Cyclic arginine-glycine-aspartic acid-modified red blood cells for drug delivery:Synthesis and in vitro evaluation

Red blood cells(RBCs)are an excellent choice for cell preparation research because of their biocompatibility,high drug loading,and long half-life.In this study,doxorubicin(DOX)was encapsulated with RBCs as the carrier.The biotin-avidin system binding principle was used to modify biotinylated cyclic arginine-glycine-aspartic acid(cRGD)onto RBC surfaces for accurate targeting,high drug loading,and sustained drug release.The RBC drug delivery system(DDS)was characterized,and the concentration of surface sulfur in the energy spectrum was 6.330%.The physical and chemical properties of RBC DDS were as follows:drug content,0.857 mg/mL;particle size,3339 nm;potential value,12.5 mV;and cumulative release rate,81.35%.There was no significant change in RBC morphology for up to seven days.The results of the targeting and cytotoxicity studies of RBC DDS showed that many RBCs covered the surfaces of U251 cells,and the fluorescence intensity was higher than that of MCF-7 cells.The IC50 value of unmodified drug-loaded RBCs was 2.5 times higher than that of targeted modified drug-loaded RBCs,indicating that the targeting of cancer cells produced satisfactory inhibition.This study confirms that the RBC DDS has the characteristics of accurate targeting,high drug loading,and slow drug release,which increases its likelihood of becoming a clinical cancer treatment in the future.

Let G be a simple graph and G~σ be the oriented graph with G as its underlying graph and orientation σ.The rank of the adjacency matrix of G is called the rank of G and is denoted by r(G).The rank of the skew-adjacency matrix of G~σ is called the skew-rank of G~σ and is denoted by sr(G~σ).Let V(G)be the vertex set and E(G) be the edge set of G.The cyclomatic number of G,denoted by c(G),is equal to |E(G)|-|V(G)|+ω(G),where ω(G) is the number of the components of G.It is proved for any oriented graph G~σ that-2c(G)≤sr(G~σ)-r(G)≤2c(G).In this paper,we prove that there is no oriented graph G~σwith sr(G~σ)-r(G)=2c(G)-1,and in addition,there are infinitely many oriented graphs G~σ with connected underlying graphs such that c(G)=k and sr(G~σ)-r(G)=2c(G)-l for every integers k,l satisfying 0 ≤l≤4k and l≠1.

The Impact of Tibetan Plateau Snow Cover on the Summer Temperature in Central Asia

The current work examines the impact of the snow cover extent(SCE)of the Tibetan Plateau(TP)on the interannual variation in the summer(June−July−August)surface air temperature(SAT)over Central Asia(CA)(SAT_CA)during the 1979−2019 period.The leading mode of the summer SAT_CA features a same-sign temperature anomalies in CA and explains 62%of the total variance in SAT_CA.The atmospheric circulation associated with a warming SAT_CA is characterized by a pronounced high-pressure system dominating CA.The high-pressure system is accompanied by warm advection as well as descending motion over CA,favoring the warming of the SAT_CA.Analysis shows that the interannual variation in the summer SAT_CA is significantly positively correlated with the April SCE over the central-eastern TP.In April,higher than normal SCE over the central-eastern TP has a pronounced cooling effect on the column of the atmosphere above the TP and can persist until the following early summer.Negative and positive height anomalies appear above and to the west of the TP.In the following months,the perturbation forcing generated by the TP SCE anomalies lies near the western center of the Asian subtropical westerly jet(SWJ),which promotes atmospheric waves in the zonal direction guided by the Asian SWJ.Associated with this atmospheric wave,in the following summer,a significant high-pressure system dominates CA,which is a favorable condition for a warm summer SAT_CA.

Max-min rate optimization for multi-user MISO-OFDM systems assisted by RIS with a wideband model

Reconfigurable intelligent surfaces(RISs)have the capability to change the wireless environment smartly Considering the attenuation of subchannels and crowding users involved in the wideband system,we introduce RISs into the multi-user multi-input single-output(MU-MISO)system with orthogonal frequency division multiplexing(OFDM)for performance enhancement.Maximizing the minimum rate of dense users in an MU-MISO-OFDM system assisted by RIS with an approximate practical model is formulated as the joint optimization problem involving subcarrier allocation,transmit precoding(TPC)matrices at the base station,and RIS passive beamforming.A coalition-game subcarrier allocation(CSA)algorithm is proposed to solve space–frequency resource allocation on subcarriers,which reforms the interference topology among dense users.Fractional programming and convex optimization method are used to optimize the TPC matrices and the RIS passive beamforming,which improves the spectral efficiency synthetically across all subchannels in the wideband system.Simulation results indicate that the CSA algorithm provides a significant gain for dense users.Besides,the proposed joint optimization method shows the considerable advantage of the RISs in the MU-MISO-OFDM system.

Effects of Electroacupuncture on Metabolic Changes in Motor Cortex and Striatum of 6-Hydroxydopamine-Induced Parkinsonian Rats

Objective:To explore the possible underlying mechanism by investigating the effect of electroacupuncture(EA)treatment on the primary motor cortex and striatum in a unilateral 6-hydroxydopamine(6-0HDA)induced rat Parkinson's disease(PD)model.Methods:Male Sprague-Dawley rats were randomly divided into sham group(n=16),model group(n=14),and EA group(n=14).EA stimulation at Dazhui(GV 14)and Baihui(GV20)was applied to PD rats in the EA group for 4 weeks.Behavioral tests were conducted to evaluate the effectiveness of EA treatment.Metabolites were detected by 7.0 T proton nuclear magnetic resonance.Results:Following 4 weeks of EA treatment in PD model rats,the abnormal behavioral impairment induced by 6-0HDA was alleviated.In monitoring changes in metabolic activity,ratios of myoinositol/creatine(Cr)and N-acetyl aspartate(NAA)/Cr in the primary motor cortex were significantly lower at the injected side than the non-injected side in PD rats(P=0.024 and 0.020).The ratios of glutamate+glutamine(Glx)/Cr and NAA/Cr in the striatum were higher and lower,respectively,at the injected side than the non-injected side(P=0.046 and 0.008).EA treatment restored the balance of metabolic activity in the primary motor cortex and striatum.In addition,the taurine/Cr ratio and GIx/Cr ratio were elevated in the striatum of PD model rats compared to sham-lesioned rats(P=0.026 and 0.000).EA treatment alleviated the excessive glutamatergic transmission by down-regulating the striatal Glx/Cr ratio(P=0.001).The Glx/Cr ratio was negatively correlated with floor plane spontaneous locomotion in PD rats(P=0.027 and P=0.0007).Conclusions:EA treatment is able to normalize the metabolic balance in the primary motor cortex and striatum of PD rats,which may contribute to its therapeutic effect on motor deficits.The striatal GIx/Cr ratio may serve as a potential indicator of PD and a therapeutic target of EA treatment.

Energy absorption characteristics of novel high-strength and hightoughness steels used for rock support

Nowadays,the development of novel metallic materials for rock support have attracted research interests since they can significantly improve the deformation and energy absorption capacities of rock bolts.Although previous studies proved the importance and mechanical advantages of utilizing high-strength and high-toughness(HSHT)steels in rock support,there is no systematic analysis to reveal the essential energy absorption parameter and the guidelines for further development of metallic rock support materials.This paper analyzes the energy absorption characteristics of novel HSHT steels(negative Poisson’s ratio(NPR)and twinning-induced plasticity(TWIP)steels)in comparison with conventional rock support materials.A physically based crystal plasticity(CP)model was set up and calibrated to study the effect of strain hardening rate(SHR).Meanwhile,the roles of underlying physical mechanisms,i.***.the dislocation density and twin volume fraction,were studied.The results show that the improvement of energy absorption density(EAD)is essential for further development of rock support materials,besides the increase of energy absorption rate(EAR)for previous development of conventional rock support materials.The increase of EAD requires increases of both strength and deformation capacity of materials.For HSHT steels,the decrease of SHR has a positive effect on the improvement of EAD.In addition,the increase of EAD is followed by the increase of twin volume fraction and the decrease of plastic Poisson’s ratio which can promote deformation plasticity of materials.Meanwhile,the increase of EAR is correlated with the accumulation of dislocation density,which can increase the strength of materials.This paper provides the theoretical basis and guidelines for developing rock support materials in deep underground engineering and other related fields.

Precision Milling of Integrated Turbine Based on a Non-Contact On-Machine Measurement System

The machining accuracy of the curved surfaces of integrated turbine blades directly determines the performance and service life of the turbojet engine system.In this paper,a non-contact on-machine measurement system is developed for precision milling of integrated turbines to reduce the impact of workpiece deformation,overcutting,tool chatter,and material work hardening.Milling with the on-machine measurement system obtained high-quality integrated turbine surfaces.The geometric accuracy error(PV)is below 3μm,and the surface roughness(Ra)is less than 2μm.The processed integrated turbine blade can achieve the accuracy requirements in the design and manufacturing and can be practically applied to the entire turbojet engine.

Enterococcus faecalis provides protection during scavenging in carrion crow (Corvus corone)

The gut microbiota significantly influences host physiology and provides essential ecosystem services.While diet can affect the composition of the gut microbiota,the gut microbiota can also help the host adapt to specific dietary habits.The carrion crow(Corvus corone),an urban facultative scavenger bird,hosts an abundance of pathogens due to its scavenging behavior.Despite this,carrion crows infrequently exhibit illness,a phenomenon related to their unique physiological adaptability.At present,however,the role of the gut microbiota remains incompletely understood.In this study,we performed a comparative analysis using 16S rRNA amplicon sequencing technology to assess colonic content in carrion crows and 16 other bird species with different diets in Beijing,China.Our findings revealed that the dominant gut microbiota in carrion crows was primarily composed of Proteobacteria(75.51%)and Firmicutes(22.37%).Significant differences were observed in the relative abundance of Enterococcus faecalis among groups,highlighting its potential as a biomarker of facultative scavenging behavior in carrion crows.Subsequently,E.faecalis isolated from carrion crows was transplanted into model mice to explore the protective effects of this bacterial community against Salmonella enterica infection.Results showed that E.faecalis down-regulated the expression of pro-inflammatory cytokines tumor necrosis factor alpha(TNF-α),interferon gamma(IFN-γ),and interleukin 6(IL-6),prevented S.enterica colonization,and regulated the composition of gut microbiota in mice,thereby modulating the host’s immune regulatory capacity.Therefore,E.faecalis exerts immunoregulatory and anti-pathogenic functions in carrion crows engaged in scavenging behavior,offering a representative case of how the gut microbiota contributes to the protection of hosts with specialized diets.

Value and prognostic factors of repeat hepatectomy for recurrent colorectal liver metastasis

Background:More than 50%of patients with colorectal cancer develop liver metastases.Hepatectomy is the preferred treatment for resectable liver metastases.This review provides a perspective on the utility and relevant prognostic factors of repeat hepatectomy in recurrent colorectal liver metastasis(CRLM).Data sources:The keywords“recurrent colorectal liver metastases”,“recurrent hepatic metastases from colorectal cancer”,“liver metastases of colorectal cancer”,“repeat hepatectomy”,“repeat hepatic resec-tion”,“second hepatic resection”,and“prognostic factors”were used to retrieve articles published in the PubMed database up to August 2020.Additional articles were identified by a manual search of references from key articles.Results:Despite improvements in surgical methods and perioperative chemotherapy,recurrence remains common in 37%-68%of patients.Standards or guidelines for the treatment of recurrent liver metastases are lacking.Repeat hepatectomy appears to be the best option for patients with resectable metastases.The commonly reported prognostic factors after repeat hepatectomy were R0 resection,carcinoembryonic antigen level,the presence of extrahepatic disease,a short disease-free interval between initial and repeat hepatectomy,the number(>1)and size(≥5 cm)of hepatic lesions,requiring blood transfusion,and no adjuvant chemotherapy after initial hepatectomy.The median overall survival after repeat hepatectomy ranged from 19.3 to 62 months,and the 5-year overall survival ranged from 21%to 73%.Chemotherapy can act as a test for the biological behavior of tumors with the goal of avoiding unnecessary surgery,and a multimodal approach involving aggressive chemotherapy and repeat hepatectomy might be the treatment of choice for patients with early recurrent CRLM.Conclusions:Repeat hepatectomy is a relatively safe and effective treatment for resectable recurrent CRLM.The presence or absence of prognostic factors might facilitate patient selection to improve short-and long-term outcomes.

The Immune Function of Keratinocytes in Anti-Pathogen Infection in the Skin

Keratinocytes,located in the outer part of human skin,are the main epidermal cell type and play an essential role in skins defense against infection.Besides creating a physical barrier between the environment and the internal body,keratinocytes exert powerful immune function in anti-pathogen infection in the skin.At the recognition stage,pattern recognition receptors(PRRs)expressed by keratinocytes sense pathogen-associated molecular patterns(PAMPs)existing in pathogens.Toll like receptors(TLRs)are the most important PRRs in keratinocytes.Other PRRs such as dectin-1 and nucleotide-binding oligomerization domain(NOD)-like receptors(NLRs)are also found to participate in this process.Activated PRRs enhance the secretion of cytokines,chemokines and the production of antimicrobial peptides(AMPs).Proinflammatory cytokines tumor necrosis factor-α,interleukin(IL)-1α,IL-6,IL-1βand IL-18,chemokines(C-X-Cmotif)ligand(CXCL)1,CXCL2,CCL20,CCL2 and IL-8,AMPs human β-defensin(HBD)2,HBD3 and LL37 are the main molecules expressed in this procedure.Thymic stromal lymphopoietin(TSLP),IL-36γ,IL-17 family member IL-17C and anti-inflammatory cytokine IL-10 can also be secreted.Some molecules produced by keratinocytes such as ribonuclease 5 and 7,S100 proteins own antimicrobial properties.Keratinocytes defense responses can be regulated by internal and external factors.This review summarizes recent advances on the innate immune function of keratinocytes against infection,promoting the finding of a new direction for avoiding severe skin infection as well as the potential treatment of keratinocyte-associated inflammatory dermatosis.
