Additive manufactured osseointegrated screws with hierarchical design

Bone screws are devices used to fix implants or bones to bones.However,conventional screws are mechanically fixed with thread and often face long-term failure due to poor osseointegration.To improve osseointegration,screws are evolving from solid and smooth to porous and rough.Additive manufacturing(AM)offers a high degree of manufacturing freedom,enabling the preparation of predesigned screws that are porous and rough.This paper provides an overview of the problems currently faced by bone screws:long-term loosening and screw breakage.Next,advances in osseointegrated screws are summarized hierarchically(sub-micro,micro,and macro).At the sub-microscale level,we describe surface-modification techniques for enhancing osseointegration.At the micro level,we summarize the micro-design parameters that affect the mechanical and biological properties of porous osseointegrated screws,including porosity,pore size,and pore shape.In addition,we highlight three promising pore shapes:triply periodic minimal surface,auxetic structure with negative Poisson ratio,and the Voronoi structure.At the macro level,we outline the strategies of graded design,gradient design,and topology optimization design to improve the mechanical strength of porous osseointegrated screws.Simultaneously,this paper outlines advances in AM technology for enhancing the mechanical properties of porous osseointegrated screws.AM osseointegrated screws with hierarchical design are expected to provide excellent long-term fixation and the required mechanical strength.

Liquid metal biomaterials: translational medicines,challenges and perspectives

Until now, significant healthcare challenges and growing urgent clinical requirements remain incompletely addressed by presently available biomedical materials. This is due to their inadequate mechanical compatibility, suboptimal physical and chemical properties, susceptibility to immune rejection, and concerns about long-term biological safety. As an alternative, liquid metal(LM) opens up a promising class of biomaterials with unique advantages like biocompatibility, flexibility, excellent electrical conductivity,and ease of functionalization. However, despite the unique advantages and successful explorations of LM in biomedical fields, widespread clinical translations and applications of LM-based medical products remain limited. This article summarizes the current status and future prospects of LM biomaterials, interprets their applications in healthcare, medical imaging, bone repair, nerve interface, and tumor therapy, etc.Opportunities to translate LM materials into medicine and obstacles encountered in practices are discussed.Following that, we outline a blueprint for LM clinics, emphasizing their potential in making new-generation artificial organs. Last, the core challenges of LM biomaterials in clinical translation, including bio-safety,material stability, and ethical concerns are also discussed. Overall, the current progress, translational medicine bottlenecks, and perspectives of LM biomaterials signify their immense potential to drive future medical breakthroughs and thus open up novel avenues for upcoming clinical practices.

In vivo label-free measurement of blood flow velocity symmetry based on dual line scanning third-harmonic generation microscopy excited at the 1700 nm window

Measurement of bloodflow velocity is key to understanding physiology and pathology in vivo.While most measurements are performed at the middle of the blood vessel,little research has been done on characterizing the instantaneous bloodflow velocity distribution.This is mainly due to the lack of measurement technology with high spatial and temporal resolution.Here,we tackle this problem with our recently developed dual-wavelength line-scan third-harmonic generation(THG)imaging technology.Simultaneous acquisition of dual-wavelength THG line-scanning signals enables measurement of bloodflow velocities at two radially symmetric positions in both venules and arterioles in mouse brain in vivo.Our results clearly show that the instantaneous bloodflow velocity is not symmetric under general conditions.

Chlorinated butyl rubber/two-step modified montmorillonite nanocomposites:Mechanical and damping properties

Montmorillonite(MMT) was modified by ultrasound and castor oil quaternary ammonium salt intercalation method to prepare a new type of organic montmorillonite(OMMT). The surface structure, particle morphology, interlayer distance, and thermal behavior of the samples obtained were characterized. The modified OMMT was then added to chlorinated butyl rubber(CIIR) by mechanical blending, and a composite material with excellent damping properties was obtained. The mechanical experiment results of CIIR nanocomposites showed that the addition of OMMT improved their tensile strength, hardness,and stress relaxation rate. Compared with pure CIIR, when the content of OMMT was 5 phr(part per hundred of rubber), the tensile strength of the nanocomposite was increased by 677% and the elongation at break was also increased by 105.4%. The enhancement of this performance was mainly due to the dispersion of the nanosheets in CIIR rubber and the chemical interaction between the organoclay and the polymer matrix, which was confirmed by morphology and spectral analysis. OMMT also endowed a positive effect on the damping properties of CIIR nanocomposites. After adding 5 phr of OMMT, the nanocomposite owned the best damping performance, and the damping factor, tanδmax, was 37.9% higher than that of pure CIIR. Therefore, the good damping and mechanical properties of these CIIR nanocomposites provided some novel and promising methods for preparing high-damping rubber in a wide temperature range.

Continuous adaptive gaits manipulation for three-fingered robotic hands via bioinspired fingertip contact events

The remarkable skill of changing its grasp status and relocating its fingers to perform continuous in-hand manipulation is essential for a multifingered anthropomorphic hand.A commonly utilized method of manipulation involves a series of basic movements executed by a high-level controller.However,it remains unclear how these primitives evolve into sophisticated finger gaits during manipulation.Here,we propose an adaptive finger gait-based manipulation method that offers real-time regulation by dynamically changing the primitive interval to ensure the force/moment balance of the object.Successful manipulation relies on contact events that act as triggers for real-time online replanning of multifinger manipulation.We identify four basic motion primitives of finger gaits and create a heuristic finger gait that enables the continuous object rotation of a round cup.Our experimental results verify the effectiveness of the proposed method.Despite the constant breaking and reengaging of contact between the fingers and the object during manipulation,the robotic hand can reliably manipulate the object without failure.Even when the object is subjected to interfering forces,the proposed method demonstrates robustness in managing interference.This work has great potential for application to the dexterous operation of anthropomorphic multifingered hands.

Full-parameter constrained parsimonious subspace identification with steady-state information for DC–DC converters

A full-parameter constrained parsimonious subspace identification method that incorporates the steady-state a priori infor-mation of the system is proposed to model the DC-DC converters.A parsimonious model with fewer parameters is used to represent the system,and then an optimal weighted methods is used to estimate the system parameters matrices by taking into account both dynamical data and steady-state data.Compared with traditional data-driven methods for DC-DC convert-ers,the subspace-based method can simultaneously estimate model structure and parameter with appropriate computational complexity.Moreover,compared with the traditional full-parameter constrained subspace approach,the proposed algorithm can accurately estimate the system parameters with a smaller variance.The experimental results on a DC-DC synchronous buck converter verify the effectiveness and superiority of the proposed method.

Kink oscillations,which are frequently observed in coronal loops and prominences,are often accompanied by extreme-ultraviolet(EUV)waves.However,much more needs to be explored regarding the causal relationships between kink oscillations and EUV waves.In this article,we report the simultaneous detection of kink oscillations and EUV waves that are both associated with an X2.1 flare on 2023 March 03(SOL2023-03-03T17:39).The kink oscillations,which are almost perpendicular to the axes of loop-like structures,are observed in three coronal loops and one prominence.One short loop shows in-phase oscillation within the same period of 5.2 min at three positions.This oscillation could be triggered by the pushing of an expanding loop and interpreted as the standing kink wave.Time lags are found between the kink oscillations of the short loop and two long loops,suggesting that the kink wave travels in different loops.The kink oscillations of one long loop and the prominence are possibly driven by the disturbance of the coronal mass ejection(CME),and that of another long loop might be attributed to the interaction of the EUV wave.The onset time of the kink oscillation of the short loop is nearly same as the beginning of an EUV wave.This fact demonstrates that they are almost simultaneous.The EUV wave is most likely excited by the expanding loop structure and shows two components.The leading component is a fast coronal wave,and the trailing one could be due to the stretching magnetic field lines.

Correlative Mn-Co catalyst excels Pt in oxygen reduction reaction of quasi-solid-state zinc-air batteries

Zn-air batteries(ZABs)as a class of promising energy storage setups are generally powered by efficient and robust catalysts at the oxygen-involving cathode.Although the existing non-noble catalysts have outperformed noble Pt benchmark in the alkaline liquid-state ZABs,to the best of our knowledge few have excelled Pt in quasi-solid-state(QSS)ZABs.Herein,we found that an integrated Mn-Co cathode derived from the bimetallic Mn/Co metal organic frameworks generates a 1.4-fold greater power density in the QSS ZABs than a Pt cathode while its power density in liquid-state ZABs is only 0.8-fold of the latter.Moreover,such Mn-Co catalyst delivers high-rate oxygen reduction reaction(ORR)capability with half-wave potential of 0.84 V.The in-depth characterizations and analyses have demonstrated that the Co and Mn species show the specific affinity towards H_(2)O and O_(2),respectively,synergizing the ORR process in the water-deficient environment of QSS ZABs.This work has enlightened the rational design of non-noble metal catalysts to improve the power density of QSS ZABs.

Opportunities and challenges of incretin-based hypoglycemic agents treating type 2 diabetes mellitus from the perspective of physiological disposition

The treatment of patients with diabetes mellitus, which is characterized by defective insulin secretion and/or the inability of tissues to respond to insulin, has been studied for decades. Many studies have focused on the use of incretin-based hypoglycemic agents in treating type 2 diabetes mellitus(T2DM). These drugs are classified as GLP-1 receptor agonists, which mimic the function of GLP-1,and DPP-4 inhibitors, which avoid GLP-1 degradation. Many incretin-based hypoglycemic agents have been approved and are widely used, and their physiological disposition and structural characteristics are crucial in the discovery of more effective drugs and provide guidance for clinical treatment of T2DM.Here, we summarize the functional mechanisms and other information of the drugs that are currently approved or under research for T2DM treatment. In addition, their physiological disposition, including metabolism, excretion, and potential drug drug interactions, is thoroughly reviewed. We also discuss similarities and differences in metabolism and excretion between GLP-1 receptor agonists and DPP-4 inhibitors. This review may facilitate clinical decision making based on patients' physical conditions and the avoidance of drug drug interactions. Moreover, the identification and development of novel drugs with appropriate physiological dispositions might be inspired.

Multi-omics analysis of adamantinomatous craniopharyngiomas reveals distinct molecular subgroups with prognostic and treatment response significance

Background: Adamantinomatous craniopharyngioma (ACP) is the commonest pediatric sellar tumor. No effective drug is available and interpatient heterogeneity is prominent. This study aimed to identify distinct molecular subgroups of ACP based on the multi-omics profiles, imaging findings, and histological features, in order to predict the response to anti-inflammatory treatment and immunotherapies.Methods: Totally 142 Chinese cases diagnosed with craniopharyngiomas were profiled, including 119 ACPs and 23 papillary craniopharyngiomas. Whole-exome sequencing (151 tumors, including recurrent ones), RNA sequencing (84 tumors), and DNA methylome profiling (95 tumors) were performed. Consensus clustering and non-negative matrix factorization were used for subgrouping, and Cox regression were utilized for prognostic evaluation, respectively.Results: Three distinct molecular subgroups were identified: WNT, ImA, and ImB. The WNT subgroup showed higher Wnt/β-catenin pathway activity, with a greater number of epithelial cells and more predominantly solid tumors. The ImA and ImB subgroups had activated inflammatory and interferon response pathways, with enhanced immune cell infiltration and more predominantly cystic tumors. Mitogen-activated protein kinases (MEK/MAPK) signaling was activated only in ImA samples, while IL-6 and epithelial-mesenchymal transition biomarkers were highly expressed in the ImB group, mostly consisting of children. The degree of astrogliosis was significantly elevated in the ImA group, with severe finger-like protrusions at the invasive front of the tumor. The molecular subgrouping was an independent prognostic factor, with the WNT group having longer event-free survival than ImB (Cox,P = 0.04). ImA/ImB cases were more likely to respond to immune checkpoint blockade (ICB) therapy than the WNT group (P <0.01).
