Potential of Zimbabwean commercial probiotic products and strains of Lactobacillus plantarum as prophylaxis and therapy against diarrhoea caused by Escherichia coli in children

Objective: To evaluate the potential of commercial fermented products sold in the country,and strains of Lactobacillus plantarum(L,plantarum)as prophylaxis and therapy against diarrhoea in children,Methods: The antimicrobial potential of cultures of lactobacilli enriched from 4 Zimbabwean commercial food/beverage products: Dairibord Lacto sour milk(DLSM),Probrand sour milk(PSM),Kefalos Vuka cheese(KVC) and Chibuku opaque beer(COB); and four strains of L,plantarum obtained from Balkan traditional cheeses against clinical strains of Escherichia coli(E,coli) was assayed using the well diffusion method,Three commercial paediatric antidiarrhoeal drug products: Biogaia(BG),Prolife(PL) and Probio Junior(PJ) and a mutant strain of E,coli [strain 11105(ATCC)- a vitamin B-12 auxotroph and penicillin G acylase-producing strain] were used as controls,An agar diffusion assay and a competitive exclusion assay were carried out on Mueller Hinton agar,Results: Crude cultures of putative lactobacillus strains obtained from Zimbabwean dairy products(Probrand sour milk,Kefalos Vuka vuka cheese and Chibuku opaque beer) had significantly higher antimicrobial activities against clinical strains of E,coli than strains of L,plantarum isolated from Balkan cheeses(CLP1,CLP2 or CLP3) and crude microbial cultures from commercial paediatric probiotic products(BG,PJ and PL) of a culture of Lactobacillus rhamnosus LGG(P<0.05),Conclusions: The putative Lactobacilli from four commercial Zimbabwean dairy products(Probrand sour milk,Kefalos Vuka vuka cheese and Chibuku opaque beer),and three strains of L,plantarum from Balkan cheeses(CLP1,CLP2 or CLP3) exhibited high antibacterial activities that can be harnessed to control paediatric diarrhoea that is caused by pathogenic strains of E,coli,Studies to characterise the probiotic potential of the live cultures in the products and the new strains of L,plantarum are underway.

Mycobacterium tuberculosis(MTB) and human immunodeficiency virus(HIV) co-infections have remained a major public health concern worldwide,particularly in Southern *** our understanding of the molecular interactions between the pathogens has remained poor,primarily due to lack of suitable preclinical models for such *** reviewed the use,this far,of mammalian cell culture models in HIV-MTB interaction *** have described the use of primary human cell cultures,including monocyte-derived macrophage(MDM) fractions of peripheral blood mononuclear cell(PBMC),alveolar macrophages(AM),cell lines such as the monocyte-derived macrophage cell line(U937),T lymphocyte cell lines(CEMx174,ESAT-6-specific CD4+ T-cells) and an alveolar epithelial cell line(A549) and special models such as stem cells,three dimensional(3D) or organoid cell models [including a blood-brain barrier(BBB) cell model] in HIV-MTB interaction *** use of cell cultures from other mammals,including:mouse cell lines [macrophage cell lines RAW 264.7 and J774.2,fibroblast cell lines(NIH 3T3,C3 H clones),embryonic fibroblast cell lines and T-lymphoma cell lines(***,TIMI.4 and R1.1)]; rat(T cells:Rat2,RGE,XC and HH16,and alveolar cells:NR8383) and primary guinea pigs derived AMs,in HIV-MTB studies is also *** the spectrum of the models available,cell cultures offer great potential for host-HIV-MTB interactions studies.

BACKGROUND Abernethy syndrome is a congenital vascular anomaly in which the portal blood completely or partially bypasses the liver through a congenital portosystemic *** the number of recognized and reported cases is gradually increasing,Abernethy syndrome is still a rare disease entity,with an estimated prevalence between 1 per 30000 to 1 per 50000 *** this case series,we aimed to contribute to the growing knowledge of potential clinical presentations,course and complications of congenital portosystemic shunts(CPSS)in *** SUMMARY Five children are presented in this case series:One female and four males,two with an intrahepatic CPSS and three with an extrahepatic *** first patient,who was diagnosed with an intrahepatic CPSS,presented with gastrointestinal bleeding,abdominal pain and hyperammonaemia at six years of *** underwent a percutaneous embolization of his shunt and has remained asymptomatic ever *** second patient presented with direct hyperbilirubinemia in the neonatal period and his intrahepatic CPSS later spontaneously *** third patient had pulmonary hypertension and hyperammonaemia due to complete portal vein agenesis and underwent liver transplantation at five years of *** fourth patient was diagnosed immediately after birth,when evaluated due to another congenital vascular anomaly,and the last patient presented as a teenager with recurrent bone fractures associated with severe *** addition,the last two patients are characterised by benign liver nodules;however,they are clinically stable on symptomatic *** Abernethy syndrome is a rare anomaly with diverse clinical features,affecting almost all organ systems and presenting at any age.

Urban life has led to the creation of human-made environments that,from a microbiological perspective,provide extreme life *** non-ubiquitous microorganisms such as thermophilic members of the black yeast genus Exophiala are enriched within these habitats for which no counterpart is known in *** consistently accumulate a number of specific black,white and red yeasts on the rubber seals of doors and in stagnant water at the *** of these yeasts are primarily known as agents of human opportunistic *** this review,the literature data are supported by a screening study involving 937 households in 15 cities in *** were detected in 230 samples(24.5%).Using rDNA sequencing,the prevalent species were identified as Exophiala dermatitidis(n=116),Candida parapsilosis(n=44),***(n=35),Magnusiomyces capitatus(n=22),Rhodotorula mucilaginosa(n=15),and ***(n=14).The possible role of dishwashers in transmitting disease is discussed.

Background: Fundus autofluorescence (AF) in some patients with retinitis pigmentosa is characterized by a parafoveal ring of increased AF which surrounds the centre as hypofluorescent changes appear at the periphery. The aim of this study was to evaluate the AF patterns in relation to retinal function measured by ele ctroretinography and visual fields. Methods: Thirty-two patients with RP were included in the study. AF imaging of the macular area was performed with the scanning laser ophthalmoscope. Patients were divided in two groups according to their fundus AF patterns. All patients from group 1 had a ring of increased AF of different size but no atrophic areas inside vascular arcades. Patients with a ring of increased AF and round atrophic changes at different eccentricities from their fovea were selected in group 2. Visual fields were tested with kinetic, automated perimetry and microperimetry; the radius of the hyperfluorescent ring and the smallest distance of hypofluorescent areas from the fovea were compared to visual fields, PERG P50 and N95 and mfERG P1 amplitudes of the inner three rings. Results: A linear relationship was found in group 1 between the radius of the ring of increased AF and both the automated (r=0.82) and kinetic perimetry (r=0.80). The radius of the AF ring correlated highlywith the PERG P50 (r=0.72) and N95 (r=0.74) amplitudes. In all patients, mfERG responses were reduced at all retinal locations, more pronounced at periphery. There was a good correlation between the ring of increased AF and the P1 amplitude of ring 2 of mfERG (r=0.62). Pati ents from group 2 had significantly reduced or non-recordable PERGs and mfERGs. The eccentricities of hypofluorescent changes did not correlate with any type of perimetry. Conclusions: Our results show that in stages of retinitis pigmentosa, before atrophic lesions spread inside the vascular arcades, the pattern of fundus autofluorescence correlates well with functional tests such as perimetry and

Purpose: To evaluate retinal sensitivity and fixation patterns in patients with Best's dystrophy by microperimetry (MP) and to correlate the results with static perimetry and retinal morphology seen by autofluorescence (AF). Methods: Central 10°visual fields in 11 patients with Best's dystrophy (VA: 0.5±0.38)were recorded by the Octopus M2 TOP program and by MP (MP1, Nidek Technologies). AF was recorded by HRA (Heidelberg Engineering). Results: High correlation (R=0.75, -0.76,-0. 48)was found between static perimetry (MS,MD and CLV indices) and *** on MP and AF results, three groups of patients were formed. Patients in the first two groups fixated inside the central nonuniform hypo-and hyperfluorescent AF ring area, next to relative (Group 1) or absolute scotoma (Group 2). Inner parts of the retina close to the fovea were most affected, whereas regions closer to the periphery of the 10°visual field showed near normal function. As the disease progressed, there was an evident shift of fixation to preferential retinal locus (PRL) in eight eyes with visual acuity 0.2 or less (Group 3). Fixation shift was superior in four eyes, temporal in two eyes, and nasal in two eyes. Conclusion: MP enabled a highly sensitive topographic monitoring of retinal function, showing central or pericentral fixation in the early stages, until loss of central function, in eyes with VA 0.2 or less, caused evident shift of fixation to PRL. PRL was never situated inside the central uniform hypofluorescent area,but corresponded with the hyperfluorescent ring seen with AF imaging.
