Full T-cell activation and function in teleosts require collaboration of first and co-stimulatory signals

Mammalian T-cell responses require synergism between the first signal and co-stimulatory signal.However,whether and how dual signaling regulates the T-cell response in early vertebrates remains unknown.In the present study,we discovered that the Nile tilapia(Oreochromis niloticus)encodes key components of the LAT signalosome,namely,LAT,ITK,GRB2,VAV1,SLP-76,GADS,and PLC-γ1.These components are evolutionarily conserved,and CD3εmAb-induced T-cell activation markedly increased their expression.Additionally,at least ITK,GRB2,and VAV1 were found to interact with LAT for signalosome formation.Downstream of the first signal,the NF-κB,MAPK/ERK,and PI3K-AKT pathways were activated upon CD3εmAb stimulation.Furthermore,treatment of lymphocytes with CD28 mAbs triggered the AKT-mTORC1 pathway downstream of the co-stimulatory signal.Combined CD3εand CD28 mAb stimulation enhanced ERK1/2 and S6 phosphorylation and elevated NFAT1,c-Fos,IL-2,CD122,and CD44 expression,thereby signifying T-cell activation.Moreover,rather than relying on the first or co-stimulatory signal alone,both signals were required for T-cell proliferation.Full T-cell activation was accompanied by marked apoptosis and cytotoxic responses.These findings suggest that tilapia relies on dual signaling to maintain an optimal T-cell response,providing a novel perspective for understanding the evolution of the adaptive immune system.

Single-Cell Sequencing Technologies: Current and Future

Intensively developed in the last few years, single-cell sequencing technologies now present numerous advantages over traditional sequencing methods for solving the problems of biological heterogeneity and low quantities of available biological materials. The application of single-cell sequencing technologies has profoundly changed our understanding of a series of biological phenomena, including gene transcription, embryo development, and carcinogenesis. However, before single-cell sequencing technologies can be used extensively, researchers face the serious challenge of overcoming inherent issues of high amplification bias, low accuracy and repro- ducibility. Here, we simply summarize the techniques used for single-cell isolation, and review the current technologies used in single-cell genomic, transcriptomic, and epigenomic sequencing, We discuss the merits, defects, and scope of application of single-cell sequencing technologies and then speculate on the direction of future developments.

Primary biliary cholangitis(PBC) is an autoimmune disease involving dysregulation of a broad array of homeostatic and metabolic processes. Although considerable single-nucleotide polymorphisms have been unveiled, a large fraction of risk factors remains enigmatic. Candidate genes with rare mutations that tend to confer more deleterious effects need to be identified. To help pinpoint cellular and developmental mechanisms beyond common noncoding variants, we integrate whole exome sequencing with integrative network analysis to investigate genes harboring de novo mutations. Prominent convergence has been revealed on a network of disease-specific co-expression comprised of 55 genes associated with homeostasis and metabolism. The transcription factor gene MEF2 D and the DNA repair gene PARP2 are highlighted as hub genes and identified to be up-and down-regulated, respectively, in peripheral blood data set. Enrichment analysis demonstrates that altered expression of MEF2 D and PARP2 may trigger a series of molecular and cellular processes with pivotal roles in PBC pathophysiology. Our study identifies genes with de novo mutations in PBC and suggests that a subset of genes in homeostasis and metabolism tend to act in synergy through converging on co-expression network, providing novel insights into the etiology of PBC and expanding the pool of molecular candidates for discovering clinically actionable biomarkers.

Flow characteristics of mixed particles with a simulated cold state in moving bed reactor

The funnel flow of high-temperature circulating ash and coal in moving bed with height restrictions directly influences the efficiency of coal pyrolysis and scale-up design of reactor.It is one of the technical bottlenecks in the use of moving bed.In order to provide data support for the particle flow and pyrolysis model close to the actual working conditions in the future,this study describes the flow characteristics of solid-solid mixed particles in a cold two-dimensional moving bed.The results indicate that flow char-acteristics of mixed particles in the quartz sand-coal system are better than those in the cold circulating ash-coal system.The optimized conditions were obtained,the insert half angle is 20° and a heat carrier to coal ratio of 6:1.As the mixture progressed downstream,secondary separation of the heat carrier and coal become apparent.Furthermore,mixture residence time has been investigated to explore the relationship between regional residence time and to predict accurately the actual pyrolysis progress in pyrolyzer.

氟化氩(ArF)浸没式光刻需在光刻胶表面形成抗水涂层,阻挡光刻胶和水之间组分交换。抗水涂层对光刻胶的分辨率、工艺窗口、低缺陷要求起着重要作用,平衡抗水涂层的疏水性和碱溶性是设计聚合物结构的重点和难点。分析了形成抗水涂层的方法和成膜机理,对比了不同方法的优缺点,根据聚合物所含官能团及其碱溶性,对抗水涂层聚合物进行了分类总结,重点阐述抗水涂层聚合物的结构和性能要求,尤其关注了聚合物侧链的位阻效应和氢键作用对涂层的疏水性影响。最后对抗水涂层的应用和发展进行了展望。

Tailoring the in-plane and out-of-plane stiffness of soft fingers by endoskeleton topology optimization for stable grasping

The intrinsic compliance of soft materials endows soft robots with great advantages to achieve large deformation and adaptive interactions in grasping tasks.However,current soft grippers usually focus on the in-plane large deformation and load capacity but ignore the effect of out-of-plane external loads,which may lead to instability in practical scenarios.This problem calls for stiffness design along multiple directions to withstand not only in-plane interacting forces with objects,but also unexpected outof-plane loads.In this paper,we design a new type of soft finger by embedding an endoskeleton inside the widely-used PneuNets actuator,and the endoskeleton layout is optimized to achieve a remarkable bending deflection and limited lateral deflection under combined external in-plane and out-of-plane loads.Based on the multi-objective topology optimization approach,the key structural features of the optimized endoskeleton are extracted and parameterized.The multi-material soft fingers are fabricated by the silicone compound mold method.Static and dynamic experiment results validate that the soft gripper with endoskeleton embedded exhibits remarkably improved out-of-plane stiffness,without sacrificing the in-plane bending flexibility,and leads to more stable grasping.

Experimental Study on the Performance and Emission of Chinese Small Agricultural Diesel Engine Fuelled with Methanol/Biodiesel/DTBP

Diesel engine alternative fuels, such as methanol and biodiesel, are beneficial to reduce diesel engine emission. In order to study the influence of methanol and biodiesel on the performance, economy and emission of small agricultural diesel engine, the physical-chemical properties(cetane number, lower heat value(LHV), viscosity, etc.) of methanol and biodiesel were analyzed. The methanol and biodiesel showed good complementary property to some extent. When a large proportion of methanol was added into biodiesel, the cetane number of the methanol/biodiesel blend will be greatly reduced. Since the cetane number of the blend fuel has great influence on the combustion process of diesel engine, after testing for blending ratio of methanol/biodiesel, the blend was prepared with 5%(BM5), 10%(BM10) and 15%(BM15) methanol, respectively. Di-Tert-Butyl Peroxide(DTBP) was chosen as a cetane number improver to be added into methanol/biodiesel blend. 0.25%, 0.50% and 0.75% of DTBP was added into BM15. The bench test was carried out on a 186 FA diesel engine to study the effect of methanol and DTBP on the engine performance and emissions. The results show that, at rated condition, compared with biodiesel, the NO;concentration of BM5, BM10 and BM15 is reduced by 5.02%, 33.85% and 21.24%, and smoke is reduced by 5.56%, 22.22% and 55.56%. However, the engine power is also reduced by 5.77%, 14.23% and 25.41%, and the brake specific energy consumption is increased by 3.31%, 7.78% and 6.37%. The addition of DTBP in methanol/biodiesel could recover the engine power to the level of diesel. DTBP shows good effect on the reduction of the brake specific energy consumption and NO_(x), CO, HC concentration, but a little increase of exhaust smoke.

目的基于分子模拟技术筛选具有潜在治疗非小细胞肺癌的ROS1激酶抑制化合物。方法本研究构建了肉瘤致癌因子ROS1受体及其抑制剂之间的药效团模型,通过化合物数据库筛选、分子对接、分子动力学模拟等方法,找到一种具有抑制ROS1功能的先导化合物,最后对该抑制剂的先导化合物与受体蛋白结合作用的模式进行了分析。结果筛选得到的75号化合物在与ROS1蛋白的分子动力学模拟过程中,显示出了比较稳定的相互作用。结论75号化合物可以作为潜在的抑制ROS1的先导化合物。

Zinc-anode-based electrochromic devices(ZECDs) are emerging as the next-generation energy-e cient transparent electronics. We report anatase W-doped TiO_(2) nanocrystals(NCs) as a Zn^(2+) active electrochromic material. It demonstrates that the W doping in TiO_(2) highly reduces the Zn^(2+) intercalation energy,thus triggering the electrochromism. The prototype ZECDs based on W-doped TiO_(2) NCs deliver a high optical modulation(66% at 550 nm),fast spectral response times(9/2.7 s at 550 nm for coloration/bleaching),and good electrochemical stability(8.2% optical modulation loss after 1000 cycles).

Preparation and Characterization of Potassium Monopersulfate/Ethyl Cellulose Microcapsules and Their Sustained Release Performance

Environmental cleaning is an important aspect of bacteria control.Ethyl cellulose microcapsules containing potassium monopersulfate(PMCM)were prepared by emulsified solvent diffusion method.The chemical structure and microstructure of the obtained PMCM was characterized by methods of Fourier transform infrared spectroscopy(FT-IR),optical microscopy,scanning electron microscopy and X-ACT energy dispersive X-ray spectroscopy.The SEM micrographs of the PMCM containing 21.6%of C,46.8%of O,10.7%of S and 19.4%of K was relatively smooth.Thermal stability,sustained release performance,and antimicrobial activity of PMCM were investigated.The results showed that the drug loading and encapsulation efficiency of PMCM were 30.3%and 42.6%respectively.Potassium monopersulfate was fully released after 8 h,following a Fickian diffusion mechanism.Results showed that the microcapsules prepared with a high concentration of potassium monopersulfate solution showed a good antimicrobial effect.The microcapsule wall of the resulting PMCM increased with increasing ethyl cellulose content and had high thermal stability from the data of 69%residue rate.The excellent thermal stability and high sustained release performance of PMCM showed high application value.
