Neuroprotective and anti-inflammatory effects of eicosane on glutamate and NMDA-induced retinal ganglion cell injury

AIM:To investigate the protective effects,antioxidant potential,and anti-inflammatory mechanisms of eicosane on glutamate-induced cell damage and on N-methyl-Daspartate(NMDA)-induced retinal ganglion cell(RGC)injury in a mouse model of glaucoma.METHODS:The protective effects of eicosane on the rat R28 retinal precursor cell line were assessed using cell counting kit-8 assays and Hoechst-propidium iodide staining.Intracellular reactive oxygen species(ROS)production was measured using the fluorescent probe 2'-7'-dichlorofluorescin diacetate and flow cytometry.The protective role of eicosane on NMDA-induced RGC injury in a mouse glaucoma model was determined by immunostaining of frozen sections of retina.The effects of eicosane on the metabolome of the retina in mice with NMDA-induced RGC damage were evaluated by liquid chromatographymass spectroscopy(LC-MS)and untargeted metabolomics analyses.RESULTS:Eicosane treatment significantly attenuated glutamate-induced damage to R28 cells in vitro.Eicosane also protected RGCs against NMDA-induced injury in a mouse glaucoma model.Untargeted metabolomics analyses showed that eicosane increased multiple metabolites,including L-arginine and L-carnitine,in the retina.CONCLUSION:Eicosane has protective effects,antioxidant potential,and anti-inflammatory properties in an in vitro model of glutamate-induced cell damage and in an in vivo model of NMDA-induced RGC injury in mouse glaucoma through modulation of L-arginine and/or L-carnitine metabolism.

Fatigue reliability-based design optimization of aeroengine structures involves multiple repeated calculations of reliability degree and large-scale calls of implicit high-nonlinearity limit state function,leading to the traditional direct Monte Claro and surrogate methods prone to unacceptable computing efficiency and accuracy.In this case,by fusing the random subspace strategy and weight allocation technology into bagging ensemble theory,a random forest(RF)model is presented to enhance the computing efficiency of reliability degree;moreover,by embedding the RF model into multilevel optimization model,an efficient RF-assisted fatigue reliability-based design optimization framework is developed.Regarding the low-cycle fatigue reliability-based design optimization of aeroengine turbine disc as a case,the effectiveness of the presented framework is validated.The reliabilitybased design optimization results exhibit that the proposed framework holds high computing accuracy and computing efficiency.The current efforts shed a light on the theory/method development of reliability-based design optimization of complex engineering structures.

Spider silks are well known for their exceptional mechanical properties that are tougher than Kevlar and steel.However,the restricted production amounts from their native sources limit applications of spider silks.Over the decades,there have been significant interests in fabricating man-made silk fibers with comparable performance to natural silks,inspiring many efforts both for biosynthesizing recombinant spider silk proteins(spidroins)in amenable heterologous hosts and biomimetic spinning of artificial spider silks.These strategies provide promising routes to produce high-performance and functionally optimized fibers with diverse applications.Herein,we summarize the hosts that have been applied to produce recombinant spidroins.In addition,the fabrication and mechanical properties of recombinant spidroin fibers and their composite fibers are also introduced.Furthermore,we demonstrate the applications of recombinant spidroin-based fibers.Finally,facing the challenges in biosynthesis,scalable production,and hierarchical assembly of high-performance recombinant spidroins,we give a summary and perspective on future development.

The respiratory system's complex cellular heterogeneity presents unique challenges to researchers in this field.Although bulk RNA sequencing and single-cell RNA sequencing(scRNA-seq)have provided insights into cell types and heterogeneity in the respiratory system,the relevant specific spatial localization and cellular interactions have not been clearly elucidated.Spatial transcriptomics(ST)has filled this gap and has been widely used in respiratory studies.This review focuses on the latest iterative technology of ST in recent years,summarizing how ST can be applied to the physiological and pathological processes of the respiratory system,with emphasis on the lungs.Finally,the current challenges and potential development directions are proposed,including high-throughput full-length transcriptome,integration of multi-omics,temporal and spatial omics,bioinformatics analysis,etc.These viewpoints are expected to advance the study of systematic mechanisms,including respiratory studies.

Added Benefit of the Early-Morning-Orbit Satellite Fengyun-3E on the Global Microwave Sounding of the Three-Orbit Constellation

The three-orbit constellation can comprehensively increase the spatial coverage of polar-orbiting satellites,but the polar-orbiting satellites currently in operation are only mid-morning-orbit and afternoon-orbit satellites.Fengyun-3E(FY-3E)was launched successfully on 5 July 2021 in China.As an early-morning-orbit satellite,FY-3E can help form a complete three-orbit observation system together with the mid-morning and afternoon satellites in the current mainstream operational system.In this study,we investigate the added benefit of FY-3E microwave sounding observations to the midmorning-orbit Meteorological Operational satellite-B(Met Op-B)and afternoon-orbit Fengyun-3D(FY-3D)microwave observations in the Chinese Meteorological Administration global forecast system(CMA-GFS).The results show that the additional FY-3E microwave temperature sounder-3(MWTS-3)and microwave humidity sounder-2(MWHS-2)data can increase the global coverage of microwave temperature and humidity sounding data by 14.8% and 10.6%,respectively.It enables the CMA-GFS to achieve nearly 100% global coverage of microwave-sounding observations at each analysis time.Furthermore,after effective quality control and bias correction,the global biases and standard deviations of the differences between observations and model simulations are also reduced.Based on the Advanced Microwave Sounding Unit A and the Microwave Humidity Sounder onboard Met Op-B,and the MWTS-2 and MWHS-2 onboard FY-3D,adding the microwave sounding data of FY-3E can further reduce the errors of analysis results and improve the global prediction skills of CMA-GFS,especially for the southern-hemisphere forecasts within 96 hours,all of which are significant at the 95% confidence level.

Enhanced bright green luminescence from GdOF:Ho^(3+) up-conversion phosphor via Yb^(3+) doping

In this paper,a series of GdOF-based green phosphors doped with Ho^(3+) and Yb^(3+) ions was prepared by a simple co-precipitation method.The crystal structure s and optical prope rties were investigated in detail.The intensity of the emission of GdOF:Yb^(3+),Ho^(3+) phosphors is enhanced by the effective energy transfer from Yb^(3+) to Ho^(3+),and the optimal luminescence is achieved with the Ho^(3+)doping amount of 0.1 mol%as well as Yb^(3+) doping amount of 5 mol%.Tunable UC emission of samples is achieved by adjusting the doping concentration of Ho^(3+) or Yb^(3+).The purity of the green UC emission is further improved by enhancing the excitation pumping power.In addition,logarithmic functions of green,red and nearinfrared radiation intensities were calculated versus logarithmic function of pump power for further study.A possible UC luminescence mechanism between Yb^(3+) and Ho^(3+) is discussed based on powerdependent UC luminescence spectrum and decay lifetimes.These results suggest that GdOF:Yb^(3+),Ho^(3+)phosphors with bright and high-purity green UC emission have potential applications in the fields of bioimaging,display and illumination.

Performance Enhancement of Parabolic Trough Collector by Using Homogenizer and Spiral

In conventional parabolic trough collectors(PTCs),sunlight is concentrated at the bottom of the absorber tube,resulting in a significant circumferential temperature gradient across the absorber tube,heat loss and thermal deformation,which affects the safety and thermal performance of PTCs.In this study,a new receiver with homogenizer and spiral(RHS) is proposed,achieving the optical and thermal synergy to ameliorate the thermal deformation of the absorber tube and enhance thermal efficiency.A plane structure homogenizer is designed to improve uniformity of the concentrated solar flux of absorber tube through second reflection.In combination with the spiral,it improves the optical-thermal efficiency of the PTC by enhancing heat exchange between the fluid and the backlight side of the absorber tube.The performance of the collector is numerically studied by building a three-dimensional coupled light-thermal-structure model.The results show that the thermal deformation of the RHS is reduced by more than 96% and the optical-thermal efficiency is improved by 1.2%-0.63% compared with conventional receivers(CRs) under the same inlet temperature conditions.The proposed receiver is validated to be effective in reducing thermal deformation and improving optical-thermal efficiency.

Resolution Enhancement of SOHO/MDI Magnetograms

Research on the solar magnetic field and its effects on solar dynamo mechanisms and space weather events has benefited from the continual improvements in instrument resolution and measurement frequency. The augmentation and assimilation of historical observational data timelines also play a significant role in understanding the patterns of solar magnetic field variation. Within the realm of astronomical data processing,super-resolution(SR) reconstruction refers to the process of using a substantial corpus of training data to learn the nonlinear mapping between low-resolution(LR) and high-resolution(HR) images, thereby achieving higherresolution astronomical images. This paper is an application study in high-dimensional nonlinear regression. Deep learning models were employed to perform SR modeling on SOHO/MDI magnetograms and SDO/HMI magnetograms, thus reliably achieving resolution enhancement of full-disk SOHO/MDI magnetograms and enhancing the image resolution to obtain more detailed information. For this study, a data set comprising 9717pairs of data from 2010 April to 2011 February was used as the training set, 1332 pairs from 2011 March were used as the validation set and 1034 pairs from 2011 April were used as the test set. After data preprocessing, we randomly cropped 128 × 128 sub-images as the LR cases from the full-disk MDI magnetograms, and the corresponding 512 × 512 sub-images as HR ones from the HMI full-disk magnetograms for model training. The tests conducted have shown that the study successfully produced reliable 4× SR reconstruction of full-disk MDI magnetograms. The MESR model's results(0.911) were highly correlated with the target HMI magnetographs as indicated by the correlation coefficient values. Furthermore, the method achieved the best PSNR, SSIM, MAE and RMSE values, indicating that the MESR model can effectively reconstruct magnetograms.

Learning accurate template matching with differentiable coarseto-fine correspondence refinement

Template matching is a fundamental task in computer vision and has been studied for decades.It plays an essential role in manufacturing industry for estimating the poses of different parts,facilitating downstream tasks such as robotic grasping.Existing methods fail when the template and source images have different modalities,cluttered backgrounds,or weak textures.They also rarely consider geometric transformations via homographies,which commonly exist even for planar industrial parts.To tackle the challenges,we propose an accurate template matching method based on differentiable coarse-tofine correspondence refinement.We use an edge-aware module to overcome the domain gap between the mask template and the grayscale image,allowing robust matching.An initial warp is estimated using coarse correspondences based on novel structure-aware information provided by transformers.This initial alignment is passed to a refinement network using references and aligned images to obtain sub-pixel level correspondences which are used to give the final geometric transformation.Extensive evaluation shows that our method to be significantly better than state-of-the-art methods and baselines,providing good generalization ability and visually plausible results even on unseen real data.

Effect of aflibercept combined with triamcinolone acetonide on aqueous humor growth factor and inflammatory mediators in diabetic macular edema

AIM:To investigate the efficacy of aflibercept combined with sub-tenon injection of triamcinolone acetonide(TA)in treating diabetic macular edema(DME)and to examine changes in growth factors and inflammatory mediator levels in aqueous humor after injection.METHODS:Totally 67 DME patients(67 eyes)and 30 cataract patients(32 eyes)were enrolled as the DME group and the control group,respectively.The DME group was divided into the aflibercept group(34 cases)and the aflibercept combined with TA group(combined group,33 cases).The aqueous humor of both groups was collected during the study period.The aqueous levels of vascular endothelial growth factor(VEGF),monocyte chemoattractant protein-1(MCP-1),interleukin-6(IL-6),interleukin-8(IL-8),and interleukin-1β(IL-1β)were detected using a microsphere suspension array technology(Luminex 200TM).Aqueous cytokines,best-corrected visual acuity(BCVA),central macular thickness(CMT),and complications before and after treatment were compared between the aflibercept group and combined group.RESULTS:The concentrations of VEGF,MCP-1,IL-6,and IL-8 in the aqueous humor were significantly higher in the DME group than those of the control group(all P0.05).CONCLUSION:The cytokines VEGF,MCP-1,IL-6,and IL-8 in the aqueous humor of DME patients are significantly increased.Aflibercept and aflibercept combined with TA have good efficacy in DME patients,can effectively reduce CMT,improve the patient’s vision,and have high safety.Aflibercept combined with TA can quickly downregulate the aqueous humor cytokines and help to relieve macular edema rapidly.However,the long-term efficacy is comparable to that of aflibercept alone
