Effect of Curcumol on NOD-Like Receptor Thermoprotein Domain 3 Inflammasomes in Liver Fibrosis of Mice

Objective:To investigate the effect of curcumol on NOD-like receptor thermoprotein domain 3(NLRP3)inflammasomes,and analyze the mechanism underlying curcumol against liver fibrosis.Methods:Thirty Kunming mice were divided into a control group,a model group and a curcumol group according to a random number table,10 mice in each group.Mice were intraperitoneally injected with 40% carbon tetrachloride(CCl4:peanut oil,2:3 preparation)at 5 m L/kg for 6 weeks,twice a week,for developing a liver fibrosis model.The mice in the control group were given the same amount of peanut oil,twice a week for 6 weeks.The mice in the curcumol group were given curcumol(30 m L/kg)intragastrically,and the mice in the model and control groups were given the same amount of normal saline,once a day for 6 weeks.Changes in liver structure were observed by hematoxylin and eosin(HE)and Masson staining.Liver function,liver fiber indices,and the expression of interleukin(IL)-10 and tumor necrosis factor-α(TNF-α)levels were determined by automatic biochemical analyzer and enzyme linked immunosorbent assay kit.Immunoblotting and reverse transcription-quantitative PCR(RT-qPCR)were performed to detect the expression of NLRP3 inflammasome-related molecules,TGF-β and collagen.Results:HE and Masson staining results showed that the hepatocytes of the model group were arranged irregularly with pseudo-lobular structure and a large amount of collagen deposition.The mice in the curcumol group had a significant decrease in liver function and liver fibers indices compared with the model group(Plusion:A potential anti-liver fibrosis mechanism of curcumol may be associated wi

Chronic hepatitis B virus(HBV)infection remains a major public health concern globally,and T cell responses are widely believed to play a pivotal role in mediating HBV clearance.Accordingly,research on the characteristics of HBV-specific T cell responses,from activation to exhaustion,has advanced rapidly.Here,we summarize recent developments in characterizing T cell immunity in HBV infection by reviewing basic and clinical research published in the last five years.We provide a comprehensive summary of the mechanisms that induce effective anti-HBV T cell immunity,as well as the latest developments in understanding T cell dysfunction in chronic HBV infection.Furthermore,we briefly discuss current novel treatment strategies aimed at restoring anti-HBV T cell responses.

Alzheimer's disease(AD)is affected by genetic factors.Polymorphisms in the glutathione S-transfe rase omega-1(Gsto1)gene have been shown by genetic correlation analyses performed in different ethnic populations to be genetic risk factors for AD.Gene expression profile data from BXD recombinant inbred mice were used in combination with genetic and bioinformatic analyses to chara cterize the mechanisms underlying regulation of Gstol variation regulation and to identify network membe rs that may contribute to AD risk or progression.Allele-specific assays confirmed that variation in Gstol expression is controlled by cis-expression quantitative trait loci.We found that Gstol mRNA levels were related to several central nervous system traits,such as glial acidic fibrillary protein levels in the caudate putamen,co rtical gray matter volume,and hippocampus mossy fiber pathway volume.We identified 2168 genes whose expression was highly correlated with that of Gsto1.Some genes were enriched for the most common neurodegenerative diseases.Some Gsto1-related genes identified in this study had previously been identified as susceptibility genes for AD,such as APP,Grin2 b,Ide,and Psenen.To evaluate the relationships between Gstol and candidate network members,we transfected astrocytes with Gstol siRNA and assessed the effect on putative downstream effecto rs.We confirmed that knockdown of Gstol had a significant influence on Pa2g4 expression,suggesting that Pa2g4 may be a downstream effector of Gstol,and that both genes intera ct with other genes in a network during AD pathogenesis.

20μm Li metal modified with phosphate rich polymer-inorganic interphase applied in commercial carbonate electrolyte

Li metal batteries are supposed to reach real application in order to fulfill the high-energy density requirement of energy storage system.Unfortunately,the commonly used carbonate electrolyte react with pristine Li,which result in short lifetime of lithium metal battery.To alleviate the side reactions of Li metal with liquid electrolyte,here we propose a phosphate rich polymer-inorganic layer as an interphase.Due to the inert properties of lithium phosphate derived from LiPO_(2)F_(2)and poly-ether,the side-reaction of carbonate solvent are prevented.As a result,lithium metal anode sustains for 800 cycles in symmetrical cell test under 1 m A cm^(-2).Even under strict condition(20μm Li,capacity ratio N/P=2.3,electrolyte/active material=3μL mg^(-1)),coin cell test still runs stable for 150 cycles with high Coulombic efficiency.Furthermore,both LiFePO_(4)and LiNi_(0.8)Co_(0.1)Mn_(0.1)O_(2)pouch cell under 5μL m A^(-1)h^(-1)condition also exhibit good stability at 0.5 C and 2 C rate.With this approach,high-energy and high-power Li metal batteries are approaching to real application in the near future.

Evolution of vortex structure around a wall-mounted rough hemisphere

In this paper,the large eddy simulation(LES)method is used to calculate Reynolds numbers the range in the range of 3000–30000(based on the diameter of the hemisphere D),and then the statistical analysis method is used to study the influence of Re on the characteristics of the flow field around the rough hemisphere and the evolution law of the tail vortex.The effect of the Reynolds number on the structure of the rough hemisphere flow field and the evolution of vortex structure is studied by vorticity distribution,POD method,and correlation analysis.It can be observed that with the increase of Reynolds number,the shear in the recirculating region of the rough hemisphere is enhanced,the large-scale structure is reduced,the small-scale structure is increased,and the angle between the hairpin vortex and the wall is reduced.λ_(ci)vortex identification and the Q-criterion are used to capture the evolution of a periodic vortex structure.Q-criterion gives the three-dimensional vortex structure behind the hemispherical flow.It is found that the horseshoe vortex and arch vortex generated by the incoming flow after passing through the hemisphere will interact with the wall surface to form a hairpin vortex,and the hairpin vortex structure will continue to evolve downstream to form a new vortex structure.

Functional analysis of UG7207D3 associated with abamectin resistance in Tetranychus cinnabarinus(Boisduval)

Uridine diphosphate(UDP)-glycosyltransferases(UGTS)are widely distributed within living organisms and share roles in biotransformation of various lipophilic endo-and xenobiotics with activated UDP sugars.In this study,it was found that the activity of UGTs in abamectin-resistant(AbR)strain was significantly higher(2.3-fod)than that in susceptible strain(SS)of Tetramychus cinnabarinus.Further analysis showed that 5-nitrouracil,the inhibitor ofUGTs,could enhance the lethal effect of abamectin on mites.From the previous microarray results,we found an UGT gene(UGT201D3)overexpressed in AbR strain.Quantitative PCR analysis showed that UGT20ID3 was highly expressed and more inducible with abamectin exposure in the AbR strain.After silencing the tran-scription of UGT201D3,the activity of UGTs was decreased and the susceptibility to abamectin was increased in AbR strain whereas it was not in sS.Furthermore,UGT201D3 gene was then succefully expressed in Escherichia coli.The recombinant UGT20ID3 exhibited a-naphthol activity(2.810.43 nmol mg protein/min),and the enzyme activity could be inhibited by abamectin(inhibitory concentration at 50%:57.50±3.54 μmol/L).High-performance liquid chromatography analysis demonstrated that the recombinant UGT201D3 could efctively deplete abamectin(15.77%±3.72%)incubating with 150 ug protein for 6 h.These results provided direct evidence that UGT201D3 was involved in abamectin resistance in T cinnabarinus.

Epigenetic regulation of energy metabolism in obesity

Obesity has reached epidemic proportions globally.Although modern adoption of a sedentary lifestyle coupled with energy-dense nutrition is considered to be the main cause of obesity epidemic,genetic preposition contributes significantly to the imbalanced energy metabolism in obesity.However,the variants of genetic loci identified from large-scale genetic studies do not appear to fully explain the rapid increase in obesity epidemic in the last four to five decades.Recent advancements of next-generation sequencing technologies and studies of tissue-specific effects of epigenetic factors in metabolic organs have significantly advanced our understanding of epigenetic regulation of energy metabolism in obesity.The epigenome,including DNA methylation,histone modifications,and RNA-mediated processes,is characterized as mitotically or meiotically heritable changes in gene function without alteration of DNA sequence.Importantly,epigenetic modifications are reversible.Therefore,comprehensively understanding the landscape of epigenetic regulation of energy metabolism could unravel novel molecular targets for obesity treatment.In this review,we summarize the current knowledge on the roles of DNA methylation,histone modifications such as methylation and acetylation,and RNA-mediated processes in regulating energy metabolism.We also discuss the effects of lifestyle modifications and therapeutic agents on epigenetic regulation of energy metabolism in obesity.

Transcriptome analysis of Porphyridium purpureum under salinities of 0 and 68

The most prominent biological characteristic of Porphyridium purpureum is their unique extracellular sulfated polysaccharides that plays an important protective role against salt stress.Adaptation to stress is associated with metabolic adjustments.However,the molecular mechanisms underlying such metabolic changes remain elusive.This study presents the fi rst transcriptome profi ling of P.purpureum.A total of 8019 assembled transcripts were identifi ed,annotated,and classifi ed into diff erent biological categories and pathways based on a BLAST analysis against various databases.The in-depth analysis revealed that many of the diff erentially expressed genes in P.purpureum under salinities of 68 and 0 involved polysaccharide metabolism.A comparison of the gene expression levels under diff erent salinities revealed that the polysaccharide synthetic pathway was signifi cantly upregulated under the 68 salinity condition.The increased expression of the FBP,pfkA,CS,pgm,USP,UGP2,galE,and MPI transcripts in the polysaccharide synthetic pathway and the increase in ATP2 and ATP6 transcripts in the energy metabolic pathway revealed the molecular mechanism of high-salt adaptation.This sequencing dataset and analysis could serve as a valuable resource to study the mechanisms involved in abiotic stress tolerance in Rhodophyta.

DNA hypermethylation is an epigenetic modification that plays a critical role in the oncogenesis of myelodysplastic syndromes(MDS).Aberrant DNA methylation represses the transcription of promotors of tumor suppressor genes,inducing gene silencing.Realgar(α-As_(4)S_(4))is a traditional medicine used for the treatment of various diseases in the ancient time.Realgar was reported to have efficacy for acute promyelocytic leukemia(APL).It has been demonstrated that realgar could efficiently reduce DNA hypermethylation of MDS.This review discusses the mechanisms of realgar on inhibiting DNA hypermethylation of MDS,as well as the species and metabolisms of arsenic in vivo.

Microstructure and secondary phases in epitaxial LaBaCo_2O_(5.5 + δ) thin films

Aberration-corrected scanning transmission electron microscopy was employed to investigate the microstructures and secondary phases in LaBaCo2O5.5＋δ（LBCO） thin films grown on SrTiO3 （STO） substrates. The as-grown films showed an epitaxial growth on the substrates with atomically sharp interfaces and orientation relationships of [100]LBCO//[100]STO and （001）LBCO//（001）STO. Secondary phases were observed in the films, which strongly depended on the sample fabrication conditions. In the film prepared at a temperature of 900 ℃, nano-scale CoO pillars nucleated on the substrate, and grew along the [001] direction of the film. In the film grown at a temperature of 1000 ℃, isolated nano-scale C0304 particles appeared, which promoted the growth of {111 } twinning structures in the film. The orientation relationships and the interfaces between the secondary phases and the films were illustrated, and the growth mechanism of the film was discussed.
