Data driven analysis reveals prognostic genes and immunological targets in human sepsis-associated acute kidney injury

BACKGROUND:The molecular mechanism of sepsis-associated acute kidney injury(SA-AKI)is unclear.We analyzed co-differentially expressed genes(co-DEGs)to elucidate the underlying mechanism and intervention targets of SA-AKI.METHODS:The microarray datasets GSE65682,GSE30718,and GSE174220 were downloaded from the Gene Expression Omnibus(GEO)database.We identified the co-DEGs and constructed a gene co-expression network to screen the hub genes.We analyzed immune correlations and disease correlations and performed functional annotation of the hub genes.We also performed single-cell and microenvironment analyses and investigated the enrichment pathways and the main transcription factors.Finally,we conducted a correlation analysis to evaluate the role of the hub genes.RESULTS:Interleukin 32(IL32)was identified as the hub gene in SA-AKI,and the main enriched signaling pathways were associated with hemopoiesis,cellular response to cytokine stimulus,inflammatory response,and regulation of kidney development.Additionally,IL32 was significantly associated with mortality in SA-AKI patients.Monocytes,macrophages,T cells,and NK cells were closely related to IL32 and were involved in the immune microenvironment in SA-AKI patients.IL32 expression increased significantly in the kidney of septic mouse.Toll-like receptor 2(TLR2)was significantly and negatively correlated with IL32.CONCLUSION:IL32 is the key gene involved in SA-AKI and is significantly associated with prognosis.TLR2 and relevant immune cells are closely related to key genes.

A small organic molecule strategy for remedying oxygen vacancies by bismuth defects in BiOBr nanosheet with excellent photocatalytic CO_(2)reduction

Defect modulation currently plays a decisive role in addressing the poor photoabsorption,sluggish electron hole separation,and high CO_(2)activation barrier in photocatalytic CO_(2)reduction.However,hunting for a straightforward strategy to balance the concentration of oxygen vacancy and metal cation defect in one photocatalyst is still a great challenge.Herein,a bismuth vacancies BiOBr nanosheets(BiOBr-1)on the exposed[001]facets were constructed via an acetic acid molecule modification strategy,which can repair oxygen defect by bismuth vacancy in low-temperature solid-state chemical method.Benefiting from the formed bismuth defects that can not only broaden light absorption and elevate charge separation efficiency,but also enhance adsorption and activation of CO_(2)molecules,the evolution rates of photocatalytic CO_(2)conversion into CO(71.23μmol·g^(-1)·h^(-1))and CH4(8.90μmol·g^(-1)·h^(-1))attained by BiOBr-1 are superior 7.1 and 11 times to that of plate-like BiOBr.The photocatalytic mechanisms including adsorption concentration and activation process of CO_(2)are further revealed by the in situ diffuse reflectance infrared flourier transform spectra(DRIFTS).This finding of the existence of distinct defects in ultrathin nanosheets undoubtedly leads to new possibilities for photocatalyst design using two-dimensional materials with high solar-driven photocatalytic activity.

The poor reversibility of Zn anodes induced by dendrite growth,surface passivation,and corrosion,severely hinders the practical applicability of Zn metal batteries.To address these issues,a plasmaassisted aerogel（PAG） interface engineering was proposed as efficient ion transport modulator that can simultaneously regulate uniform Zn2+flux and desolvation behavior during battery operation.The PAG with ordered mesopores acted as an ion sieve to homogenize Zn deposition and accelerate Zn2+flux,which is favorable for corrosion resistance and dendrite suppression.Importantly,the plasma-assisted aerogel with abundant hydrophilic groups can facilitate the desolvation kinetics of Zn2+due to the multiple hydrogen-bonding interaction with the activated water molecules,thus accelerating the Zn2+migration kinetics.Consequently,the Zn/Zn cell assembled with PAG-modified separator demonstrates stable plating and stripping behavior(over 1400 h at 1 mA cm-2) and high Coulombic efficiency(99.8% at1 mA cm-2after 1100 cycles),and the Zn‖MnO2full cell shows excellent long-term cycling stability and maintains a high capacity of 154.9 mA h g-1after 1000 cycles at 1 A g-1.This study provides a feasible approach for the large-scale fabrication of aerogel functionalized separators to realize ultra-stable Zn metal batteries.

Interval-censored failure time data arise frequently in periodical follow-up studies including clinical trials and epidemiological surveys. In addition, some covariates may be subject to measurement errors due to the instrumental contamination, biological variation or other reasons. For the analysis of interval-censored data with mis-measured covariates, the existing methods either assume a parametric model or rely on the availability of replicated surrogate measurements for the error-prone covariate, which both have obvious limitations. To overcome these shortcomings, the authors propose a simulation-extrapolation estimation procedure under a general class of transformation models. The resulting estimators are shown to be consistent and asymptotically normal. The numerical results obtained from a simulation study indicate that the proposed method performs reasonably well in practice.In particular, the proposed method can reduce the estimation bias given by the naive method that does not take measurement errors into account. Finally, the proposed method is applied to a real data set on hypobaric decompression sickness.

The glucose metabolism is crucial for sustained brain activity as it provides energy and is a carbon source for multiple biomacromolecules;glucose metabolism decreases dramatically in Alzheimer’s disease(AD)and may be a fundamental cause for its development.Recent studies reveal that the alternative splicing events of certain genes effectively regulate several processes in glucose metabolism including insulin receptor,insulin-degrading enzyme,pyruvate kinase M,receptor for advanced glycation endproducts,and others,thereby,influencing glucose uptake,glycolysis,and advanced glycation end-products-mediated signaling pathways.Indeed,the discovery of aberrant alternative splicing that changes the proteomic diversity and protein activity in glucose metabolism has been pivotal in our understanding of AD development.In this review,we summarize the alternative splicing events of the glucose metabolism-related genes in AD pathology and highlight the crucial regulatory roles of splicing factors in the alternative splicing process.We also discuss the emerging therapeutic approaches for targeting splicing factors for AD treatment.

Background:Inhibitor of NF-κB kinase-interacting protein(IKIP)is known to promote proliferation of glioblastoma(GBM)cells,but how it affects migration and invasion by those cells is unclear.Methods:We compared levels of IKIP between glioma tissues and normal brain tissue in clinical samples and public databases.We examined the effects of IKIP overexpression and knockdown on the migration and invasion of GBM using transwell and wound healing assays,and we compared the transcriptomes under these different conditions to identify the molecular mechanisms involved.Results:Based on data from our clinical samples and from public databases,IKIP was overexpressed in GBM tumors,and its expression level correlated inversely with survival.IKIP overexpression in GBM cells inhibited migration and invasion in transwell and wound healing assays,whereas IKIP knockdown exerted the opposite effects.IKIP overexpression in GBM cells that were injected into mouse brain promoted tumor growth but inhibited tumor invasion of surrounding tissue.The effects of IKIP were associated with downregulation of THBS1 mRNA and concomitant inhibition of THBS1/FAK signaling.Conclusions:IKIP inhibits THBS1/FAK signaling to suppress migration and invasion of GBM cells.

Plant viruses are a group of intracellular pathogens that persistently threaten global food security.Significant advances in plant virology have been achieved by Chinese scientists over the last 20 years,including basic research and technologies for preventing and controlling plant viral diseases.Here,we review these milestones and advances,including the identification of new crop-infecting viruses,dissection of pathogenic mechanisms of multiple viruses,examination of multilayered interactions among viruses,their host plants,and virus-transmitting arthropod vectors,and in-depth interrogation of plantencoded resistance and susceptibility determinants.Notably,various plant virus-based vectors have also been successfully developed for gene function studies and target gene expression in plants.We also recommend future plant virology studies in China.

Competing endogenous RNAs(ce RNAs)containing micro RNA response elements can competitively interact with micro RNA via mi RNA response elements,which can combine non-coding RNAs with protein-coding RNAs through complex ce RNA networks.Ce RNAs include non-coding RNAs(long non-coding RNAs,circular RNAs,and transcribed pseudogenes)and protein-coding RNAs(m RNAs).Molecular interactions in ce RNA networks can coordinate many biological processes;however,they may also lead to ce RNA network imbalance and thus contribute to cancer occurrence when disturbed.Recent studies indicate that many dysregulated RNAs derived from lung cancer may function as ce RNAs to regulate multitudinous biological functions for lung cancer,including tumor cell proliferation,apoptosis,growth,invasion,migration,and metastasis.This study therefore reviewed the research progress in the field of non-coding and protein-coding RNAs as ce RNAs in lung cancer,and highlighted validated ce RNAs involved in biological lung cancer functions.Furthermore,the roles of ce RNAs as novel prognostic and diagnostic biomarkers were also discussed.Interpreting the involvement of ce RNAs networks in lung cancer will provide new insight into cancer pathogenesis and treatment strategies.

OBJECTIVE: To study the anticancer mechanism of polyphyllin I (PPI), a Traditional Chinese Medicine, on the ovarian cancer cell line HO-8910PM invitro. METHODS: Transwell chamber invasive assays were used to investigate the inhibitory capacity of PPI on HO-8910PM metastasis. Gene expression profiling chips was used to screen differentially expressed genes between experiment group and control group. Reverse transcription PCR and Western blotting were used to determine mRNA and protein levels. RESULTS: With increasing PPI concentration, the metastatic capacity of cells decreased, with significance differences between the experimental and control groups (P<0.01) as well as between two concentration groups. Gene expression profiling identified 123 differentially expressed genes, of which 70 were downregulated and 53 were upregulated. The genes were involved in multiple signal transduction pathways, including apoptosis, proliferation and metastasis. Real-time PCR (RT-PCR) showed that differential genes PIK3C2B, Caspase 9and Wnt5A were downregulated with increasing PPI, showing an evident dose-effect relationship. The c-Jun was an exception. As the PPI dosage increased and the exposure time was extended, c-Jun relative expression showed an upward trend. There were significant differences between the experiment and control (P<0.05). Western blot analyses showed that PPI treatment decreased levels of Caspase 9, Wnt5A and PIK3C2B and increased activatedCaspase9,c-Junandp-c-Junexpressionlevels. CONCLUSION: PPI has strong antitumor and anti transfer activity. It can activate c-Jun expression and the JNK signaling pathway, elicit cell apoptosis via the mitochondrial-mediated Caspase activation pathway, and finally inhibit tumor growth and mi- gration in vitro. The downregulation of PIK3C2B and Wnt5A jointly inhibit the proliferation and metastasis of HO-8910PM. PPI may be a novel treatment for ovarian cancer.
