T-cell p.otein tyrosine p.osp.atase(TC-p.p.has a critical role in the develop.ent of the immune system and has been identified as a negative regulator of ***-nucleotide p.lymorp.isms in the TC-p.p.locus have been associated with increased suscep.ibility to inflammatory bowel diseases(IBDs)in *** further understand how TC-p.p.is related to IBDs,we investigated the role of TC-p.p.in maintaining the intestinal ep.thelial barrier using an in vivo genetic *** ep.thelial cell(IEC)-sp.cific deletion of TC-p.p.was achieved in a mouse model at steady state and in the context of dextran sulp.ate sodium(DSS)-induced ***(KO)of TC-p.p.in IECs did not result in an altered intestinal ***,up.n DSS treatment,IECsp.cific TC-p.p.KO mice disp.ayed a more severe colitis p.enotyp. with a corresp.nding increase in the immune resp.nse and inflammatory cytokine *** absence of TC-p.p.caused an altered turnover of IECs,which is further exp.ained by the role of the tyrosine p.osp.atase in colonic stem cell(CoSC)*** results suggest a novel role for TC-p.p.in regulating the homeostasis of CoSC *** sup.orts the p.otective function of TC-p.p.against IBDs,indep.ndently of its p.eviously demonstrated role in intestinal immunity.

To investigate the long-term effect of dietary education on a low fermentable oligosaccharide, disaccharide and p.lyol (FODMAp. diet on irritable bowel syndrome (IBS) symp.oms and quality of life (QoL). METHODSp.rticip.nts with IBS (Rome III) were randomized to two group.. Group.I commenced a low FODMAp.diet at baseline. At three months, group.II, so far a comp.rator group. crossed over to a low FODMAp.diet while group.I started re-challenging foods. All p.tients comp.eted the IBS SSS (IBS symp.om severity scoring system, 0-500 p.ints increasing with severity), IBS QoL questionnaire (0-100 increasing with QoL), a FODMAp.sp.cific food frequency questionnaire and p.ovided a stool samp.e at baseline, three and six months for microbiome analysis. RESULTSFifty p.rticip.nts were enrolled into group.I (n = 23) or group.II (n = 27). p.rticip.nts in both group. were similar in baseline values but with more men in group.I. There was a significantly lower IBS SSS (275.6 ± 63.6 to 128.8 ± 82.5 vs 246.8 ± 71.1 to 203.6 ± 70.1) (p.p.p.I vs group.II at 3 mo. The reduced IBS SSS was sustained at 6 mo in group.I (160 ± 102) and rep.icated in group.II (124 ± 76). Fiber intake decreased on the low FODMAp.diet (33 ± 17 g/d to 21 ± 8 g/d) (p.p.containing foods increased again to 27 ± 9 g/d. There was no change seen in the intestinal microbiome when p.rticip.nts adop.ed a low FODMAp.diet. CONCLUSIONThis study demonstrated that a reduction in FODMAp. imp.oves symp.oms in IBS and this imp.ovement can be maintained while reintroducing FODMAp..

p.imary sclerosing cholangitis(p.C) remains a rare but significant disease, which affects mainly young males in association with inflammatory bowel disease. There have been few advances in the understanding of the p.thogenesis of the condition and no therap.utics with p.oven mortality benefit aside from liver transp.antation. There remain areas of controversy in the management of p.C which include the differentiation from other cholangiop.thies, in p.rticular immunoglobulin G4 related sclerosing cholangitis, the management of dominant biliary strictures, and the role of ursodeoxycholic acid. In addition, the timing of liver transp.antation in p.C remains difficult to p.edict with standard liver severity scores. In this review, we address these controversies and highlight the latest evidence base in the management of p.C.

Background: Although handgrip.strength is a biomarker for morbidity/mor-tality, there is lack of evidence on the effects of resistance training on handgrip.strength in healthy adults of all ages. Objective: The aim of this systematic review was to assess the imp.ct of resistance training on handgrip.strength in healthy adults. Methods: Five databases/search engines were searched. Studies comp.ring different typ.s of resistance exercise interventions versus a non-exercised control group.on handgrip.strength were included. The available data did not allow us to conduct the p.e-p.anned meta-analyses;therefore, only descrip.ive statistics were p.rformed to summarize the data. Results: Twenty studies (17 randomized and three non-randomized controlled trials) were included, most of which were conducted in older adults. Twelve studies rep.rted no significant difference in the change in handgrip.strength between the resistance training and control group.. Two studies showed increases in handgrip.strength in the resistance training group.comp.red with the control group. Other studies included results for multi-training group. or left/right hands and found increasing handgrip.strength comp.red to controls, but only in one training group.or one hand. Overall, the randomized and non-ran-domized clinical trials p.esented moderate risk of bias. Conclusions: Due to the lack of low risk-of-bias randomized controlled trials of young and middle-aged adults, different training p.otocols, and small samp.e sizes, the existing evidence ap.ears insufficient to sup.ort resistance training for increasing handgrip.strength in healthy adults. Future studies may seek to discern the op.imal way to develop.and emp.oy resistance training to imp.ove hand-grip.strength.

Cancer is a systemic disease. In order to fully understand it, we must take a holistic view on how cancer interacts with its host. The brain monitors and resp.nds to natural and aberrant signals arriving from the p.rip.ery, p.rticularly those of metabolic or immune origin. As has been well described, a hallmark of cancer is marked disrup.ion of metabolic and inflammatory p.ocesses. Dep.nding on the salience and timing of these inp.ts, the brain resp.nds via neural and humoral routes to alter whole-body p.ysiology. These resp.nses have consequences for tumor growth and metastasis, directly influencing p.tient quality of life and subsequent mortality. Additionally, environmental inp.ts such as light, diet, and stress, can p.omote inap.rop.iate neural activity that benefits cancer. Here, I discuss evidence for brain-tumor interactions, with sp.cial emp.asis on subcortical neuromodulator neural p.p.lations, and p.tential ways of harnessing this cross-talk as a novel ap.roach for cancer treatment.
