Demethylase-assisted site-specific detection of n^(1)-methyladenosine in RnA

The dynamic RnA modifications have been viewed as new posttranscriptional regulator in modulating gene expression as well as in a broad range of physiological processes.n^(1)-methyladenosine(m^(1)A)is one of the most prevalent modifications existing in multiple types of ***-depth investigation of the functions of m^(1)A requires the site-specific assessment of m^(1)A stoichiometry in ***,we established a demethylase-assisted method(DA-m^(1)A)for the site-specific detection and quantification of m^(1)A in RnA.n^(1)-methyl group in m^(1)A could result in the stalling of reverse transcription at m^(1)A site,thus producing the truncated *** AlkB is a demethylase that can demethylate m^(1)A to produce adenine in RnA,thus generating full-length cDnA from AlkB-treated *** of the produced amounts of full-length cDnA by quantitative real-time PCR can achieve the site-specific detection and quantification of m^(1)A in *** the DA-m^(1)A method,we examined and successfully confirmed the previously well-characterized m^(1)A sites in various types of RnAs with low false positive *** addition,we found that the level of m^(1)A was significantly decreased at the bromodomain containing 2(BRD2)mRnA position 1674 and CST telomere replication complex component 1(CTC1)mRnA position 5643 in human hepatocellular carcinoma *** results suggest that these two m^(1)A sites in mRnA may be involved in liver *** together,the DA-m^(1)A method is simple and enables the rapid,cost-effective,and site-specific detection and quantification of m^(1)A in RnA,which provides a valuable tool to decipher the functions of m^(1)A in human diseases.

This study evaluated the effects of flavonoids from mulberry leaves(FML)on plasma biochemical indices,serum activities of lipid metabolism-related enzymes,fat morphology,fatty acid composition,and lipid metabolism in different adipose tissues of finishing *** used 120 Chinese hybrid barrows of Berkshire and Bama mini-pigs with an average initial body weight of 45.11±4.23 *** pigs were randomly assigned to five treatment groups and fed a control diet based on corn,soybean meal,and wheat bran or a control diet supplemented with 0.02%,0.04%,0.08%,or 0.16%*** experimental group had six replicates(pens),with four pigs per *** a 7-d adaptation period,the feeding trial was conducted for 58 *** and adipose tissue samples were collected from 30 pigs(one pig per pen)at the end of the *** results showed that FML supplementation significantly decreased the feed intake to body gain ratio,the plasma concentrations of total cholesterol and free fatty acids,and the serum activity of 3-hydroxy-3-methylglutaryl coenzyme A reductase(linear or quadratic effects,Pnd decreased the plasma triglyceride concentration(quadratic,P=0.07).Increasing FML supplementation increased the average daily gain and serum activities of lipoprotein lipase(linear and quadratic effects,Pnd adipose triglyceride lipase(linear,Pntation decreased the adipocyte area in the dorsal subcutaneous adipose(DSA)tissue of finishing pigs(linear,P=0.05)and increased the adipocyte area in the visceral adipose tissue(quadratic,Pncreasing FML supplementation decreased the C20:1 content in DSA,abdominal subcutaneous adipose,and visceral adipose tissues of finishing pigs(Pnd increased the C18:3n3 and n-3 PUfa contents(Pnes were regulated by the PPARγ-LXRa-ABCA1 signaling pathway,and their expressions differed in different adipose *** findings suggest that FML improved growth performance,regulated lipid met

Perioperative outcomes of transvaginal specimen extraction laparoscopic total gastrectomy and conventional laparoscopicassisted total gastrectomy

Tracing nitrate sources in one of the world's largest eutrophicated bays(Hangzhou Bay):insights from nitrogen and oxygen isotopes

Eutrophication caused by inputs of excess nitrogen(n) has become a serious environmental problem in Hangzhou Bay(China),but the sources of this nitrogen are not well *** this study,the August 2019 distributions of salinity,nutrients [nitrate(nO_(3)^(-)),nitrite,ammonium,and phosphate],and the stable isotopic composition of nO_(3)^(-)(δ^(15)n and δ^(18)O) were used to investigate sources of dissolved inorganic nitrogen(DIn) to Hangzhou B *** distributions of nitrate,salinity,and nitrate δ^(18)O indicate that the Qiantang River,the Changji.ng River,and nearshore coastal waters may all contribute nitrate to the *** on the isotopic compositions of nitrate in these potential source waters and conservative mixing of nitrate in our study area,we suggest that the nO_(3)^(- )in Hangzhou B ay was likely derived mainly from soils,synthetic n fertilizer,and manure and ***-member modeling indicates that in the upper half of the bay,the Qiantang River was a very important DIn source,possibly contributing more than 50% of DIn in the bay head *** the lower half of the bay,DIn was sourced mainly from strongly intruding coastal *** coming directly from the Changji.ng River made a relatively small contribution to Hangzhou Bay DIn in August 2019.
