Ethanol extract of Cyathulae Radix inhibits osteoclast differentiation and bone loss

Cyathulae Radix,a traditional Chinese medicine and a common vegetable,boasts a history spanning millennia.It enhances bone density,boosts metabolism,and effectively alleviates osteoporosis-induced pain.Despite its historical use,the molecular mechanisms behind Cyathulae Radix’s impact on osteoporosis remain unexplored.In this study,we investigated the effects and mechanisms of Cyathulae Radix ethanol extract(CEE)in inhibiting osteoporosis and osteoclastogenesis.Eight-week-old female mice underwent ovariectomy and were treated with CEE for eight weeks.Micro-computed tomography(micro-CT)assessed histomorphometric parameters,bone tissue staining observed distal femur histomorphology,and three-point bending tests evaluated tibia mechanical properties.Enzyme-linked immunosorbent assay(ELISA)measured serum estradiol(E2),receptor activator for nuclear factor B ligand(RANKL),and osteoprotegerin(OPG)levels.Osteoclastogenesis-related markers were analyzed via Western blotting(WB)and quantitative real-time polymerase chain reaction(qRT-PCR).Additionally,CEE effects on RANKL-induced osteoclast formation and bone resorption were investigated in vitro using tartrate-resistant acid phosphatase(TRAP)staining,qRT-PCR,and WB assay.Compared with the ovariectomy(OVX)group,CEE treatment enhanced trabecular bone density,maximal load-bearing capacity,and various histomorphometric parameters.Serum E2 and OPG levels significantly increased,while Receptor activator of nuclear factor-κB(RANK)decreased in the CEE group.CEE downregulated matrix metallopeptidase 9(MMP-9),Cathepsin K(CTSK),and TRAP gene and protein expression.In bone marrow macrophages(BMMs),CEE reduced mature osteoclasts,bone resorption pit areas,and MMP-9,CTSK,and TRAP expression during osteoclast differentiation.Compared with DMSO treatment,CEE markedly inhibited RANK,TNF receptor associated factor 6(TRAF6),Proto-oncogene c-Fos(c-Fos),Nuclear factor of activated T-cells cytoplasmic 1(NFATc1)expressions,and Extracellular regulated protein kinases(ERK),c-Jun N-terminal kinase

Nanostructure photodetectors,as the core component of optoelectronic devices,are mainly focused on the precise preparation of mixed-component nano-heterostructures and the realization of zero power consumption devices.Herein,we successfully fabricated n-GaN/p-ZnTe core/shell nanopillar array and realized self-power ultraviolet/violet photodetection.The radial heterojunction nanodevice reveals high light-dark current ratio of 104 at 0 V bias,indicating effective carriers’separation.And more,by integrating plasmonic effect,the responsivity and detectivity of the Au nanoparticles decorated device are increased from 3.85 to 148.83 mA/W and 4.45×1011 to 2.33×1012 Jones under 325 nm UV light irradiation.While the rise and the fall time are decreased 1.3 times and 6.8 times under 520 nm visible light irradiation at 0 V bias.The high photocurrent gain is derived from that the oscillating high-energy hot electrons in Au nanoparticles spontaneously inject into the ZnTe conduction band to involve the photodetection process.This work presents an effective route to prepare high-performance self-power photodetector and provides a promising blueprint to realize different functional photoelectronic devices based on core/shell nanostructure.

Graphene-based nanomaterials(GBNMs)has been thoroughly investigated and extensively used in many biomedical fields,especially cancer therapy and bacteria-induced infectious diseases treatment,which have attracted more and more attentions due to the improved therapeutic efficacy and reduced reverse effect.GBNMs,as classic two-dimensional(2D)nanomaterials,have unique structure and excellent physicochemical properties,exhibiting tremendous potential in cancer therapy and bacteria-induced infectious diseases treatment.In this review,we first introduced the recent advances in development of GBNMs and GBNMs-based treatment strategies for cancer,including photothermal therapy(PTT),photodynamic therapy(PDT)and multiple combination therapies.Then,we surveyed the research progress of applications of GBNMs in anti-infection such as antimicrobial resistance,wound healing and removal of biofilm.The mechanism of GBNMs was also expounded.Finally,we concluded and discussed the advantages,challenges/limitations and perspective about the development of GBNMs and GBNMs-based therapies.Collectively,we think that GBNMs could be potential in clinic to promote the improvement of cancer therapy and infections treatment.

Nowadays,education and teaching have become a hot topic,and teaching in colleges and universities is facing a brand-new development direction.Principles of Concrete Structure Design,as one of the main courses,transmits professional knowledge for students,enhances the students’professional ability,and further carries out in-depth research on the course to bring a better teaching effect for students.The article mainly focuses on the research of the principles of concrete structure design course,conducts an analysis of the teaching characteristics of the principles of concrete structure design course,and reasonably sets the teaching content from the optimization of the course teaching objectives;innovative course teaching methods can deepen the effect of knowledge understanding;reform of experimental practice teaching can lay down the effect of the internalization of knowledge,etc.The in-depth description and discussion of the relevant aspects of the research aim to provide guidelines for related research.

Antimicrobial peptides act on the rumen the performance of castrated bulls

Background Many countries have already banned the use of antibiotics in animal husbandry,making it extremely difficult to maintain animal health in livestock breeding.In the livestock industry,there is an urgent need to develop alternatives to antibiotics which will not lead to drug resistance on prolonged use.In this study,eighteen castrated bulls were randomly divided into two groups.The control group(CK)was fed the basal diet,while the antimicrobial peptide group(AP)was fed the basal diet supplemented with 8 g of antimicrobial peptides in the basal diet for the experimental period of 270 d.They were then slaughtered to measure production performance,and the ruminal contents were isolated for metagenomic and metabolome sequencing analysis.Result The results showed that antimicrobial peptides could improve the daily weight,carcass weight,and net meat weight of the experimental animals.Additionally,the rumen papillae diameter and the micropapillary density in the AP were significantly greater than those in the CK.Furthermore,the determination of digestive enzymes and fermentation parameters showed that the contents of protease,xylanase,andβ-glucoside in the AP were greater than those in the CK.However,lipase content in the CK was greater than that in the AP.Moreover,the content of acetate,propionate,butyrate,and valerate was found to be greater in AP than those in CK.The metagenomic analysis annotated 1993 differential microorganisms at the species level.The KEGG enrichment of these microorganisms revealed that the enrichment of drug resistance-related pathways was dramatically decreased in the AP,whereas the enrichment of immune-related pathways was significantly increased.There was also a significant reduction in the types of viruses in the AP.187 probiotics with significant differences were found,135 of which were higher in AP than in CK.It was also found that the antimicrobial mechanism of the antimicrobial peptides was quite specific.Seven low-abundance microorganisms(Acinetobactersp.Ac1271,Aequorivita soeso

隧道围岩变形和松动圈厚度是隧道结构设计的重要考虑因素和安全运营的重要参考依据,现有的松动圈识别方法和隧道围岩变形测量手段多停留在检测层面,缺乏长期实时监测方法.为此,研发内嵌光纤的自感知纤维增强复合材料(FRP)锚杆,提出基于内嵌光纤自感知FRP锚杆的隧道围岩智能监测系统,可以实现对围岩变形立体化、全天候的实时监测;结合监测数据与理论分析,提出隧道围岩松动圈的识别方法,并将该智能监测方法应用于广汕高铁陈塘隧道.研究结果表明:自感知FRP锚杆能够精准探知现场施工对围岩变形的影响规律,型钢钢架对隧道围岩的支护作用较为突出;监测数据可以实时反映自感知锚杆的受力规律,从而准确识别隧道不同位置围岩的松动圈厚度;基于自感知FRP锚杆的隧道围岩智能监测系统将在隧道运营过程中持续、实时监测隧道围岩的变形,为隧道全生命周期的结构安全提供高技术保障.

目的:探究长链非编码RNA(lncRNA)NORAD调控肺腺癌(LUAD)发生发展及其可能的作用机制。方法:RT-PCR检测19例肺腺癌与邻近正常组织NORAD的表达,分析NORAD表达量与临床病理分期关系;RT-PCR检测NORAD在A549和NCI-H1299细胞表达情况,si-NORAD干扰沉默A549和NCI-H1299细胞,检测si-NORAD对细胞增殖、迁移和侵袭能力的影响;Western Blot检测MMP9、MMP2、Vimentin、N-Cadherin及E-Cadherin表达情况;基于TCGA数据库提取LUAD转录组数据行加权基因共表达网络分析(WGCNA),进一步对促进LUAD可能的信号通路进行基因集富集分析(GSEA)NORAD,再行Western Blot验证。结果:NORAD在肺腺癌组织中高表达,且更易发生淋巴结转移;NORAD在A549和NCI-H1299细胞明显过表达,敲低NORAD能明显降低A549和NCI-H1299细胞增殖、迁移和侵袭能力;敲低NORAD,MMP9、MMP2、Vimentin、N-Cadherin表达上调,E-Cadherin表达下调;WGCNA、GSEA及Western Blot实验证明NORAD通过PI3K/AKT信号通路促进LUAD的发生发展。结论:NORAD的过表达可能导致LUAD患者的不良预后。NORAD可通过PI3K-AKT-mTOR信号通路影响LUAD细胞株的基因组稳定性,从而促进LUAD的发生发展和转移。

A Study on the Influence of Social Media Use on Psychological Anxiety among Young Women

To explore the relationship between social influence,social comparison,clarity of self-concept,and psychological anxiety among young women during their usage of social networking sites,our study selected 338 young women aged 14-34 from the social site platforms of Little Red Book and Weibo for questionnaire surveys.The Passive Social Network Utilization Scale,Social Comparison Scale(SCS),Social Influence Questionnaire,Self-Concept Clarity Scale(SCCS),and Generalized Anxiety Disorder Scale(GAD-7)were employed to measure the subjects.Our results show that the frequency of passive social media use is positively related to the level of psychological anxiety.Social comparison,social influence,and unclear self-concepts under social media use are negatively predictive of psychological anxiety.The chain mediation effects indicate that social comparison and social influence under social media use negatively predict the clarity of self-concept,thus having a negative impact on the psychological health of young women.Therefore,young women should strengthen their self-concepts,control their frequency of social media usage,avoid addiction,and pay special attention to their frequency of passive use,in order to protect their psychological health.Our study provides some practical implications and insights regarding the relationship between young women’s social media use and psychological health.
