the production and secretion of antibodies by human plasma cells(PCs)are two essential processes of humoral *** secretion process relies on a group of proteins known as soluble N-ethylmaleimide-sensitive factor attachment protein receptors(SNAREs),which are located in the plasma membrane(t-SNAREs)and in the antibody-carrying vesicle membrane(v-SNARE),and mediate the fusion of both *** have previously shown that SNAP23 and StX4 are the t-SNAREs responsible for antibody ***,using human PCs and antibody-secreting cell lines,we studied and characterized the expression and subcellular distribution of vesicle associated membrane protein(VAMP)isoforms,demonstrating that all isoforms(with the exception of VAMP1)are expressed by the referenced ***,the functional role in antibody secretion of each expressed VAMP isoform was tested using *** results show that VAMP2 may be the v-SNARE involved in vesicular antibody *** further support this conclusion,we used tetanus toxin light chain to cleave VAMP2,conducted experiments to verify co-localization of VAMP2 in antibody-carrying vesicles,and demonstrated the coimmunoprecipitation of VAMP2 with StX4 and SNAP23 and the in situ interaction of VAMP2 with *** together,these findings implicate VAMP2 as the main VAMP isoform functionally involved in antibody secretion.

the service life of refractories in ladle walls is limited by corrosion,abrasion,thermal shock and structural spalling mechanisms. When the ladle is lined with bricks they typically need to be completely removed after a certain number of heats to be replaced by new bricks of the same size as the original bricks. Not so for monolithic ladle linings. At the end of their service life the remains of the castable can almost completely be recycled as they can in fact stay in place. Only the worn-out part of the lining has to be replaced by a new castable. 20%-50% of the initial ladle lining can be recycled ＂insitu＂. the installation can efficiently be done by shotcreting technics. But it requires a castable that resists slag penetration very well. Castables based on a calcium magnesium aluminate bond provide the required penetration resistance. Pumping and shotcreting is very well adapted for repairs of blast furnace shafts,torpedo cars,hot metal and steel ladle linings[1]. However,very little is published about how a good pumping and shotcreting performance can be achieved when the installation has to be done under extreme weather conditions. At high ambient temperature the challenge is to ensure a good pumping result without early castable stiffening. Blockage of the pump would be the consequence. At low ambient temperature the difficulty is to de-activate the highly efficient deflocculant fast enough with a gelling agent added intothe wet mix at the nozzle to prevent the gunned material slipping off the wall. this paper studies strategies how to achieve good installed properties even at extreme ambient temperatures. the interactions between deflocculants,retarders,gelling agents,and calcium magnesium aluminate binder as a function of temperature are studied for an alumina- spinel ladle castable. Effect of laser shock processing on erosive resistance of 6061-t6 aluminum

Application of laser shock processing （LSP） on 6061-t6 aluminum was made in order to evaluate its response to the erosive wear by silica sand. Impact angles of 15° , 30° , 60° and 90° were tested, two particle speeds （37 and 58 m/s） and two LSP irradiation conditions were used. Erosion marks were characterized by 3D profilometry and SEM analysis was conducted to identify the erosion mechanisms for each tested angle. the results showed a maximum erosive wear at low impact angles （ductile type behavior）. Erosion strength and the erosion mechanisms were not affected by the application of LSP and they were attributed to the high strain rate of the erosion phenomena. A few differences encountered on the erosion plots were explained on the basis of the surface roughness left by the LSP process. the maximum mass loss and the maximum erosion penetration happened in different impact angles （15° and 30° , respectively）. Finally, a well-defined erosion mechanism transition was observed, from cutting action at low impact angle, to crater formation at 90° of incidence.

Purpose A portable fast-neutron imaging system is being developed to provide complementary information to field X-ray *** include inspection of vehicles and infrastructure for corrosion,measurement of material levels in containers,and inspection of munitions and suspicious *** fast-neuron imaging generally provides lower imaging resolution compared to X-rays,fast-neutron interaction cross-sections have a weak dependence on material *** enables imaging of low-Z materials inside high-Z ***,we discuss the limitations and current improvements in fast-neuron *** Limitations in portable fast-neutron imaging systems include low D-t neutron generator output,low light pro-duction in ZnS(Cu)imaging scintillators,low resolution due to scintillator thickness and D-t spot size,and digital-panel darknoise that varies in time and position and that can be 100×larger than the neutron *** have made improvements in these areas through development of a segmented high light yield scintillator,panel noise mitigation techniques,and testing of new high-output,small spot size D-t neutron *** the segmented high light yield fast-neutron scintillator demonstrated 5×increase in light compared to ZnS(Cu).An additional 2×improvement in signal-to-noise was demonstrated with panel-noise mitigation *** MCNP calculations also show good agreement with neutron imaging results Conclusions We have demonstrated improvements in fast-neutron imaging through development of a segmented high light yield neutron scintillator,mitigation of digital panel noise,and preliminary testing with new high-output,small spot size D-t neutron *** have also demonstrated good results modeling fast-neutron images and scatter effects using MCNP.

BACKGROUND Oxidative damage of DNA and RNA has been associated with mortality of patients with different ***,there is no published data on the potential use of DNA and RNA oxidative damage to predict the prognosis of patients with hepatocellular carcinoma(HCC)undergoing liver transplantation(Lt).AIM to determine whether patients with increased DNA and RNA oxidative damage prior to Lt for HCC have a poor Lt *** Patients with HCC who underwent Lt were included in this observational and retrospective *** levels of all three oxidized guanine species(OGS)were measured prior to Lt since guanine is the nucleobase that forms DNA and RNA most prone to *** mortality at 1 year was the end-point *** Surviving patients(n=101)showed lower serum OGS levels(P=0.01)and lower age of the liver donor(P=0.03)than non-surviving patients(n=13).An association between serum OGS levels prior to Lt and 1-year Lt(odds ratio=2.079;95%confidence interval=1.356-3.189;P=0.001)was found in the logistic regression *** the main new finding was that high serum OGS concentration prior to Lt was associated with the mortality 1 year after Lt in HCC patients.

【目的】制备兔抗鼠三结构域蛋白59(tripartite motif-containing 59,tRIM59)多克隆抗体。【方法】利用PCR方法得到小鼠tRIM59基因片段,将其克隆至pGEX-2t原核表达载体中,转入大肠杆菌BL21,IPtG诱导表达GSt-tRIM59融合蛋白。GSt-tRIM59融合蛋白经亲和层析纯化浓缩后免疫新西兰大耳白兔,制备多克隆抗体,通过ELISA测定兔抗鼠tRIM59多克隆抗体效价,用Western blot测定其特异性。用免疫组织化学方法检测tRIM59在小鼠前列腺癌组织中的表达。【结果】成功构建了pGEX-2t-tRIM59原核表达重组质粒,并诱导表达出GSt-tRIM59融合蛋白,免疫新西兰大耳白兔5周后获得多克隆抗体。ELISA测定结果表明,tRIM59多克隆抗体效价为1∶106以上;Western blot分析表明,tRIM59多克隆抗体具有良好的特异性。免疫组织化学检测发现,tRIM59在小鼠前列腺癌组织细胞中高表达。【结论】成功地制备了特异性良好的兔抗鼠tRIM59多克隆抗体,其具有良好的免疫学活性,能够用于相关研究。
