A new method of synthesis of 2-(1,2-diferrocenylvinyl)benz- and azabenzimidazoles (3a-f), (4a-f) and 1’H,3’H(Me)-spiro-[(aza)benzimidazoline-2’,3-(1,2-diferrocenylcyclopropenes)] (5a-f) via reactions of diferrocenyl(methylsulfanyl)cyclopropenylium iodide (1) with aromatic o-diamines (2a-f) in the presence of Et3N (80°C - 82°C) is described. The structures of the resultant compounds are established using IR, 1H and 13C NMR spectroscopy, mass spectrometry and elemental analysis. The structure of one compound, cis-2-(1,2-diferrocenylvinyl)-1-methylbenzimidazole (3b), is confirmed by X-ray diffraction analysis. The electrochemical properties of compounds 3a, 3b, 3d and 5f are investigated using cyclic square wave voltammetry. Two electrochemical processes (I-II), attributed to oxidation of the ferrocene moieties, and the values of E0’(I), E0’(II), DE0’(II-I) and comporportionation constant Kcom are reported. The bioactivities of seven compounds 3a, 3c-f, 5d, 5f are evaluated. Compound 5f is the most active compound with a modest cytotoxic activity against six human cancer cell lines: U-251 (glioma), PC-3 (prostate cancer), K-562 (leukemia), HCT-15 (colon cancer), MCF-7 (breast cancer) and sKLU-1 (lung cancer).

HIIT对高脂喂养ApoE敲除鼠骨骼肌氧化应激的影响及肌肉因子BAIBA的保护作用

目的:探讨高强度间歇训练(high-intensity interval training,HIIT)对高脂喂养的载脂蛋白E(ApoE)敲除(Apo^(E-/-))小鼠骨骼肌氧化应激的改善作用以及肌肉因子β-氨基异丁酸(β-aminoisobutyic acid,BAIBA)在其中的可能调节机制。方法:9周龄健康雄性C57BL/6j.鼠和Apo^(E-/-)小鼠分别喂食标准和高脂饲料,其中C57BL/6j.鼠为基线对照组(WT,n=6),Apo^(E-/-)小鼠经高脂饲料喂养12周后随机分为Apo^(E-/-)安静组(Apo^(E-/-),n=10)和Apo^(E-/-)高强度间歇训练组(Apo^(E-/-)HIIT,n=10),Apo^(E-/-)HIIT组进行6周HIIT,WT和Apo^(E-/-)组在笼中自由活动。最后一次运动后48 h取小鼠两侧腿部腓肠肌进行氧化损伤、抗氧化及BAIBA生成相关指标的检测。随后采用L-BAIBA预孵育C2C12细胞,后用过氧化氢(H_(2)O_(2))诱导氧化应激,结合干预条件随机分为Control、H_(2)O_(2)和L-BAIBA+H_(2)O_(2)组。相差显微镜观察细胞形态,并检测细胞ROs水平和Nrf2介导的抗氧化通路的相关指标。结果:1)与WT组相比,Apo^(E-/-)组小鼠骨骼肌氧化损伤相关的4-HNE修饰蛋白、蛋白质羰基含量和ROs水平显著升高,而抗氧化相关的p-Nrf2蛋白表达和GsH含量显著降低;而与Apo^(E-/-)组相比,Apo^(E-/-)HIIT组小鼠骨骼肌4-HNE修饰蛋白和蛋白质羰基含量明显降低,p-Nrf2蛋白表达和GsH含量明显升高。2)Apo^(E-/-)HIIT组小鼠骨骼肌生成BAIBA的相关酶基因Acads和Hadha mRNA的表达水平显著高于Apo^(E-/-)组。3)与Control组相比,H_(2)O_(2)干预后C2C12细胞明显皱缩,活细胞数量降低,ROs水平显著增加;而与H_(2)O_(2)组相比,L-BAIBA+H_(2)O_(2)组活细胞数量增加,形态较饱满,ROs水平明显降低。4)与Control组相比,H_(2)O_(2)显著降低了细胞Nrf2、p-Nrf2及其介导的抗氧化酶sOD1、NQO1和CAT蛋白以及合成GsH的相关酶基因Gss、Gsr和Gclm mRNA的表达水平,并显著降低了细胞GsH含量;而L-BAIBA+H_(2)O_(2)组较H_(2)O_(2)组则显著增加了p-Nrf2、sOD1和NQO1蛋白以及Gss、Gsr、Gclc和Gclm mRNA的表达量,同时细胞GsH含量也显著增加。结论:6周HIIT降低了高脂喂养的Apo^(E-/-)小鼠骨骼肌的氧化损伤,其中肌肉因子BAIBA对Nrf2介导的抗氧化信号的激活可能起着一定的作用。

Immuno-oncology combinations： raising the tail of the survival curve

There have been exponential gains in immuno-oncology in recent times through the development of immune checkpoint inhibitors. Already approved by the U.s. Food and Drug Administration for advanced melanoma and non-small cell lung cancer,immune checkpoint inhibitors also appears to have significant antitumor activity in multiple other tumor types. An exciting component of immunotherapy is the durability of antitumor responses observed, with some patients achieving disease control for many years. Nevertheless, not all patients benefit, and efforts should thus now focus on improving the efficacy of immunotherapy through the use of combination approaches and predictive biomarkers of response and resistance. There are multiple potential rational combinations using an immunotherapy backbone, including existing treatments such as radiotherapy, chemotherapy or molecularly targeted agents, as well as other immunotherapeutics. The aim of such antitumor strategies will be to raise the tail on the survival curve by increasing the number of long term survivors, while managing any additive or synergistic toxicities that may arise with immunotherapy combinations. Rational trial designs based on a clear understanding of tumor biology and drug pharmacology remain paramount. This article reviews the biology underpinning immuno-oncology, discusses existing and novel immunotherapeutic combinations currently in development, the challenges of predictive biomarkers of response and resistance and the impact of immuno-oncology on early phase clinical trial design.

Evaluation of the virulence of entomopathogenic fungus, Isaria fumosorosea isolates against subterranean termites Coptotermes spp.(Isoptera:Rhinotermitidae)

The entomopathogenic fungus Isaria fumosorosea Wize, formerly known as Paecilomyces fumosoroseus is reported as a promising biocontrol agent for controlling subterranean termites, particularly those belonging to the family Rhinotermitidae. In Malaysia, the family Rhinotermitidae includes two species of subterranean termites with extremely high economic importance; namely Coptotermes curvignathus Holmgren, and the Asian sLubterranean Termite (Coptotermes gestroi Wasmann). To comprehend the potential control of this soil-dwelling fungus against these subterranean termites in Malaysia, an investigation was carried out by testing the pathogenecity of 11 isolates against these termite species. All isolates showed pathogenic potential against the termite (Mortality rate of C. curvignathus:84.4%; C. gestroi:67.3%). IsolatePF49 was the most effective against both species of termites and was further tested for its virulence and *** LC_(50) values of PF49 against C. curvignathus and *** were 7.55×103 and 1.09×102 conidia/ml,respectively. The average number of days required to complete the mycosis process in C. curvignathus and *** were 4.7 and 8 days, respectively. These fungi are believed useful for protecting living trees, plants, wood,wood structures, and other cellulosic materials susceptible to termite infestation and damage.

Preference for uninfected mates is presumed beneficial as it minimizes one＇s risk of contracting an infection and infecting one＇s offspring. In avian systems, visual ornaments are often used to indi- cate parasite burdens and facilitate mate choice. However, in mammals, olfactory cues have been proposed to act as a mechanism allowing potential mates to be discriminated by infection status. The effect of infection upon mammalian mate choice is mainly studied in captive rodents where ex- perimental trials support preference for the odors of uninfected mates and some data suggest scent marking is reduced in individuals with high infection burdens. Nevertheless, whether such effects occur in nonmodel and wild systems remains poorly understood. Here, we investigate the interplay between parasite load （estimated using fecal egg counts） and scent marking behavior in a wild population of banded mongooses Mungos mungo. Focusing on a costly protozoan parasite of the genus Isospora and the nematode worm Toxocara, we first show that banded mongooses that engage in frequent, intensive scent marking have lower Isospora loads, suggesting marking behavior may be an indicator trait regarding infection status. We then use odor presentations to demonstrate that banded mongooses mark less in response to odors of opposite sexed individuals with high Isospora and Toxocara loads. As both of these parasites are known to have detrimental effects upon the health of preweaned young in other species, they would appear key targets to avoid during mate choice. Results provide support for scent as an important ornament and mechanism for advertising parasitic infection within wild mammals.
