Full T-cell activation and function in teleosts require collaboration of first and co-stimulatory signals

Mammalian T-cell responses require synergism between the first signal and co-stimulatory signal.However,whether and how dual signaling regulates the T-cell response in early vertebrates remains unknown.In the present study,we discovered that the Nile tilapia(Oreochromis niloticus)encodes key components of the LAT signalosome,namely,LAT,ITK,GRB2,VAV1,SLP-76,GADS,and PLC-γ1.These components are evolutionarily conserved,and CD3εmAb-induced T-cell activation markedly increased their expression.Additionally,at least ITK,GRB2,and VAV1 were found to interact with LAT for signalosome formation.Downstream of the first signal,the NF-κB,MAPK/ERK,and PI3K-AKT pathways were activated upon CD3εmAb stimulation.Furthermore,treatment of lymphocytes with CD28 mAbs triggered the AKT-mTORC1 pathway downstream of the co-stimulatory signal.Combined CD3εand CD28 mAb stimulation enhanced ERK1/2 and S6 phosphorylation and elevated NFAT1,c-Fos,IL-2,CD122,and CD44 expression,thereby signifying T-cell activation.Moreover,rather than relying on the first or co-stimulatory signal alone,both signals were required for T-cell proliferation.Full T-cell activation was accompanied by marked apoptosis and cytotoxic responses.These findings suggest that tilapia relies on dual signaling to maintain an optimal T-cell response,providing a novel perspective for understanding the evolution of the adaptive immune system.

Wear law in mixed lubrication based on stress-promoted thermal activation

Although several empirical wear formulas have been proposed,theoretical approaches for predicting surface topography evolution during sliding wear are limited.In this study,we propose a novel wear-prediction method,wherein the energy-based Arrhenius equation is combined with a mixed elastohydrodynamic lubrication(EHL)model to predict the point-contact wear process in mixed lubrication.The surface flash temperature and contact pressure are considered in the wear model.Simulation results are compared with the experimental results to verify the theory.The surface topography evolutions are observed during the wear process.The influences of load and speed on wear are investigated.The simulation results based on the Arrhenius equation relationship shows good agreement with the results of experiments as well as the Archard wear formula.However,the Arrhenius equation is more accurate than the Archard wear theory in some aspects,such as under high-temperature conditions.The results indicate that combining the wear formulas with the mixed EHL simulation models is an effective method to study the wear behavior over time.

高硅超细铁精矿熔剂性球团矿固结行为及还原特性

高炉高比例球团矿冶炼技术是减轻炼铁系统碳排放和环保压力的重要方案。将高硅超细铁精矿资源作为球团矿生产原料,有助于拓宽中国国内铁矿资源来源,加大中国国内贫矿应用力度。系统地探讨了碱度对高硅超细铁精矿球团矿固结行为和还原特性的影响。通过热力学计算、矿相分析、扫描电镜-能谱分析等方法对其固结行为和还原特性进行了研究。研究结果表明,随着碱度的增加,高硅超细铁精矿球团矿抗压强度先增加后降低,孔隙率先降低后增加,碱度为0.06~0.73时,球团矿的抗压强度均大于4000 N/个,碱度为0.24时的球团矿抗压强度最高,孔隙率也最低。碱度的增加会降低高硅超细铁精矿球团矿中的二氧化硅相含量,促进铁酸钙相和硅酸盐相的生成。在碱度为1.20、温度为1300℃条件下,球团矿中液相比例可达24.38%。碱度大于0.73时,球团矿中的液相量在大于1250℃后会迅速增加。随着碱度的增加,高硅超细铁精矿球团矿渣相和硅酸盐相中的钙元素含量会逐渐增加,此外,碱度越高,赤铁矿晶粒越容易呈现多边形结构且晶粒间填充的渣相量越多。随着碱度增加,高硅超细铁精矿球团矿还原度和还原膨胀率先降低后增加,球团矿抗还原粉化性能减弱。碱度大于0.24后,球团矿内部孔隙率和铁酸钙相含量的增加使得还原气体能更容易扩散至球团矿内部并促进还原反应的进行,可以有效提高球团矿的还原度,碱度为1.20时的球团矿还原度比碱度为0.06时提高了34.9%。

电力巡检机械臂系统通常具有较强的非线性和不确定性,包括负载变化、摩擦和摆动等因素。这些非线性和不确定性对控制系统的稳定性造成影响,导致传统的控制设计方法难以实现精准控制,且存在一定误差。为此,提出电力巡检机械臂终端滑模鲁棒控制方法。将电力巡检机械臂投影至笛卡尔坐标系,得到机械臂末端执行器位姿信息,将其作为滑模控制器的参数,结合模糊自适应算法与李雅普诺夫(Lyapunov)算法,构建机械臂滑模控制器控制律,获取控制信号。利用径向基函数(RBF)网络逼近不确定项,获取其控制律,根据鲁棒项,输出RBF网络鲁棒控制结果。实验表明,所提方法能够在实现电力巡检机械臂精准作业控制的同时,避免因未知扰动造成的机械臂抖动,且关节转角控制误差小、响应快及超调量低。

Smoothed-Particle Hydrodynamics Simulation of Ship Motion and Tank Sloshing under the Effect of Regular Waves

Predicting the response of liquefied natural gas(LNG)contained in vessels subjected to external waves is extremely important to ensure the safety of the transportation process.In this study,the coupled behavior due to ship motion and liquid tank sloshing has been simulated by the Smoothed-Particle Hydrodynamics(SPH)method.Firstly,the sloshing flow in a rectangular tank was simulated and the related loads were analyzed to verify and validate the accuracy of the present SPH solver.Then,a three-dimensional simplified LNG carrier model,including two prismatic liquid tanks and a wave tank,was introduced.Different conditions were examined corresponding to different wave lengths,wave heights,wave heading angles,and tank loading rates.Finally,the effects of liquid tank loading rate on LNG ship motions and sloshing loading were analyzed,thereby showing that the SPH method can effectively provide useful indications for the design of liquid cargo ships.

A Joint Entity Relation Extraction Model Based on Relation Semantic Template Automatically Constructed

The joint entity relation extraction model which integrates the semantic information of relation is favored by relevant researchers because of its effectiveness in solving the overlapping of entities,and the method of defining the semantic template of relation manually is particularly prominent in the extraction effect because it can obtain the deep semantic information of relation.However,this method has some problems,such as relying on expert experience and poor portability.Inspired by the rule-based entity relation extraction method,this paper proposes a joint entity relation extraction model based on a relation semantic template automatically constructed,which is abbreviated as RSTAC.This model refines the extraction rules of relation semantic templates from relation corpus through dependency parsing and realizes the automatic construction of relation semantic templates.Based on the relation semantic template,the process of relation classification and triplet extraction is constrained,and finally,the entity relation triplet is obtained.The experimental results on the three major Chinese datasets of DuIE,SanWen,and FinRE showthat the RSTAC model successfully obtains rich deep semantics of relation,improves the extraction effect of entity relation triples,and the F1 scores are increased by an average of 0.96% compared with classical joint extraction models such as CasRel,TPLinker,and RFBFN.

By conducting a two-dimensional experimental study,this paper aims to enhance the understanding of the mechanism of sand convective motions in the vicinity of a wall subjected to long-term cyclic lateral loadings.The experimental tests were conducted in a rectangular sandbox with a transparent front-wall,through which the process of sand particle motions could be recorded by using a high-resolution digital camera.The images were processed with a high time-resolved PIV(Particle Image Velocimetry)system.Based on the experimental data,this work(1)presents the sand flow field in the convective zones;(2)provides means to describe the convection mechanism;(3)proposes the relationships between the loading conditions and dimensions of the region with intense sand movement;and(4)elaborates the similarity of the sand flow velocity structure within the sand convective zones.

Modulating the alloying mode in the doping-induced synthesis of Au-Pd nanowires

Heterogeneous doping is one effective strategy for synthesizing metal alloy nanowires.Herein,the heterogeneous doping processes of Pd on the ultrathin Au nanowires were systematically modulated and investigated.Au-Pd alloy nanowires with various morphologies and lattice structures can be obtained by adjusting the morphology of the precursor Au nanowires and the kinetics of the heterogeneous doping processes.The effects of the rate of Pd reduction and the concentration of the ligand oleylamine(OAm)on the Pd deposition and alloying mode were articulated.Generally,as the Pd deposition rate decreases,the Pd deposition and alloying mode switches from the island-forming Stransky–Krastanov(SK)mode to the epitaxial Frank-van der Merwe(FM)mode,and eventually to an unconventional twisting alloying mode,where the interdiffusion of Pd and Au causes drastic rearrangement of the lattice structure and formation of helical structures.The kinetics-related variation of alloying mode could also be observed in the Au-Ag nanowires,demonstrating a general design principle for the synthesis of alloy nanostructures.In addition,the electrocatalytic performance of various Au-Pd nanowires was evaluated,and the alloy nanowire formed via the SK mode was found to be an excellent electrocatalyst for oxygen reduction and ethanol oxidation.

Enhancing low-resource cross-lingual summarization from noisy data with fine-grained reinforcement learning

Cross-lingual summarization(CLS)is the task of generating a summary in a target language from a document in a source language.Recently,end-to-end CLS models have achieved impressive results using large-scale,high-quality datasets typically constructed by translating monolingual summary corpora into CLS corpora.However,due to the limited performance of low-resource language translation models,translation noise can seriously degrade the performance of these models.In this paper,we propose a fine-grained reinforcement learning approach to address low-resource CLS based on noisy data.We introduce the source language summary as a gold signal to alleviate the impact of the translated noisy target summary.Specifically,we design a reinforcement reward by calculating the word correlation and word missing degree between the source language summary and the generated target language summary,and combine it with cross-entropy loss to optimize the CLS model.To validate the performance of our proposed model,we construct Chinese-Vietnamese and Vietnamese-Chinese CLS datasets.Experimental results show that our proposed model outperforms the baselines in terms of both the ROUGE score and BERTScore.
