The member of Rho family of small GTPases cdc42 plays important and conserved roles in cell polarity and motility. The cdc42ep family proteins have been identified to bind to cdc42, yet how they interact with cdc42 to regulate cell migration remains to be elucidated. In this study, we focus on cdc42epl, which is expressed predominantly in the highly migratory neural crest ceils in frog embryos. Through morpholino-mediated knockdown, we show that cdc42epl is required for the migration of cranial neural crest cells. Loss of cdc42epl leads to rounder cell shapes and the formation of membrane blebs, consistent with the observed disruption in actin organization and focal adhesion alignment. As a result, cdc42ep1 is critical for neural crest cells to apply traction forces at the correct place to migrate efficiently. We further show that cdc42ep1 is localized to two areas in neural crest celts： in membrane protrusions together with cdc42 and in perinuciear patches where cdc42 is absent. cdc42 directly interacts with cdc42epl （through the cRIB domain） and changes in cdc42 level shift the distribution of cdc42epl between these two subcellular locations, controlling the formation of membrane protrusions and directionality of migration as a consequence. These results suggest that cdc42ep1 elaborates cdc42 activity in neural crest cells to promote their efficient migration.

Rice Hull Mulch Affects Germination of Bittercress and creeping Woodsorrel in container Plant culture

Mulches are commonly used to control weeds in container nursery crops, especially in sites where preemergence herbicides are either not labeled or potentially phytotoxic to the crop. Parboiled rice hulls have been shown to provide effective weed control when applied 1.25 to 2.5 cm deep over the container substrate surface. The objective of this research was to determine if weed seed placement, above or below the mulch layer, affects flexuous bittercress or creeping woodsorrel establishment. Seeds of both species were placed either above or below rice hull mulch layers 0, 0.6, 1.3, or 2.5 cm deep in nursery containers with a 80 pine bark: 20 sphagnum peat moss substrate. Establishment of both weeds decreased with increasing mulch depth. Establishment of both species was generally greater from beneath the mulch compared to when seed were applied above the mulch. Light penetration through varying depths of rice hulls was determined with a spectroradiometer. Photosynthetically active radiation (PAR) decreased exponentially with increasing rice hull depth, and was less than 1 μmol·m-2·s-1 beneath depths greater than 1 cm. Germination of both species was determined in Petri dishes placed beneath varying densities of shade cloth. Flexuous bittercress germination responded quadratically to decreasing light level, but still germinated (13%) in complete darkness after 3 weeks. creeping woodsorrel germination was not affected by light level and was high (92%) after 3 weeks. The role of light exclusion by rice hulls as a mechanism for controlling buried weed seed is discussed. Water retention immediately after irrigation, and for 24 hr following irrigation, was determined for a 2.5 cm layer of rice hulls, sphagnum peat moss, and pine bark. Rice hulls retained less water, and dried more quickly than peat moss or pine bark. 目的　探讨TNF α能否诱导成骨类细胞凋亡及OPG能否抑制其凋亡 ,开辟一条防治骨质疏松的新路。方法　将人间充质干细胞在分化培养液中培养 2 1天使之成为成骨细胞 ,与MG6 3一起用于凋亡实验。鼠WEHI 16 4细胞系用于阳性对照。AnnexinV和TUNEL分析方法检测成骨类细胞凋亡。结果　和FasL诱导细胞凋亡一样 ,TNF α能引起MG6 3骨肉瘤细胞、间充质干细胞和成骨细胞的凋亡 ,并表现为明显的浓度依赖和时间依赖。低浓度TNF α(170～ 5 0 0 pmol/L)作用于细胞 2～ 4h就显示了较明显的凋亡 ,而 0 4 5～1 5nmol/LOPG几乎完全抑制了 5 0 0 pmol/LTNF α诱导的凋亡。结论　OPG能够抑制TNF α诱导的成骨类细胞凋亡。

Dose of alendronate directly increases trabeculae expansivity without altering bone volume in rat femurs

AIM To evaluate the effects of sodium alendronate on bone repair in fractures created in appendicular bones. METHODS Wistar rats(n = 36) were allocated into three distinct groups: group c(control), group B1(received 1 mg/kg of alendronate), and group B2(received 3 mg/kg of alendronate). The rats underwent femoral transversal linear fracture surgery using stable internal fixation with a 2.0 mm plate and screw system. Each animal randomly received intraperitoneal applications of sodium alendronate at a dose corresponding to group B1 or B2 three times a week, while the control group received a 0.9% saline solution. Drug administration was performed until euthanasia at 45 d. The femurs were removed and each surgical piece was sent for radiographic, tomographic and microtomographic analysis. Data were submitted to descriptive and inferential statistical analysis(95% confidence interval).RESULTS Quantitative evaluations of bone neoformation did not show differences among the groups in the radiographic(P = 0.341), microtomographic(P = 0.581) and tomographic evaluations(P = 0.171). In the qualitative microtomographic analysis, a smaller distance was observed between the internal bone trabeculae in the groups that used alendronate(P = 0.05). On the other hand, group B2 had a higher amount of bone trabeculae per unit length when compared to the other groups(P = 0.04).cONcLUSION It is likely that the use of alendronate did not have a direct influence on the amount of bone neoformation, however it did influence the bone quality in a dosedependent manner, ultimately affecting the distance and quantity of the trabeculae.
