Chronic renal failure(CRF) is a worldwide public health issue with poor outcomes and high *** years, some plant-derived medicines,such as rhubarb,revealed desirable prospects to treat ***,the safeness of rhubarb is controversial for the reported nephrotoxicity caused by the anthraquinones(AQs) contained in *** this study,the renal risk of rhubarb was investigated by comparatively evaluating renal protection and concurrent lesion in both CRF and normal animals with histopathologic,biochemical and toxicokinetic *** histopathologic results showed that there was evidence of a treatment-related renal protection of rhubarb on CRF rats and no evidence of a treatment-related renal lesion on normal *** was the main anthraquinones absorbed after oral administration of rhubarb and was intensively investigated to evaluate the *** same dosage,the area under curve(AUC) and maximal blood concentration of CRF groups were lower than those of normal *** the elimination in CRF groups was faster than normal *** results showed that there is no unambiguous evidence of renal toxicity of rhubarb no matter to normal or CRF rats successively administered for 3 weeks.

自身免疫性肝炎(autoimmune hepatitis,AIH)、原发性胆汁性胆管炎(primary biliary cholangitis,PBC)及其重叠综合征(overlap syndrome,OS)是临床常见的3种自身免疫性肝病,由于其发病机制的复杂性和混杂性,给临床准确区分诊断带来了挑战.本研究采用非靶向代谢组学方法,尝试对42例AIH、103例PBC和57例AIH-PBC OS患者血清筛选可辅助区分诊断3种肝病的代谢组学特征标志物.结果表明,在与AIH和PBC相关的所有变量中共鉴定出101个特征代谢物,并进一步筛选出富集于胆汁、磷脂和氨基酸代谢途径的29个代谢物,作为对AIH和PBC区分能力最强的指纹性代谢物.从指纹性代谢物中压缩降维得到的特征代谢物向量,显示出良好的区分诊断AIH与PBC的能力,受试者工作特征曲线下面积达到0.797(P<0.0001).基于指纹性代谢物主成分分析的可视化分布图,PBC和AIH样本呈现出3个区域:单纯PBC样本区、单纯AIH样本区和混合区.而将AIH-PBC OS样本放入该模型后发现,AIH-PBC OS样本在上述3个区域均有分布,提示目前诊断标准下的AIH-PBC OS患者实际上具有不同的代谢特征和病因倾向,需要更为精确的区分诊断.基于指纹性代谢物构建的区分诊断模型,可以辅助用于自身免疫性肝病患者的精确诊断和管理.

Intratumoral injection is the palliative treatment to get further improvements in survival and quality of life for advanced or recurrent carcinomas patients, or the patients with severe comorbidities or a poor performance status. In this study, a solvent-injection method was used to prepare paclitaxel–cholesterol complex loaded lecithin-chitosan nanoparticles(PTX-CH-loaded LCSPs) for intratumoral injection therapy, and the physicochemical properties of NPs were well characterized. Release behavior of PTX from PTX-CH-loaded LCSPs showed a pH-sensitive pattern. The result of cell uptake assay indicated that PTXCH-loaded LCSPs could effectively penetrate cells through the energy-dependent caveolae-mediated endocytosis and macropinocytosis via Golgi apparatus. Meanwhile, PTX-CH-loaded LCSPs had superior ability to induce cell apoptosis comparing with paclitaxel solution. The in vivo anti-tumor results suggested that PTX-CH-loaded LCSPs effectively inhibited mouse mammary cancer growth and metastasis to distant organs and significantly improved survival ratio of tumor-bearing mice by intratumoral administration. In general, our study demonstrated that PTX-CH-loaded LCSPs used for palliative treatment by intratumoral injection exhibited enhanced safety and antitumor efficacy, and provided an alternative choice in the field of palliative chemotherapy.
