High-density lipoprotein （HDL） serves as a natural nanoparticle with compositional and functional heterogeneity and contributes to the maintenance of lipid metabolism and various biological functions. HDL also transports endogenous microRNAs, vitamins, hormones, and proteins through blood and interstitial fluids to various organs. These unique and diverse features of HDL have encouraged its applications for the transport of therapeutics and diagnostic reagents in the last decade. In this review, we describe the heterogeneous characteristics and biological functions of HDL and highlight HDL mimetic approaches, including apolipoprotein mimetic peptides and reconstituted HDL nanoparticles. Given the potential of HDL as a nanocarrier delivering various therapeutic agents, we discuss the current representative studies of HDL mimetic nanotherapeutics for cardiovascular and neurodegenerative diseases and analyze the current challenges and future perspective.

Background:Immune checkpoint inhibitors(ICIs)are effective in a subset of patients with metastatic solid ***,the patients who would benefit most from ICIs in biliary tract cancer(BTC)are still *** and methods:We molecularly characterized tissues and blood from 32 patients with metastatic BTC treated with the ICI pembrolizumab as second-line ***:All patients had microsatellite stable(MSS)type *** of the 32 patients achieved partial response(PR),with an objective response rate(ORR)of 9.4%(95%confidence interval[CI],2.0–25.2)and nine showed stable disease(SD),exhibiting a disease control rate(DCR)of 37.5%(95%CI,21.1–56.3).For the 31 patients who had access to PD-1 ligand 1(PD-L1)combined positive score(CPS)testing(cut-off value≥1%),the ORR was not different between those who had PD-L1-positive(PD-L1+;1/11,9.1%)and PDL1-(2/20,10.0%)tumors(p=1.000).The tumor mutational burden(TMB)of PD-L1+BTC was comparable to that of PD-L1-BTC(p=0.630).TMB and any exonic somatic mutations were also not predictive of pembrolizumab *** analysis of blood and tumor samples demonstrated a relatively high natural killer(NK)cell proportion in the peripheral blood before pembrolizumab treatment in patients who achieved tumor ***,the tumors of these patients presented high enrichment scores for NK cells,antitumor cytokines,and Th1 signatures,and a low enrichment score for cancer-associated ***:This study shows the molecular characteristics associated with the efficacy of pembrolizumab in BTC of the MSS type.

With the rise of remote collaboration,the demand for advanced storage and collaboration tools has rapidly ***,traditional collaboration tools primarily rely on access control,leaving data stored on cloud servers vulnerable due to insufficient *** paper introduces a novel mechanism that encrypts data in‘bundle’units,designed to meet the dual requirements of efficiency and security for frequently updated collaborative *** bundle includes updated information,allowing only the updated portions to be reencrypted when changes *** encryption method proposed in this paper addresses the inefficiencies of traditional encryption modes,such as Cipher Block Chaining(CBC)and Counter(CTR),which require decrypting and re-encrypting the entire dataset whenever updates *** proposed method leverages update-specific information embedded within data bundles and metadata that maps the relationship between these bundles and the plaintext *** utilizing this information,the method accurately identifies the modified portions and applies algorithms to selectively re-encrypt only those *** approach significantly enhances the efficiency of data updates while maintaining high performance,particularly in large-scale data *** validate this approach,we conducted experiments measuring execution time as both the size of the modified data and the total dataset size *** show that the proposed method significantly outperforms CBC and CTR modes in execution speed,with greater performance gains as data size ***,our security evaluation confirms that this method provides robust protection against both passive and active attacks.

Biomaterial-based drug delivery systems have been developed to expedite cartilage regeneration;however,challenges related to drug recovery,validation,and efficient drug delivery *** instance,compound K(CK)is a major metabolite of ginsenosides that is known to protect against joint degeneration by inhibiting the production of inflammatory cytokines and the activation of immune ***,its effects on cartilage degradation and tissue regeneration remain ***,tissue-adhesive drug delivery depots that stably adhere to cartilage defects are required for CK *** this study,CK-loaded adhesive patches were reported to seal cartilage defects and deliver CK to defect sites,preventing cartilage degradation and accelerating cartilage tissue *** patches are stable and suitable for application in surgical procedures under physiological conditions and show excellent adhesiveness to cartilage *** addition,there were no significant differences in the adhesive polymeric networks before and after CK ***-loaded hydrocaffeic acid-conjugated chitosan patches significantly inhibited the stimulation of cartilage-degrading enzymes and apoptosis in osteoarthritic cartilage by releasing CK in cartilage ***,the NFkB signaling pathway of released CK from the adhesive patches in the treatment of osteoarthritis is ***,the CK-loaded adhesive patches are expected to significantly contribute to cartilage regeneration.

Observation and Numerical Simulations with Radar and Surface Data Assimilation for Heavy Rainfall over Central Korea

This study investigated the impact of multiple-Doppler radar data and surface data assimilation on forecasts of heavy rainfall over the central Korean Peninsula;the Weather Research and Forecasting(WRF) model and its three-dimensional variational data assimilation system(3DVAR) were used for this purpose. During data assimilation,the WRF 3DVAR cycling mode with incremental analysis updates(IAU) was used. A maximum rainfall of 335.0 mm occurred during a 12-h period from 2100 UTC 11 July 2006 to 0900 UTC 12 July *** radar data showed that the heavy rainfall was due to the back-building formation of mesoscale convective systems(MCSs).New convective cells were continuously formed in the upstream region,which was characterized by a strong southwesterly low-level jet(LLJ).The LLJ also facilitated strong convergence due to horizontal wind shear,which resulted in maintenance of the *** assimilation of both multiple-Doppler radar and surface data improved the accuracy of precipitation forecasts and had a more positive impact on quantitative forecasting(QPF) than the assimilation of either radar data or surface data *** back-building characteristic was successfully forecasted when the multiple-Doppler radar data and surface data were *** data assimilation experiments,the radar data helped forecast the development of convective storms responsible for heavy rainfall,and the surface data contributed to the occurrence of intensified low-level *** surface data played a significant role in enhancing the thermal gradient and modulating the planetary boundary layer of the model,which resulted in favorable conditions for convection.

The present article reports the experimental Particle Image Velocimetry （PIV） investigation and the corresponding numerical simulation results about the water flow over the oscillating hydrofoil and its unsteady dynamic characters. The experimental study focuses on the effect of mean angles of attack. The comparison between the PIV results and numerical prediction about the flow field using Fluent well demonstrates the capability of CFD on the simulation of the water flow around the pitching hydrofoil. The numerical results indicate that the forced oscillating frequencies have evident effects on the flow separation and vortex shedding. The simulations about the hydrodynamic drag and lift coefficients were also performed.

A novel approach is proposed for improving adaptive feedback cancellation using a variable step-size affine projection algorithm(VSS-APA) based on global speech absence probability(GSAP).The variable step-size of the proposed VSS-APA is adjusted according to the GSAP of the current *** weight vector of the adaptive filter is updated by the probability of the speech *** performance measure of acoustic feedback cancellation is evaluated using normalized *** results demonstrate that the proposed approach has better performance than the normalized least mean square(NLMS) and the constant step-size affine projection algorithms.

Objective:To elucidate the potential anti-inflammatory mechanisms of Rhamnus crenata leaf extracts using RAW264.7 ***:We used 3-[4,5-dimethylthiazol-2-yl]-2,5 diphenyl tetrazolium bromide assay to measure cell *** oxide(NO)production was measured using Griess *** blotting and RT-PCR assays were carried out for analyzing the protein and gene expressions of pro-inflammatory mediators,***,PD98059(ERK1/2 inhibitor),SB203580(p38 inhibitor),SP600125(JNK inhibitor),and BAY11-7082(NF-κB inhibitor)were used to evaluate the anti-inflammatory mechanism of Rhamnus crenata leaf ***:Rhamnus crenata leaf extracts significantly inhibited the production of the pro-inflammatory mediators such as NO,iNOS,COX-2,IL-1β,and TNF-αin lipopolysaccharide(LPS)-stimulated RAW264.7 *** crenata leaf extracts also suppressed LPS-induced degradation of IκB-αand nuclear accumulation of p65,which resulted in the inhibition of NF-κB activation in RAW264.7 ***,the extracts attenuated the phosphorylation of p38,ERK1/2,and JNK in LPS-stimulated RAW264.7 ***,HO-1 expression induced by Rhamnus crenata leaf extracts was significantly downregulated by SB230580,PD98059,SP600125 and ***:Rhamnus crenata leaf extract may upregulate HO-1 expression through inhibition of p38,ERK1/2,and NF-κB activation,which may contribute to the anti-inflammatory activity of the *** crenata leaf extracts may have great potential for the development of anti-inflammatory drugs to treat acute and chronic inflammatory diseases.

Clinical bone-morphogenetic protein 2(BMP2)treatment for bone regeneration,often resulting in complications like soft tissue inflammation and ectopic ossification due to high dosages and non-specific delivery systems,necessitates research into improved biomaterials for better BMP2 stability and *** tackle this challenge,we introduced a groundbreaking bone-targeted,lipoplex-loaded,three-dimensional bioprinted bilayer scaffold,termed the polycaprolactone-bioink-nanoparticle(PBN)scaffold,aimed at boosting bone *** encapsulated BMP2 within the fibroin nanoparticle based lipoplex(Fibroplex)and functionalized it with DSS6 for bone tissue-specific targeting.3D printing technology enables customized,porous PCL scaffolds for bone healing and soft tissue growth,with a two-step bioprinting process creating a cellular lattice structure and a bioink grid using gelatin-alginate hydrogel and DSS6-Fibroplex,shown to support effective nutrient exchange and cell growth at specific pore *** PBN scaffold is predicted through in silico analysis to exhibit biased BMP2 release between bone and soft tissue,a finding validated by in vitro osteogenic differentiation *** PBN scaffold was evaluated for critical calvarial defects,focusing on sustained BMP2 delivery,prevention of soft tissue cell infiltration and controlled fiber membrane pore size in *** PBN scaffold demonstrated a more than eight times longer BMP2 release time than that of the collagen sponge,promoting osteogenic differentiation and bone regeneration in a calvarial defect *** findings suggest that the PBN scaffold enhanced the local concentration of BMP2 in bone defects through sustained release and improved the spatial arrangement of bone formation,thereby reducing the risk of heterotopic ossification.
