AIM To investigate the anti-fibrotic effects of the traditional oriental herbal medicine Daikenchuto(DKT) associated with transient receptor potential ankyrin 1(TRPA1) channels in intestinal myofibroblasts. METHODS Inflammatory and fibrotic changes were detected in a2,4,6-trinitrobenzenesulfonic acid(TNBS) chronic colitis model of wild-type and TRPA1-knockout(TRPA1-KO) mice via pathological staining and immunoblotting ***^(2+) imaging experiments examined the effects of DKT and its components/ingredients on intestinal myofibroblast(In Myo Fib) cell TRPA1 channel *** factors and transforming growth factor (TGF) -β1-associated signaling were tested in an In Myo Fib cell line by q PCR and immunoblotting *** from non-stenotic and stenotic regions of the intestines of patients with Crohn’s disease (CD) were used for pathological analysis. RESULTS Chronic treatment with TNBS caused more severe inflammation and fibrotic changes in TRPA1-KO than in wild-type mice.A one-week enema administration of DKT reduced fibrotic lesions in wild-type but not in TRPA1-KO *** active ingredients of DKT,i.e.,hydroxyα-sanshool and 6-shogaol,induced Ca^(2+) influxes in In Myo Fib,and this was antagonized by co-treatment with a selective TRPA1 channel blocker,*** counteracted TGF-β1-induced expression of TypeⅠcollagen andα-smooth muscle actin (α-SMA) ,which were accompanied by a reduction in the phosphorylation of Smad-2 and p38-mitogen-activated protein kinase (p38-MAPK) and the expression of ***,24-h incubation with a DKT active component Japanese Pepper increased the m RNA and protein expression levels of TRPA1 in In Myo Fibs,which in turn negatively regulated collagen *** the stenotic regions of the intestines of CD patients,TRPA1 expression was significantly *** The effects of DKT on the expression and activation of the TRPA1 channel could be advantageous for suppressing intestinal fibrosis

The Tetrahedral Constellation Gravitational Wave Observatory(TEGO) [1] represents a significant leap forward in gravitational wave detection, advancing our understanding of gravitational theory. By introducing a fourth spacecraft to form a tetrahedral configuration, TEGO enhances traditional triangular setups, enabling the simultaneous sensitivity to all six gravitational wave polarization modes, including those not predicted by general relativity.

With the increasing demand for water in hydroponic systems and agricultural irrigation,viral diseases have seriously affected the yield and quality of *** removing plant viruses in water environments,virus transmission can be prevented and agricultural production and ecosystems can be *** so far,there have been few reports on the removal of plant viruses in water ***,in this study,easily recyclable biomass-based carbon nanotubes catalysts were synthesized with varying metal activities to activate peroxymonosulfate(PMS).Among them,the magnetic 0.125Fe@NCNTs-1/PMS system showed the best overall removal performance against pepper mild mottle virus,with a 5.9 log_(10)removal within 1 ***,the key reactive species in the 0.125Fe@NCNTs-1/PMS system is^(1)O_(2),which can maintain good removal effect in real water matrices(river water and tap water).Through RNA fragment analyses and label free analysis,it was found that this system could effectively cleave virus particles,destroy viral proteins and expose their *** capsid protein of pepper mild mottle virus was effectively decomposed where serine may be the main attacking sites by^(1)O_(2).Long viral RNA fragments(3349 and 1642 nt)were cut into smaller fragments(∼160 nt)and caused their *** summary,this study contributes to controlling the spread of plant viruses in real water environment,which will potentially help protect agricultural production and food safety,and improve the health and sustainability of ecosystems.

By regarding the Newton constant GNand cosmological constant Λ as variables, in this paper we study the thermodynamics and phase transition of the Reissner-Nordstr o m anti-de Sitter(RN-Ad S) black hole with a global monopole within the framework of Ad S/CFT correspondence. We find interesting critical phenomena and phase behavior in the(grand)canonical ensembles of fixed( Q,V,C),( Φ,V,C) and( Q,V,μ). When the other parameters are fixed, the free energy decreases with the global monopole increases. In the( Q,V,C) ensemble,the range of the unstable region decreases with the increase of the global monopole. In the( Φ,V,C) ensemble, when Φ < Φc, the free energy appears as two branches, where the upper and lower branches correspond to low and high entropy, respectively. When( Q,V,μ) is fixed, a new zero-order phase transition occurs in the high-entropy phase and the low-entropy phase at certainμ-dependent temperatures. When μ increases to a certain value, this zero-order phase transition disappears. This certain value is negatively related to the magnitude of the global ***, we find that p-V criticality does not appear with the change of global ***, it is important to note that the CFT states of charged black holes with global monopoles do not correspond to van der Waals fluids. Finally, we find that charged black holes with global monopoles can better reflect thermodynamic phase transitions and critical phenomena under the Ad S/CFT correspondence. By adjusting the change of the global monopole, the thermodynamic phase transition will also change.

Objective:To explore the mechanism of action of asperosaponinⅥ(AⅥ)in the treatment of rheumatoid arthritis(RA)and validate it in ex vivo experiments using network pharmacology and molecular docking ***:The predicted targets of AⅥwere obtained from PharmMaper,UniProt and Swiss Target Prediction platforms,the disease targets were collected from Online Mendelian Inheritance in Man,Therapeutic Target Database and Gene Cards databases,the intersection targets of AⅥand RA were obtained from Venny 2.1.0,and the protein-protein interaction(PPI)network was obtained from STRING database,which was analyzed by Cytoscape software and screened to obtain the core *** software was used to analyze PPI network and screen the core *** on the Database for Annotation,Visualization and Integrated Discovery database,Gene Ontology functional and Kyoto Encyclopedia of Genes and Genomes pathway enrichment analysis were performed,and Cytoscape software was used to construct the"DiseasePathway-Target-Drug"network,which was finally verified by molecular docking and animal ***:Network pharmacological studies showed that AⅥwas able to modulate 289 targets,with 102 targets for the potential treatment of RA,with the core pathway being the AKT/PI3K signaling pathway,and the core targets being the epidermal growth factor receptor(EGFR)and matrix metalloproteinase 9(MMP9).Molecular docking results showed that AⅥcould produce strong binding with both of the 2 core *** vitro cellular experiments showed that AⅥreduced nitric oxide,prostaglandin E_2,tumor necrosis factor-alpha(TNF-α),interleukin-6(IL-6),and IL-1βlevels(P<0.05)and inhibited cyclooxygenase-2,nitric oxide synthase,EGFR,MMP9,phosphorylated phosphoinositide 3-kinase(p-PI3K),and phosphorylated serine-threonine kinase(p-AKT)proteins(P<0.05).The results of in vivo studies showed that AⅥimproved RA score and foot swelling thickness and decreased TNF-α,IL-6,p-PI3K and p-AKT levels in RA rat

Inexact proximal gradient algorithm with random reshuffling for nonsmooth optimization

Proximal gradient algorithms are popularly implemented to achieve convex optimization with nonsmooth regularization. Obtaining the exact solution of the proximal operator for nonsmooth regularization is challenging because errors exist in the computation of the gradient; consequently, the design and application of inexact proximal gradient algorithms have attracted considerable attention from researchers. This paper proposes computationally efficient basic and inexact proximal gradient descent algorithms with random reshuffling. The proposed stochastic algorithms take randomly reshuffled data to perform successive gradient descents and implement only one proximal operator after all data pass through. We prove the convergence results of the proposed proximal gradient algorithms under the sampling-without-replacement reshuffling *** computational errors exist in gradients and proximal operations, the proposed inexact proximal gradient algorithms can converge to an optimal solution neighborhood. Finally, we apply the proposed algorithms to compressed sensing and compare their efficiency with some popular algorithms.
