Pathogen-induced methylglyoxal negatively regulates rice bacterial blight resistance by inhibiting OsCDR1 protease activity

Xanthomonas oryzae pv.oryzae(Xoo)causes bacterial blight(BB),a globally devastating disease of rice(Oryza sativa)that is responsible for significant crop loss.Sugars and sugar metabolites are important for pathogen infection,providing energy and regulating events associated with defense responses;howev-er,the mechanisms by which they regulate such events in BB are unclear.As an inevitable sugar metabolite,methylglyoxal(MG)is involved in plant growth and responses to various abiotic stresses,but the underlying mechanisms remain enigmatic.Whether and how MG functions in plant biotic stress responses is almost completely unknown.Here,we report that the Xoo strain PXO99 induces OsWRKY62.1 to repress transcrip-tion of OsGLY Il genes by directly binding to their promoters,resulting in overaccumulation of MG.MG negatively regulates rice resistance against Pxo99:osglyll2 mutants with higher MG levels are more sus-ceptible to the pathogen,whereas OsGLYIl2-overexpressing plants with lower MG content show greater resistance than the wild type.Overexpression of OsGLYll2 to prevent excessive MG accumulation confers broad-spectrum resistance against the biotrophic bacterial pathogens Xoo and Xanthomonas oryzae pv.oryzicola and the necrotrophic fungal pathogen Rhizoctonia solani,which causes rice sheath blight.Further evidence shows that MG reduces rice resistance against PXO99 through CONSTITUTIVE DISEASE RESISTANCE 1(OsCDR1).MGmodifies the Arg97 residue of OsCDR1to inhibit its aspartic protease activ-ity,which is essential for OsCDR1-enhanced immunity.Taken together,these findings illustrate how Xoo promotes infection by hijacking a sugar metabolite in the host plant.

Background:Yiqi Peiyuan(YQPY)prescription,a composite prescription of traditional Chinese medicine,has been used to prevent or delay the continued deterioration of renal function after acute kidney injury(AKI)in some institutions and has shown considerable efficacy.Objective:This is the first randomized controlled trial to assess efficacy and safety of YQPY for improving short-term prognosis in adult patients with AKI.Design,setting,participants and interventions:This is a prospective,double-blind,multicenter,randomized,and placebo-controlled clinical trial.A total of 144 enrolled participants were randomly allocated to two groups according to a randomization schedule.Participants,caregivers and investigators assessing the outcomes were blinded to group assignment.Patients in the YQPY group received 36 g YQPY granules twice a day for 28 days.Patients in the placebo group received a placebo in the same dose as the YQPY granules.Main outcome measures:The primary outcome was the change in the estimated glomerular filtration rate(eGFR)between baseline and after 4 and 24 weeks of treatment.The secondary outcomes were the change of serum creatinine(Scr)level between baseline and after treatment,and the incidence of endpoint events,defined as eGFR increasing by more than 25%above baseline,eGFR>75 mL/min per1.73 m2or the composite endpoint,which was defined as the sum of patients meeting either of the above criteria.Results:Data from a total of 114 patients(59 in the YQPY group and 55 in the control group)were analyzed.The mean changes in eGFR and Scr in weeks 4 and 24 had no difference between the two groups.In further subgroup analysis(22 in the YQPY group and 31 in the control group),the mean change in eGFR after treatment for 4 weeks was 27.39 mL/min per 1.73 m2in the YQPY group and 5.78 mL/min per1.73 m2in the placebo group,and the mean difference between groups was 21.61 mL/min per 1.73 m2(P<0.001).Thirteen(59.1%)patients in the YQPY group and 5(16.1%)in the placebo group reached the composite endpoints(P=0.002).Du

BACKGROUND Cyclin-dependent kinase 9(CDK9)expression and autophagy in colorectal cancer(CRC)tissues has not been widely studied.CDK9,a key regulator of transcription,may influence the occurrence and progression of CRC.The expression of auto-phagy-related genes BECN1 and drug resistance factor ABCG2 may also play a role in CRC.Under normal physiological conditions,autophagy can inhibit tumorigenesis,but once a tumor forms,autophagy may promote tumor growth.Therefore,understanding the relationship between autophagy and cancer,partic-ularly how autophagy promotes tumor growth after its formation,is a key motivation for this research.AIM To investigate the relationship between CDK9 expression and autophagy in CRC,assess differences in autophagy between left and right colon cancer,and analyze the associations of autophagy-related genes with clinical features and prognosis.METHODS We collected tumor tissues and paracarcinoma tissues from colon cancer patients with liver metastasis to observe the level of autophagy in tissues with high levels of CDK9 and low levels of CDK9.We also collected primary tissue from left and right colon cancer patients with liver metastasis to compare the autophagy levels and the expression of BECN1 and ABCG2 in the tumor and paracarcinoma tissues.RESULTS The incidence of autophagy and the expression of BECN1 and ABCG2 were different in left and right colon cancer,and autophagy might be involved in the occurrence of chemotherapy resistance.Further analysis of the rela-tionship between the expression of autophagy-related genes CDK9,ABCG2,and BECN1 and the clinical features and prognosis of colorectal cancer showed that the high expression of CDK9 indicated a poor prognosis in colorectal cancer.CONCluSION This study laid the foundation for further research on the combination of CDK9 inhibitors and autophagy inhibitors in the treatment of patients with CRC.

Nasopharyngeal carcinoma(NPC),a highly invasive and metastatic cancer,is widely prevalent in South China,Southeast Asia,and North Africa.NPC patients with stages I and II are often asymptomatic.Therefore,NPC patients are usually diagnosed with advanced-stage(stages III and IV)at their first visit.Detection of NPC at early stage is the key to improve the survival of patients.Epstein-Barr virus(EBV)plays an essential role in NPC occurrence in endemic areas.EBV-based biomarkers,including EBV antibody in serum and EBV DNA in plasma,have been specifically proposed for NPC early detection.Herein,we summarize recent advances in biomarkers for the early detection of NPC.

Seismic migration and inversion are closely related techniques to portray subsurface images and identify hydrocarbon reservoirs.Seismic migration aims at obtaining structural images of subsurface geologic discontinuities.More specifically,seismic migration estimates the reflectivity function(stacked average reflectivity or pre-stack angle-dependent reflectivity)from seismic reflection data.On the other hand,seismic inversion quantitatively estimates the intrinsic rock properties of subsurface formulations.Such seismic inversion methods are applicable to detect hydrocarbon reservoirs that may exhibit lateral variations in the inverted parameters.Although there exist many differences,pre-stack seismic migration is similar with the first iteration of the general linearized seismic inversion.Usually,seismic migration and inversion techniques assume an acoustic or isotropic elastic medium.Unconventional reservoirs such as shale and tight sand formation have notable anisotropic property.We present a linearized waveform inversion(LWI)scheme for weakly anisotropic elastic media with vertical transversely isotropic(VTI)symmetry.It is based on two-way anisotropic elastic wave equation and simultaneously inverts for the localized perturbations(ΔVp_(0)/Vp_(0)/Vs_(0)/Vs_(0)/,Δ∈,Δδ)from the long-wavelength reference model.Our proposed VTI-elastic LWI is an iterative method that requires a forward and an adjoint operator acting on vectors in each iteration.We derive the forward Born approximation operator by perturbation theory and adjoint operator via adjoint-state method.The inversion has improved the quality of the images and reduces the multi-parameter crosstalk comparing with the adjoint-based images.We have observed that the multi-parameter crosstalk problem is more prominent in the inversion images for Thomsen anisotropy parameters.Especially,the Thomsen parameter is the most difficult to resolve.We also analyze the multi-parameter crosstalk using scattering radiation patterns.The linearized waveform inversion for VTI-el

Detection of Novel BEST1 Variations in Autosomal Recessive Bestrophinopathy Using Third-generation Sequencing

Objective:Autosomal recessive bestrophinopathy(ARB),a retinal degenerative disease,is characterized by central visual loss,yellowish multifocal diffuse subretinal deposits,and a dramatic decrease in the light peak on electrooculogram.The potential pathogenic mechanism involves mutations in the BEST1 gene,which encodes Ca2+-activated Cl−channels in the retinal pigment epithelium(RPE),resulting in degeneration of RPE and photoreceptor.In this study,the complete clinical characteristics of two Chinese ARB families were summarized.Methods:Pacific Biosciences(PacBio)single-molecule real-time(SMRT)sequencing was performed on the probands to screen for disease-causing gene mutations,and Sanger sequencing was applied to validate variants in the patients and their family members.Results:Two novel mutations,c.202T>C(chr11:61722628,p.Y68H)and c.867+97G>A,in the BEST1 gene were identified in the two Chinese ARB families.The novel missense mutation BEST1 c.202T>C(p.Y68H)resulted in the substitution of tyrosine with histidine in the N-terminal region of transmembrane domain 2 of bestrophin-1.Another novel variant,BEST1 c.867+97G>A(chr11:61725867),located in intron 7,might be considered a regulatory variant that changes allele-specific binding affinity based on motifs of important transcriptional regulators.Conclusion:Our findings represent the first use of third-generation sequencing(TGS)to identify novel BEST1 mutations in patients with ARB,indicating that TGS can be a more accurate and efficient tool for identifying mutations in specific genes.The novel variants identified further broaden the mutation spectrum of BEST1 in the Chinese population.

Hepatitis B virus(HBV)produces and releases various particle types,including complete virions,subviral particles with envelope proteins,and naked capsids.Recent studies demonstrate that HBV exploits distinct intracellular membrane trafficking pathways,including the endosomal vesicle trafficking and autophagy pathway,to assemble and release viral and subviral particles.Herein,we summarize the findings about the distinct roles of autophagy and endosomal membrane trafficking and the interaction of both pathways in HBV replication,assembly,and release.

Role of phosphorus on the biogeochemical behavior of cadmium in the contaminated soil under leaching and pot experiments

Phosphorus(P)is involved in various biochemical reactions in plant growth,so it is beneficial to plants growing in soils contaminated by metals,including cadmium(Cd).However,few studies have reported on the mechanistic roles of P in mitigating Cd toxicity to ryegrass root,and especially in alleviating the disruption of the mitochondrial function of living cells.In this study,the physiological and biochemical mechanisms associated with ryegrass growth under various Cd and P treatments were investigated using leaching and pot systems.The concentration of Cd in soil leachates showed a significantly positive relationship with redox potential(Plues and dissolved organic carbon(DOC),indicating that exogenous P addition(as H_(2)PO_(4)*)may decrease Cd leaching from contaminated soil.Compared to the control(without P addition),the cumulative Cd content was reduced by 53.3%and 64.5%in the soil leachate with exogenous P application(20 mg/L and 80 mg/L),respectively.Notably,application of P decreased the Cd concentrations in the symplastic fractions and increased the Cd concentrations in the apoplastic fractions in root tips,which may help to alleviate Cd stress to the protoplast.Moreover,exogenous P was found to play a positive role in mitochondrial function and Ca^(2+)variation in root cells under Cd stress,which provides novel insights into the mechanisms of exogenous P in alleviating plant Cd injury.

From non-carbon host toward carbon-free lithium-sulfur batteries

Lithium-sulfur(Li-S)batteries with advantages of high energy densities(2600 Wh·kg^(-1)/2800 Wh·L^(-1))and sulfur abundance are regarded as promising candidates for next-generation high-energy batteries.However,the conventional carbon host used in sulfur cathodes suffers from poor chemical adsorption towards Li-polysulfides(LPS)in liquid electrolyte and sluggish redox kinetics,leading to low capacity and rate capability.Besides,carbon host used in Li metal anode with the intrinsic property of poor lithiophilicity and high Li-nucleation barrier gives rise to uncontrollable dendrite growth and further battery failure.Therefore,non-carbon hosts with chemical adsorption toward LPS and catalytic activity for accelerating LPS redox conversion as well as lithiophilic property for guiding uniform Li deposition are proposed and demonstrated a high efficiency in both sulfur cathodes and Li metal anodes.In this review,the principle and challenges of Li-S batteries are first presented,then recent work using non-carbon hosts in Li-S batteries is summarized comprehensively,and the mechanism of non-carbon host in improving sulfur utilization and stabilizing Li metal anode is discussed in detail.Furthermore,remaining challenges and outlook on the implementation of non-carbon host for practical carbon-free Li-S batteries are also provided.

Origin and evolution of false brinelling and fretting corrosion under rolling conditions

Rolling bearings working in oscillating applications often suffer from false brinelling and fretting corrosion that limit their service life.At present,the study of oscillation-induced failures is primarily dominated by a large number of sliding and bench tests.However,most of the previous work are not able to fully describe the two closely related but different failure mechanisms.In this study,the origin and evolution of the two failures are thoroughly investigated under strictly controlled oscillating rolling conditions.The results show that failure evolves from false brinelling to fretting corrosion under dry friction conditions regardless of the oscillation amplitude.This process is accompanied by a large area peeling and serious oxidation,and the oxygen content increases from 2.50 wt%to 21.22 wt%.Oxidation is a crucial factor in the evolution of false brinelling into fretting corrosion.However,the distribution of microwear and oxygen content depend on the oscillation amplitude.Under lubrication conditions,the friction coefficient(COF)is smaller in the initial stage and led to local slip or transient instability of rolling elements.Therefore,the size of the wear marks and COF increased and appeared to be larger than those under a dry friction condition.However,grease can separate the contact surfaces,only surface deformation owing to false brinelling and slight damage at the roughness level occurred.The evolution of false brinelling and fretting corrosion is also closely related to residual stress.False brinelling often occurrs with residual stress accomulation,meanwhile,the residual stress increases from 120 to 300 MPa.When peeling occurs,residual stress is released.The failure pattern transformed from false brinelling to fretting corrosion and is accompanied by oxidation.So far,none of investigation is able to show satisfactory evolution of oscillating-induced failures,this study may contribute to more scientific understanding of rolling bearings against long-run reciprocating oscillating wear.
