AIM: To observe the biologic behavior of pancreatic cancer cells in vitro and in vivo, and to explore the potential value of angiostatin gene therapy for pancreatic ***: The recombinant vector pcDNA3.1(+)-angiostatin was transfected into human pancreatic cancer cells PC-3 with Lipofectamine 2000, and paralleled with the vector and mock control. Angiostatin transcription and protein expression were determined by immunofluorescence and Western blot. The stable cell line was selected by G418. The supernatant was collected to treat endothelial cells. Cell proliferation and growth in vitro were observed under microscope. Cell growth curves were *** troms-fected or untroms-fected cells overexpressing angiostatin vector were implanted subcutaneously into nude mice. AIM： To explore the influence of angiostatin up-regulation on the biologic behavior of gallbladder carcinoma cells in vitro and in vitro, and the potential value of angiostatin gene therapy for gallbladder carcinoma. METHODS： A eukaryotic expression vector of pcDNA3.1（＋） containing murine angiostatin was constructed and identified by restriction endonuclease digestion and sequencing. The recombinant vector pcDNA3.1-angiostatin was transfected into human gallbladder carcinoma cell line GBC-SD with Upofectamine 2000, and paralleled with the vector and mock control. The resistant clone was screened by G418 filtration. Angiostatin transcription and protein expression were examined by RT-PCR, immunofluorescence and Western-blot. The supernatant was collected to treat endothelial cells. Cell proliferation and growth in vitro were observed under microscope. RESULTS： Murine angiostatin cDNA was successfully cloned into the eukaryotic expression vector pcDNA3.1 （＋）. AFter 14 d of transfection and selection with G418, macroscopic resistant cell cloning was formed in the experimental group transfected with pcDNA 3.1（＋）-angiostatin and vector control. But untreated cells died in the mock control. Angiostatin was detected by RT-PCR and protein expression was detected in the experimental group by immunofluorescence and Western-blot. Cell proliferation and growth in v/tro in the three groups were observed respectively under microscope. No significant difference was observed in the growth speed of GBC- SD cells between groups that were transfected with and without angiostatin. After treatment with supernatant, significant differences were observed in endothelial cell （ECV-304） growth in vitro. The cell proliferation and growth were inhibited. CONCLUSION： Angiostatin does not directly inhibit human gallbladder carcinoma cell proliferation and growth in vitro, but the secretion of angiostatin inhabits endothelial cell proliferation and growth.

Cadmium （Cd） is a reproductive toxicant that induces germ cell apoptosis in the testes. Previous studies have demonstrated that endoplasmic reticulum （ER） stress is involved in Cd-induced germ cell apoptosis. The aim of the present study was to investigate the effects of N-acetylcysteine （NAC）, an antioxidant, on Cd-induced ER stress and germ cell apoptosis in the testes. Male CD-1 mice were intraperitoneally injected with CdCl2 （2.0 mg kg^-1）. As expected, acute Cd exposure induced germ cell apoptosis in the testes, as determined by terminal dUTP nick-end labelling （TUNEL）. However, the administration of NAC alleviated Cd-induced germ ceil apoptosis in the testes. Further analysis showed that NAC attenuated the Cd-induced upregulation of testicular glucose-regulated protein 78 （GRP78）, an important ER molecular chaperone. Moreover, NAC inhibited the Cd-induced phosphorylation of testicular eukaryotic translation initiation factor 2a （elF2a）, a downstream target of the double-stranded RNA-activated kinase-like ER kinase （PERK） pathway. In addition, NAC blocked the Cd-induced activation of testicular X binding protein （XBP）-1, indicating that NAC attenuates the Cd-induced ER stress and the unfolded protein response （UPR）. Interestingly, NAC almost completely prevented the Cd-induced elevation of C/EBP homologous protein （CHOP） and phosphorylation of c-Jun N-terminal kinase （.INK）, two components of the ER stress-mediated apoptotic pathway. In conclusion, NAC protects against Cd-induced germ cell apoptosis by inhibiting endoplasmic reticulum stress in the testes.

Growth Mechanism of Vertically Aligned Carbon Nanotube Arrays

Vertically aligned multi-walled carbon nanotube arrays grown on quartz substrate are obtained by co-pyrolysis of xylene and ferrocene at 850 °C in a tube furnace. Raman spectroscopy and high resolution transmission electron microscopy measurements show that the single-walled carbon nanotubes are only present on top of vertically aligned multi-walled carbon nanotube arrays. It has been revealed that isolated single-walled carbon nanotubes are only present in those floating catalyst generated materials. It thus suggests that the single-walled carbon nanotubes here are also generated by floating catalyst. Vertically aligned carbon nanotube arrays on the quartz substrate have shown good orientation and good graphitization. Meanwhile, to investigate the growth mechanism, two bi-layers carbon nanotube films with different thickness have been synthesized and analyzed by Raman spectroscopy. The results show that the two-layer vertically aligned carbon nanotube films grow “bottom-up”. There are distinguished Raman scattering signals for the second layer itself, surface of the first layer, interface between the first and second layer, side wall and bottom surface. It indicates that the obtained carbon nanotubes follow the base-growth mechanism, and the single-walled carbon nanotubes grow from their base at the growth beginning when iron catalyst particles have small size. Those carbon nanotubes with few walls (typically <5 walls) have similar properties, which also agree with the base-growth mechanism.

A composite material model for investigation of micro-fracture mechanism of brittle rock subjected to uniaxial compression

A two phase model of rock was proposed in order to investigate the mechanism of brittle fracture due to uniaxial compression, in which rock was considered to be a composite material consisting of hard grains and colloids. The stress state in colloid region near grains was calculated using Finite Element Method (FEM). The influence of the tensile stresses on the crack initiation and failure process of brittle rock subjected to uniaxial compression was investigated by numerical experiments. The FE results show that tensile stresses are induced easily in the neighboring area of hard grains with the maximum value near grain boundaries. The distribution of tensile stresses depends on the relative position of hard grains. The cracks initiated just near the boundary area of hard grains, which was governed by tensile stress. These results clearly reveal the micro fracture mechanism of brittle rock loaded by uniaxial compression. It can be concluded that the failure mode of brittle rock under uniaxial compression is still tensile fracture from the point view of microstructure. However, since the wide colloid region is still under compressive stress state, further propagation of boundary cracks through this region obviously needs more external load, thus causing the uniaxial compressive strength of rock much higher than its tensile strength obtained via Brazilian (splitting)

Effect of specimen thickness on Mode Ⅱ fracture toughness of rock

Anti symmetric four point bending specimens with different thickness, without and with guiding grooves, were used to conduct Mode Ⅱ fracture test and study the effect of specimen thickness on Mode Ⅱ fracture toughness of rock. Numerical calculations show that the occurrence of Mode Ⅱ fracture in the specimens without guiding grooves (when the inner and outer loading points are moved close to the notch plane) and with guiding grooves is attributed to a favorable stress condition created for Mode Ⅱ fracture, i.e. tensile stress at the notch tip is depressed to be lower than the tensile strength or to be compressive stress, and the ratio of shear stress to tensile stress at notch tip is very high. The measured value of Mode Ⅱ fracture toughness K ⅡC decreases with the increase of the specimen thickness or the net thickness of specimen. This is because a thick specimen promotes a plane strain state and thus results in a relatively small fracture toughness.
