Yeast diversity and screening of biocontrol Yarrawia lipolitica against ochratoxin A biosynthesis by Aspergillus Niger in Panxian ham

Nature fermented dry-cured hams may be contaminated by ochratoxin A (OTA) due to the colonization of OTA-producing fungi. Yeast plays essential roles in the formation of the special flavor of ham as well as the potential to retard OTA biosynthesis. In this study， the yeast diversity of Panxian hams fermented from 1 year to 2 years was investigated， revealing a total of 9 yeast genera. The dominant genus switched from Debaryomyces hansenii to Yamadazyma triangularis， while the yeast diversity increased with the prolongation of the fermentation period. Notably， Yarrawia lipolitica GZPX-3Y-1 was identified as a biocontrol yeast strain that can effectively inhibit the OTA biosynthesis up to 82% in OTA-producing modeling media and 55% in ham media when cocultivated with a previously identified OTA producing fungi from Panxian ham， specifically Aspergillus niger A-8. The double-plate methods experiment unveiled that Yarrawia lipolitica GZPX-3Y-1 can only inhibit the growth of Aspergillus niger A-8 under the anoxic conditions. Characteristics of Yarrawia lipolitica GZPX-3Y-1 were explored， showing its ability to grow at wide range of temperature (18℃ ～33℃) and pH levels (3-12). Additionally， it exhibited tolerance to high salt conditions (8%) and demonstrated lipolysis ability without hemolytic ability. These findings indicate the great potential of Yarrawia lipolitica GZPX-3Y-1 to be applied in the fermentation of Panxian ham as a biocontrol strain.

The present study aims to examine the suitability of two commonly used assumptions that simplify modelling metoceanconditions for designing offshore wind turbines in the South China Sea (SCS). The first assumption assumes thatjoint N-year extreme wind and wave events can be independently estimated and subsequently combined. The secondone assumes peak wind and waves can be modelled as occurring simultaneously during a tropical cyclone (TC) event.To better understand the potential TC activity, a set of 10000 years synthetic TC events are generated. The wind fieldmodel and the Mike 21 spectral wave model are employed to model the TC-induced hazards. Subsequently, theeffect of the assumptions is evaluated by analyzing the peak structural response of both monopile and semisubmersibleoffshore wind turbines during TC events. The results demonstrate that the examined assumptions are generally accurate.By assessing the implications of these assumptions, valuable insights are obtained, which can inform andimprove the modelling of TC-induced hazards in the SCS region.

BACKGROUND The SETD1B gene is instrumental in human intelligence and nerve development.Mutations in the SETD1B gene have been linked in recent studies to neurodevelopmental disorders,seizures,and language delay.CASE SUMMARY This study aimed to analyze the clinical manifestations and treatment of three patients suffering from mental retardation,epilepsy,and language delay resulting from a new mutation in the SETD1B gene.Three individuals with these symptoms were selected,and their clinical symptoms,gene test results,and treatment were analyzed.This article discusses the impact of the SETD1B gene mutation on patients and outlines the treatment approach.Among the three patients(two females and one male,aged 8,4,and 1,respectively),all exhibited psychomotor retardation,attention deficit,and hyperactivity disorder,and two had epilepsy.Antiepileptic treatment with sodium tripolyvalproate halted the seizures in the affected child,although mental development remained somewhat delayed.Whole exome sequencing revealed new mutations in the SETD1B gene for all patients,specifically with c.5473C>T(p.Arg1825trp),c.4120C>T(p.Gln1374*,593),c.14_15insC(p.His5Hisfs*33).CONCLUSION Possessing the SETD1B gene mutation may cause mental retardation accompanied by seizures and language delay.Although the exact mechanism is not fully understood,interventions such as drug therapy,rehabilitation training,and family support can assist patients in managing their symptoms and enhancing their quality of life.Furthermore,genetic testing supplies healthcare providers with more precise diagnostic and therapeutic guidance,informs families about genetic disease risks,and contributes to understanding disease pathogenesis and drug research and development.

Internal curing agents (ICA) based on super absorbent polymer have poor alkali tolerance and reduce the early strength of concrete.An alkali tolerate internal curing agent (CAA-ICA) was designed and prepared by using sodium carboxymethyl starch (CMS) with high hydrophilicity,acrylic acid (AA) containing anionic carboxylic group and acrylamide (AM) containing non-ionic amide group as the main raw materials.The results show that the ratio of CAA-ICA alkali absorption solution is higher than that existing ICA,which solves the low water absorption ratio of the ICA in alkali environment.The water absorption ratio of CAA-ICA in saturated Ca(OH)_(2) solution is 95.8 g·g^(-1),and the alkali tolerance coefficient is 3.4.The application of CAA-ICA in cement-based materials can increase the internal relative humidity and miniaturize the pore structure.The compressive strength of mortar increases up to 12.95%at 28 d,which provids a solution to overcome the reduction of the early strength.

Study of the pressure transient behavior of directional wells considering the effect of non-uniform flux distribution

During the production, the fluid in the vicinity of the directional well enters the wellbore with different rates, leading to non-uniform flux distribution along the directional well. However, in all existing studies,it is oversimplified to a uniform flux distribution, which can result in inaccurate results for field applications. Therefore, this paper proposes a semi-analytical model of a directional well based on the assumption of non-uniform flux distribution. Specifically, the direction well is discretized into a carefully chosen series of linear sources, such that the complex well trajectory can be captured and the nonuniform flux distribution along the wellbore can be considered to model the three-dimensional flow behavior. By using the finite difference method, we can obtain the numerical solutions of the transient flow within the wellbore. With the aid of Green's function method, we can obtain the analytical solutions of the transient flow from the matrix to the wellbore. The complete flow behavior of a directional well is perfectly represented by coupling the above two types of transient flow. Subsequently, on the basis of the proposed model, we conduct a comprehensive analysis of the pressure transient behavior of a directional well. The computation results show that the flux variation along the direction well has a significant effect on pressure responses. In addition, the directional well in an infinite reservoir may exhibit the following flow regimes: wellbore afterflow, transition flow, inclined radial flow, elliptical flow, horizontal linear flow, and horizontal radial flow. The horizontal linear flow can be observed only if the formation thickness is much smaller than the well length. Furthermore, a dip region that appears on the pressure derivative curve indicates the three-dimensional flow behavior near the wellbore.

Functional divergences of natural variations of TaNAM-A1 in controlling leaf senescence during wheat grain filling

Leaf senescence is an essential physiological process related to grain yield potential and nutritional quality. Green leaf duration(GLD) after anthesis directly reflects the leaf senescence process and exhibits large genotypic differences in common wheat; however, the underlying gene regulatory mechanism is still lacking. Here,we identified TaNAM-A1 as the causal gene of the major loci q GLD-6A for GLD during grain filling by map-based cloning. Transgenic assays and TILLING mutant analyses demonstrated that TaNAM-A1 played a critical role in regulating leaf senescence, and also affected spike length and grain size. Furthermore, the functional divergences among the three haplotypes of TaNAM-A1 were systematically evaluated. Wheat varieties with TaNAM-A1d(containing two mutations in the coding DNA sequence of TaNAM-A1) exhibited a longer GLD and superior yield-related traits compared to those with the wild type TaNAM-A1a. All three haplotypes were functional in activating the expression of genes involved in macromolecule degradation and mineral nutrient remobilization,with TaNAM-A1a showing the strongest activity and TaNAM-A1d the weakest. TaNAM-A1 also modulated the expression of the senescencerelated transcription factors TaNAC-S-7A and TaNAC016-3A. TaNAC016-3A enhanced the transcriptional activation ability of TaNAM-A1a by protein–protein interaction, thereby promoting the senescence process. Our study offers new insights into the fine-tuning of the leaf functional period and grain yield formation for wheat breeding under various geographical climatic conditions.

The hot deformation behaviors of sulfur-containing gear steel 20MnCr5 containing three different contents of Nb and B(0,0.021%Nb,and 0.024%Nb-0.0022%B)were investigated.Hot compression and tenssion tests were carried out by Gleeble3800 at the austenite region from 850 to 1150℃and the adverse effects of Nb and B were analyzed by the fracture,microstructure and precipitate observations.Hot compression tests showed that the proportions of instable area in hot processing maps of 0.021%Nb and Nb-B steels were higher and the deformability of Nb free steel was better.The tensile deformation experiments showed that the reduction areas of Nb free,0.021%Nb and Nb-B steels were 92%-99%,84%-98%and 67%-97%,respectively.The addition of Nb or Nb and B inhibited the dynamic recrystallization during hot deformation,and consequently,more deformed grains were then formed in 0.021%Nb and Nb-B steels thus to obtain the microstructure with worse uniformity and then deteriorate the deformability.In addition,the interaction between inclusions and microalloyed elements was also significant.NbC particles of 0.021%Nb and Nb-B steels dynamically precipitated during deformation and precipitated together with MnS thus to worsen the deformability,resulting in the decrease of reduction area.

BACKGROUND The incidence of chronic kidney disease among patients with diabetes mellitus(DM)remains a global concern.Long-term obesity is known to possibly influence the development of type 2 diabetes mellitus.However,no previous meta-analysis has assessed the effects of body mass index(BMI)on adverse kidney events in patients with DM.AIM To determine the impact of BMI on adverse kidney events in patients with DM.METHODS A systematic literature search was performed on the PubMed,ISI Web of Science,Scopus,Ovid,Google Scholar,EMBASE,and BMJ databases.We included trials with the following characteristics:(1)Type of study:Prospective,retrospective,randomized,and non-randomized in design;(2)participants:Restricted to patients with DM aged≥18 years;(3)intervention:No intervention;and(4)kidney adverse events:Onset of diabetic kidney disease[estimated glomerular filtration rate(eGFR)of<60 mL/min/1.73 m2 and/or microalbuminuria value of≥30 mg/g Cr],serum creatinine increase of more than double the baseline or end-stage renal disease(eGFR<15 mL/min/1.73 m2 or dialysis),or death.RESULTS Overall,11 studies involving 801 patients with DM were included.High BMI(≥25 kg/m2)was significantly associated with higher blood pressure(BP)[systolic BP by 0.20,95%confidence interval(CI):0.15–0.25,P<0.00001;diastolic BP by 0.21 mmHg,95%CI:0.04–0.37,P=0.010],serum albumin,triglycerides[standard mean difference(SMD)=0.35,95%CI:0.29–0.41,P<0.00001],low-density lipoprotein(SMD=0.12,95%CI:0.04–0.20,P=0.030),and lower high-density lipoprotein(SMD=–0.36,95%CI:–0.51 to–0.21,P<0.00001)in patients with DM compared with those with low BMIs(<25 kg/m2).Our analysis showed that high BMI was associated with a higher risk ratio of adverse kidney events than low BMI(RR:1.22,95%CI:1.01–1.43,P=0.036).CONCLUSION The present analysis suggested that high BMI was a risk factor for adverse kidney events in patients with DM.

The NAC transcription factor LuNAC61 negatively regulates fiber development in flax (Linum usitatissimum L.)

Flax is a crucial fiber crop that exhibits excellent textile properties and serves as a model plant for investigating phloem fiber development. The regulation of multiple genes significantly influences fiber development, notably involving NAC(NAM, ATAF1/2, CUC2) transcription factors in forming the fiber secondary cell wall(SCW).Overexpression of LuNAC61 in flax resulted in sparse top meristematic zone leaves and significantly reduced stem cellulose content. Scanning electron microscopy and staining observations revealed a significant reduction in fiber bundles. β-Glucuronidase(GUS) staining analysis demonstrated high activity of the LuNAC61 promoter in the bast fibers of the flax stem. Additionally, several members of the LuPLATZ and LuCesA families exhibited significant coexpression with LuNAC61. Subcellular localization indicated the presence of LuPLATZ24 protein in the nucleus and cytoplasm, LuNAC61 protein exclusively in the nucleus, and LuCesA10 in the nucleus and endoplasmic reticulum. LuPLATZ24 positively regulates LuNAC61, whereas LuNAC61 negatively affects LuCesA10, suggesting the involvement of a metabolic network in regulating flax fiber development. In conclusion, this study provides a critical opportunity for a comprehensive and in-depth analysis of the mechanisms governing flax fiber development and the potential use of biotechnology to enhance flax fiber yield.
