Substance use,specifically the use of prescription and non-prescription opioids among pregnant women,is a major public health issue and chief contributor to the opioid *** prevalence of Neonatal Opioid Withdrawal Syndrome has risen 5-fold in the past decade,and is a well-recognized consequence of perinatal opioid *** contrast,the long-term damage to the developing brain from opioid medications is just beginning to be recognized as a serious *** data suggest that opioid exposure commencing in utero negatively affects the maturation of the neural-immune system,and trajectory of central nervous system *** induces peripheral immune hyper-reactivity,lasting structural and microstructural brain injury,and significant deficits in executive function and cognitive control in adult animals following in utero ***,to address the cascading public health crisis stemming from the multitude of infants with in utero opioid exposure who will grow up with altered neurodevelopmental trajectories,rigorous preclinical,mechanistic studies are *** studies will define the long-term sequelae of prenatal opioid exposure in an effort to develop appropriate and targeted ***,the development of novel fluid,neuroimaging and biobehavioral biomarkers will be the most useful to aid in early identification and treatment of opioid exposed infants with the greatest risk of poor clinical *** studies will be essential to understand how in utero insults determine brain structure and function in adulthood,and what targeted interventions will be required to improve longterm outcomes in the countless children being born exposed to opioids each year.

Genotype×year interaction of pod and seed mass and stability of Pongamia pinnata families in a semi-arid region

Sixteen pongamia families were evaluated in a field experiment for eight consecutive years in dryland conditions to identify stable,high-yielding *** trial was conducted in a randomized complete block design with three *** family,consisting of nine trees per replication,was planted at a spacing of3 m x 3 *** stability was analyzed using(1)Eberhart and Russel’s regression coefficient(β_i)and deviation from regression(S_d^2),(2)Wrike’s ecovalence(W_i);(3)Shukla stability variance(σ_i^2);and(4)Piepho and Lotito’s stability index(L_i).Families were also analyzed for adaptability and stability using AMMI and GGE biplots graphical *** study revealed significant variances due to family and family x year interaction for pod and seed *** performed differently and ranked differently across *** performance of families was influenced by both genetic factor and environmental conditions in different *** families tested,TNMP20,Acc14,TNMP14 and Acc30 were high yielders for pods,and Acc14,Acc30,TNMP6,RAK19 and TNMP14 were high for seed *** to the Eberhart and Russell model,Acc30,TNMP14 and TNMP3 were stable across *** the graphical view of family x year interaction based on AMMI methods,TNMP3,TNMP4 and TNMP14 had greater stability with moderate seed yield,and Acc14 and Acc30 had moderate stability with high seed *** the other hand,GGE biplots revealed Acc14,Acc30 and TNMP14 as high yielders with moderate *** and GGE biplots were able to capture nonlinear parts of the family x year interaction that were not be captured by the Eberhart and Russel model while also identifying stable *** on different methodologies,Acc14,Acc30 and TNMP14 were identified as high yielding and stable families for promoting pongamia cultivation as a biofuel crop for semi-arid regions.

China’s coupled human-environment system(CHES) is assessed here via a systems schema that emphasizes the complex interactions of components and their attributes. In addition to the human and environment components, we identified two other components to evaluate the relationship. The four components are human activity intensity, resource carrying capacity, ecological constraints and system’s openness. Based on their interactions, we derived a cognitive schema for classifying the level of strain or stress of an area. The analysis draws on 11 indicators and 29 sub-indicators including remote sensing data and statistical data that are used to estimate the four components. The findings indicate that human activities are highly intense in a few geographical areas, particularly large urban systems and trade and investment zones on the eastern coastal areas. Nonetheless, these areas are also well-endowed in water resources and fertile soils although urban systems are increasingly stressed from negative pollution externalities. They are also open systems which allow them to bear a higher level of pressure and adjust accordingly. Desertification and soil erosion point to relatively fragile biophysical systems in the west and southwest, but human activities are still relatively less intense compared to their coastal counterparts. As a whole, only 14% of areas may be said to be relatively or highly strained. This however belies another one-third of areas that are currently unstable, and likely to become strained and thereby vulnerable in the near future.

NF-κB promotes the stem-like properties of leukemia cells by activation of LIN28B

AIM To examine whether nuclear factor kappa B(NF-κB) activity regulates LIN28 B expression and their roles in leukemia stem cell(LSC)-like properties. METHODS We used pharmacological inhibitor and cell viability assays to examine the relation between NF-κB and LIN28 B. Western blot and q RT-PCR was employed to determine their protein and m RNA levels. Luciferase reporter was constructed and applied to explore the transcriptional regulation of LIN28 B. We manipulated LIN28 B level in acute myeloid leukemia(AML) cells and investigated LSC-like properties with colony forming and serial replating assays. RESULTS This study revealed the relationship between NF-κB and LIN28 B in AML cells through drug inhibition and overexpression experiments. Notably,inhibition of NF-κB by pharmacological inhibitors reduced LIN28 B expression and decreased cell proliferation. We demonstrated that NF-κB binds to the-819 to-811 region of LIN28 B promoter,and transcriptionally regulates LIN28 B expression. LIN28 B protein was significantly elevated in NFκB1 transfected cells compared to vector control. Importantly,ectopic expression of LIN28 B partially rescued the self-renewal capacity impaired by pharmacological inhibition of NF-κB activity. CONCLUSION These results uncover a regulatory signaling,NF-κB/LIN28 B,which plays a pivotal role in leukemia stem cell-like properties and it could serve as a promising intervening target for effective treatment of AML disease.
