Considering the uncertain and stochastic of intermittent distributed generations（DGS） in active distribution network（ADN）, a scenario method using Wasserstein distance metric and K-means cluster scenes reduction technology to generate optimal scene is proposed in this paper. So the stochastic problem is transformed into a deterministic problem. The multi-scenario tree models of wind-photovoltaic-load are built. A multi-objective optimization mathematical model of active distribution network containing intermittent DGS is established, which includes objectives that are the annual profits, the total active power losses and the voltage deviations of the bus, and considering active management characteristic. The tabu search artificial bee colony algorithm is used to solve the optimization problems. The simulation results show that the optimal scenes based on the Wasserstein distance indicators and K-means cluster technique reflect the random feature of distributed generation active power output more accurately. Finally, the simulation analysis of IEEE33-bus distribution test system is carried out to verify the effectiveness and feasibility of the proposed method.

Volatile organic compounds(VOCs)play key roles in plant–plant communication,especially in response to pest attack.E-2-hexenal is an important component of VOCs,but it is unclear whether it can induce endog-enous plant resistance to ***,we show that E-2-hexenal activates early signaling events in Ara-bidopsis(Arabidopsis thaliana)mesophyll cells,including an H2O2 burst at the plasma membrane,the directedflow of calcium ions,and an increase in cytosolic calcium *** of wild-type Arabidopsis plants with E-2-hexenal increases their resistance when challenged with the diamond-back moth Plutella xylostella L.,and this phenomenon is largely lost in the wrky46 ***,E-2-hexenal induces the expression of WRKY46 and MYC2,and the physical interaction of their encoded proteins was verified by yeast two-hybrid,firefly luciferase complementation imaging,and in vitro pull-down *** WRKY46–MYC2 complex directly binds to the promoter of RBOHD to promote its expres-sion,as demonstrated by luciferase reporter,yeast one-hybrid,chromatin immunoprecipitation,and electrophoretic mobility shift *** module also positively regulates the expression of E-2-hexenal-induced naringenin biosynthesis genes(TT4 and CHIL)and the accumulation of totalflavonoids,thereby modulating plant tolerance to ***,our results highlight an important role for the WRKY46–MYC2 module in the E-2-hexenal-induced defense response of Arabidopsis,providing new in-sights into the mechanisms by which VOCs trigger plant defense responses.

番茄愈伤组织的诱导与遗传转化体系的建立

番茄是植物研究领域常用的模式植物之一,番茄高效稳定的遗传转化体系是研究番茄基因生物学功能的基础,但常规番茄遗传转化耗时过长。以番茄愈伤组织为侵染对象,最终获得转基因番茄愈伤组织,可极大地缩短获得番茄转基因材料的时间。本文以番茄Micro-Tom(Solanum lycopersicum cv. Micro-Tom)为实验材料,探索番茄愈伤诱导条件,建立标准的番茄愈伤组织遗传转化体系。结果表明,胚根为外植体、激素配比2,4-二氯苯氧乙酸(2,4-Dichlorophenoxyacetic acid, 2,4-D)1 mg/L+6-苄氨基腺嘌呤(6-Benzylaminopurine, 6-BA)1 mg/L、蔗糖浓度30 g/L、光照强度3 000 lx为最佳诱导条件。标准番茄愈伤遗传转化体系包括愈伤组织培养、悬浮细胞建立、农杆菌侵染、共培养、抗性愈伤筛选、核酸鉴定等。该实验结果能够为以番茄为研究对象的分子机理研究及其所涉及的免疫沉淀、高通量测序、亚细胞定位等实验提供科研实验材料和方法参考。

Abstract Vegetable soybean and cowpea are related warm-season legumes showing contrasting leaf water use behaviors under similar root drought stresses,whose mechanisms are not well *** we conducted an integrative phenomic-transcriptomic study on the two crops grown in a feedback irrigation system that enabled precise control of soil water *** transpiration rate monitoring demonstrated that cowpea used water more conservatively under earlier soil drought stages,but tended to maintain higher transpiration under prolonged ***,we observed a soybean-specific transpiration rate increase accompanied by phase shift under moderate soil ***-series transcriptomic analysis suggested a dehydration avoidance mechanism of cowpea at early soil drought stage,in which the VuHAI3 and VuTIP2;3 genes were suggested to be *** gene clustering analysis revealed different responsiveness of genes to drought,time of day and their interactions between the two crops,which involved species-dependent regulation of the circadian clock *** network analysis identified two co-expression modules each associated with transpiration rate in cowpea and soybean,including a pair of negatively correlated modules between *** hub genes,including the ABA-degrading gene GmCYP707A4 and the trehalose-phosphatase/synthase gene VuTPS9 were *** network analysis revealed putative co-players of the hub *** analyses verified the role of VuTPS9 in regulating transpiration rate under osmotic *** findings propose that species-specific transcriptomic reprograming in leaves of the two crops suffering similar soil drought was not only a result of the different drought resistance level,but a cause of it.

顺式还原酮加双氧酶(acireductone dioxygenase, ARD)是Cupins蛋白家族的一员,在原核生物和真核生物中广泛存在,主要参与甲硫氨酸代谢途径的催化;另外,植物中甲硫氨酸代谢途径与乙烯和多胺等的合成密切相关,且乙烯和多胺参与了植物生长发育和逆境胁迫响应等多种生物学过程。植物中关于ARD的研究报道有限,尤其是园艺作物。番茄在人们的日常饮食结构中占有重要比例,也是植物研究领域常用的模式植物之一。本研究以番茄(Solanum lycopersicum)为研究对象,鉴定番茄基因组中包含的SlARD成员,并利用生物信息学分析手段、定量PCR技术和亚细胞定位等实验,对番茄SlARD基因家族进行分析及功能鉴定。结果表明,番茄基因组中共包含2个SlARD成员,分别命名为SlARD1和SlARD2,二者均定位于番茄第9号染色体,与拟南芥(Arabidopsis thalina)AtARD家族成员存在一定的系统发育和进化关系;SlARD1/2均只包含1个ARD结构域;番茄SlARD1/2在根、茎、叶中均有表达,能够响应水涝胁迫,定位于细胞质和细胞核。本研究还对结合SlARD1和SlARD2启动子的转录因子和SlARD1/2的互作蛋白网络进行了预测。上述实验结果能够为番茄SlARD的功能和分子机理研究提供理论参考和实验依据。

Long-range sequencing grants insight into additional genetic information beyond what can be accessed by both short reads and modern longread *** new sequencing technologies,such as“Hi-C”and“Linked Reads”,produce long-range datasets for high-throughput and high-resolution genome analyses,which are rapidly advancing the field of genome assembly,genome scaffolding,and more comprehensive variant *** this review,we focused on five major long-range sequencing technologies:high-throughput chromosome conformation capture(Hi-C),10X Genomics Linked Reads,haplotagging,transposase enzyme linked long-read sequencing(TELL-seq),and singletube long fragment read(stLFR).We detailed the mechanisms and data products of the five platforms and their important applications,evaluated the quality of sequencing data from different platforms,and discussed the currently available bioinformatics *** work will benefit the selection of appropriate long-range technology for specific biological studies.

Surgery remains the standard treatment for spinal ***,uncontrolled intraoperative bleeding poses a significant challenge for adequate surgical resection and compromises surgical *** this study,we develop a thrombin(Thr)-loaded nanorobothydrogel hybrid superstructure by incorporating nanorobots into regenerated silk fibroin nanofibril *** superstructure with superior thixotropic properties is injected percutaneously and dispersed into the spinal metastasis of hepatocellular carcinoma(HCC)with easy bleeding characteristics,before spinal surgery in a mouse *** near-infrared irradiation,the self-motile nanorobots penetrate into the deep spinal tumor,releasing Thr in a controlled ***-induced thrombosis effectively blocks the tumor vasculature and reduces bleeding,inhibiting tumor growth and postoperative recurrence with Au nanorod-mediated photothermal *** minimally invasive treatment platform provides a novel preoperative therapeutic strategy for HCC spinal metastasis effectively controlling intraoperative bleeding and tumor growth,with potentially reduced surgical complications and enhanced operative outcomes.

Injectable materials show their special merits in regeneration of damaged/degenerated bones in minimally-invasive *** calcium phosphate bone cement(CPC)has attracted broad attention for its bioactivity,as compared to non-degradable polymethyl methacrylate ***,its brittleness,poor anti-washout property and uncontrollable biodegradability are the main challenges to limit its further clinical application mainly because of its stone-like dense structure and fragile inorganic-salt ***,we developed a kind of injectable CPC bone cement with porous structure and improved robustness by incorporating poly(lactide-co-glycolic acid)(PLGA)nanofiber into CPC,with carboxymethyl cellulose(CMC)to offer good injectability as well as anti-wash-out ***,the introduction of PLGA and CMC also enabled a formation of initial porous structure in the cements,where PLGA nanofiber endowed the cement with a dynamically controllable biodegradability which provided room for cell movement and bone ***-estingly,the reinforced biodegradable cement afforded a sustainable provision of Ca^(2+)bioactive components,together with its porous structure,to improve synergistically new bone formation and osteo-integration in vivo by using a rat model of femur condyle *** study on regenerative mechanisms indicated that the good minimally-invasive bone regeneration may come from the synergistic enhanced osteogenic effect of calcium ion enrichment and the improved revascularization capacity contributed from the porosity as well as the lactic acid released from PLGA *** results indicate the injectable bone cement with high strength,anti-washout property and controllable biodegradability is a promising candidate for bone regeneration in a minimally-invasive approach.
