Direct in vivo conversion of astrocytes into functional new neurons induced by neural transcription factors has been recognized as a potential new therapeutic intervention for neural injury and degenerative disorders. However, a few recent studies have claimed that neural transcription factors cannot convert astrocytes into neurons, attributing the converted neurons to pre-existing neurons mis-expressing transgenes. In this study, we overexpressed three distinct neural transcription factors––NeuroD1, Ascl1, and Dlx2––in reactive astrocytes in mouse cortices subjected to stab injury, resulting in a series of significant changes in astrocyte properties. Initially, the three neural transcription factors were exclusively expressed in the nuclei of astrocytes. Over time, however, these astrocytes gradually adopted neuronal morphology, and the neural transcription factors was gradually observed in the nuclei of neuron-like cells instead of astrocytes. Furthermore,we noted that transcription factor-infected astrocytes showed a progressive decrease in the expression of astrocytic markers AQP4(astrocyte endfeet signal), CX43(gap junction signal), and S100β. Importantly, none of these changes could be attributed to transgene leakage into preexisting neurons. Therefore, our findings suggest that neural transcription factors such as NeuroD1, Ascl1, and Dlx2 can effectively convert reactive astrocytes into neurons in the adult mammalian brain.

Objective To investigate the effects of bisdemethoxycurcumin （BDMC） on non-small cell lung cancer （NSCLC） cell line, A549, and the highly metastatic lung cancer 95D cells. Methods CCK-8 assay was used to assess the effect of BDMC on cytotoxicity. Flow cytometry was used to evaluate apoptosis. Western blot analysis, electron microscopy, and quantification of GFP-LC3 punctuates were used to test the effect of BDMC on autophagy and apoptosis of lung cancer cells. Results BDMC inhibited the viability of NSCLC cells, but had no cytotoxic effects on lung small airway epithelial cells （SAECs）. The apoptotic cell death induced by BDMC was accompanied with the induction of autophagy in NSCLC cells. Blockage of autophagy by the autophagy inhibitor 3-methyladenine （3-MA） repressed the growth inhibitory effects and induction of apoptosis by BDMC. In addition, BDMC treatment significantly decreased smoothened （SMO） and the transcription factor glioma-associated oncogene 1 （G1il） expression. Furthermore, depletion of Gill by siRNA and cyclopamine （a specific SMO inhibitor） induced autophagy. Conclusion Aberrant activation of hedgehog （Hh） signaling has been implicated in several human cancers, including lung cancers. The present findings provide direct evidence that BDMC-induced autophagy plays a pro-death role in NSCLC, in part, by inhibiting hedgehog signaling.

Au nanocomposite enhanced electret film for triboelectric nanogenerator

A triboelectric nanogenerator （TENG） with an organic nanocomposite electret thin film as the triboelectric layer for mechanical energy harvesting was investigated systematically. In combination with corona charging, a TENG was fabricated by using embedded-nanocapacitor-structure polytetrafluoroethylene （PTFE） impregnated with gold nanoparticles （Au-NPs）. The output performances, stability, and durability of the TENGs with Au-PTFE nanocomposite films were characterized after being washed in water. It was found that the output current increases by 70% and the equivalent surface charge density （ESCD） reaches 85 μC/m^2 in comparison to the virgin PTFE film. Such outstanding performance is likely due to the equivalent nanocapacitors between the Au-NPs and PTFE molecules, which serve as nano charge traps in the nanocomposite electret film under negative high-voltage corona charging. This work not only expands the practical applications of TENGs, but also opens up new possibilities for the development of high performance triboelectric materials.

Transient and weak protein–protein interactions are essential to many biochemical reactions, yet are technically challenging to study. Chemical cross-linking of proteins coupled with mass spectrometry analysis(CXMS) provides a powerful tool in the analysis of such interactions. Central to this technology are chemical cross-linkers. here, using two transient heterodimeric complexes EIN/HPr and EIIAGlc/EIIBGlc as our model systems, we evaluated the effects of two amine-specific homo-bifunctional cross-linkers with different reactivities. We showed previously that DOPA2(di-ortho-phthalaldehyde with a di-ethylene glycol spacer arm) cross-links proteins 60–120 times faster than DSS(disuccinimidyl suberate). We found that though most of the intermolecular cross-links of either cross-linker are consistent with the encounter complexes(ECs), an ensemble of short-lived binding intermediates,more DOPA2 intermolecular cross-links could be assigned to the stereospecific complex(SC), the final lowest-energy conformational state for the two interacting proteins. Our finding suggests that faster cross-linking captures the SC more effectively and cross-linkers of different reactivities potentially probe protein–protein interaction dynamics across multiple timescales.

Relationships among the Stem,Aboveground and Total Biomass across Chinese Forests

Forest biomass plays a key role in the global carbon cycle. In the present study, a general allometric model was derived to predict the relationships among the stem biomass Ms, aboveground biomass MA and total biomass MT, based on previously developed scaling relationships for leaf, stem and root standing biomass. The model predicted complex scaling exponents for MT and/or MA with respect to Ms. Because annual biomass accumulation in the stem, root and branch far exceeded the annual increase in standing leaf biomass, we can predict that MT ∝MA ∝ Ms as a simple result of the model. Although slight variations existed in different phyletic affiliations （i.e. conifers versus angiosperms）, empirical results using Model Type Ⅱ （reduced major axis） regression supported the model＇s predictions. The predictive formulas among stem, aboveground and total biomass were obtained using Model Type I （ordinary least squares） regression to estimate forest biomass. Given the low mean percentage prediction errors for aboveground （and total biomass） based on the stem biomass, the results provided a reasonable method to estimate the biomass of forests at the individual level, which was insensitive to the variation in local environmental conditions （e.g. precipitation, temperature, etc.）.

Establishment of a multi-cycle generalized Weng model and its application in forecasts of global oil supply

Low oil prices under the influence of economic structure transformation and slow economic growth have hit the existing markets of traditional big oil suppliers and upgraded the conflict of oil production capacity and interest between OPEC producers and other big oil supplier countries such as the USA and Russia. Forecasting global oil production is significant for all countries for energy strategy planning, although many past forecasts have later been proved to be very seriously incorrect. In this paper,the original generalized Weng model is expanded to a multi-cycle generalized Weng model to better reflect the multi-cycle phenomena caused by political, economic and technological factors. This is used to forecast global oil production based on parameter selection from a large sample, depletion rate of remaining resources, constraints on oil reserves and cycle number determination. This research suggests that the world will reach its peak oil production in 2022, at about 4340×10~6 tonnes. China needs to plan for oil import diversity, a domestic oil production structure based on the supply pattern of large oil suppliers worldwide and the oil demand for China＇s own development.
