Phenotype and metabolism alterations in PCB-degrading Rhodococcus biphenylivorans TG9^(T) under acid stress

Environmental acidification impairs microorganism diversity and their functions on substance transformation.Rhodococcus is a ubiquitously distributed genus for contaminant detoxification in the environment,and it can also adapt a certain range of pH.This work interpreted the acid responses from both phenotype and metabolism in strain Rhodococcus biphenylivorans TG9^(T)(TG9)induced at pH 3.The phenotype alterations were described with the number of culturable and viable cells,intracellular ATP concentrations,cell shape and entocyte,degradation efficiency of polychlorinated biphenyl(PCB)31 and biphenyl.The number of culturable cells maintained rather stable within the first 10 days,even though the other phenotypes had noticeable alterations,indicating that TG9 possesses certain capacities to survive under acid stress.The metabolism responses were interpreted based on transcription analyses with four treatments including log phase(LP),acid-induced(PER),early recovery after removing acid(RE)and later recovery(REL).With the overview on the expression regulations among the 4 treatments,the RE sample presented more upregulated and less downregulated genes,suggesting that its metabolism was somehow more active after recovering from acid stress.In addition,the response mechanism was interpreted on 10 individual metabolism pathways mainly covering protein modification,antioxidation,antipermeability,H+consumption,neutralization and extrusion.Furthermore,the transcription variations were verified with RT-qPCR on 8 genes with 24-hr,48-hr and 72-hr acid treatment.Taken together,TG9 possesses comprehensive metabolism strategies defending against acid stress.Consequently,a model was built to provide an integrate insight to understand the acid resistance/tolerance metabolisms in microorganisms.

Multi-scale numerical simulation of fluidized beds: Model applicability assessment

In the past few decades,multi-scale numerical methods have been developed to model dense gas-solidflow in fluidized beds with different resolutions,accuracies,and efficiencies.However,ambiguity needsto be clarified in the multi-scale numerical simulation of fluidized beds:(i)the selection of the submodels,parameters,and numerical resolution;(ii)the multivariate coupling of operating conditions,bed configurations,polydispersity,and additional forces.Accordingly,a state-of-the-art review is performed to assess the applicability of multi-scale numerical methods in predicting dense gas-solid flow influidized beds at specific fluidization regimes(e.g.,bubbling fluidization region,fast fluidization regime),with a focus on the inter-particle collision models,inter-phase interaction models,collision parameters,and polydispersity effect.A mutual restriction exists between resolution and efficiency.Higherresolution methods need more computational resources and thus are suitable for smaller-scale simulations to provide a database for closure development.Lower-resolution methods require fewercomputational resources and thus underpin large-scale simulations to explore macro-scale phenomena.Model validations need to be further conducted under multiple flow conditions and comprehensivemetrics(e.g.,velocity profiles at different heights,bubbles,or cluster characteristics)for furtherimprovement of the applicability of each numerical method.

Study of biomass gasification in an industrial-scale dual circulating fluidized bed(DCFB)using the Eulerian-Lagrangian method

Dual circulating fluidized bed(DCFB)has emerged as an efficient reactor for biomass gasification due to its unique feature of high gas-solid contact efficiency and separated reactions in two reactors,yet the understanding of complex in-furnace phenomena is still lacking.In this study,biomass gasification in an industrial-scale DCFB system was numerically studied using a multiphase particle-in-cell(MP-PIC)method featuring thermochemical sub-models(e.g.,heat transfer,heterogeneous reactions,and homogeneous reactions)under the Eulerian-Lagrangian framework.After model validation,the hydrodynamics and thermochemical characteristics(i.e.,pressure,temperature,and species)in the DCFB are comprehensively investigated.The results show that size-/density-induced segregation makes solid fuels concentrate on the bed surface.Interphase momentum exchange leads to the continuous decrease of the gas pressure axially.In the gasifier and combustor,the lower heating value(LHV)of the gas products is 5.56 MJ/Nm^(3)and 0.2 MJ/Nm^(3)and the combustible gas concentration(CGC)is 65.5%and 1.86%,respectively.The temperature in the combustor is about 100 K higher than that in the gasifier.A higher solid concentration results in a smaller value of particle heat transfer coefficient(HTC).The HTCs range from 50 to 150 W/(m^(2) K)for a solid concentration larger than 0.3 in the combustor while the HTCs range from 100 to 200 W/(m^(2 )K)in the gasifier.The Reynolds number of biomass particles is two orders of magnitude larger than that of the sand particle.The numerical results shed light on the reactor design and process optimization of biomass gasification in DCFBs.

Conformally anchoring nanocatalyst onto quartz fibers enables versatile microreactor platforms for continuous-flow catalysis

Continuous-flow microreactors offer increased reactivity and reusability via unique reaction pathways to address a wide range of practical nanocatalysis problems.However,only limited platforms exist to employ these microreactors for versatile nanocatalytic reactions.In this work,we conformally anchored nickel oxide(Ni O)nanosheets onto quartz fibers(QFs),which exhibited a high catalytic activity using the hydrogenation of 4-nitrophenol(4-NP)as a model reaction in a batch reaction study.More importantly,we demonstrated that fiber-based QF@Ni O composites(e.g.,cotton,fabric,belt,felt)can be integrated as versatile platforms to develop microreactors for continuous-flow catalytic applications including hydrogenation reactions and dyecatalyzed degradation.This fiber-based three-dimensional(3 D)nanocatalyst architecture effectively drives continuous-flow catalytic reactions with unprecedented efficiency due to the easy diffusion of reactant molecules into the fibrous structure,allowing contact with catalytic active sites.Our approach to continuous-flow microreactor design uses surface hybridization as a guideline to immobilize nanocatalysts onto the QFs.These QF-based platforms,coupled with rational design,are expected to be applied to a wide range of nanocatalytic reactions.

S-scheme heterojunctions have promising applications in photocatalytic CO_(2) reduction due to their unique structure and interfacial interactions,but improving their carrier separation efficiency and CO_(2) adsorption capacity remains a challenge.In this work,highly dispersed MOF-BiOBr/Mn_(0.2) Cd_(0.8) S(MOF-BiOBr/MCS)S-scheme heterojunctions with high photocatalytic CO_(2) reduction performance were constructed.The intimate contact between the MCS nano-spheres and the nanosheet-assembled MOF-BiOBr rods,driven by the internal electric field,accelerates the charge transfer along the S-scheme pathway.Moreover,the high specific surface area of MOFs is preserved to provide abundant active sites for reaction/adsorption.The formation of MOF-BiOBr/MCS S-scheme heterojunction is confirmed by theoretical calculations.The optimum MOF-BiOBr/MCS shows excellent activity in CO_(2) reduction,affording a high CO evolution rate of 60.59µmol h^(−1) g^(−1).The present work can inspire the exploration for the construction of effective heterostructure photocatalysts for photoreduction CO_(2).

Sirt1-ROS-TRAF6 Signaling-Induced Pyroptosis Contributes to Early Injury in Ischemic Mice

Stroke is an acute cerebro-vascular disease with high incidence and poor prognosis,most commonly ischemic in nature.In recent years,increasing attention has been paid to inflammatory reactions as symptoms of a stroke.However,the role of inflammation in stroke and its underlying mechanisms require exploration.In this study,we evaluated the inflammatory reactions induced by acute ischemia and found that pyroptosis occurred after acute ischemia both in vivo and in vitro,as determined by interleukin-1β,apoptosis-associated speck-like protein,and caspase-1.The early inflammation resulted in irreversible ischemic injury,indicating that it deserves thorough investigation.Meanwhile,acute ischemia decreased the Sirtuin 1(Sirtl)protein levels,and increased the TRAF6(TNF receptor associated factor 6)protein and reactive oxygen species(ROS)levels.In further exploration,both Sirtl suppression and TRAF6 activation were found to contribute to this pyroptosis.Reduced Sirtl levels were responsible for the production of ROS and increased TRAF6 protein levels after ischemic exposure.Moreover,N-acetyl-L-cysteine,an ROS scavenger,suppressed the TRAF6 accumulation induced by oxygen-glucose deprivation via suppression of ROS bursts.These phenomena indicate that Sirtl is upstream of ROS,and ROS bursts result in increased TRAF6 levels.Further,the activation of Sirtl during the period of ischemia reduced ischemiainduced injury after 72 h of reperfusion in mice with middle cerebral artery occlusion.In sum,these results indicate that pyroptosis-dependent machinery contributes to the neural injury during acute ischemia via the Sirt1-ROS-TRAF6 signaling pathway.We propose that inflammatory reactions occur soon after oxidative stress and are detrimental to neuronal survival;this provides a promising therapeutic target against ischemic injuries such as a stroke.

Alum split applications strengthened phosphorus fixation and phosphate sorption in high legacy phosphorus calcareous soil

High phosphorus(P)saturation arising from historic P inputs to protected vegetable fields(PVFs)drives high P mobilisation to waterbodies.Amendment of soils with alum has shown potential in terms of fixing labile P and protecting water quality.The present 15 month pot experiment investigated P stabilisation across single alum application(Alum-1 treatment,20 g alum/kg soil incorporated into soil before the maize was sown),alum split applications(Alum-4 treatment,5 g alum/kg soil incorporated into soil before each crop was sown i.e.4×5 g/kg)and soil only treatment(Control).Results showed that the Alum-1 treatment caused the strongest stabilisation of soil labile P after maize plant removal,whereas the P stabilisation effect was gradually weakened due to the transformation of soil non-labile P to labile P and the reduced active Al^(3+)in soil solution.For the Alum-4 treatment,soil labile P decreased gradually with each crop planting and was lower than the Alum-1 treatment at the end of the final crop removal,without any impairment on plant growth.The better P stabilisation at the end of Alum-4 treatment was closely correlated with a progressive supply of Al^(3+)and a gradual decrease of pH,which resulted in higher contents of poorlycrystalline Al,Fe and exchangeable Ca.These aspects were conducive to increasing the soil P stabilisation and phosphate sorption.In terms of management,growers in continuous cropping systems could utilise split alum applications as a strategy to alleviate P losses in high-P enriched calcareous soil.

Membrane separation technology has been taken up for use in diverse applications such as water treatment,pharmaceutical,petroleum,and energy-related industries.Compared with the design of membrane materials,the innovation of membrane preparation technique is more urgent for the development of membrane separation technology,because it not only affects physicochemical properties and separation performance of the fabricated membranes,but also determines their potential in industrialized application.Among the various membrane preparation methods,spray technique has recently gained increasing attention because of its low cost,rapidity,scalability,minimum of environmental burden,and viability for nearly unlimited range of materials.In this Review article,we summarized and discussed the recent developments in separation membranes using the spray technique,including the fundamentals,important features and applications.The present challenges and future considerations have been touched to provide inspired insights for developing the sprayed separation membranes.

在世行新营商环境评估项目下,“金融服务”主题旨在考察企业运营与扩张中的金融准入因素,“动产担保交易与登记系统的运营”是其下围绕经济体的动产担保法制进行考察的领域。在“统一的动产担保交易法律框架”指标下,通过对我国民法典相关规则的展开解释,可以将动产让与担保纳入现有动产担保制度体系内,并使应收账款的担保性让与和完全让与平等地适用规则,但未来应当赋予非基于保理的应收账款完全让与以登记能力。在“动产担保物与担保债权的类型”指标下,我国的失分点是抵押权的效力不能自动及于抵押物所生孳息,担保物权的效力不能自动及于担保物的转让对价,解决方案是通过当事人的合意实现动产担保物权效力的自动延伸。在“动产担保物权的优先顺位与权利实现”指标下,我国的问题是担保债权与税收债权的优先顺位需要更明确、合理的规则。虽然流担保契约的效力受到限制,但我国承认了担保物权庭外实现约定的效力。在“动产担保登记系统的运营”指标下,我国的动产融资统一登记公示系统能够最大程度地满足世行要求,但我国未来的制度设计须考虑到如何降低相对人的信息获取成本、避免担保权人虚假登记。

融资租赁交易的功能化,所着眼的是出租人与第三人的权利冲突如何协调、出租人的利益如何实现,其路径是将出租人所有权的物权保护与动产抵押权同等对待。在《民法典》体系下,应当依据案件客观事实,识别普通租赁与融资租赁,再适用具体的规则。普通动产的融资租赁应在动产融资统一登记公示系统进行登记,特殊动产的融资租赁登记则应继续在原有权利登记系统展开。在出租人与第三人就租赁物产生权利冲突时,应当依据《民法典》第745条,参照适用动产抵押权的登记对抗规则。至于出租人所有权与其他担保权的竞存顺位,则参照适用动产抵押权的顺位规则,并以《民法典》第416条作为特别适用的规则。在承租人违约后,出租人可以准用担保债权的实现路径,即在主张租金加速到期的前提下,可通过普通民事诉讼程序或“实现担保物权案件”的特别程序,同时主张就租赁物价值受偿。同时,出租人还可以依据《民法典》第752条,选择解除合同、收回租赁物。在承租人破产时,出租人无权行使破产取回权,但在其权利已登记时,可以申报担保债权,否则只能申报普通债权。
