Coronaviruses RNA interacts with host miR-500a-5p and miR-501-5p to regulate multiple pathways

The Coronavirus disease is the contagious disease. Clinical manifests vary from no symptoms, upper and lower respiratory tract infections, and even severe acute respiratory distress syndrome and multiple organs failure. Recently, three of new beta-coronaviruses have emerged to cause serious and widespread illness and death, such as the Severe Acute Respiratory Syndrome Coronaviruses (SARS-CoV), Middle East Respiratory Syndrome Coronaviruses (MERS-CoV), and 2019-nCoV (the causative pathogen of Coronavirus Disease 2019 (COVID-19). The COVID-19 is continuing to spread globally. However, the detailed molecular mechanisms have not yet been fully clarified. Since the similarity of the pathological features among 2019-nCoV, SARS-CoV and MERS-CoV, we could speculate the biological features of current COVID-19 by researching those of MERS-CoV and SARS-CoV. Therefore, we analyzed expression levels of miRNA and mRNA profiles of MERS-CoV, SARS-CoV and 2019-nCoV from five public GEO datasets (Table S1). As a result, we found 16 miRNAs potentially target a group of hub genes clustering together to tune the immune response. Notably, viral RNAs have the ability to sponge miR-500a-5p and miR-501-5p, indicating viral RNAs sequestering these two miRNAs to trigger genes expression and contribute to pathogenesis. We hope this study will pave the way to better understand 2019-nCoV pathogenesis and develop the innovative therapeutics.

Numerical modeling of scour and deposition around permeable cylindrical structures

Flow past wall-mounted cylindrical structures is commonly encountered in natural rivers where piers of bridge crossings or vegetation stalks are common within channels.In the current study,the influence of cylindrical structures on flow/bathymetric alterations for three different permeabilities is explored via two-dimensional numerical modeling.In model construction processes,the structure permeability is varied with the surface void ratio along the perimeter of the cylinder,i.***.the density of emergent and submerged solid elements is used to delineate the cylinder boundaries.The validation of this model is guaranteed through careful comparison with experimental data obtained for similar hydrodynamic conditions and cylinder properties.The validated model then is applied to investigate flow properties and scour and deposition patterns with structure permeabilities of 0.0,0.38,and 0.62.Simulated results show that a permeable structure has less impeding effects on flow than a solid cylinder.The wake velocity reduction decreases 38% with a 63% increase in the structure permeability due to increasing intensity of the bleeding flow through surface voids,causing less flow contraction and diversion,lower turbulent kinetic energy,and lower lee-side scour around the permeable structure and less deposition downstream under live-bed conditions.

Tuning Polymer Molecular Weight Distribution in Cationic RAFT Polymerization by Mixing Chain Transfer Agents

Polymer dispersity (Đ) or molecular weight distribution (MWD) is a basic but vital parameter for the properties of polymeric materials. Developing new methodologies for controlling polymer MWD is emerging as a research hotspot. However, the methods to tune polymer MWD in cationic polymerization are still not well explored. Herein, we present a simple method to control the dispersity of poly(isobutyl vinyl ether) (PIBVE) by mixing two different chain transfer agents in batch visible light induced cationic RAFT polymerization. A broad dispersity range (Đ ≈ 1.16—1.80) was successfully achieved while maintaining monomodal MWD. Moreover, chain extension of PIBVE through both cationic polymerization and radical polymerization has been studied, which also provides a method to tune polymer MWD in mechanism transformation polymerization.

Nonlinear simultaneous inversion of pore structure and physical parameters based on elastic impedance

Carbonate reservoirs have complex pore structures,which not only significantly affect the elastic properties and seismic responses of the reservoirs but also affect the accuracy of the prediction of the physical parameters.The existing rockphysics inversion methods are mainly designed for clastic rocks,and the inversion objects are generally porosity and water saturation.The data used are primarily based on the elastic parameters,and the inversion methods are mainly linear approximations.To date,there has been a lack of a simultaneous pore structure and physical parameter inversion method for carbonate reservoirs.To solve these problems,a new Bayesian nonlinear simultaneous inversion method based on elastic impedance is proposed.This method integrates the differential effective medium model of multiple-porosity rocks,Gassmann equation,Amplitude Versus Offset(AVO)theory,Bayesian theory,and a nonlinear inversion algorithm to achieve the simultaneous quantitative prediction of the pore structure and physical parameters of complex porous reservoirs.The forward modeling indicates that the contribution of the pore structure,i.e.,the pore aspect ratio,to the AVO response and elastic impedance is second only to that of porosity and is far greater than that of water saturation.The application to real data shows that the new inversion method for determining the pore structure and physical parameters directly from pre-stack data can accurately predict a reservoir's porosity and water saturation and can evaluate the pore structure of the effective reservoir.

Plastome-wide comparison reveals new SNV resources for the authentication of Dendrobium huoshanense and its corresponding medicinal slice(Huoshan Fengdou)

Dendrobium species and their corresponding medicinal slices have been extensively used as traditional Chinese medicine(TCM) in many Asian countries. However, it is extremely difficult to identify Dendrobium species based on their morphological and chemical features. In this study, the plastomes of D. huoshanense were used as a model system to investigate the hypothesis that plastomic mutational hotspot regions could provide a useful single nucleotide variants(SNVs) resource for authentication studies. We surveyed the plastomes of 17 Dendrobium species, including the newly sequenced plastome of D. huoshanense. A total of 19 SNVs that could be used for the authentication of D. huoshanense were detected. On the basis of this comprehensive comparison, we identified the four most informative hotspot regions in the Dendrobium plastome that encompass ccsA to ndhF, matK to 3′trnG, rpoB to psbD, and trnT to rbcL. Furthermore, to established a simple and accurate method for the authentication of D. huoshanense and its medicinal slices, a total of 127 samples from 20 Dendrobium species including their corresponding medicinal slices(Fengdous) were used in this study. Our results suggest that D. huoshanense and its medicinal slices can be rapidly and unequivocally identified using this method that combines real-time PCR with the amplification refractory mutation system(ARMS).

Lateral shear performance of sheathed post-and-beam wooden structures with small panels

Sheathed post-and-beam wooden structures are distinct from light-wood structures.They allow for using sheathing panels that are smaller(0.91 m×1.82 m)than standard-sized panels(1.22 m×2.44 m or 2.44 m×2.44 m).Evidence indicates that nail spacing and panel thickness determine the lateral capacity of the wood frame shear walls.To verify the lateral shear performance of wood frame shear walls with smaller panels,we subjected 13 shear walls,measuring 0.91 m in width and 2.925 m in height,to a low-cycle cyclic loading test with three kinds of nail spacing and three panel thicknesses.A nonlinear numerical simulation analysis of the wall was conducted using ABAQUS finite element(FE)software,where a custom nonlinear spring element was used to simulate the sheathing-frame connection.The results indicate that the hysteretic performance of the walls was mainly determined by the hysteretic performance of the sheathing-frame connection.When same nail specifications were adopted,the stiffness and bearing capacity of the walls were inversely related to the nail spacing and directly related to the panel thickness.The shear wall remained in the elastic stage when the drift was 1/250 rad and ductility coefficients were all greater than 2.5,which satisfied the deformation requirements of residential structures.Based on the test and FE analysis results,the shear strength of the post-and-beam wooden structures with sheathed walls was determined.

Recycling of waste aluminum scraps to fabricate sulfidated zero-valent iron-aluminum particles for enhanced chromate removal

Massive waste aluminum scraps produced from the spent aluminum products have high electron capacity and can be recycled as an attractive alternative to materials based on zerovalent iron(Fe^(0))for the removal of oxidative contaminants from wastewater.This study thus proposed an approach to fabricate micron-sized sulfidated zero-valent iron-aluminum particles(S-Al^(0)@Fe^(0))with high reactivity,electron selectivity and capacity using recycled waste aluminum scraps.S-Al^(0)@Fe^(0)with a three-layer structure contained zero-valent aluminum(Al^(0))core,Fe^(0) middle layer and iron sulfide(FeS)shell.The rates of chromate(Cr(Ⅵ))removal by S-Al^(0)@Fe^(0)at pH 5.0-9.0 were 1.6-5.9 times greater than that by sulfidated zero-valent iron(S-Fe^(0)).The Cr(Ⅵ)removal capacity of S-Al^(0)@Fe^(0)was 8.2-,11.3-and 46.9-fold greater than those of S-Fe0,zero-valent iron-aluminum(Al^(0)-Fe^(0))and Fe^(0),respectively.The chemical cost of S-Al^(0)@Fe^(0) for the equivalent Cr(Ⅵ)removal was 78.5%lower than that of S-Fe^(0).Negligible release of soluble aluminum during the Cr(Ⅵ)removal was observed.The significant enhancement in the reactivity and capacity of S-Al^(0)@Fe^(0)was partially ascribed to the higher reactivity and electron density of the Al0core than Fe^(0).More importantly,S-Al^(0)@Fe^(0) served as an electric cell to harness the persistent and selective electron transfer from the Al^(0)-Fe^(0) core to Cr(Ⅵ)at the surface via coupling Fe^(0)-Fe^(2+)-Fe^(3+)redox cycles,resulting in a higher electron utilization efficiency.Therefore,S-Al^(0)@Fe^(0) fabricated using recycled waste aluminum scraps can be a cost-effective and environmentally-friendly alternative to S-Fe^(0) for the enhanced removal of oxidative contaminants in industrial wastewater.

Metabolic plasticity of T cell fate decision

The efficacy of adaptive immune responses in cancer treatment relies heavily on the state of the T cells. Upon antigen exposure, T cells undergo metabolic reprogramming, leading to the development of functional effectors or memory populations. However, within the tumor microenvironment (TME), metabolic stress impairs CD8+ T cell anti-tumor immunity, resulting in exhausted differentiation. Recent studies suggested that targeting T cell metabolism could offer promising therapeutic opportunities to enhance T cell immunotherapy. In this review, we provide a comprehensive summary of the intrinsic and extrinsic factors necessary for metabolic reprogramming during the development of effector and memory T cells in response to acute and chronic inflammatory conditions. Furthermore, we delved into the different metabolic switches that occur during T cell exhaustion, exploring how prolonged metabolic stress within the TME triggers alterations in cellular metabolism and the epigenetic landscape that contribute to T cell exhaustion, ultimately leading to a persistently exhausted state. Understanding the intricate relationship between T cell metabolism and cancer immunotherapy can lead to the development of novel approaches to improve the efficacy of T cell-based treatments against cancer.

High temperature electrochemical discharge performance of LaFeO3 coated with C/Ni as anode material for NiMH batteries

Perovskite LaFeO3is considered as a promising new anode material for nickel/metal hydride batteries due to its low cost,environmental friendliness and high temperature resistance.However,the poor conductivity of LaFeO3material restricts the discharge ability,which is problematic for its future widespread application.To solve the above issue,in this study,we prepared C/Ni-coated LaFeO3composite in view of the excellent electrical conductivity of carbon and nickel metal.Results show that the C/Ni-coated LaFeO3composite delivers remarkably increased discharge capacity of ～345 mAh g-1at 60℃ in contrast to ～267 mAh g-1for pure LaFeO3.Furthermore,the carbon and nickel not only increase the electrical conductivity of the LaFeO3but also reduces the agglomeration of the LaFeO3,therefore,the C/Ni-coated LaFeO3composite serves superior long cycle-life,which maintains 60.9% after 100 cycles(52.9% for the LaFeO3sample).In overall,the electrochemical behavior of the C/Ni-coated LaFeO3composite confirms its high potential as nickel/metal hydride batteries for energy storage applications.
