TDED) is established with R134a as the cooling fluid. Based on the minimization of a complex function(CF) composed of linear weighting sum of maximum temperature difference and pumping power consumption, constructal design of the TDED is conducted first; and then, maximum temperature difference and pumping power consumption are minimized by non-dominated sorting genetic algorithm-II methods. The results reveal that there exist an optimal mass flow rate(0.0012 kg/s) and a quadratic optimal aspect ratio(AR)(0.39) of the microchannel which lead to quadratic minimum CF(0.817). Compared with the original value, the CF after optimization is reduced by 18.34%. Reducing the inlet temperature of cooling fluid and microchannel number appropriately can help to enhance the overall performance of TDED. By using the artificial neural network and genetic algorithms in the toolboxes of Matlab software, the optimal AR gained in the Pareto solution set is located between 0.2–0.45. The smallest deviation index among three discussed strategies is 0.346, and the corresponding optimal AR is 0.413, which is selected as the optimal design strategy of the microchannel in the TDED under multiple requirements. The findings in this study can serve as theoretical guides for thermal designs of electronic devices.

Simultaneous improvement to solubility and bioavailability of active natural compound isosteviol using cyclodextrin metal-organic frameworks

Cyclodextrin metal-organic framework(CD-MOF)as a highly porous supramolecular carrier could be one of the solutions to the insolubility of isosteviol(STV).The solubility of STV was lower than20.00 ng/mL at pH 1.0 and pH 4.5,whilst its solubility increased to 20,074.30 ng/mL at pH 6.8 and129.58 ng/mL in water with a significant pH-dependence.The in vitro release profiles of STV from STV@CD-MOF(0.5:1)were pH-independent in distinct pH media and closed to be thoroughly released but no such release profiles were observed for STV@CD-MOF(1:1)owing to nanoclusters formation.The bioavailability of STV@CD-MOF(1:1)in rats was 8.67-fold higher than that of STV,and was1.32-and 1.27-fold higher than that of STV@CD and STV@CD-MOF(0.5:1).Our results indicated that the inclusion mechanism played a primary role when STV in CD-MOF was at a low loading ratio,while the increasement in bioavailability at a high loading ratio,which was attributed to the nanocluster mechanism.This was confirmed by molecular simulation.In conclusion,CD-MOF is a promising system for STV loading,overcoming the insolubility and to improve the bioavailability of this natural compound.
