Evolutionary mechanisms and practical significance of reproductive success and clonal diversity in unisexual vertebrate polyploids

Unisexual reproduction is generally relevant to polyploidy, and unisexual vertebrates are often considered an evolutionary “dead end” due to the accumulation of deleterious mutations and absence of genetic diversity. However, some unisexual polyploids have developed strategies to avoid genomic decay, and thus provide ideal models to unveil unexplored evolutionary mechanisms, from the reproductive success to clonal diversity creation. This article reviews the evolutionary mechanisms for overcoming meiotic barrier and generating genetic diversity in unisexual vertebrates, and summarizes recent research advancements in the polyploid Carassius complex. Gynogenetic gibel carp(Carassius gibelio) is a unique amphitriploid that has undergone a recurrent autotriploidy and has overcome the bottleneck of triploid sterility via gynogenesis. Recently, an efficient strategy in which ploidy changes, including from amphitriploid to amphitetraploid, then from amphitetraploid to novel amphitriploid, drive unisexual-sexual-unisexual reproduction transition and clonal diversity has been *** on this new discovery, multigenomic reconstruction biotechnology has been used to breed a novel strain with superior growth and stronger disease resistance. Moreover, a unique reproduction mode that combines both abilities of ameiotic oogenesis and sperm-egg fusion,termed as ameio-fusiongensis, has been discovered, and it provides an efficient approach to synthesize sterile allopolyploids. In order to avoid ecological risks upon escape and protect the sustainable property rights of the aquaculture seed industry, a controllable fertility biotechnology approach for precise breeding is being developed by integrating sterile allopolyploid synthesis and gene-editing *** review provides novel insights into the origin and evolution of unisexual vertebrates and into the attempts being made to exploit new breeding biotechnologies in aquaculture.

Aquaculture is one of the most efficient modes of animal protein production and plays an important role in global food *** animals exhibit extraordinarily diverse sexual phenotypes and underlying mechanisms,providing an ideal system to perform sex determination research,one of the important areas in life ***,sex is also one of the most valuable traits because sexual dimorphism in growth,size,and other economic characteristics commonly exist in aquaculture ***,we synthesize current knowledge of sex determination mechanisms,sex chromosome evolution,reproduction strategies,and sexual dimorphism,and also review several approaches for sex control in aquaculture animals,including artificial gynogenesis,application of sex-specific or sex chromosome-linked markers,artificial sex reversal,as well as gene *** anticipate that better understanding of sex determination mechanisms and innovation of sex control approaches will facilitate sustainable development of aquaculture.

Rethinking fish biology and biotechnologies in the challenge era for burgeoning genome resources and strengthening food security

Fish biology has been developed for more than 100 years,but some important breakthroughs have been made in the last *** studies commonly concentrated on morphology,phylogenetics,development,growth,reproduction manipulation,and disease *** studies have mostly focused on genetics,molecular biology,genomics,and genome biotechnologies,which have provided a solid foundation for enhancing aquaculture to ensure food security and improving aquatic environments to sustain ecosystem ***,we review research advances in five major areas:(1)biological innovations and genomic evolution of four significant fish lineages including non-teleost ray-finned fishes,northern hemisphere sticklebacks,East African cichlid fishes,and East Asian cyprinid fishes;(2)evolutionary fates and consequences of natural polyploid fishes;(3)biological consequences of fish domestication and selection;(4)development and innovation of fish breeding biotechnologies;and(5)applicable approaches and potential of fish genetic breeding ***,five precision breeding biotechniques are examined and discussed in detail including gene editing for the introgression or removal of beneficial or detrimental alleles,use of sex-specific markers for the production of mono-sex populations,controllable primordial germ cell on-off strategy for producing sterile offspring,surrogate broodstock-based strategies to accelerate breeding,and genome incorporation and sexual reproduction regainbased approach to create synthetic *** on these scientific and technological advances,we propose a blueprint for genetic improvement and new breed creation for aquaculture species and analyze the potential of these new breeding strategies for improving aquaculture seed industry and strengthening food security.

jian-Kang Liu： A pioneer of sex determination studies in vertebrates

Sixty-eight years ago, Dr. W.S. Bullough, a Fellow of the Royal Society, published an article ＂Hermaphroditism in the lower vertebrates＂ in Nature （Bullough, 1947） and thereby commented a significant finding in lower vertebrate sex mechanism fulfilled by a young Chinese scholar jian-Kang Liu （刘健康, C.K. Liu）（Fig. 1 ）. At the beginning of the article, Dr. Bullough respectfully acknowledged that：＂By the publi- cation in 1944 of a description of the gonads of Monopterus javanensis Lac. （Symbranchii; Teleostei）（Liu, 1944a）, Liu has furnished new and interesting evidence concerning the mechanism of sex determination in the lower vertebrates, and has opened a fresh field for research into this subject＂（Bullough, 1947）.

Diverse and variable sex determination mechanisms in vertebrates

Sex is prevalent in nature and sex determination is one of the most fundamental biological processes, while the way of initiating female and male development exhibits remarkable diversity and variability across vertebrates. The knowledge on why and how sex determination mechanisms evolve unusual plasticity remains limited. Here, we summarize sex determination systems,master sex-determining genes and gene-regulatory networks among vertebrates. Recent research advancements on sex determination system transition are also introduced and discussed in some non-model animals with multiple sex determination mechanisms. This review will provide insights into the origin, transition and evolutionary adaption of different sex determination strategies in vertebrates, as well as clues for future perspectives in this field.

Diversity,evolutionary contribution and ecological roles of aquatic viruses

Aquatic viruses include infected viruses in aquatic animals, plants and microorganisms, and free-floating viruses(virioplankton)in water environments. In the last three decades, a huge number of aquatic viruses, especially diverse free-floating viruses,including cyanophages, phycoviruses, archaea viruses, giant viruses, and even virophages, have been identified by virological experiments and metagenomic analyses. Based on a comprehensive introduction of aquatic virus classification and their morphological and genetic diversity, here, we summarize and outline main virus species, their evolutionary contribution to aquatic communities through horizontal gene transfer, and their ecological roles for cyanobacterial bloom termination and global biogeochemical cycling in freshwater and marine ecosystems. Thereby, some novel insights of aquatic viruses and virus-host interactions, especially their evolutionary contribution and ecological rolesin diverse aquatic communities and ecosystems, are highlighted in this review.

Most vertebrates present two sexes, and females and males are determined via two diverse strategies including geno-typic sex determination （GSD） and environmental sex de-termination （ESD）（Mei and gui, 2015; Ma et al., 2016）. The most common form of ESD is temperature-dependent sex determination （TSD）. Although several master sex-de-termining genes and their molecular pathways have been elucidated in vertebrates with GSD, the molecular mechan-ism underlying TSD remains unclear （Bachtrog et al., 2014; Capel, 2017）.

Origin and chromatin remodeling of young X/Y sex chromosomes in catfish with sexual plasticity

Assembly of a complete Y chromosome is a significant challenge in animals with an XX/XY sex-determination *** cently,we created YY-supermale yellow catfish by crossing XY males with sex-reversed XY females,providing a valuable model for Y-chromosome assembly and ***,we assembled highly homomorphic Y and X chromosomes by sequencing genomes of the YY supermale and XX female in yellow catfish,reve aling their nucleotide divergences with only less than 1% and with the same gene *** sex-determining region(SDR) was identified to locate within a physical distance of0.3 Mb by ***,the incipient sex chromosomes were revealed to originate via autosome-autosome fusion and were characterized by a highly rearranged region with an SDR downstream of the fusion *** found that the Y chromosome was at a very early stage of differentiation,as no clear evidence of evolutionary strata and classical structure features of recombination suppression for a rather late stage of Y-chromosome evolution were ***,a number of sex-antagonistic mutations and the accumulation of repetitive elements were discovered in the SDR,which might be the main driver of the initial establishment of recombination suppression between young X and Y ***,distinct three-dimensional chromatin organizations of the Y and X chromosomes were identified in the YY supermales and XX females,as the X chromosome exhibited denser chromatin structure than the Y chromosome,while they respectively have significantly spatial interactions with female-and male-related genes compared with other *** chromatin configuration of the sex chromosomes as well as the nucleus spatial organization of the XX neomale were remodeled after sex reversal and similar to those in YY supermales,and a male-specific loop containing the SDR was found in the open chromatin *** results elucidate the origin of young sex chromosomes and the chromatin remodel

Viral co-infection has been found in animals;however,the mechanisms of co-infection are *** abundance and diversity of viruses in water make fish highly susceptible to ***,we reported a coinfection in fish,which resulted in reduced host lethality and illustrated the intracellular molecular mechanism of viral *** spring viremia of carp virus(SVCV)is a highly lethal virus that infects Cyprinidae,such as *** mortality of SVCV infection was significantly reduced when co-infected with the grass carp reovirus(GCRV).The severity of tissue damage and viral proliferation of SVCV was also reduced in co-infection with *** transcriptome bioinformatics analysis demonstrated that the effect on the host transcripts in response to SVCV infection was significantly reduced in *** excluding the extracellular interactions of these two viruses,the intracellular mechanisms were *** found that the GCRV NS38 remarkably decreased SVCV infection and viral *** interaction between GCRV NS38 and SVCV nucleoprotein(N)and phosphoprotein(P)proteins was identified,and NS38 downregulated both N and P *** analysis demonstrated that the N protein was degraded by NS38 indispensable of the autophagy receptor,sequestosome 1(p62).Meanwhile,K63-linked ubiquitination of the P protein was reduced by NS38,leading to ubiquitinated degradation of the P *** results reveal that the intracellular viral protein interactions are a crucial mechanism of co-infection and influence the host pathology and expand our understanding in intracellular viral interactions co-infection.
