Toughening additive manufactured Zr-based bulk metallic glass composites by martensite phase transformation

Additive manufacturing technology based on laser powder bed fusion(LPBF)offers a novel approach for fabricating bulk metallic glass(BMG)products without restriction in size and geometry.Nevertheless,the BMGs prepared by LPBF usually suffered from less plasticity and poorer fracture toughness as compared to their cast counterparts due to partial crystallization in heat-affected zones(HAZs).Since crystallization in HAZs is hard to avoid completely in LPBF BMGs,it is desirable to design a suitable alloy system,in which only ductile crystalline phase,instead of brittle intermetallics,is formed in HAZs.This unique structure could effectively increase the toughness/plasticity of the LPBF BMGs.To achieve this goal,a quaternary BMG system with a composition of Zr_(47.5)Cu_(45.5)Al_(5)Co_(2)is adopted and subjected to LPBF.It is found that nearly a single phase of B_(2)-ZrCu is precipitated in HAZs,while a fully amorphous phase is formed in molten pools(MPs).This B_(2)phase reinforced BMG composite exhibits excellent mechanical properties with enhanced plasticity and toughness.Furthermore,it is easy to modulate the mechanical properties by altering the amount of the B_(2)phase via adjusting the laser energy input.Finally,the best combination of strength,plasticity,and notch toughness is obtained in the BMG composite containing 27.4%B_(2)phase and 72.6%amorphous phase,which exhibits yield strength(σ_(s))of 1423 MPa,plastic strain(ε_(p))of 4.65%,and notch toughness(K_(q))of 53.9 MPa m 1/2.Furthermore,a notable strain-hardening is also observed.The improvement of plasticity/toughness and appearance of strain-hardening behavior are mainly due to the martensite phase transformation from the B_(2)phase to the Cm phase during plastic deformation(i.e.,the phase transformation-induced plasticity effect).The current work provides a guide for making advanced BMGs and BMG composites by additive manufacturing.

Predictive value of bilirubin and serum γ-glutamyltranspeptidase levels in type-2 diabetes mellitus patients with acute coronary syndrome

BACKGROUND Cardiovascular disease is a major complication of diabetes mellitus(DM).Type-2 DM(T2DM)is associated with an increased risk of cardiovascular events and mortality,while serum biomarkers may facilitate the prediction of these outcomes.Early differential diagnosis of T2DM complicated with acute coronary syndrome(ACS)plays an important role in controlling disease progression and improving safety.AIM To investigate the correlation of serum bilirubin andγ-glutamyltranspeptidase(γ-GGT)with major adverse cardiovascular events(MACEs)in T2DM patients with ACS.METHODS The clinical data of inpatients from January 2022 to December 2022 were analyzed retrospectively.According to different conditions,they were divided into the T2DM complicated with ACS group(T2DM+ACS,n=96),simple T2DM group(T2DM,n=85),and simple ACS group(ACS,n=90).The clinical data and laboratory indices were compared among the three groups,and the correlations of serum total bilirubin(TBIL)levels and serumγ-GGT levels with other indices were discussed.T2DM+ACS patients received a 90-day follow-up after discharge and were divided into event(n=15)and nonevent(n=81)groups according to the occurrence of MACEs;Univariate and multivariate analyses were further used to screen the independent influencing factors of MACEs in patients.RESULTS The T2DM+ACS group showed higherγ-GGT,total cholesterol,low-density lipoprotein cholesterol(LDL-C)and glycosylated hemoglobin(HbA1c)and lower TBIL and high-density lipoprotein cholesterol levels than the T2DM and ACS groups(P<0.05).Based on univariate analysis,the event and nonevent groups were significantly different in age(t=3.3612,P=0.0011),TBIL level(t=3.0742,P=0.0028),γ-GGT level(t=2.6887,P=0.0085),LDL-C level(t=2.0816,P=0.0401),HbA1c level(t=2.7862,P=0.0065)and left ventricular ejection fraction(LEVF)levels(t=3.2047,P=0.0018).Multivariate logistic regression analysis further identified that TBIL level and LEVF level were protective factor for MACEs,and age andγ-GGT level were risk factors(P<0.05).CONCLUSION Serum

Integrated two-phase free radical hydrogel:Safe,ultra-fast tooth whitening and antibacterial activity

The growing demand for brilliant smile and tooth whitening has led to the increasing significance of tooth whitening methods.However,the development of tooth whitening materials with high efficiency and safety has been considered to be an urgent challenge.Here,we report a novel tooth whitening strategy,which overcomes the limitation of conventional methods,shows excellent tooth whitening efficiency and safety,and even exhibits outstanding antibacterial activity.Inspired by stomata and chloroplasts in green leaves,we have developed an integrated two-phase free radical hydrogel(TP-GEL),which integrates solid-phase and liquid-phase free radical functional components fabricated by cold plasma technology.TP-GEL demonstrated safe and ultrafast tooth-whitening effect with or without visible light,which can be more significant than that of 30% H_(2)O_(2),the commonly whitening material in dental clinical practice.More importantly,we also avoid the damage to enamel,such as microhardness,roughness,surface morphology.Benefiting from free radicals,much higher antibacterial properties than commercial products(Chlorhexidine)was obtained.This research provides fundamental design criteria for the development of dual functional oral health care materials with advanced tooth whitening and antibacterial activity.

A novel shape memory alloy actuated soft gripper imitated hand behavior

The limited length shrinkage of shape memory alloy(SMA)wire seriously limits the motion range of SMA-based gripper.In this paper,a new soft finger without silicone gel was designed based on pre bent SMA wire,and the finger was back to its original shape by heating SMA wire,rather than relying only on heat exchange with the environment.Through imitating palm movement,a structure with adjustable spacing between fingers was made using SMA spring and rigid spring.The hook structure design at the fingertip can form self-locking to further improve the load capacity of gripper.Through the long thin rod model,the relationship of the initial pre bent angle on the bending angle and output force of the finger was analyzed.The stress-strain model of SMA spring was established for the selection of rigid spring.Three grasping modes were proposed to adapt to the weight of the objects.Through the test of the gripper,it was proved that the gripper had large bending amplitude,bending force,and response rate.The design provides a new idea for the lightweight design and convenient design of soft gripper based on SMA.

Chemotherapy remains the mainstay of cancer treatment,benefiting millions of patients each year,but the side effects of chemotherapy drugs severely limit their clinical use.Doxorubicin(DOX)can cause various side effects such as heart damage and treatment-related tumors.The effective use of active and passive targeting will improve the clinical application of DOX.Here,TPGS3350 and bioactive peptides were utilized to construct a micelle-based stage-by-stage impelled efficient system(missiles)for DOX delivery(DOX missiles).By taking advantage of the EPR effect,DOX missiles are efficiently enriched at the tumor site.After being cleaved by matrix metalloproteinase2(MMP2),the peptide(VRGD)targets tumor cells to facilitate uptake of the missiles by the tumor cells via receptor-mediated endocytosis.The intracellular activated caspase-3-catalyzed explosion of DOX missiles further enables efficient tumor killing.This study provides an efficient approach for DOX delivery and toxicity reduction.

A REF6-dependent H3K27me3-depleted state facilitates gene activation during germination in Arabidopsis

Seed germination is a critical developmental switch from a quiescent state to active growth,which involves extensive changes in metabolism,gene expression,and cellular identity.However,our understanding of epigenetic and transcriptional reprogramming during this process is limited.The histone H3 lysine 27 trimethylation(H3K27me3)plays a key role in regulating gene repression and cell fate specification.Here,we profile H3K27me3 dynamics and dissect the function of H3K27 demethylation during germination in Arabidopsis.Our temporal genome-wide profiling of H3K27me3 and transcription reveals delayed H3K27me3reprogramming compared with transcriptomic changes during germination,with H3K27me3 changes mainly occurring when the embryo is entering into vegetative development.RELATIVE OF EARLY FLOWERING 6(REF6)-mediated H3K27 demethylation is necessary for robust germination but does not significantly contribute to H3K27me3 dynamics during germination,but rather stably establishes an H3K27me3-depleted state that facilitates the activation of hormone-related and expansin-coding genes important for germination.We also show that the REF6 chromatin occupancy is gradually established during germination to counteract increased Polycomb repressive complex 2(PRC2).Our study provides key insights into the H3K27me3 dynamics during germination and suggests the function of H3K27me3 in facilitating cell fate switch.Furthermore,we reveal the importance of H3K27 demethylation-established transcriptional competence in gene activation during germination and likely other developmental processes.

T cells, including both CD4^(+) and CD8^(+)T cells, play a pivotal role in mediating various inflammation and immune disorders. A long-standing challenge in T cell-based immunotherapy is to precisely inactivate or delete the pathogenic T cells in inflammation and autoimmune diseases, or to selectively expand the immunocompetent T cell in tumor or other immune compromised situations, without inducing global immunosuppression or zealous immune activation respectively. To achieve this, a specific marker is needed to differentiate the pathogenic or immunocompetent T cell among the rests. Indeed,recent progress of immunology strongly suggests that CXC chemokine receptor 6(CXCR6, CD186) is such a kind of marker. Here, we review the emerging role of CXCR6 as a novel target for immunotherapy and discuss the underlying mechanism. We propose that CXCR6-based immunotherapy will play a significant role in autoimmune, nonalcoholic steatohepatitis(NASH), tumor, coronavirus disease 2019(COVID-19) and even ageing-related inflammatory infliction.

Targeting protein phosphatases for the treatment of inflammation-related diseases:From signaling to therapy

Inflammation is the common pathological basis of autoimmune diseases,metabolic diseases,malignant tumors,and other major chronic diseases.Inflammation plays an important role in tissue homeostasis.On one hand,inflammation can sense changes in the tissue environment,induce imbalance of tissue homeostasis,and cause tissue damage.On the other hand,inflammation can also initiate tissue damage repair and maintain normal tissue function by resolving injury and restoring homeostasis.These opposing functions emphasize the significance of accurate regulation of inflammatory homeostasis to ameliorate inflammation-related diseases.Potential mechanisms involve protein phosphorylation modifications by kinases and phosphatases,which have a crucial role in inflammatory homeostasis.The mechanisms by which many kinases resolve inflammation have been well reviewed,whereas a systematic summary of the functions of protein phosphatases in regulating inflammatory homeostasis is lacking.The molecular knowledge of protein phosphatases,and especially the unique biochemical traits of each family member,will be of critical importance for developing drugs that target phosphatases.Here,we provide a comprehensive summary of the structure,the“double-edged sword”function,and the extensive signaling pathways of all protein phosphatases in inflammation-related diseases,as well as their potential inhibitors or activators that can be used in therapeutic interventions in preclinical or clinical trials.We provide an integrated perspective on the current understanding of all the protein phosphatases associated with inflammationrelated diseases,with the aim of facilitating the development of drugs that target protein phosphatases for the treatment of inflammation-related diseases.
