Myotis bat STINg attenuates aging-related inflammation in female mice

Bats,notable as the only flying mammals,serve as natural reservoir hosts for various highly pathogenic viruses in humans(e.g.,SARS-CoV and Ebola virus).Furthermore,bats exhibit an unparalleled longevity among mammals relative to their size,particularly the Myotis bats,which can live up to 40 ***,the mechanisms underlying these distinctive traits remain incompletely *** our prior research,we demonstrated that bats exhibit dampened STINg-interferon activation,potentially conferring upon them the capacity to mitigate virus-or aging-induced *** substantiate this hypothesis,we established the first in vivo bat-mouse model for aging studies by integrating Myotis davidii bat STINg(MdSTINg)into the mouse *** monitored the genotypes of these mice and performed a longitudinal comparative transcriptomic analysis on MdSTINg and wild-type mice over a 3-year aging *** transcriptomic analysis indicated a reduction in aging-related inflammation in female MdSTINg mice,as evidenced by significantly lower levels of pro-inflammatory cytokines and chemokines,immunopathology,and neutrophil recruitment in aged female MdSTINg mice compared to aged wild-type mice in *** results indicated that MdSTINg knock-in attenuates the aging-related inflammatory response and may also improve the healthspan in mice in a sex-dependent *** the underlying mechanism awaits further study,this research has critical implications for bat longevity research,potentially contributing to our comprehension of healthy aging in humans.

Rare earth fluorescent nanoprobes with minimal side effects enable tumor microenvironment activation for chemotherapy

Chemotherapy,the use of antitumor drugs to kill cancer cells,is currently one of the most effective treatments for ***,serious toxic side effects caused by long-term drug accumulation can cause significant damage to the body,which limits the clinical application of antitumor *** this study,a novel RENPs@DOX-Fe nanoprobe(NP) was constructed by coating the surface of rare earth nanomaterials(NaLuF_(4):Yb,Er) with a complex formed by doxorubicin(DOX) and iron ion(Ⅲ).Due to the low toxicity of anthracycline-metal complexes,the damage to normal cells is *** unique acidic microenvironment in tumor cells facilitates the decomposition and gradual release of DOX from the DOX-Fe *** addition,the DOX-Fe complex can convert near-infrared(NIR) light into heat energy,which promotes the decomposition of the complex,further enhancing the release of DOX in the tumor *** change of ratio fluorescence of rare earth nanomaterials at 660 and 1550 nm after DOX release enables visual monitoring of drug release,which can potentially improve the chemotherapeutic *** vitro experiments established that RENPs@DOX-Fe NPs with NIR illumination had good therapeutic efficacy in *** work provides new insights into designing tumor microenvironment-responsive nanoprobes for chemotherapy with minimal side effects.

Bayesian inference of neutron-skin thickness and neutron-star observables based on effective nuclear interactions

We have obtained the constraints on the density dependence of the symmetry energy from neutron-skin thickness data by parityviolating electron scatterings and neutron-star observables using a Bayesian approach,based on the standard Skyrme-HartreeFock(SHF)model and its extension as well as the relativistic mean-field(RMF)*** the neutron-skin thickness data(neutron-star observables)mostly constrain the symmetry energy at subsaturation(suprasaturation)densities,they may more or less constrain the behavior of the symmetry energy at suprasaturation(subsaturation)densities,depending on the energy-density functional *** showing the final posterior density dependence of the symmetry energy,we also compare the slope parameters of the symmetry energy at 0.10 fm-3as well as the values of the symmetry energy at twice saturation density from three effective nuclear *** present work serves as a comparison study based on relativistic and non-relativistic energy-density functionals,for constraining the nuclear symmetry energy from low to high densities using a Bayesian approach.

Downscaling Seasonal Precipitation Forecasts over East Africa with Deep Convolutional Neural Networks

This study assesses the suitability of convolutional neural networks(CNNs) for downscaling precipitation over East Africa in the context of seasonal forecasting. To achieve this, we design a set of experiments that compare different CNN configurations and deployed the best-performing architecture to downscale one-month lead seasonal forecasts of June–July–August–September(JJAS) precipitation from the Nanjing University of Information Science and Technology Climate Forecast System version 1.0(NUIST-CFS1.0) for 1982–2020. We also perform hyper-parameter optimization and introduce predictors over a larger area to include information about the main large-scale circulations that drive precipitation over the East Africa region, which improves the downscaling results. Finally, we validate the raw model and downscaled forecasts in terms of both deterministic and probabilistic verification metrics, as well as their ability to reproduce the observed precipitation extreme and spell indicator indices. The results show that the CNN-based downscaling consistently improves the raw model forecasts, with lower bias and more accurate representations of the observed mean and extreme precipitation spatial patterns. Besides, CNN-based downscaling yields a much more accurate forecast of extreme and spell indicators and reduces the significant relative biases exhibited by the raw model predictions. Moreover, our results show that CNN-based downscaling yields better skill scores than the raw model forecasts over most portions of East Africa. The results demonstrate the potential usefulness of CNN in downscaling seasonal precipitation predictions over East Africa,particularly in providing improved forecast products which are essential for end users.

The Strategic Analysis of Demand Forecast-Sharing in a Hybrid-Format Online Platform Supply Chain

This paper examines the optimal forecast-sharing strategy in a hybrid-format online platform supply chain where a supplier sells a product through agency format and reselling format provided by a platform retailer who possesses demand forecasts from two *** asymmetry and co-opetitive relationship arise between the platform retailer and the supplier,which affect their operational decisions and the supply chain’s *** improve supply chain efficiency,we compare different forecast-sharing strategies(i.e.,no forecast sharing,sharing a single forecast,and sharing two forecasts),and analyze the effects of co-opetitive parameters on the optimal forecast-sharing *** analysis shows that forecast sharing is always beneficial to the supplier,and sharing two forecasts is more beneficial than sharing a single *** for the platform retailer and the whole supply chain,forecast sharing is beneficial only under certain conditions,depending on the co-opetitive *** optimal forecast-sharing strategy is the result of a combination of the negative effect of double marginalization in reselling channel and the positive effect of responding pricing to demand uncertainty in agency *** illustrate the parameter regions of the platform retailer’s voluntary sharing,contract sharing,and no sharing,and also find that higher channel competition intensity,higher market share of agency channel,and higher commission rate can promote the platform retailer’s voluntary *** study extends the research scope of demand forecast-sharing and sheds light on the decision-making processes for managing a hybrid-format online platform supply chain.
