LncRNA HIF1A-AS2 accelerates malignant phenotypes of renal carcinoma by modulating miR-30a-5p/SOX4 axis as a ceRNA

Objective:Several reports have proposed that lnc RNAs,as potential biomarkers,participate in the progression and growth of malignant ***1 A-AS2 is a novel lnc RNA and potential biomarker,involved in the genesis and development of ***,the molecular mechanism of HIF1 A-AS2 in renal carcinoma is ***:The relative expression levels of HIF1 A-AS2 and miR-30 a-5 p were detected using RT-qPCR in renal carcinoma tissues and cell *** loss-of-function and overexpression,the biological effects of HIF1 A-AS2 and miR-30 a-5 p in kidney carcinoma progression were *** luciferase reporter gene analysis and Western blot were used to detect the potential mechanism of HIF1 A-AS2 in renal ***:HIF1 A-AS2 was upregulated in kidney carcinoma tissues when compared with para-carcinoma tissues(P<0.05).In addition,tumor size,tumor node mestastasis stage and differentiation were identified as being closely associated with HIF1 A-AS2 expression(P<0.05).Knockdown or overexpression of HIF1 A-AS2 either restrained or promoted the malignant phenotype and WNT/β-catenin signaling in renal carcinoma cells(P<0.05).Mi R-30 a-5 p was downregulated in renal cancers and partially reversed HIF1 A-AS2 functions in malignant renal tumor ***1 A-AS2 acted as a micro RNA sponge that actively regulated the relative expression of SOX4 in sponging miR-30 a-5 p and subsequently increased the malignant phenotypes of renal ***1 A-AS2 showed a carcinogenic effect and miR-30 a-5 p acted as an antagonist of the anti-oncogene effects in the pathogenesis of renal ***:The HIF1 A-AS2-miR-30 a-5 p-SOX4 axis was associated with the malignant progression and development of renal *** relative expression of HIF1 A-AS2 was negatively correlated with the expression of miR-30 a-5 p,and was closely correlated with SOX4 mRNA levels in renal cancers.

Recent studies suggested that LIM and SH3 protein 1(LASP-1)is a promising therapeutic target for renal cell cancer(RCC).This study aimed to explore the role of LASP-1 in *** this purpose,LASP-1 expression in RCC tissues was analyzed by immunohistochemistry and Western blot *** proliferation,migration,invasion,and gene expression were detected by CCK-8 assay,Transwell assay,and Western blot *** results showed that LASP-1 was highly expressed in RCC,and its expression level,t was positively correlated with lymph node metastasis and tumor,nodes,and metastases(TNM)*** knockdown of LASP-1 expression significantly inhibited the proliferation of RCC cells,increased the apoptosis rate,and inhibited RCC cell invasion and migration by inhibiting epithelial–mesenchymal *** conclude that LASP-1 promotes RCC progression and metastasis and is a promising therapeutic target for RCC.

Background Rat renal transplantation is an essential experimental model and requires greater microsurgical ***,training novices to perform quick and reliable microvascular anastomosis is of vital importance for rat renal *** this study,we developed and evaluated a staged microvascular anastomosis training program for novices,harvesting and transplanting both kidneys from one rat *** Five trainees without any prior microsurgical experience underwent a training program in which the goals were staged according to *** trainee had to achieve satisfactory results as evaluated by a mentor before entering the next *** renal transplantation was accomplished by end-to-end technique with a bladder *** the intensive rat renal transplantation stage,the trainees required an average of 20 independent attempts at isotransplantation as final training *** After 2 months of intensive practice,all trainees had achieved stable and reproducible rat renal transplantation,with a satisfactory survival rate of 85.9% at postoperative Day *** total mean operative time was 78.0 minutes and the mean hot ischemia time was 26.2 *** experience increasing,the operative time for each trainee showed a decreasing trend,from 90-100 minutes to 60-70 *** 20 cases,the mean operative time of the trainees was not statistically significantly different from that of the *** Harvesting and transplanting both kidneys from one rat donor after a staged microvascular anastomosis training program is feasible for novices without any prior microsurgical skills.

Myeloid-derived suppressor cells(MDSC) potently inhibit antitumor immune responses, and thereby promoting tumor progression and metastasis. However, the nature of human tumor-infiltrating MDSC remains poorly characterized. Here, we found that B7-H3 was exclusively expressed on a subset of intratumoral CD14+HLA-DR-/low MDSC but not on those from adjacent normal lung tissues of the cancer patients. Cytokine analysis revealed that B7-H3+CD14+HLA-DR-/low MDSC(B7-H3+MDSC) produced higher levels of IL-10 and TNF-α but lower levels of the IL-1β and IL-6 when compared with B7-H3-CD14+HLA-DR-/low MDSC(B7-H3-MDSC). In a murine lung cancer model, B7-H3+MDSC were found only in the tumor microenvironment and their frequencies increased during tumor progression. Clinical data analysis indicated that higher frequencies of B7-H3+MDSC were associated with reduced recurrence-free survival in patients with NSCLC. Taken together, we identify a novel subset of MDSC within the tumor microenvironment that leads to tumor progression.

Establishment of orthotopic transplantation model of human bladder cancer and detection by MRI

Objective: To establish an orthotopic bladder cancer model bearing human bladder cancer for experimental research, and monitor tumor progression by magnetic resonance imaging (MRI). Methods: The mucosa was mechanically damaged transurethrally under direct vision, and then human bladder cancer cell line T24 was inoculated into the bladders of BALB/c nude mice to establish orthotopic bladder cancer model. To find a suitable concentration of Gd-DTPA for this re- search. MRI was performed weekly to assess tumor growth, using Gd-DTPA as contrast agent. The pathologic morphology of the bladders and other specimens were observed with HE stain. Results: All the 25 mice developed bladder cancer after inoculation. The best concentration of Gd-DTPA was 1.408 mg/mL. On MRI, no change in the bladders was observed on day 7 after inoculation, filling defect in the bladders, accordant to actual tumor size, was detected on days 14, 21 and 28. Pathologic examination showed that tumor grew in the mucosa or superficial muscle of bladder on day 7, confined in muscle layer on days 14–28, and invaded serosa on day 35. Conclusion: Transurethrally damaged bladder mucosa under direct vision and instilled bladder cancer cell T24, we successfully established an orthotopic bladder cancer model. Tumor growth simulated the progression of human bladder cancer approximately. MRI was a reliable way for dynamic detection of murine orthotopic bladder tumor.

Objective:The aim of the study was to investigate the expressions of matrix metalloproteinase-2(MMP-2) and tissue inhibitor of MMP-2(TIMP-2) mRNA in transitional cell carcinomas of bladder and discuss their clinical ***:Using RT-PCR and real time quantitative PCR(RQ-PCR) technique,the expressions of MMP-2 and TIMP-2 mRNA of 45 cases of bladder carcinoma(tumor group) and 10 cases of normal bladder tissue(control group) were ***:MMP-2 and TIMP-2 were not expressed in control ***-2 was expressed in 30 cases tumor samples and TIMP-2 was expressed in 26 *** expression of MMP-2 mRNA in non-muscle invasive bladder cancers and grade I cancers was lower than that in muscle invasive bladder cancers and grades II-III cancers *** differences were statistically significant(P0.05).Conclusion:These results suggest that MMP-2 and TIMP-2 play an important role in the invasion step of transitional cell carcinoma of ***-2 may become a new approach to the diagnosis of bladder carcinoma.

Background The Cylex Immuknow(IK) assay provides a rapid and quantitative assessment of T-cell-mediated immune function. 目的比较经皮肾镜气压弹道碎石(PCNL)、体外震波碎石(ESWL)、开放手术治疗复杂性肾结石的疗效。方法132例复杂性肾结石患者,分别采用PCNL43例,ESWL47例,开放手术42例。治疗后应用腹部平片评估其疗效,了解结石排净率,观察并记录不良反映和并发症。随访3个月。结果开放手术组结石清除率为92%,残留率8%,并发症26例;PCNL组结石清除率为89%,残留率11%,并发症10例;ESWL组结石清除率为87%,残留率13%,并发症12例,均无严重并发症。结论PCNL治疗复杂性肾结石优于ESWL和开放手术,可作为首选;ESWL治疗复杂性肾结石是较为安全、有效的治疗方法,但结石残留率较高,多次震波碎石易致肾脏并发症;开放手术可作为适当的补充,但创伤大,住院时间长。
