Exploring the abundance and influencing factors of antimicrobial resistance genes in manure plasmidome from swine farms

Plasmids play a critical role in the dissemination of antimicrobial resistance genes(ARGs),however,a systematical understanding of ARGs originated from plasmids in swine production is currently lacking.Herein,quantitative polymerase chain reaction was applied to determine the prevalence of ten ARGs and the class1 integron gene intI1 of plasmid source in swinemanure from 44 farms in Sichuan,Hubei and Hebei provinces,China.All assayed ARGs were observed in plasmid DNA samples,and the average absolute abundance of aac(6’)-Ib-cr,blaNDM,blaCTX-M,optrA,ermB,floR,mcr-1,qnrS,tetM,sul1 and intI1 were 7.09,2.90,4.67,6.62,7.55,7.14,4.08,4.85,7.16,7.11 and 8.07 of 10 log copies/gram,respectively.IntI1 showed a high correlation(r>0.8,P<0.01)with the abundance of aac(6’)-Ib-cr and sul1 in swine manure.Moreover,the farm scale(i.e.,herd population)and geographical location were not found to be critical factors influencing the absolute abundance of ARGs of plasmid DNA in swine farms.However,the concentrations of florfenicol,Cu,Zn,Fe,total phosphorus(TP)and total potassium(TK)demonstrated a significant correlation with the abundance of several ARGs.Particularly,Cu and Zn had high correlations with optrA and blaCTX-M,respectively.Our results demonstrated that antibiotics,heavy metals and environmental nutrients are likely jointly contributing to the long-term persistence of ARGs in swine production.This study provides insights into the abundance and influencing factors of ARGs from swine manure,which is of significance for assessing and reducing the public health risks in livestock production.

Antimicrobial resistance characteristics and phylogenetic relationships of pleuromutilin-resistant Enterococcus isolates from different environmental samples along a laying hen production chain

Antimicrobial resistance in the laying hen production industry has become a serious public health problem.The antimicrobial resistance and phylogenetic relationships of the common conditional pathogen Enterococcus along the laying hen production chain have not been systematically clarified.105 Enterococcus isolates were obtained from 115 environmental samples (air,dust,feces,fies,sewage,and soil) collected along the laying hen production chain (breeding chicken,chick,young chicken,and commercial laying hen).These Enterococcus isolates exhibited resistance to some clinically relevant antibiotics,such as tetracycline (92.4%),streptomycin (92.4%),and erythromycin (91.4%),and all strains had multidrug resistance phenotypes.Whole genome sequencing characterized 29 acquired antibiotic resistance genes (ARGs) that conferred resistance to 11 classes of antibiotics in 51pleuromutilin-resistant Enterococcus isolates,and lsa(E),which mediates resistance to pleuromutilins,always co-occurred with lnu(B).Alignments with the Mobile Genetic Elements database identified four transposons (Tn554,Tn558,Tn6261,and Tn6674) with several ARGs(erm(A),ant(9)-la,fex(A),and optrA) that mediated resistance to many clinically important antibiotics.Moreover,we identified two new transposons that carried ARGs in the Tn554 family designated as Tn7508 and Tn7492.A complementary approach based on conventional multi-locus sequence typing and whole genome single nucleotide polymorphism analysis showed that phylogenetically related pleuromutilin-resistant Enterococcus isolates were widely distributed in various environments on different production farms.Our results indicate that environmental contamination by antimicrobial-resistant Enterococcus requires greater attention,and they highlight the risk of pleuromutilin-resistant Enterococcus and ARGs disseminating along the laying hen production chain,thereby warranting effective disinfection.

Dear editor,Implementation attacks such as fault attack(FA)[1] and side-channel analysis (SCA)[2] are significant threats to the cryptographic device security. Infective countermeasure is a promising strategy to resist FAs. Its principle is to scramble the data path of the encryption and make the faulty ciphertext useless in the key retrieval. The scrambling here is called infection. It can be executed either once at the end of the algorithm or multiple times between different encryption operations.

Emotional Mechanisms in Supervisor-Student Relationship:Evidence from Machine Learning and Investigation

How to cultivate innovative talents has become an important educational issue nowadays.In China’s long-term mentorship education environment,supervisor-student relationship often affects students’creativity.From the perspective of students’psychology,we explore the influence mechanism of supervisor-student relationship on creativity by machine learning and questionnaire survey.In Study 1,based on video interviews with 16 postgraduate students,we use the machine learning method to analyze the emotional states exhibited by the postgraduate students in the videos when associating them with the supervisor-student interaction scenario,finding that students have negative emotions in bad supervisor-student relationship.Subsequently,we further explore the impact of supervisor-student relationship on postgraduate students’development in supervisor-student interaction scenarios at the affective level.In Study 2,a questionnaire survey is conducted to explore the relationship between relevant variables,finding that a good supervisor-student relationship can significantly reduce power stereotype threat,decrease emotional labor surface behaviors,and promote creativity expression.The above results theoretically reveal the internal psychological processes by which supervisor-student relationship affects creativity,and have important implications for reducing emotional labor and enhancing creativity expression of postgraduate students.

Synergistic effects of lipopolysaccharide and rotenone on dopamine neuronal damage in rats

OBJECTIVE The etiology of Parkinson disease(PD) is still unknown. Until now,oxidative stress and neuroinflammation play a crucial role in the pathogenesis of PD. However, the specific synergistic role of oxidative stress and neuroinflammation in the occurrence and development of PD remains unclear. METHODS The changes in motor behavior, dopamine(DA) neurons quantification and their mitochondrial respiratory chain,glial cells activation and secreted cytokines, Nrf2 signaling pathway, and redox balance in the brain of rats were evaluated. RESULTS Lipopolysaccharide(LPS)-induced neuroinflammation and rotenone(ROT)-induced oxidative stress synergistically aggravated motor dysfunction, DA neuron damage, activation of glial cells, and release of related mediators, activation of Nrf2 signaling and destruction of oxidative balance. In addition, further studies indicated that after ROT-induced oxidative stress caused direct damage to DA neurons, LPSinduced inflammatory effects had stronger promoting neurotoxic effects on the above aspects. CONCLUSION Neuroinflammation and oxidative stress synergistically aggravated DA neuronal loss. Furtherly, oxidative stress followed by neuroinflammation caused more DA neuronal loss than neuroinflammation followed by oxidative stress.

Research on Coupling Transfer Characteristics of Vibration Energy of Free Piston Linear Generator

In order to clarify the mechanism and main influencing factors of the vibration energy coupling transmission with a dual-piston structure,a thermodynamic and dynamic coupling model of the free piston linear generator(FPLG)was established.The system energy conversion,vibration energy coupling transmission,and influencing factors were studied in detail.The coupling transmission paths and the secondary influence mechanism from in-cylinder combustion on vibration energy transmission were obtained.In addition,the influence of the movement characteristics of the dual-piston on the vibration energy transmission was studied,and the typical parameter variation law was obtained,which provides theoretical guidance for the subsequent vibration reduction design of the FPLG.

Underground ground control monitoring and interpretation,and numerical modeling, and shield capacity design

Mine or longwall panel layout is a 3D structure with highly non-uniform stress distribution. Recognition of such fact will facilitate underground problem identification/investigation and solving by numerical modeling through proper model construction. Due to its versatility, numerical modeling is the most popular method for ground control design and problem solving. However numerical modeling results require highly experienced professionals to interpret its validity/applicability to actual mining operations due to complicated mining and geological conditions. Underground ground control monitoring is routinely performed to predict roof behavior such as weighting and weighting interval without matching observation of face mining condition while the mining pressures are being monitored, resulting in unrealistic interpretation of the obtained data on mining pressure. The importance of ground control pressure monitoring and simultaneous observation of mining and geological conditions is illustrated by an example of shield leg pressure monitoring and interpretation in an U.S. longwall coal mine: it was found that the roof strata act like a plate, not an individual block of the size of a shield dimension, as commonly assumed by all researchers and shield capacity is not a fixed property for a longwall panel or a mine or a coal seam. A new mechanism on the interaction between shield's hydraulic leg pressure and roof strata for shield loading is proposed.

Automation in U.S. longwall coal mining: A state-of-the-art review

This paper reviews the development of U.S. longwall mining from an unknown to became the world standard in the past five decades with emphasis on automation. Large scale longwall face equipment were imported from Germany and United Kingdom to increase production in the 1970 s and great effort was made to improve them to suit U.S. conditions, rather than domestic market. Automation began with the development of electrohydraulic shields in 1984 and continue to present. Introduction of first generation semi-automated longwall system occurred in 1995 and step-to-step improvement continues to present following the development of sensor technology and internet of things(IOT). Since then, emphasis on new development has been concentrated on the improvement of equipment reliability, miner's health and safety as well as production, including dust control techniques, proximity sensor, anti-collision and remote control. Automation is classified into two categories: automation of individual face equipment and automation of longwall system. The automation development of longwall system is divided into three stages: shearer-initiated-shield-advance(SISA), semi-automated longwall system, and remote control shearer.

Graphene-Gold Nanoparticle-modified Electrochemical Sensor for Detection of Kanamycin Based on Target-induced Aptamer Displacement

A highly sensitive and selective label-free electrochemical sensor was developed for the determination of kanamycin. To improve the sensitivity of the electrochemical sensor, graphene-gold nanoparticles were prepared by a one-step electrochemical coreduction process and were modified on the surface of a glassy carbon electrode. The double-stranded DNA（ds-DNA） duplex probe was immobilized onto the graphene-gold nanoparticle-modified elec-trode. The introduction of target kanamycin induced the displacement of aptamer from the ds-DNA duplex into the solution. Methylene blue（MB） as a redox indicator monitored the current change using differential pulse voltammetry Under optimal conditions, the designed electrochemical aptasensor exhibited a wide linear range from 0. i pmol/L to 10 pmol/L with a detection limit of 0.03 pmol/L for kanamycin. The experimental strategy enabled the direct analysis of milk samples, and the results showed high sensitivity and good selectivity.

A Combined Countermeasure Against Side-Channel and Fault Attack with Threshold Implementation Technique

Side-channel attack(SCA) and fault attack(FA) are two classical physical attacks against cryptographic implementation. In order to resist them, we present a combined countermeasure scheme which can resist both SCA and FA. The scheme combines threshold implementation and duplication-based exchange techniques. The exchange technique can confuse the fault propagation path and randomize the faulty values. The threshold implementation technique can ensure a provable security against SCA. Moreover, it can also help to resist the FA by its incomplete property and random numbers. Compared with other methods, the proposed scheme has simple structure, which can be easily implemented in hardware and result in a low implementation cost. Finally, we present a detailed design for the block cipher light encryption device(LED) and implement it.The hardware cost evaluation shows our scheme has the minimum overhead factor.
