MRI of magnetically labeled mesenchymal stem cells in hepatic failure model

AIM:To track intravascularly transplanted mesenchymal stem cells (MSCs) labeled with superparamagnetic iron oxide (SPIO) by using magnetic resonance imaging (MRI) in an experimental rabbit model of hepatic ***:Human MSCs labeled with FDA-approved SPIO particles (Feridex) were transplanted via the mes-enteric vein into rabbits (n=16) with carbon tetrachloride-induced hepatic *** resonance (MR) examinations were performed with a 3.0 T clinical scanner immediately before and 2 h and 1,3,and 7 d after *** intensity (SI) changes on T2weighted MRI were measured,and correlation between MR findings and histomorphologic findings was also ***:SI on T2-weighted MRI decreased significantly in the liver 2 h after injection of human MSCs and returned gradually to the levels found before injection in 7 *** in SI in the liver at 2 h,1,3,and 7 d were 41.87% ± 9.63%,10.42% ± 4.3%,5.12% ± 1.9%,3.75% ± 1.2%,respectively (P < 0.001).Histologic analyses confirmed the presence of MSCs in the liver,localized mainly in the sinusoids in early period (2 h and 1 d) and concentrated to the border zone in late period (3 and 7 d).The number of iron-positive cells in the liver at 2 h and on 1,3 and 7 d after transplantation was 29.2 ± 4.8,10.1 ± 3.7,6.7 ± 2.2,and 5.8 ± 2.1,respectively (P=0.013).CONCLUSION:Intravascularly injected SPIO-labeled MSCs in an experimental rabbit model of hepatic failure can be detected and followed with MRI.

Currently, medical linear accelerators （LINACs） are the most common method of radiotherapy for tumor treatments and more than 90 LINACs are installed and operated in Korea in 2005. To increase the treatment efficiency, LINACs are trending toward higher energy（more than 15MV） nowadays. Meanwhile, the increased photon energy above about 8MV can produce undesired photo neutrons through photonuclear reaction between photon and surrounding materials in LINAC. In this study, the neutron generation and flux around the head geometry of a Varian Clinac 2100C LINAC has been simulated by a Monte Carlo calculation to evaluate photo neutron dose. Monte Carlo calculation using the MCNPX code has been performed for two different photons （generated from Varian 10MV, Varian 15MV） for three different field sizes （0ì 0 cm2, 10ì 10 cm2,20ì 20 cm2）. To compare with the calculation results, additional experimental measurements have been conducted by bubbledetectors on neutron dose and neutron energy spectrum.

Load-Measuring Pot Bearing with Built-In Load Cell —Part II: Fatigue Performance and Experimental Temperature Correction

This paper presents the results of fatigue performance tests performed up to 10 million cycles on a load-measuring pot bearing with built-in load cell to verify its field applicability and proposes an empirical temperature correction formula. In Part I of this work, various measurement performances of the load-measuring pot bearing were evaluated through static and dynamic loading tests. Bridge bearings are subjected to the effect of fatigue caused by the repeated application of moving loads and exposed to harsh site conditions including cold and hot weathers differently to laboratory conditions. Accordingly, the durability of the load-measuring pot bearing with built-in load cell shall be secured and the environmental effects like temperature shall be minimized for its application on field. This study conducted fatigue tests up to 10 million cycles on a load-measuring pot bearing with the capacity of 1000 kN to examine eventual degradation of the measurement accuracy with respect to the number of fatigue loading cycles. In addition, the experimental temperature correction procedure is proposed to obtain the temperature correction formula enabling to correct the effect of temperature on the load measurement.

AIM： To investigate the effect of Garden/a fasm/noides （GJ） on cerulein-induced acute pancreatitis （AP） in mice. METHODS： C57BL/6 mice weighing 18-20 g were divided into three groups. （1） Normal salinetreated group, （2） treatment with GJ at a dose of 0.1 g/kg, （3） treatment with GJ at a dose of 1 g/kg. GJ was administered orally （n = 6 per group） for 1 wk. Three hours later, the mice were given anintraperitoneal injection of cerulein （50 μg/kg）, a stable cholecystokinin （CCK） analogue, every hour for a total of 6h as described previously. The mice were sacrificed at 6 h after completion of cerulein injections. Blood samples were obtained to determine serum amylase, lipase and cytokine levels. The pancreas was rapidly removed for morphologic examination and scoring. A portion of pancreas was stored at -70℃ and prepared for the measurement of tissue myeloperoxidase （MPO） activity, an indicator of neutrophil sequestration, and for reverse-transcriptase PCR （RT-PCR） and real-time PCR measurements. RESULTS： Treatment with G.1 decreased significantly the severity of pancreatitis and pancreatitis-associated lung injury. Treatment with G.1 attenuated the severity of AP compared with saline-treated mice, as shown by reduction in pancreatic edema, neutrophil infiltration, serum amylase and lipase levels, serum cytokine levels, and mRNA expression of multiple inflammatory mediators. CONCLUSION： These results suggest that GJ attenuated the severity of AP as well as pancreatitisassociated lung injury.

Load-Measuring Pot Bearing with Built-In Load Cell —Part I: Design and Performance

This paper presents the underlying principle and the results of various performance evaluations for a load-measuring pot bearing with built-in load cell. The pot bearing composed of a pot made of steel in which an elastomer disk is inserted is a bearing supporting larger loads than the elastomeric bearing and accommodating rotational movement. Owing to a Poisson’s ratio close to 0.5, elastomer withstands hydrostatic pressure when confined in a rigid body. Accounting for this principle, the vertical load applied on the pot bearing can be obtained by converting the pressure acting on the elastomer. Therefore, a load-measuring pot bearing is developed in this study by embedding a load cell exhibiting remarkable durability in the base plate of the bearing. The details for the insertion of the load cell in the base plate of the pot were improved through finite element analysis to secure sufficient measurement accuracy. The evaluation of the static performance of the pot bearing applying these improved details verified that the bearing exhibited sufficient accuracy for the intended measurement purpose. The dynamic performance evaluation results indicated that accurate measurement of the dynamic load was also achieved without time lag.

Dendritic cell-based immunotherapy for colon cancer using an HLA-A＊0201-restricted cytotoxic T-lymphocyte epitope from tumor-associated antigen 90K

Tumor-associated antigen 90K is implicated in cell-cell and cell-extracellular matrix adhesion through its interaction with galectin-3 and integrin-β 1 and is highly expressed in malignant tissues, making it a novel target for the development of new immunotherapies. We investigated a potential immunotherapy treatment for colon cancer using 90K-specific cytotoxic T lymphocytes induced by autologous dendritic cells and pulsed with 90K peptides. We selected three peptides （90K3s1, 90K5 and 90K523） that bind to HLA-A＊0201 molecules on the basis of their binding affinity, as determined by a peptide-T2 binding assay. Dendritic cells pulsed with 90K peptides resulted in the efficient generation of mature dendritic cells and exhibited enhanced T-cell stimulation and polarization of naive T cells toward Thl. Dendritic cells pulsed with 90K peptides generated potent cytotoxic T-lymphocytes that lysed T2 cells loaded with each 90K peptide, and 90K＋/HLA-A2＋ colon cancer cell lines, including HCT116 and SW480, in a dose-dependent and HLA-A＊O2Ol-restricted manner. No killing was observed in 90K＋/HLA-A2- DLD1 or 90K-/HLA-A2- K562 cells. Therefore, we believe that cytotoxic T-lymphocytes stimulated by 90K peptide-pulsed dendritic cells naturally recognize the 90K peptide presented by colon cancer cells in the context of HLA-A2, and kill 90K-positive tumor cells. Dendritic cells pulsed with 90K peptides led to the induction of granzyme B and perforin positive CD8＋ T cells against HCT116 and SW480 cells, but not DLD1 cells. In conclusion, 90K-specific cytotoxic T lymphocytes, generated by stimulating T cells with 90K peptide-pulsed dendritic cells, could be useful effector cells for the immunotherapv treatment of colon cancer.

Iodized oil uptake assessment with cone-beam CT in chemoembolization of small hepatocellular carcinomas

AIM:To evaluate the utility of assessing iodized oil uptake with cone-beam computed tomography(CT)in transarterial chemoembolization(TACE)for small he-patocellular carcinoma(HCC).METHODS:Cone-beam CT provided by a biplane flat-panel detector angiography suite was performed on eighteen patients(sixteen men and two women;41-76 years;mean age,58.9 years)directly after TACE for small HCC(26 nodules under 30 mm;mean diam-eter,11.9 mm;range,5-28 mm).The pre-procedural locations of the tumors were evaluated using tripha-sic multi-detector row helical computed tomography(MDCT).The tumor locations on MDCT and the iodized oil uptake by the tumors were analyzed on cone-beam CT and on spot image directly after the ***:All lesions on preprocedural MDCT were de-tected using iodized oil uptake in the lesions on cone-beam CT(sensitivity 100%,26/26).Spot image depictediodized oil uptake in 22 of the lesions(sensitivity 85%).The degree of iodized oil uptake was overestimated(9%,2/22)or underestimated(14%,3/22)on spot image in f ive nodules compared with that of cone-beam ***:Cone-beam CT is a useful and conve-nient tool for assessing the iodized oil uptake of small hepatic tumors(< 3 cm)directly after TACE.

Differential diagnosis of left-sided abdominal pain:Primary epiploic appendagitis vs colonic diverticulitis

AIM:To investigate the clinical characteristics of left primary epiploic appendagitis and to compare them with those of left colonic ***:We retrospectively reviewed the clinical records and radiologic images of the patients who presented with left-sided acute abdominal pain and had computer tomography(CT) performed at the time of presentation showing radiological signs of left primary epiploic appendagitis(PEA) or left acute colonic diverticulitis(ACD) between January 2001 and December 2011. A total of 53 consecutive patients were enrolled and evaluated. We also compared the clinical characteristics,laboratory findings,treatments,and clinical results of left PEA with those of left ***:Twenty-eight patients and twenty-five patients were diagnosed with symptomatic left PEA and ACD,respectively. The patients with left PEA had focal abdominal tenderness on the left lower quadrant(82.1%). On CT examination,most(89.3%) of the patients with left PEA were found to have an oval fatty mass with a hyperattenuated ring sign. In cases of left ACD,the patients presented with a more diffuse abdominal tenderness throughout the left side(52.0%vs 14.3%; P = 0.003). The patients with left ACD had fever and rebound tenderness more often than those with left PEA(40.0% vs 7.1%,P = 0.004; 52.0% vs14.3%,P = 0.003,respectively). Laboratory abnormalities such as leukocytosis were also more frequently observed in left ACD(52.0% vs 15.4%,P = 0.006).CONCLUSION:If patients have left-sided localized abdominal pain without associated symptoms or laboratory abnormalities,clinicians should suspect the diagnosis of PEA and consider a CT scan.
