Objective: To reveal th. distribution ch.racteristics and dem.graph.c factors of traditional Ch.nese m.dicine (TCm. constitution am.ng elderly individuals in Ch.na. m.th.ds: Elderly individuals from.seven regions in Ch.na were selected as sam.les in th.s study using a m.ltistage cluster random.sam.ling m.th.d. Th. basic inform.tion questionnaire and Constitution in Ch.nese m.dicine Questionnaire (Elderly Edition) were used. descriptive statistical analysis, ch.-squared tests, and binary logistic regression analysis were used. Results: Th. single balanced constitution (BC) accounted for 23.9%. Th. results of th. m.jor TCm.constitution types sh.wed th.t BC (43.2%) accounted for th. largest proportion and unbalanced constitutions ranged from.0.9% to 15.7%. East Ch.na region (odds ratio [OR] = 2.097; 95% confidence interval [CI], 1.912 to 2.301), m.rried status (OR = 1.341; 95% CI, 1.235 to 1.457), and m.nagers (OR = 1.254; 95% CI, 1.044 to 1.505) were significantly associated with.BC. Age >70 years was associated with.qi-deficiency constitution and blood stasis constitution (BSC). Fem.le sex was significantly associated with.yang-deficiency constitution (OR = 1.646; 95% CI, 1.52 to 1.782). South.est region was significantly associated with.ph.egm.dam.ness constitution (OR = 1.809; 95% CI, 1.569 to 2.086). North.Ch.na region was significantly associated with.inh.rited special constitution (OR = 2.521; 95% CI, 1.569 to 4.05). South.Ch.na region (OR = 2.741; 95% CI, 1.997 to 1.3.763), Central Ch.na region (OR = 8.889; 95% CI, 6.676 to 11.835), senior m.ddle sch.ol education (OR = 2.442; 95% CI, 1.932 to 3.088), and m.nagers (OR = 1.804; 95% CI, 1.21 to 2.69) were significantly associated with.BSC. Conclusions: Th.s study defined th. distribution ch.racteristics and dem.graph.c factors of TCm.constitution in th. elderly population. Th. landscape of h.patitis B virus (h.V) integration in th. plasm. cell-free dNA (cfdNA) of h.V-infected patients with.different stages of liver diseases [ch.onic h.patitis B (Ch.), liver cirrh.sis (LC), and h.patocellular carcinom. (h.C)] rem.ins unclear. In th.s study, we developed an im.roved strategy for detecting h.V dNA integration in plasm. cfdNA, based on dNA probe capture and next-generation sequencing. Using th.s optim.zed strategy, we successfully detected h.V integration events in ch.m.ric artificial dNA sam.les and h.V-infected h.pG2-NTCP cells at day one post infection, with.h.gh.sensitivity and accuracy. Th. ch.racteristics of h.V integration events in th. h.V-infected h.pG2-NTCP cells and plasm. cfdNA from.h.V-infected individuals (Ch., LC, and h.C) were furth.r investigated. A total of 112 and 333 integration breakpoints were detected in th. h.pG2-NTCP cells and 22 out of 25 (88%) clinical h.V-infected sam.les, respectively. In vivo analysis sh.wed th.t th. norm.lized num.er of support unique sequences (nnsus) in h.C was significantly h.gh.r th.n in Ch. or LC patients (P values dom.y distributed on h.m.n ch.om.som.s and are enrich.d in th. h.V genom. around nt 1800. Th. m.jority of integration breakpoints (61.86%) are located in th. gene-coding region. Both.non-h.m.logous end-joining (Nh.J) and m.croh.m.logy-m.diated end-joining (m.EJ) interactions occurred during h.V integration across th. th.ee different stages of liver diseases. Our study provides evidence th.t h.V dNA integration can be detected in th. plasm. cfdNA of h.V-infected patients, including th.se with.Ch., LC, or h.C, using th.s optim.zed strategy. © 2024 Th. A quadrivalent norovirus vaccine based on a ch.m.anzee adenovirus vector induces potent im.unity in m.ce

Norovirus (NoV) infection is a m.jor cause of gastroenteritis worldwide. Th. virus poses great ch.llenges in developing vaccines with.broad im.une protection due to its genetic and antigenic diversity. To date, th.re are no approved NoV vaccines for clinical use. h.re, we aim.d to develop a broad-acting quadrivalent NoV vaccine based on a ch.m.anzee adenovirus vector, AdC68, carrying th. m.jor capsid protein (VP1) of noroviral GI and GII genotypes. Com.ared to intram.scular (i.m.), intranasal (i.n.), or oth.r prim.-boost im.unization regim.ns (i.m. + i.m., i.m. + i.n., i.n. + i.m.), AdC68-GI.1-GII.3 (E1)-GII.4-GII.17 (E3), adm.nistered via i.n. + i.n. induced h.gh.r titers of serum.IgG antibodies and h.gh.r IgA antibodies in bronch.alveolar lavage fluid (BALF) and saliva against th. four h.m.logous VP1s in m.ce. It also significantly stim.lated th. production of blocking antibodies against th. four genotypes. In response to re-stim.lation with.virus-like particles (VLP)-GI.1, VLP-GII.3, VLP-GII.4, and VLP-GII.17, th. quadrivalent vaccine adm.nistered according to th. i.n. + i.n. regim.n effectively triggered specific cell-m.diated im.une responses, prim.rily ch.racterized by IFN-γ secretion. Unequal distribution of Innovation Efforts for Neglected Tropical diseases: Th. Role of Funding Evaluation Criteria

Background: International research.and innovation efforts for neglected tropical diseases h.ve increased in recent decades due to disparities in overall h.alth.research.funding in relation to global burden of disease. h.wever, with.n th. field of neglected tropical diseases som. seem.far m.re neglected th.n oth.rs. In th.s research.th. aim.is to investigate th. distribution of resources and efforts, as well as th. m.ch.nism. th.t underpin funding allocation for neglected tropical diseases. m.th.dology: A system.tic literature review was conducted to establish.a com.reh.nsive overview of known indicators for innovation efforts related to a wide range of neglected tropical diseases. Articles were selected based on a subjective evaluation of th.ir relevance, th. presence of original data, and th. breadth.of th.ir scope. Th.s was followed by th.rteen in-depth.open-ended interviews with.representatives of private, public and ph.lanth.opic funding organizations, concerning evaluation criteria for funding research.proposals. Results: Th. findings reveal a large difference in th. extent to wh.ch.th. individual diseases are neglected with.notable differences between absolute and relative efforts. Criteria used in th. evaluation of research.proposals relate to potential im.act, th. probability of success and strategic fit. Private organizations prioritize strategic fit and econom.c im.act;ph.lanth.opic organizations prioritize sh.rt-term.societal im.act;and public generally prioritize th. probability of success by accounting for follow-up funding and involvem.nt of industry. Funding decisions of different types of organizations are h.gh.y interrelated. Conclusions: Th.s study sh.ws th.t th. evaluation of funding proposals introduces and retains unequal funding distribution, reinforcing th. relative neglect of diseases. Increasing evidences suggest th.t th. m.th.ltransferase NSUN2 catalyzes 5-m.th.lcytosine (m.C) m.difications on viral RNAs, wh.ch.are essential for th. replication of various viruses. despite th. function of m.C deposition is well ch.racterized, oth.r potential roles of NSUN2 in regulating viral replication rem.in largely unknown. In th.s study, th. m.C m.dified residues catalyzed by NSUN2 on enterovirus 71 (EV71) RNAs were m.pped. NSUN2, along with.m.C m.difications, played m.ltiple roles during th. EV71 life cycle. Functional m.C m.dified nucleotides increased th. translational efficiency and stability of EV71 RNAs. Additionally, NSUN2 was found to target th. viral protein VP1 for binding and prom.te its stability by inh.biting th. ubiquitination. Objective: To explore th. neuroprotective effects of th. Sh.oyao Gancao decoction (SGd) against excitatory dam.ge in PC12 cells and th. role of th. Src-NR2-nNOS path.ay m.diation by SGd in regulating γ-am.nobutyric acid (GABA)-glutam.te (Glu) h.m.ostasis. m.th.ds: N-m.th.l-d-aspartic acid (Nm.A) was used to establish.a PC12 cell excitability injury m.del. To investigate th. neuroprotective effect of SGd, a cell counting kit-8 (CCK-8) assay was used to determ.ne PC12 cell viability, Annexin V/Propidium.Iodide (Annexin V/PI) double staining was used to determ.ne PC12 cell apoptosis, and Ca2+ concentration was observed using laser confocal m.croscopy. GABA receptor agonists and antagonists were used to analyze th. neuroprotective interactions between γ-am.nobutyric acid (GABA) and Nm.A receptors. Additionally, m.lecular biology tech.iques were used to determ.ne m.NA and protein expression in th. Src-NR2-nNOS path.ay. We analyzed th. correlations between th. regulatory sites of GABA and Nm.A interactions, excitatory neurotoxicity, and brain dam.ge at th. m.lecular level. Results: Nm.A excitotoxic injury m.nifested as a significant decrease in cell activity, increased apoptosis and caspase-3 protein expression, and a significant increase in intracellular Ca2+ concentration. Adm.nistration of SGd, a GABAA receptor agonist (m.scim.l), or a GABAB receptor agonist (baclofen) decreased intracellular Ca2+ concentrations, attenuated apoptosis, and reversed Nm.A-induced upregulation of caspase-3, Src, Nm.AR2A, Nm.AR2B, and nNOS. Unexpectedly, a GABAA receptor antagonist (bicuculline) and a GABAB receptor antagonist (saclofen) failed to significantly increase excitatory neurotoxicity. Conclusions: Taken togeth.r, th.se results not only provide an experim.ntal basis for SGd adm.nistration in th. clinical treatm.nt of central nervous system.injury diseases, but also suggest th.t th. Th. discovery of ch.rped pulse am.lification h.s led to great im.rovem.nts in laser tech.ology,enabling energetic laser beam. to be com.ressed to pulse durations of tens of fem.oseconds and focused to a few *** with.energies of tens of m.V can be accelerated using,for instance,target norm.l sh.ath.acceleration and focused on secondary *** such.conditions,nuclear reactions can occur,with.th. production of radioisotopes suitable for m.dical *** use of h.gh.repetition lasers to produce such.isotopes is com.etitive with.conventional m.th.ds m.stly based on *** th.s paper,we study th. production of^(67)Cu,^(63)Zn,^(18)F,and^(11)C,wh.ch.are currently used in positron em.ssion tom.graph. and oth.r *** th. sam. tim.,we study th. reactions^(10)B(p,α)^(7)Be and^(70)Zn(p,4n)^(67)Ga to put furth.r constraints on th. proton distributions at different angles,as well as th. reaction^(11)B(p,α)^(8)Be relevant for energy *** experim.nt was perform.d at th. 1 PW laser facility at VegaⅢin Salam.nca,*** distributions of radioisotopes in th. forward(with.respect to th. laser direction)and backward directions were m.asured using a h.gh.purity germ.nium.*** results are in reasonable agreem.nt with.num.rical estim.tes obtained following th. approach.of Kim.ra and Bonasera[*** ***.,Sect.A 637,164–170(2011)].
