PbrARF4 contributes to calyx shedding of fruitlets in ‘Dangshan Suli’ pear by partly regulating the expression of abscission genes

Fruitlet calyx shedding in pear plants is apparently regulated via numerous pathways that involve both environmental triggers and phytohormones cues such as auxin. In this study, we found at 10 days after full bloom (DAFB) higher levels of indoleacetic acid (IAA) and tryptophan (Trp) in calyx persistence fruitlet (CPF) than calyx shedding fruitlet (CSF) ofDanshan Suli’ pear (Pyrus bretschneideri Rhed.). Consisting with this, the activity of indolealdehyde oxidase (IAAIdO), which promotes IAA synthesis, was remarkably increased, and that of peroxidase(POD), which degrades IAA, dropped markedly in CPF but not in CSF. Further, qRT-PCR results revealed that most of 31 PbrARFs (encoding auxin response factors) in Pyrus bretschneideri were highly expressed in CPF, whereas PbrARF4, PbrARF24 and PbrARF26 were significantly downregulated in CPF vis-a-vis CSF. Phylogenetic analysis revealed that 6 PbrARFs clustered in the group III, where PbrARF4 showed the closest affinity with AtARF1 that promotes organ abscission, indicating a putative role of PbrARF4 in mediating the process of calyx shedding in pear. In fact, the ectopic overexpression of PbrARF4 in Solanum lycopersicum resulted in an earlier-formed and deeper abscission layer (AL) in the transgenic plants, whose calyxes were more prone to wilt at the mature red stage (MR) compared with the control plants (wild-type). More importantly, expression levels of the abscission genes SILS and Sl Cel2 in transgenic plants overexpressing PbrARF4 were significantly upregulated in comparation with the WT, whereas those of Sl BI and Sl TAPG2 were considerably inhibited. Further, PbrJOINTLESS and PbrIDA,the two genes related to calyx shedding in pear, were up-regulated more in CSF than CPF. The findings contribute to a better understanding of PbrARFs involved in fruitlet calyx shedding of pear, which could prove beneficial to improving the quality of pear fruit.

Hexokinase 1 and 2 mediates glucose utilization to regulate the synthesis of kappa casein via ribosome protein subunit 6 kinase 1 in bovine mammary epithelial cells

Glucose plays a vital part in milk protein synthesis through the mTOR signaling pathway in bovine mammary epithelial cells (BMEC). The objectives of this study were to determine how glucose affects hexokinase (HK) activity in BMEC and investigate the regulatory effect of HK in kappa casein (CSN3) synthesis via the mechanistic target of rapamycin complex 1 (mTORC1) signaling pathway in BMEC. For this,HK1 andHK2 were knocked out in BMEC using the CRISPR/Cas9 system. The gene and protein expression, glucose uptake, and cell proliferation were measured. We found that glucose uptake, cell proliferation,CSN3 gene expression levels, and expression of HK1 and HK2 increased with increasing glucose concentrations. Notably, glucose uptake was significantly reduced inHK2 knockout (HK2KO) BMEC treated with 17.5 mM glucose. Moreover, under the same glucose treatment conditions, the proliferative ability and abundance of CSN3 were significantly diminished in bothHK1 knockout (HK1KO) and HK2KO BMEC compared with that in wild-type BEMC. We further observed that the phosphorylation levels of ribosome protein subunit 6 kinase 1 (S6K1) were reduced in HK1KO and HK2KO BMEC following treatment with 17.5 mM glucose. As expected, the levels of glucose-6-phosphate and the mRNA expression levels of glycolysis-related genes were decreased in both HK1KO and HK2KO BMEC following glucose treatment. Combining network pharmacology， molecular docking， and experimental verification to investigate the anti-rheumatoid arthritis effect and mechanism of Baeckea frutescens leaves

The aromatic Baeckea frutescens leaves (BFL) known as “Gang-Song-Cha” is a popular drink (Tea) in South China. To explore the healthcare value， a systematic and in-depth study on BFL alleviating rheumatoid arthritis (RA) was carried out by integrating network pharmacology， molecular docking， and experimental verification. The efficacy was first evaluated， and the BFL extract could ameliorate RA by inhibiting articular swelling， lowering immune organ index， improving the ankle pathologies， and regulating serum levels of TNF-α， IL-6， and IL-10 in CIA rats. Then the anti-RA mechanism of BFL was investigated by leveraging the high-content and active component 5-hydroxy-7-methoxy-2-isopropylchromone (Chro.)， and the potential signaling pathway and related target proteins for Chro. acting on RA were analyzed by KEGG enrichment and docking simulation. Finally， the experimental validations were conducted on RA-FLS cells， and compound Chro. could inhibit cell invasion and migration， block cell cycle at G2/M phase and induce apoptosis， and reduce the secretion of inflammatory cytokines by suppressing the PI3K-Akt pathway. These results provided pharmacological basis for the folk usage of “Gang-Song-Cha (Tea)” for intervening RA.

Polyoxometalate-based nanocomposites with electrocatalytic activity have been applied in hydrogen evolution reactions(HER).Seawater as the main water resource on the earth should be developed as the water electrolysis to prepare high-purity *** this paper,we used two synthesis strategies to prepare the nanocomposite Co_(4)-POM@Co-PGDY(Co_(4)-POM:the Kegging-type microcrystals of K_(10)[Co_(4)(PW_(9)O_(3)4)2]and Co-PGDY:cobalt-porphyrin linked graphdiyne)with excellent activity for ***-PGDY as the porous material is applied not only as the protection of microcrystals towards the metal ion in seawater but also as the co-electrocatalyst of Co_(4)-***_(4)-POM@Co-PGDY exhibits excellent HER performance in seawater electrolytes with low overpotential and high stability at high ***,we have observed a key H_(3)O+intermediate emergence on the surface of nanocomposite during hydrogen evolution process in seawater by Raman synchrotron radiation-based Fourier transform infrared(SR-FTIR).The results in this paper provide an effective strategy for preparing polyoxometalate-based electrocatalysts with high-performance toward hydrogen evolution reaction.

The response and anti-stress mechanisms of nitrifying sludge under long-term exposure to Cd Se quantum dots

The wide application of CdSe quantum dots(QDs)increases its stress risk to sewage treatment *** study evaluated the response of nitrification performance,floc characteristics and microbial community of nitrifying sludge under long-term exposure to CdSe *** showed CdSe QDs(≥1 mg/L)would decrease the activity of ammonia monooxygenase(AMO).Under the stress of 30 mg/L CdSe QDs,the activity of AMO was reduced by 66%,while the activities of hydroxylamine oxidase and nitrite oxidoreductase were enhanced by 19.1%and 26%,***,the final nitrification effects were not adversely affected,and the production rates of NO_(2)^(-)-N and NO_(3)^(-)-N were ***,CdSe QDs improved biomass concentration in sludge and maintained the stability of sludge *** throughput sequencing analysis showed that CdSe QDs evidently reduced the abundance and diversity of microbial community in nitrifying *** abundances of amino acid metabolism and lipid metabolism were ***,CdSe QDs decreased the fluorescence intensity of tryptophan-like protein from 2,326 to 1,179 *** loosely bound extracellular polymeric substances(EPSs)and from 3,792 to 3,117 *** tightly bound *** relieve CdSe QD stress,the polysaccharide content increased from0.31 to 0.61 mg/g MLSS and intracellular antioxidant defense was *** CdSe QD level increasing to 30 mg/L,the total antioxygenic capacity and the activities of catalase were enhanced up to 411%and 143.2%,***,CdSe QDs had little adverse effects on cell membrane integrity,microbial metabolism and the abundance of Nitrospirae.

Dear Editor,Extramammary Paget disease(EMPD)is a rare adenocarcinoma that originates in the apocrine gland-rich skin and usually affects the perineal skin(primary EMPD)or represents the intraepithelial spread of an underlying visceral carcinoma(secondary EMPD).Approximately half of EMPD cases are confined to the epidermis,which was defined as intraepithelial EMPD.

Metallic Sb-stabilized porous silicon with stable SEI and high electron/ion conductivity boosting lithium-ion storage performance

Silicon （Si） has mild discharge potential and high theoretical capacity,making it a highly desirable material for lithium-ion batteries （LIBs）.Nevertheless,the excessive volume expansion,poor ion/electron conductiv ity and unstable solid electrolyte interface （SEI） hinde practical application to ***,the metallic antimony（Sb） stabilized porous Si （SiDSb） composite was prepared by magnesiothermic reduction of Sb2O3and Mg2Si and chemical etching to remove the by-product of *** highly conductive Sb nanodots embedded in the Si liga ments promote not only the formation of conductive and stable LiF-rich SEI,but also the electron/ion transpor ability of *** to the outstanding bulk/interface stability,excellent conductivity,as well as ideal porous structure,the SiDSb electrode demonstrates a capacity of820 mAh·g-1after undergoing 320 turns at 1000 mA·***,it exhibits a stable capacity of 675 m Ah·g-1when tested at a higher current density of 5000 m A·*** results reveal a viable solution to solve three problems at the same time,namely the poor conductivity,inferior SEI and excessive volume expansion of Si,boding well for the design of Si-based materials for high-energy LIBs.
