Purification, Biological Activities, and Molecular Cloning of a Novel Mannose-Binding Lectin from Bulbs of Zephyranthes candida Herb （Amaryllidaceae）

A novel mannose-bindlng aggiutinln was purified from bulbs of Zephyranthes candida Herb by extraction, precipitation with 80% （NH4）2SO4, and ion-exchange chromatography on DEAE-Sepharose followed by gel flitration on Sephscryl S-100. The purified Z. candida agglutlnln （ZCA） migrated as a single band of 12 kDa on sodium dodecyi suifate-poiyecryiamide gel electrophoresis under reducing and non-reducing conditions. The apparent molecular mass of the iectln, as datermlned by gel filtration chromatography, was 48 kDa. The results Indicated that ZCA was composed of four Identical subunlts of 12 kDa each （homotetramerlc nature）. The ZCA agglutlhated rabbit erythrocytes, Escherichla coil and Saccharomyces cerevislae ceils at concentrations of 0.95, 1.90, and 31.30 μg/mL, respectively. Bloassays Indicated that ZCA has a significant effect on wheat aphid survival. Mortality after 7 d was 〉 90% at 0.26%. A degenerate primer was designed In accordance with the N-terminal partial sequence of purified ZCA. The full-length cDNA was cloned by 3＇- and 5＇-rapid amplification of cDNA ends. The full-length cDNA had 661 bp and the sequence encoded an open reading frame of 168 amino acids. The mature protein of ZCA Includes 109 amino acid residues and the molecular weight of the protein was 12.1 kDa. The result show that the zca gene encodes a protein precursor with a signal peptlde, a mature protein, and a C-terminal cleavage amino acids sequence. Molecular modeling of ZCA Indicated that Its three-dimensional atructure strongly resembies that of the snowdrop aggiutinin. Blocks＇ analysis revealed that the deduced amino acid sequence of ZCA has three functional domains specific for agglutination and three carbohydrate binding boxes （QDNY）.

MAPKs and acetyl-CoA are associated with Curvularia lunata pathogenicity and toxin production in maize

Mitogen-activated protein kinase （MAPK） cascades play an important role in extracellular signal transduction and are involved in the pathogenicity of fungal pathogens to host plants. In Curvularia lunata, the roles of two MAPK genes, Clkl and CIm 1, have already been studied. Clkl is involved in conidia formation and pathogenicity, and Clmf is closely related to pathogen cell wall formation and pathogenicity to maize leaves. In this study, a third C. lunata MAPK gene, Clhl, which is homologous to hog1, was successfully cloned. We found that a Clhl deletion mutant had lower intracellular glycerol accumulation than the wild-type stain and was unable to grow normally under osmotic stress conditions. Furthermore, the deletion mutants of three C. lunata MAPK genes （Clkl, Clml and Clhl） had lower levels of acetyI-CoA, which is an important intermediate product in the synthesis of melanin and furan toxin, and down-regulated expression of pathogenicity-associated genes. Furthermore, pathogenicity and the ability to produce toxin were restored after adding acetyI-CoA to the culture medium, suggesting that acetyI-CoA is closely involved in the pathogen MAPK signaling pathway.

Effects of the extrusion parameters on microstructure,texture and room temperature mechanical properties of extruded Mg-2.49Nd-1.82Gd-0.2Zn-0.2Zr alloy

Microstructure,texture,and mechanical properties of the extruded Mg-2.49Nd-1.82Gd-0.2Zn-0.2Zr alloy were investigated at different extrusion temperatures(260 and 320℃),extrusion ratios(10:1,15:1,and 30:1),and extrusion speeds(3 and 6 mm/s).The experimental results exhibited that the grain sizes after extrusion were much finer than that of the homogenized alloy,and the second phase showed streamline distribution along the extrusion direction(ED).With extrusion temperature increased from 260 to 320℃,the microstructure,texture,and mechanical properties of alloys changed *** dynamic recrystallization(DRX)degree and grain sizes enhanced as the extrusion ratio increased from 10:1 to 30:1,and the strength gradually decreased but elongation(EL)*** the extrusion speed increased from 3 to 6 mm/s,the grain sizes and DRX degree increased significantly,and the samples presented the typical-rare-earth(RE)*** alloy extruded at 260℃ with extrusion ratio of 10:1 and extrusion speed of 3 mm/s showed the tensile yield strength(TYS)of 213 MPa and EL of 30.6%.After quantitatively analyzing the contribution of strengthening mechanisms,it was found that the grain boundary strengthening and dislocation strengthening played major roles among strengthening *** results provide some guidelines for enlarging the industrial application of extruded Mg-RE alloy.

Several promising plasma biomarker proteins,such as amyloid-β(Aβ),tau,neurofilament light chain,and glial fibrillary acidic protein,are widely used for the diagnosis of neurodegenerative ***,little is known about the long-term stability of these biomarker proteins in plasma samples stored at-80°*** aimed to explore how storage time would affect the diagnostic accuracy of these biomarkers using a large *** samples from 229 cognitively unimpaired individuals,encompassing healthy controls and those experiencing subjective cognitive decline,as well as 99 patients with cognitive impairment,comprising those with mild cognitive impairment and dementia,were acquired from the Sino Longitudinal Study on Cognitive Decline *** samples were stored at-80°C for up to 6 years before being used in this *** results showed that plasma levels of Aβ42,Aβ40,neurofilament light chain,and glial fibrillary acidic protein were not significantly correlated with sample storage ***,the level of total tau showed a negative correlation with sample storage ***,in individuals without cognitive impairment,plasma levels of total protein and tau phosphorylated protein threonine 181(p-tau181)also showed a negative correlation with sample storage *** was not observed in individuals with cognitive ***,we speculate that the diagnostic accuracy of plasma p-tau181 and the p-tau181 to total tau ratio may be influenced by sample storage ***,caution is advised when using these plasma biomarkers for the identification of neurodegenerative diseases,such as Alzheimer's ***,in cohort studies,it is important to consider the impact of storage time on the overall results.

A low redox potential and long life organic anode material for sodium-ion batteries

Sodium-ion batteries （SIBs） with organic electrodes are an emerging research direction due to the sustainability of organic materials based on elements like C,H,O,and sodium ***,organic electrode materials for SIBs are mainly used as cathodes because of their relatively high redox potentials（>1 V）.Organic electrodes with low redox potential that can be used as anode are ***,a novel organic anode material （tetrasodium 1,4,5,8-naphthalenetetracarboxylate,Na4TDC） has been developed with low redox potential （<0.7 V） and excellent cyclic *** three-sodium storage mechanism was demonstrated with various in-situ/ex-situ spectroscopy and theoretical calculations,showing a high capacity of 208 mAh/g and an average decay rate of merely 0.022%per ***,the Na4TDC-hard carbon composite can further acquire improved capacity and cycling stability for 1200 cycles even with a high mass loading of up to 20 mg *** pairing with a thick Na3V2（PO4）3cathode (20.6 mg cm-2),the as-fabricated full cell exhibited high operating voltage （2.8 V）,excellent rate performance and cycling stability with a high capacity retention of 88.7% after 200 cycles,well highlighting the Na4TDC anode material for SIBs.

Fast three-dimensional radiation field characterization in largescale nuclear installations

The requirement of the fast three-dimensional radiation field calculation is raised during the decommissioning of large-scale nuclear installations. The most often used methods, such as the Monte Carlo and the discrete ordinates methods, have shortcomings in their simulations of such problems. The coupled Monte Carlo–point kernel computational scheme is developed to meet the requirement. The facility is separated into the source region and the bulk shielding region, with a common interface. The transport within the source region is simulated using the Monte Carlo method, which is by nature good at treating complex geometries. The radiation field in the bulk shielding region is treated by the point kernel approach to avoid the extremely expensive computation for deep penetration problems. The flow rate through the interface,which is given by the Monte Carlo simulation, is considered as the equivalent surface source for the point kernel calculation. Test calculations from simplified typical waste storage facilities have been performed to validate the coupled scheme by comparing them with the results conducted by the Monte Carlo method directly. The good agreement of the results, as well as the significant saving in computing time, indicates that the coupled method is suitable for the fast three-dimensional radiation field calculation.

Star formation associated with the infrared dust bubble N68

We investigated the environment of the infrared dust bubble N68 and searched for evidence of triggered star formation in its surroundings. We performed a multiwavelength study of the nebula with data taken from several large-scale surveys： GLIMPSE, MIPSGAL, IRAS, NVSS, GRS and JCMT. We analyzed the spectral pro- file and the distribution of the molecular gas （13CO J -- 1 - 0 and J -- 3 - 2）, and the dust in the environment of N68. The position-velocity diagram clearly shows that N68 may be expanding outward. We used two three-color images of the mid-infrared emis- sion to explore the physical environment, and one color-color diagram to investigate the distribution of young stellar objects （YSOs）. We found that the 24 p^m emission is surrounded by the 8.0 ~m emission. Morphologically, the 1.4 GHz continuum strongly correlates with the 24 gm emission, and the 13CO J -- 1 - 0 and J -- 3 - 2 emissions correlate well with the 8.0 p^m emission. We investigated two compact cores located in the shell of N68. The spectral intensity ratios of 13CO J -- 3 - 2 to J = 1 - 0 range from 5 to 0.3. In addition, YSOs, masers, IRAS and UC HII regions are distributed in the shell of the bubble. The active region may be triggered by the expansion of the bubble N68.

Radio Frequency Interference Mitigation and Statistics in the Spectral Observations of FAST

In radio astronomy,radio frequency interference(RFI)becomes more and more serious for radio observational *** RFI always influences the search and study of the interesting astronomical *** the RFI becomes an essential procedure in any survey data *** Five-hundred-meter Aperture Spherical radio Telescope(FAST)is an extremely sensitive radio *** is necessary to find out an effective and precise RFI mitigation method for FAST data *** this work,we introduce a method to mitigate the RFI in FAST spectral observation and make a statistic for the RFI using~300 h FAST *** details are as ***,according to the characteristics of FAST spectra,we propose to use the Asymmetrically Reweighted Penalized Least Squares algorithm for baseline *** test results show that it has a good ***,we flag the RFI with four strategies,which are to flag extremely strong RFI,flag long-lasting RFI,flag polarized RFI,and flag beam-combined RFI,*** test results show that all the RFI above a preset threshold could be ***,we make a statistic for the probabilities of polarized XX and YY RFI in FAST *** statistical results could tell us which frequencies are relatively *** such statistical data,we are able to avoid using such frequencies in our spectral ***,based on the~300 h FAST data,we obtained an RFI table,which is the most complete database currently for FAST.
