Although bone morphogenetic proteins(bMPs)initially showed effective induction of ectopic bone growth in muscle,it has since been determined that these proteins,as members of the TGF-b superfamily,play a diverse and critical array of biological *** roles include regulating skeletal and bone formation,angiogenesis,and development and homeostasis of multiple organ *** of the members of the TGF-b/bMP superfamily result in severe skeletal and extra-skeletal irregularities,suggesting high therapeutic potential from understanding this family of bMP *** it was once one of the least characterized bMPs,bMP9 has revealed itself to have the highest osteogenic potential across numerous experiments both in vitro and in vivo,with recent studies suggesting that the exceptional potency of bMP9 may result from unique signaling pathways that differentiate it from other *** effectiveness of bMP9 in inducing bone formation was recently revealed in promising experiments that demonstrated efficacy in the repair of critical sized cranial defects as well as compatibility with bone-inducing bio-implants,revealing the great translational promise of ***,emerging evidence indicates that,besides its osteogenic activity,bMP9 exerts a broad range of biological functions,including stem cell differentiation,angiogenesis,neurogenesis,tumorigenesis,and *** review aims to summarize our current understanding of bMP9 across biology and the body.

Use of portal pressure studies in the management of variceal haemorrhage

Portal hypertension occurs as a complication of liver cirrhosis and complications such as variceal bleeding lead to significant demands on resources. Endoscopy is the gold standard method for screening cirrhotic patients however universal endoscopic screening may mean a lot of unnecessary procedures as the presence of oesophageal varices is variable hence a large time and cost burden on endoscopy units to carry out both screening and subsequent follow up of variceal bleeds. A less invasive method to identify those at high risk of bleeding would allow earlier prophylactic measures to be applied. Hepatic venous pressure gradient (HVPG) is an acceptable indirect measurement of portal hypertension and predictor of the complications of portal hypertension in adult cirrhotics. Varices develop at a HVPG of 10-12 mmHg with the appearance of other complications with HPVG > 12 mmHg. Variceal bleeding does not occur in pressures under 12 mmHg. HPVG > 20 mmHg measured early after admission is a significant prognostic indicator of failure to control bleeding varices, indeed early transjugular intrahepatic portosystemic shunt (TIPS) in such circumstances reduces mortality significantly. HVPG can be used to identify responders to medical therapy. Patients who do not achieve the suggested reduction targets in HVPG have a high risk of rebleeding despite endoscopic ligation and may not derive significant overall mortality benefit from endoscopic intervention alone, ultimately requiring TIPS or liver transplantation. Early HVPG measurements following a variceal bleed can help to identify those at risk of treatment failure who may benefit from early intervention with TIPS. Therefore, we suggest using HVPG measurement as the investigation of choice in those with confirmed cirrhosis in place of endoscopy for intitial variceal screening and, where indicated, a trial of b-blockade, either intravenously during the initial pressure study with assessment of response or oral therapy with repea

In the design of building structures,joint efforts must be decided to resolve the depth of competent layers across the intended site to safeguard the durability of civil engineering structures and to avert the disastrous consequences of structural failure and *** this study,an integrated methodology that employed DC resistivity tomography involving 2-D and 3-D techniques and geotechnical-soil analysis was used to evaluate subsoil conditions for engineering site investigation at Okerenkoko primary school,in the Warri-southwest area of Delta State,to adduce the phenomena responsible for the visible cracks/structural failure observed in the *** results obtained brought to light the geological structure beneath the subsurface,which consists of four geoelectric layers identified as topsoil,dry/lithified upper sandy layer,wet sand(water-saturated)and peat/clay/sandy clayey soil(highly water-saturated).The deeply-seated peat/clay materials(ρ≤20Ωm)were delineated in the study area to the depths of 17.1 m and 19.8 m from 2-D and 3-D tomography respectively.3-D images presented as horizontal depth slices revealed the dominance of very low resistivity materials ***/clay/sandy clay within the fourth,fifth and sixth layers at depths ranging from 8.68-12.5 m,12.5-16.9 m and 16.9-21.9 m *** dominance of mechanically unstable peat/clay/sandy clay layers beneath the subsurface,which are highly mobile in response to volumetric changes,is responsible for the noticeable cracks/failure detected on structures within the study *** observations were validated by a geotechnical test of soil samples in the study ***’s limits of the samples revealed plasticity indices of ***,the soil samples within the depth analyzed were representatives of sandy soil that does not possess any *** methods justifiably provided relevant information on the subsurface geology beneath the study site and should be appropriated as major tools for

Through ingestion and subsequent egestion,Arctic seabirds can bioaccumulate microplastics at and around their colony breeding *** microplastics in Arctic seabirds have been well documented,it is not yet understood to what extent these particles can act as transport vehicles for plastic-associated contaminants,including legacy persistent organic pollutants(POPs),trace metals,and organic *** investigated the occurrence and pattern of organic and inorganic co-contaminants of microplastics in two seabird species from the Canadian Arctic-northern fulmar(Fulmarus glacialis)and black-legged kittiwake(Rissa tridactyla).We found that fulmars had higher levels of plastic contamination and emerging organic compounds(known to be plastic additives)than kittiwakes,whereas higher concentrations of legacy POPs were found in kittiwakes than the ***,fulmars,the species with the much larger foraging range(~200 km),had higher plastic pollution and overall contaminant burdens,indicating that birds may be acting as long-range transport vectors for plasticassociated *** results suggest a potential connection between plastic additive contamination and plastic pollution burdens in the bird stomachs,highlighting the importance of treating plastic particles and plastic-associated organic additives as co-contaminants rather than separate pollution issues.

Quantitative or complex traits are determined by the combined effects of many loci,and are affected by genetic networks or molecular *** the present study,we genotyped a total of 138 mutations,mainly single nucleotide polymorphisms derived from 71 functional genes on a Wagyu x Limousin reference *** hundred forty six F animals were measured for 5 carcass,6 eating quality and 8 fatty acid composition traits.A total of 2,280 single marker-trait association runs with 120 tagged mutations selected based on the HAPLOVIEW analysis revealed 144 significant associations(Pbut 50 of them were removed from the analysis due to the small number of animals(≤9) in one genotype group or absence of one genotype among three *** remaining 94 single-trait associations were then placed into three groups of quantitative trait modes(QTMs) with additive,dominant and overdominant *** significant markers and their QTMs associated with each of these 19 traits were involved in a linear regression model analysis,which confirmed single-gene associations for 4 traits,but revealed two-gene networks for 8 traits and three-gene networks for 5 *** genetic networks involving both genotypes and QTMs resulted in high correlations between predicted and actual values of performance,thus providing evidence that the classical Mendelian principals of inheritance can be applied in understanding genetic complexity of complex *** present study also indicated that carcass, eating quality and fatty acid composition traits rarely share genetic ***,marker-assisted selection for improvement of one category of these traits would not interfere with improvement of another.

4-Aminobutyrate accumulates in plants under abiotic ***,targeted quantitative profiling of metabolites and transcripts was conducted to monitor glutamate-and polyamine-derived 4-aminobutyrate production and its subsequent catabolism to succinate or 4-hydroxybutyrate in apple(Malus x domestica borkh.)fruit stored at 0°C with 2.5 kPa O2 and 0.03 or 5 kPa CO2 for 16 ***-temperature-induced protein hydrolysis appeared to be responsible for the enhanced availability of amino acids during early storage,and the resulting higher glutamate level stimulated 4-aminobutyrate levels more than *** CO2 increased the levels of polyamines,as well as succinate and 4-hydroxybutyrate,during early storage,and 4-aminobutyrate and 4-hydroxybutyrate over the longer *** of all of the genes likely involved in 4-aminobutyrate metabolism from glutamate/polyamines to succinate/4-hydroxybutyrate was induced in a co-ordinated ***2-regulated expression of apple GLUTAMATE DECARbOXYLASE 2,AMINE OXIDASE 1,ALDEHYDE DEHYDROGENASE 10A8 and POLYAMINE OXIDASE 2 was evident with longer term *** suggested that respiratory activities were restricted by the elevated CO2/O2 environment,and that decreasing NAD+availability and increasing NADPH and NADPH/NADP+,respectively,played key roles in the regulation of succinate and 4-hydroxybutyate ***,these findings suggest that both transcriptional and biochemical mechanisms are associated with 4-aminobutyrate and 4-hydroxybutyrate metabolism in apple fruit stored under multiple abiotic stresses.
