Spray freeze drying of small nucleic acids as inhaled powder for pulmonary delivery

The therapeutic potential of small nucleic acids such as small interfering RNA(siRNA) to treat lung diseases has been successfully demonstrated in many in vivo studies. A major barrier to their clinical application is the lack of a safe and efficient inhaled *** this study, spray freeze drying was employed to prepare dry powder of small nucleic *** and herring sperm DNA were used as bulking agent and model of small nucleic acid therapeutics, respectively. Formulations containing different solute concentration and DNA concentration were produced. The scanning electron microscope(SEM) images showed that the porosity of the particles increased as the solute concentration decreased. Powders prepared with solute concentration of 5% w/v were found to maintain a balance between porosity and robustness. Increasing concentration of DNA improved the aerosol performance of the formulation. The dry powder formulation containing 2% w/w DNA had a median diameter of 12.5 μm, and the aerosol performance study using next generation impactor(NGI) showed an emitted fraction(EF) and fine particle fraction(FPF) of 91% and 28% respectively. This formulation(5% w/v solute concentration and 2% w/w nucleic acid) was adopted subsequently to produce siRNA powder. The gel retardation and liquid chromatography assays showed that the siRNA remained intact after spray freeze drying even in the absence of delivery vector. The siRNA powder formulation exhibited a high EF of 92.4%and a modest FPF of around 20%. Further exploration of this technology to optimise inhaled siRNA powder formulation is warranted.

Axonal regeneration following surgical nerve repair is slow and often incomplete,resulting in poor functional recovery which sometimes contributes to lifelong ***,there are no FDA-approved therapies available to promote nerve *** accelerates axonal regeneration,but systemic side effects presently outweigh its potential benefits for peripheral nerve *** authors describe herein a biodegradable polyurethane-based drug delivery system for the sustained local release of tacrolimus at the nerve repair site,with suitable properties for scalable production and clinical application,aiming to promote nerve regeneration and functional recovery with minimal systemic drug *** is encapsulated into co-axially electrospun polycarbonate-urethane nanofibers to generate an implantable nerve wrap that releases therapeutic doses of bioactive tacrolimus over 31 *** and drug loading are adjustable for applications in small and large caliber nerves,and the wrap degrades within 120 days into biocompatible *** released from the nerve wrap promotes axon elongation in vitro and accelerates nerve regeneration and functional recovery in preclinical nerve repair models while off-target systemic drug exposure is reduced by 80%compared with systemic *** its surgical suitability and preclinical efficacy and safety,this system may provide a readily translatable approach to support axonal regeneration and recovery in patients undergoing nerve surgery.

A comprehensive review on bitter gourd (Momordica charantia L.) as a gold mine of functional bioactive components for therapeutic foods

Bitter gourd is a tropical wine grown mainly in India, China and South East Asia. The plant is cultivated mainly for its fruit part which is edible. Bitter gourd is unaccepted widely due to its bitter taste. Nevertheless, the fruit is a source of several key nutrients. The plant, as a whole contains, more than 60 phyto-medicines that are active against more than 30 diseases, including cancer and diabetes. Currently, the incorporation of the bioactive compounds isolated from bitter gourd into functional foods and beverages finds a new horizon. Nanoencapsulation and novel green extraction methods can be employed to improve the yield and quality of extracted compounds and their stability while incorporation into food products. The present review is an attempt to throw light to nutritional aspects, various bioactive compounds present and important nutraceutical properties of the bitter gourd plant in detail.

Ca2+ plays a pivotal role in the physiology and biochemistry of prokaryotic and mammalian *** also utilize the universal Ca2+ signal to create a specific cellular environment to achieve coexistence with the host,and to *** this paper we first describe our development of a grafting approach to understand site-specific Ca2+ binding properties of EF-hand proteins with a helix-loop-helix Ca2+ binding motif,then summarize our prediction and identification of EF-hand Ca2+ binding sites on a genome-wide scale in bacteria and virus,and next report the application of the grafting approach to probe the metal binding capability of predicted EF-hand motifs within the streptococcal hemoprotein receptor(Shr) of Streptococcus pyrogenes and the nonstructural protein 1(nsP1) of Sindbis *** methods such as the grafting approach are developed in conjunction with prediction algorithms we are better able to probe continuous Ca2+-binding sites that have been previously underrepresented due to the limitation of conventional methodology.

Laboratory assessment of Alaska aggregates using Micro-Deval test

Aggregates suitable for use in asphalt concrete （AC） pavement construction must meet durability criteria. Thus, it is critical to select appropriate tests to properly characterize aggregate durability. In Alaska, durability tests currently being used for aggregates in AC pavement include Los Angeles （LA） abrasion test, sulfate soundness test and washington degradation test. However, there have long been concerns arising over washington degradation test used as an acceptance tool, motivating pavement practitioners to seek more suitable alternatives. This paper presents a study to investigate the feasibility of using Micro-Deval test, commonly used in other states, to evaluate the durability of Alaskan aggregates in AC pavement as well as its potential to replace washington degradation test. Micro-Deval test, washington degradation test and other tests currently specified in Alaska were conducted on aggregates from 16 batches representing statewide sources. Based on the testing results, it is found that using Micro-Deval test for durability assessment of Alaska aggregates was feasible and reproducible, and a high potential was revealed to use Micro-Deval test to replace washington degradation test in Alaska. It is recommended that Micro-Deval test be considered as an additional test for a certain period, but in the long run should be used along with current LA abrasion and sulfate soundness tests to provide a more desirable durability assessment of Alaska aggregates used in AC pavement.

Reliable cell purification and determination of cell purity:crucial aspects of olfactory ensheathing cell transplantation for spinal cord repair

Connecting Race to Ethics Related to Technology:A Call for Critical Tech Ethics

Critical tech ethics is my call for action to influencers,leaders,policymakers,and educators to help move our society towards centering race,deliberately and intentionally,to tech *** too long,when“ethics”is applied broadly across different kinds of technology,ethics does not address race explicitly,including how diverse forms of technologies have contributed to violence against and the marginalization of communities of *** several years of research,I have studied online behavior to evaluate gender and racial biases.I have concluded that a way to improve technologies,including the Internet,is to create a specific type of ethics termed“critical tech ethics”that connects race to ethics related to *** article covers guiding theories for discovering critical tech ethical challenges,contemporary examples for illustrating critical tech ethical challenges,and institutional changes across business,education,and civil society actors for teaching critical tech ethics and encouraging the integration of critical tech ethics with undergraduate computer *** tech ethics has been developed with the imperative to help improve society through connecting race to ethics related to technology,so that we may reduce the propagation of racial injustices currently occurring by educational institutions,technology corporations,and civil *** aim is to improve racial equity through the development of critical tech ethics as research,teaching,and practice in social norms,higher education,policy making,and civil society.

Study of methods for endpoint aware inspection in a next generation firewall

Given the global increase in remote work with the COVID-19 pandemic and deperimeterization due to cloud deployment of next generation frewalls,the concept of a next generation firewall is at a breaking *** is becoming more dificult to define the barrier between the good and the *** provide the best security for an endpoint with minimal false positives or false negatives it is often necessary to identify the communicating endpoint application.n this study,we present an analysis of key research and methods for providing endpoint aware protection in the context of a next generation frewall we examine both academic research as well as state-of-the-art of the existing next generation firewall *** divide endpoint application identification into passive and active *** passive endpoint application identification,we study several traffc fingerprinting methods for different *** active methods we consider active scanning,endpoint metadata analysis and content injection and reference existing *** conclude that there are several open areas for future research,and that none of the considered methods is a silver bullet solution for endpoint aware inspection in the context of a next generation *** our best knowledge,this is the first study to examine current research and existing implementations of endpoint aware inspection.
