Computational identification and characterization of microRNAs and their targets in Penaeus monodon

Study on shrimp miRNAs was limited and just 7 mature miRNA sequences of Marsupenaeus japonicus are deposited in mir Base database. In this study, miRNAs and their target gene candidates were computationally identified from shrimp Penaeu s monodon and then experimentally validated. Using 39 908 expressed sequence tags(ESTs) and 21 124 genome survey sequences(GSSs) of P. monodon(pmo) as reference dataset, a comprehensive approach based on inter-species homolog search was employed to investigate the candidate miRNAs(i.e. pmo-miRNA). A total of eight miRNAs belonging to 7 families were computationally identified and five out of them were subsequently validated by PCR and sequencing. Of these, pmo-miR-4961a, pmo-miR-4961b, pmo-miR-4979 and pmo-miR-3819 were first identified from shrimps. Both the mature pmo-miRNAs and the corresponding precursors were conserved among different species. Based on perfect or near-perfect match to the target region, the target gene candidates of pmomiRNAs were predicted from 10 331 mRNA sequences of P. monodon. A total of 20 genes were predicted as the targets of pmo-miR-4961a, pmo-miR-4961b, pmo-miR-4979 and pmo-miR-6492. Experimental validation by dual luciferase reporter assay confi rmed the targeting between 3 pmo-miRNAs and one or two of their target genes, especially the pmo-miR-4979 which could significantly down-regulate the expression of target gene(JR226772). This study updates the miRNAs and their targets in P. monodon and lays a solid foundation for future RNAi study.

Simulation and experiment investigations on fabrication of Fe-based amorphous powders by a novel atomization process equipped with assisted gas nozzles

Based on computational fluid dynamics method,the effect of atomization gas pressure on the atomization efficiency of Laval nozzle was studied,and then a discrete phase model was established and combined with industrial trials to study the effect of a new type of assisted gas nozzles(AGNs)on powder size distribution and amorphous powder *** results show that increasing the atomization pressure can effectively improve the gas velocity for the Laval nozzle;however,it will decrease the aspiration pressure,and the optimal atomization pressure is 2.0 *** with this,after the application of AGNs with the inlet velocity of 200 m s^(-1),assisted gas jet can increase the velocity of overall droplets in the break-up and solidification area by 40 m s^(-1) and the maximum cooling rate is increased from 1.9×10^(4) to 2.3×10^(4) K s^(-1).The predicted particle behavior is demonstrated by the industrial trails,that is,after the application of AGNs,the median diameter of powders d50 is decreased from 28.42 to 25.56 lm,the sphericity is increased from 0.874 to 0.927,the fraction of amorphous powders is increased from 90.4% to 99.4%,and only the coercivity is increased slightly due to the accumulation of internal *** is illustrated that the AGNs can improve the yield of fine amorphous powders,which is beneficial to providing high-performance raw powders for additive manufacturing technology.

An increasing body of evidence shows that the lipid droplet,a neutral lipid storage organelle,plays a role in lipid metabolism and energy homeostasis through its interaction with ***,the cellular functions and molecular mechanisms of the interaction remain *** we present data from transmission electron microscopy,fluorescence imaging,and reconstitution assays,demonstrating that lipid droplets physically contact mitochondria in vivo and in *** a bimolecular fluorescence complementation assay in Saccharomyces cerevisiae,we generated an interactomic map of protein-protein contacts of lipid droplets with mitochondria and *** lipid droplet proteins Erg6 and Pet10 were found to be involved in 75%of the interactions ***,interactions between 3 pairs of lipid metabolic enzymes were ***,these data demonstrate that lipid droplets make physical contacts with mitochondria and peroxisomes,and reveal specific molecular interactions that suggest active participation of lipid droplets in lipid metabolism in yeast.

Radiological insights of ectopic thyroid in the porta hepatis: A case report and review of the literature

BACKGROUND Ectopic thyroid is defined as a rare developmental anomaly where thyroid tissues are atypically found in locations other than its normal anatomical position:Anterolateral to the second,third,and fourth tracheal *** intemperate descent or a migration failure of the thyroid anlage results in sub-diaphragmatic thyroid ectopia,a sparse clinical *** SUMMARY This case portrays a 63-year-old female patient presenting with chronic abdominal discomfort at a local hospital whereby a computed tomography(CT)scan revealed a well-defined mass in the hepatic *** further examination,the patient underwent a CT scan with contrast,magnetic resonance imaging(MRI),and CT-angiography(CTA)at our *** CT scan showed a welldefined and high attenuated mass measuring 43 mm×38 mm in the hepatic entrance with *** CTA revealed an additional finding:Blood supply to the mass from the right hepatic *** of the upper abdomen demonstrated a mass with mixed signal intensity on T1 and T2 weighted images in the hepatic *** patient underwent surgery with resection of the mass which was sent for *** thyroid at the level of porta hepatis with nodules was the definitive diagnosis since histopathological report revealed presence of thyroid tissue in the resected liver *** This case delivers a rare insight of pre-operative radiological imaging of an ectopic thyroid located in the *** findings can aid in narrowing down potential differential diagnosis when managing a patient with those subsequent findings.

Time-dependent changes in eye-specific segregation in the dorsal lateral geniculate nucleus and superior colliculus of postnatal mice

Eye-specific segregation in the dorsal lateral geniculate nucleus(dLGN) and superior colliculus(SC) starts from the embryonic stage and continues to develop postnatally until eye-opening in mice. However, there have been few systematic studies on the details of this developmental process. Here, we carried out time-dependent studies of eye-specific segregation in the dLGN and SC. Our results demonstrated that the development of eye-specific segregation in the SC is completed before postnatal day 12(P12), which is earlier than in the dLGN(P20). During the whole period, ipsilateral and overlapping axonal projections decreased continuously in both the dLGN and SC. On the other hand, contralateral axonal projections showed little change, except for a slight decrease between P8 and P20 in the dLGN.

A preliminary experimental investigation on the biotribocorrosion of a metal-on-polyethylene hip prosthesis in a hip simulator

Corrosion at the taper/trunnion interface of total hip replacement(THR)often results in severe ***,the underlying mechanisms of biotribocorrosion at the taper/trunnion interface during the long-term walking gait cycles remain to be fully *** this study,a hip joint simulator was therefore instrumented with an electrochemical cell for in-situ monitoring of the tribocorrosion evolution in a metal-on-polyethylene(MoP)THR during a typical long-term walking *** addition,the biotribocorrosion mechanism was investigated via surface and chemical *** experimental results confirmed that the taper/trunnion interface dominated the contemporary MoP hip joint *** cyclic variations in the open circuit potential(OCP)were observed throughout the long-term electrochemical measurements,attributed to the formation and disruption of the adsorbed protein *** corrosion exhibited an initial increase at each period,peaking at approximately 0.125 million cycles,followed by a subsequent gradual *** and chemical analyses revealed the formation of a tribochemical reaction layer(tribolayer)on the worn surface of the taper/trunnion *** surface/chemical characterizations and the electrochemical measurements indicated that the adhesion force of the adsorbed protein layer was weaker than that of the *** contrast,the opposite was true for the corrosion *** on the observations from this study,the tribocorrosion mechanism of the taper/trunnion interface under the long-term walking gait cycles is deduced.
