RNA binding proteins in spermatogenesis： an in depth focus on the musashi family

Controlled gene regulation during gamete development is vital for maintaining reproductive potential. During the complex process of mammalian spermatogenesis, male germ cells experience extended periods of the inactive transcription despite heavy translational requirements for continued growth and differentiation. Hence, spermatogenesis is highly reliant on mechanisms of posttranscriptional regulation of gene expression, facilitated by RNA binding proteins （RBPs）, which remain abundantly expressed throughout this process. One such group of proteins is the musashi family, previously identified as critical regulators of testis germ cell development and meiosis in Drosophila, and also shown to be vital to sperm development and reproductive potential in the mouse. This review describes the role and function of RBPs our recent knowledge of the musashi proteins in spermatogenesis. within the scope of male germ cell development, focusing on The functional mechanisms utilized by RBPs within the cell are outlined in depth, and the significance of sub-cellular localization and stage-specific expression in relation to the mode and impact of posttranscriptional regulation is also highlighted. We emphasize the historical role of the musashi family of RBPs in stem cell function and cell fate determination, as originally characterized in Drosophila and Xenopus, and conclude with our current understanding of the differential roles and functions of the mammalian musashi proteins, musashi-1 and musashi-2, with a primary focus on our findings in spermatogenesis. This review highlights both the essential contribution of RBPs to posttranscriptional regulation and the importance of the musashi family as master regulators of male gamete development.

Genotype×year interaction of pod and seed mass and stability of Pongamia pinnata families in a semi-arid region

Sixteen pongamia families were evaluated in a field experiment for eight consecutive years in dryland conditions to identify stable,high-yielding *** trial was conducted in a randomized complete block design with three *** family,consisting of nine trees per replication,was planted at a spacing of3 m x 3 *** stability was analyzed using(1)Eberhart and Russel’s regression coefficient(β_i)and deviation from regression(S_d^2),(2)Wrike’s ecovalence(W_i);(3)Shukla stability variance(σ_i^2);and(4)Piepho and Lotito’s stability index(L_i).Families were also analyzed for adaptability and stability using AmmI and GGE biplots graphical *** study revealed significant variances due to family and family x year interaction for pod and seed *** performed differently and ranked differently across *** performance of families was influenced by both genetic factor and environmental conditions in different *** families tested,TNmP20,Acc14,TNmP14 and Acc30 were high yielders for pods,and Acc14,Acc30,TNmP6,RAK19 and TNmP14 were high for seed *** to the Eberhart and Russell model,Acc30,TNmP14 and TNmP3 were stable across *** the graphical view of family x year interaction based on AmmI methods,TNmP3,TNmP4 and TNmP14 had greater stability with moderate seed yield,and Acc14 and Acc30 had moderate stability with high seed *** the other hand,GGE biplots revealed Acc14,Acc30 and TNmP14 as high yielders with moderate *** and GGE biplots were able to capture nonlinear parts of the family x year interaction that were not be captured by the Eberhart and Russel model while also identifying stable *** on different methodologies,Acc14,Acc30 and TNmP14 were identified as high yielding and stable families for promoting pongamia cultivation as a biofuel crop for semi-arid regions.

Impaired eye tracking is associated with symptom severity but not dynamic postural control in adolescents following concussion

NF-κB promotes the stem-like properties of leukemia cells by activation of LIN28B

AIm To examine whether nuclear factor kappa B(NF-κB) activity regulates LIN28 B expression and their roles in leukemia stem cell(LSC)-like properties. mETHODS We used pharmacological inhibitor and cell viability assays to examine the relation between NF-κB and LIN28 B. Western blot and q RT-PCR was employed to determine their protein and m RNA levels. Luciferase reporter was constructed and applied to explore the transcriptional regulation of LIN28 B. We manipulated LIN28 B level in acute myeloid leukemia(AmL) cells and investigated LSC-like properties with colony forming and serial replating assays. RESULTS This study revealed the relationship between NF-κB and LIN28 B in AmL cells through drug inhibition and overexpression experiments. Notably,inhibition of NF-κB by pharmacological inhibitors reduced LIN28 B expression and decreased cell proliferation. We demonstrated that NF-κB binds to the-819 to-811 region of LIN28 B promoter,and transcriptionally regulates LIN28 B expression. LIN28 B protein was significantly elevated in NFκB1 transfected cells compared to vector control. Importantly,ectopic expression of LIN28 B partially rescued the self-renewal capacity impaired by pharmacological inhibition of NF-κB activity. CONCLUSION These results uncover a regulatory signaling,NF-κB/LIN28 B,which plays a pivotal role in leukemia stem cell-like properties and it could serve as a promising intervening target for effective treatment of AmL disease.

Effect of Rice Bran Quality and Protein Supplement, Base mix, and/or Soybean meal on Growing Pig Performance in Cambodia

The objective of this study was to evaluate the effect of rice bran quality and dietary supplement source on growing pig performance. A total of 60 castrated male pigs (Yorkshire × Duroc, initially 16.1 kg) were used for 52-day trial. Pigs were allotted to pens in a completely randomized design (4 pigs per pen) and assigned to four dietary treatments in a 2 × 2 factorial with factors being rice bran source (high or low-quality) and source of protein, vitamins, and minerals (supplement or soybean meal and base mix). Analyzed protein and fat level for the high- and low-quality rice bran was 11.07% and 5.86% and 11.20% and 4.50%, respectively. Crude fiber content for high- and low-quality rice bran was 13.44% and 27.79%. Diets were formulated in two phases (phase 1: 16 to 27 and phase 2: 27 to 48 kg) to adjust to the pig’s nutrient requirements. There was no evidence for an interaction (P > 0.05) between rice bran quality × supplement type or main effect of supplement type (soybean meal + base mix vs protein supplement) for final body weight, average daily gain (ADG), average daily feed intake (ADFI), and feed efficiency (F/G). However, pigs fed high-quality rice bran had improved (P mpared to pigs fed low protein rice bran. In conclusion, rice bran quality as measured by protein, crude fiber, and fat influenced growing pig performance, while the type of dietary protein supplement did not. Additionally, rice bran with high protein, fat, and low fiber improved ADG and F/G.

High affinity soluble ILT2 receptor:a potent inhibitor of CD8^(+)T cell activation

Using directed mutagenesis and phage display on a soluble fragment of the human immunoglobulin superfamily receptor ILT2(synonyms:LIR1,mIR7,CD85j),we have selected a range of mutants with binding affinities enhanced by up to 168,000-fold towards the conserved region of major histocompatibility complex(mHC)class I *** in a dimeric form,either by chemical cross-linking with bivalent polyethylene glycol(PEG)derivatives or as a genetic fusion with human IgG Fc-fragment,the mutants exhibited a further increase in ligand-binding strength due to the avidity effect,with resident half-times(t1/2)on the surface of mHC I-positive cells of many *** novel compounds antagonized the interaction of CD8 co-receptor with mHC I in vitro without affecting the peptide-specific binding of T-cell receptors(TCRs).In both cytokine-release assays and cell-killing experiments the engineered receptors inhibited the activation of CD8^(+)cytotoxic T lymphocytes(CTLs)in the presence of their target cells,with subnanomolar potency and in a dose-dependent *** a selective inhibitor of CD8^(+)CTL responses,the engineered high affinity ILT2 receptor presents a new tool for studying the activation mechanism of different subsets of CTLs and could have potential for the development of novel autoimmunity therapies.
