Bermudagrass （Cynodon dactylon L. cv. Goldensun） is highly resistant to and accumulates large amounts of cadmium （Cd）. A phytochelatin synthase （PCS） cDNA （CdPCS1） was isolated from this grass by rapid amplification of cDNA ends. The putative CdPCS1 protein shared a high homology with PCS from other plants, with 79% homology at the N-terminal and 47% homology at the C-terminah However, 16 Cys residues were found at the C-terminal of CdPCS1, and among these residues, three positions were different from other PCS proteins. Semiquantitative reverse transcription-polymerase chain reaction analysis showed that Cd stress induced CdPCS1 expression in both roots and leaves in Bermudagrass. We verified that CdPCS1 plays an important role in Cd tolerance in yeast cells by expressing the gene in ABDE1, a Cdsensitive mutant. CdPCS1 was then introduced into tobacco plants. The phytochelatin level in some transgenic tobacco lines increased 3.88-fold more than in wild type plants and Cd accumulation in these transgenic plants was enhanced 3.21-fold accordingly. The results suggested that CdPCS1 could be used as a gene element for phytoremediation in the future.

为探究月季(Rosahybrida)花瓣衰老机制以及为通过分子育种等手段创制抗衰老品种提供基因储备,以切花月季‘卡罗拉’(Carola)为试材,通过月季转录组测序筛选到1个E3泛素连接酶RING结构域家族基因RhRNF185-like。qRT-PCR结果表明,在花朵开放的中后期3~5级的花瓣中RhRNF185-like的表达量显著增强,说明其可能影响了月季花瓣的衰老。将其在烟草叶肉细胞进行瞬时过表达,通过激光共聚焦扫描显微镜观察到Rh RNF185-like的GFP信号与DAPI核信号重合,说明其定位在细胞核上。利用病毒诱导的基因沉默技术将RhRNF185-like在月季花瓣中沉默,发现沉默的花瓣圆片衰老程度明显大于对照。此外,与对照相比,RhRNF185-like沉默的花瓣圆片中衰老标志基因RhSAG12的相对表达量、离子渗透率和褪色率显著升高,均说明沉默RhRNF185-like促进了月季花瓣的衰老。以上结果表明,RhRNF185-like以拮抗调节的方式影响了月季花瓣衰老。

Nucleolar GTP-binding protein 1(NOG1) is a highly conserved GTPase first reported in Trypanosoma as required for ribosome biogenesis. We characterized NbNOG1, a Nicotiana benthamiana NOG1ortholog sharing more than 45% amino acid identity with Trypanosoma,yeast, and human NOG1. N. benthamiana plants silenced for NbNOG1were stunted and produced sterile ***1is functionally interchangeable with yeast NOG1(ScNOG1), rescuing yeast lethality caused by loss of ScNOG1. Finally, NbNOG1silencing caused overaccumulation of pre-rRNA processing intermediates,and concomitant loss of mature rRNAs. Collectively,these data support a role for NbNOG1in ribosomal RNA processing.

Weather forecasting for the Zhangjiakou competition zone of the Beijing 2022 Winter Olympic Games is a challenging task due to its complex *** weather prediction models generally perform poorly for cold air pools and winds over complex terrains,due to their low spatiotemporal resolution and limitations in the description of dynamics,thermodynamics,and microphysics in mountainous *** study proposes an ensemble-learning model,named ENSL,for surface temperature and wind forecasts at the venues of the Zhangjiakou competition zone,by integrating five individual models—linear regression,random forest,gradient boosting decision tree,support vector machine,and artificial neural network(ANN),with a ridge regression as meta *** ENSL employs predictors from the high-resolution ECMWF model forecast(ECMWF-HRES) data and topography data,and targets from automatic weather station *** categories of predictors(synoptic-pattern related fields,surface element fields,terrain,and temporal features) are fed into *** results demonstrate that ENSL achieves better performance and generalization than individual *** root-mean-square error(RMSE) for the temperature and wind speed predictions is reduced by 48.2% and 28.5%,respectively,relative to *** the gust speed,the performance of ENSL is consistent with ANN(best individual model) in the whole dataset,whereas ENSL outperforms on extreme gust samples(42.7% compared with 38.7% obtained by ECMWF-HRES in terms of RMSE reduction).Sensitivity analysis of predictors in the four categories shows that ENSL fits their feature importance rankings and physical explanations effectively.

EjGASA6 promotes flowering and root elongation by enhancing gibberellin biosynthesis

Potato（Solanum tuberosum L.） is the fourth most important crop in the world and one of the top seven important crops in china. Late blight is a devastating disease of potato caused by Phytophthora infestans（Mont.） de Bary, which was the reason of the Great Irish Famine in 1845. Today, late blight remains the No. 1 threat to potato production. In previous research, it proved that it was difficult to control potato late blight just because P. infestans ranked the highest risk in the evolutionary potential and it could easily mutate its Avr genes and thus overcome potato plants carrying corresponding R genes. So, one of the promising solutions for late blight control is to release several R genes simultaneously and to artificially create a resistance polymorphism in the field, such as the strategy so called R gene polyculture. In the past years, scientists gained a great success in cloning R genes conferring late blight resistance, providing a starting point to create R gene polyculture by means of cisgenic technology.

The Gibberellic Acid-stimulated Arabidopsis(GASA)gene family is involved in the regulation of gene expression and plant growth,development,and stress *** investigate the function of loquat GASA genes in the growth and developmental regulation of plants,a loquat EjGASA6 gene homologous to Arabidopsis AtGASA6 was ***6 expression was induced by gibberellin,and ectopic transgenic plants containing this gene exhibited earlier bloom and longer primary roots since these phenotypic characteristics are related to higher gibberellin *** analysis and qRT-PCR results showed that the expression levels of GA3ox1 and GA20ox1,which encode key enzymes in gibberellin biosynthesis,were significantly ***,we confirmed that EjGASA6 could promote the expression of GA20ox1 via the luciferase reporter ***,our results suggest that EjGASA6 promotes blooming and main-root elongation by positively regulating gibberellin *** findings broaden our understanding of the role of GASAs in plant development and growth,and lay the groundwork for future research into the functions of EjGASA6 in regulating loquat growth and development.

Understanding the response of the Earth system to varying concentrations of carbon dioxide(CO_(2))is critical for projecting possible future climate change and for providing insight into mitigation and adaptation strategies in the near *** this study,we generate a dataset by conducting an experiment involving carbon dioxide removal(CDR)—a potential way to suppress global warming—using the Chinese Academy of Sciences Earth System Model version 2.0(CASESM2.0).A preliminary evaluation is *** model is integrated from 200–340 years as a 1%yr^(−1) CO_(2) concentration increase experiment,and then to~478 years as a carbon dioxide removal experiment until CO_(2) returns to its original ***,another 80 years is integrated in which CO_(2) is kept *** in the 2-m temperature,precipitation,sea surface temperature,ocean temperature,Atlantic meridional overturning circulation(AMOC),and sea surface height are all *** the ramp-up period,the global mean 2-m temperature and precipitation both increase while the AMOC *** of all the above variables change in the opposite direction in the ramp-down period,with a delayed peak relative to the CO_(2) *** CO_(2) returns to its original value,the global mean 2-m temperature is still~1 K higher than in the original state,and precipitation is~0.07 mm d^(–1) *** the end of the simulation,there is a~0.5°C increase in ocean temperature and a 1 Sv weakening of the *** model simulation produces similar results to those of comparable experiments previously reported in the literature.

谷氧还蛋白(glutaredoxin,GRX)是一类小分子氧化还原酶,在植物生长发育调控及逆境胁迫中起着重要调节作用。本研究拟对玉米MS22基因的生物信息及玉米中GRX基因家族的亲缘进化进行分析研究。通过对MS22基因的生物信息分析,发现MS22基因全长881 bp,编码159个氨基酸,属于CC型谷氧还蛋白。玉米基因组中共鉴定出22个GRX类基因,其中有6个基因为GRX-subⅠ,7个基因属于GRX-subⅡ,9个基因属于GRX-subⅢ。通过聚类分析发现MS22属于GRX-subⅢ类型,该蛋白与不同来源CC型谷氧还蛋白的氨基酸序列保守性较高,一致率介于61%~87%之间。通过该研究对玉米中该类型基因的进一步研究具有较好的理论指导意义。
